A reliable three-step synthesis of ortho-substituted 4-pyrazolyl-2-ethylamines starting from a pyrazole methyl ketone and N-tert-butanesulfinylamine is described. Strongly dehydrating conditions were required to form the N-tert-butanesulfinyl ketimines, but these intermediates were stable to aqueous work-up and chromatography. Reduction of N-tert-butanesulfinyl ketimines with L-selectride®, or Super-hydride®, in THF afforded excellent yields and diastereoselectivities of the sulfinamides, which were converted into chiral amines with methanolic HCl. The 4-pyrazolyl-2-ethylamines from these investigations are attractive building blocks for medicinal chemists, as they exhibit low molecular weight, and low calculated octanol-water partition coefficient (c Log P values), while disrupting planarity with three-dimensional character. For example, the amine products were utilized in structure-activity studies of the human transient receptor potential vanilloid 1 (TRPV1/VR1).  相似文献

The regiochemistry in the tert-butyldiphenylsilylcupration of the allene depends on the temperature and also on the nature of the electrophile. Thus, the intermediate generated by addition of the silylcuprate at −78°C reacted with electrophiles to give allylsilanes, except with oxo compounds which afforded vinylsilanes. On the other hand, the silylcopper reagent was added at −40°C leading, in all cases, to the corresponding allylsilanes. When enones were used as electrophile the vinylsilanes were the 1,2-addition products and the allylsilanes those from 1,4-addition. These functionalized vinyl or allyl tert-butyldiphenylsilanes are interesting synthons for the preparation of conjugated tert-butyldiphenylsilyltrienes and functionalized exocyclic alkylidenecyclopentenes.  相似文献

The 1,2,3,4-tetrahydro-N-pivaloyl-isoquinoline-3-carboxylic acids 1d , 2d , and 3d , derived from (R)- or (S)-phenylalanine, (S)-dopa, and (S)-α-methyldopa, respectively, are doubly deprotonated with (tert-butyl)lithium in THF and alkylated at the 1-position (products 5 – 10 ). The major diastereoisomers formed are the result of electrophilic attack from the face opposite to the carboxylate group (rel. topicity ul-1,3). Even the addition to benzaldehyd (→ 7,8 ) is highly stereoselective (one of four diastereoisomers is formed exclusively (300-MHz 1H-NMR analysis)), if MgBr2 etherate is added prior to the electrophile. Some of the obtained amino-acid derivatives are decarboxylated by anodic oxidation in MeOH (→ 11 , 12 , 17 ) and NaBH3CN reduction, and converted to the known 1-methyl- and 1-benzyltetrahydroisoquinolines ( 15 , 16 ) of > 95% ee as well as to the phthalide isoquinoline alkaloid (+)-corlumine of ≥80% ee. The synthetic approach described here is compared with other methods of synthesizing enantiomerically pure 1-substituted tetrahydroisoquinolines (and thus an important group of alkaloids, Scheme 1).  相似文献

The selective cycloboronation of enantiopure 1,1,4,4-tetrasubstituted butanetetraols was investigated, and a general preparation of chiral cyclic boron-containing bifunctional Lewis acids was discovered. Compounds (2R,3R)- or (2S,3S)-1,1,4,4-tetrasubstituted butanetetraols were reacted with ArB(OH)2 at reflux in toluene, or THF, or under solvent-free condition to furnish tricoordinated, chiral bicyclo[4.4.0]diboronic esters in high yield via selective 1,3-cycloboronation. Structural characterization and properties of the novel chiral boron compounds are also reported.  相似文献

Ethyl 1-aryl-5-(trifluoromethyl)-1H-pyrazole-4-carboxylates 2 were prepared by the condensation of arylhydrazines with ethyl 3-ethoxy-2-(trifluoroacetyl)-2-propenoate (1a) at low temperature. The corresponding acids were also synthesized. X-ray diffraction analysis of an amide derivative 4 verified the position of the trifluoromethyl group on the pyrazole ring.  相似文献

