The four-component domino reactions of (E)-3-(dimethylamino)-1-arylprop-2-en-1-ones with anilines, aromatic aldehydes, and 1,3-cyclohexanediones in an equimolar ratio in acetic acid afforded functionalized tetrahydroquinolones in good yields. This one-pot transformation involves the formation of two C–C and two C–N bonds in a single synthetic operation.  相似文献

A general, highly efficient synthesis of arylated benzenes from simple stitching of α-oxo-ketene-S,S-acetals and functionalized deoxybenzoins via a ‘lactone intermediate’ is described. This procedure offers easy access to highly functionalized arylated benzenes containing sterically demanding groups in good to excellent yields. The advantage of the procedure lies in the fabrication of arylated benzenes with desired conformational flexibility along the molecular axis at room temperature and in a transition metal-free environment through easily accessible precursors.  相似文献

Facile, four-component domino reactions of 2-hydroxy-1,4-naphthaquinone, aromatic aldehydes, methyl/ethyl acetoacetate and ammonium acetate in ethanol under microwave irradiation at 100 °C afforded tetrahydrobenzo[g]quinoline-5,10-diones regioselectively in good yields. This transformation presumably proceeds via α,β-unsaturated triketone generation/Michael addition/regioselective annulation via intramolecular condensation domino sequence.  相似文献

1-Benzenesulfonyl-5-aminoindole 5, prepared from 5-nitroindole 3, was condensed with alkyl isothiocyanates and separately with carbon disulfide and alkyl bromides/iodides to furnish efficiently the corresponding N-alkyl-thioureidoindoles 6a-c and the alkyl N-(indol-5′-yl)dithiocarbamates 9a-e, respectively. Their cyclisation using N-bromosuccinimide (NBS) and 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU), in the cold, followed by indolic N-deprotection, furnished regioselectively the 2-alkylamino- and the 2-alkylthiothiazolo[5,4-e]indoles 8a-c and 11a-e, respectively, in good overall yields.  相似文献

The stereocontrolled synthesis of the protected (2S,3S)-N-methyl-5-hydroxyisoleucine, a component of halipeptins A and B with potent anti-inflammatory activity, has been achieved. The key steps include (i) installation of a double bond to bicyclic lactam 4 using N-tert-butyl phenylsulfinimidoyl chloride, (ii) highly exo-selective Michael reaction with lithium dimethylcuprate in the presence of chlorotrimethylsilane, and (iii) Ru-catalyzed oxidative deprotection of N,O-benzylidene acetal to the acid anhydride.  相似文献

The asymmetric synthesis of (−)-(S,S)-homaline was achieved in 8 steps from commercially available starting materials using the diastereoselective conjugate addition of the novel lithium amide reagent lithium (R)-N-(3-chloropropyl)-N-(α-methyl-p-methoxybenzyl)amide to methyl cinnamate to install the correct stereochemistry. Subsequent functional group manipulation of the resultant β-amino ester and Sb(OEt)3-mediated macrolactamisation was followed by homodimerisation to give (−)-(S,S)-homaline in 18% overall yield, representing the first asymmetric, and by far the most efficient synthesis of this natural product reported to date.  相似文献

Enantiopure (S)-6-alkoxynicotine derivatives have been synthesized in two steps from (S)-nicotine via (S)-6-iodonicotine. Deprotonation and substitution at the C-5 position of the pyridine ring of (S)-6-methoxynicotine were achieved using mesityllithium as the base at 0 °C. Conditions for the C-4 lithiation/substitution of (S)-6-isopropoxynicotine and (S)-5-chloro-6-methoxynicotine were also developed.  相似文献

(1E,3Z)-1-Aryl-4-methanesulfonyl-2-nitro-1,3-butadienes (8), derived from the initial ring-opening of 3-nitrothiophene (5), have been found to undergo a facile base-induced cyclization leading to thiopyran S,S-dioxides (9), thus furnishing a further example of effective ring-enlargement from 5- to 6-membered sulfur heterocycles. Compounds 9 are obtained as single racemic mixtures in satisfactory yields; they still contain a nitrovinylic moiety, which can be exploited for further modifications targeted to new derivatives endowed with either synthetic or pharmacological potentialities e.g., in the field of L-type Ca2+-channel blockers.  相似文献

Promoted by TiCl4, a series of α-(1,3-dithiolan-2-ylidene)-β-amino carbonyl derivatives--the aza-Morita-Baylis-Hillman adducts, have been synthesized from α-oxo cyclic ketene-S,S-acetals, arylaldehydes, and nitriles in good to excellent yields. A new solvent-free, TBD-catalyzed protocol for the synthesis of 3-formyl-2H-chromenes is presented. Substituted salicylaldehydes 1 and 3-methyl-2-butenal 2 in the presence of 10 mol % of TBD underwent a domino oxa Michael/aldol reaction to effectively yield the corresponding 2H-chromene derivatives 3. The methodology represents an improvement either in terms of efficiency and sustainability as a small amount of catalyst is required and the work-up procedure is simple and straightforward.  相似文献

Axially dissymmetric P,S-heterodonor ligand L3 synthesized from BINOL is an effective promoter in the palladium(0)-catalyzed Suzuki cross-coupling reaction of phenylboronic acid with aryl bromides and iodide at 60-80 °C. On the basis of 13C and 31P NMR spectroscopic investigation and X-ray diffraction, it was revealed that N,N-dimethylthiocarbamate-phosphine ligand L3 might be a P,S-heterodonor bidentate ligand to palladium(0) center.  相似文献

16R-Bromopregnane-3S,20S-diol reacted with potassium t-butoxide to afford androst-16-en-3S-ol in a moderate yield via fragmentation reaction. The latter is a key intermediate for the synthesis of 5α-androst-16-en-3-one, as boar sex pheromone, and other steroidal drugs. [2S,3S-Hmp]-Aureobasidin L 2 has been successfully synthesised through a combination of solution- and solid-phase peptide synthesis. All of the Fmoc-protected residues including a depsidipeptide, Fmoc-MeVal-Hmp-OH, were prepared in solution phase. Chain elongation on chlorotrityl resin was undertaken using selected coupling reagents including HBTU/HOBt, HATU/HOAt and BTC/collidine. Mononuclear mixed-ligand complexes of Pd(II) containing a N,S-heterocyclic carbene (NSHC) with a secondary alkyl N-substituent and pyridyl ligand, with the general formula [PdI2(C10H11NS)L] (C10H11NS = 3-isopropylbenzothiazolin-2-ylidene; L = pyridine, 2-aminopyridine, 3-iodopyridine and 4-tert-butyl-pyridine) have been synthesized and characterized by X-ray single-crystal crystallography. Both solution and solid-state structures, as evident from their 1H NMR spectra and X-ray structures, show anagostic γ-hydrogen interactions of metal with methine of the substituent on the carbene or pyridyl ligand giving 5-membered-chelate-like structures.  相似文献

Treatment of 2,4-disubstituted glutaryl dichlorides with benzimidic acid ethyl ester hydrochloride in the presence of triethylamine did not give the expected bis(acylbenzimidates), but delivered O-acyl-N-ethoxybenzylidene-ketene-N,O-acetals in good to excellent yields. These compounds, which are stable to moisture and chromatography on silica gel, underwent an unprecedented rearrangement to cyclic enamides under stronger basic conditions. An efficient, three-component domino reaction of dimedone 1, aromatic aldehydes (2ao), and 1,3-cyclohexanedione 1a in the regio-selective synthesis of 3,3-dimethyl-9-phenyl-2H-xanthene-1,8(5H,9H)-diones (3ao) is reported.