In this paper we describe a three‐step synthesis of aryldi‐ and triboronic acids starting from phenols. Several substituted phenols (I) were converted into the corresponding aryldiethylphosphates (II) in good to excellent yields. The latter, on reaction with sodium trimethylstannide in liquid ammonia, under irradiation, afforded the aryl‐ and heteroarylpoly(trimethylstannyl) derivatives in 65–90% yield. The third step is the reaction of the organotin compounds with borane in THF, which leads to the corresponding arylpolyboronic acids in around 80% yield. In order to confirm their structure, some of the diboronic acids were converted into the corresponding pinacol esters. The results obtained in a study on the synthesis of various terphenyls through double and triple Suzuki couplings catalyzed by palladium acetate between the obtained arylpolyboronic acids and various aryl halides are also reported. These reactions proceeded with an average 65% yield, and also enabled us to confirm the structures of some of the diboronic acids. The structure of the new compounds was determined by 1H, 13C and 119Sn NMR spectroscopy, mass spectrometry and IR spectroscopy. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

A seven-step synthesis of 1-substituted 5-(2-acylaminoethyl)-1H-pyrazole-4-carboxamides 20 as the pyrazole analogues of histamine was developed. The synthesis starts with a three-step preparation of N(1)-substituted methyl 5-(2-tert-butoxycarbonylaminoethyl)-1H-pyrazole-4-carboxylates 7 from commercially available Boc-β-alanine (1). Subsequent four-step transformation of the key-intermediates 7 into the final products 20 was performed following two complementary reaction sequences comprising acidolytic removal of the Boc group, hydrolysis of the COOMe group, amidations of the COOH group, and acylations of the NH2 group. The structures of pyrazole derivatives were determined by spectroscopic methods and by X-ray diffraction.  相似文献

tert-Butyl peroxyacetate, tert-butyl peroxybutyrate, tert-butyl phenylperoxyacetate, and tert-butyl peroxyundecanoate were obtained in nearly quantitative yields by the esterification of the corresponding carboxylic acids with tert-butyl hydroperoxide in the presence of trifluoroacetic anhydride and pyridine in nonaqueous medium at 0–5°C. No tert-butyl peroxytrifluoroacetate was formed as a by-product during the process. A possible reaction mechanism is discussed.  相似文献

Treatment of 2-nitro-N-(2-methyl-1-propen-1-yl)benzenamines with potassium tert-butoxide in tert-butanol followed by the addition of an electrophile affords N-alkoxy-2H-benzimidazoles. Electrophiles including methyl iodide, allylic bromides, propargylic bromides, benzyl bromide, and acetyl chloride gave good to excellent yields of product while 1-iodo- and 2-iodo-butane afforded very low yields.  相似文献

A convenient and practical method for the preparation of enantiomerically pure α-trifluoromethylated α-propargylamines is described. A range of enantiopure α-trifluoromethylated α-propargyl sulfinamides were obtained by the addition of lithium acetylides generated in situ with n-BuLi and terminal alkynes to diverse chiral CF3-substituted (S)-N-tert-butanesulfinyl ketimines in moderate to excellent yields (56–97%) and with uniformly excellent diastereoselectivities (>99:1) by using Ti(OiPr)4 as the catalyst and THF as the polar solvent. Enantiomerically pure α-trifluoromethylated α,α-dibranched propargyl amines were then readily obtained in excellent yields (87–97%) by acidic cleavage of the tert-butanesulfinyl group.  相似文献

A reaction of potassium tert-butoxide with chloromethyltrichlorosilane in THF leads to tri(tert-butoxy)chloromethylsilane. Upon reflux in THF, it reacted with magnesium with the formation of stable [tri(tert-butoxy)silyl]methylmagnesium chloride, whose structure was confirmed by 1H, 13C, and 29Si NMR spectroscopy. In the THF solution at 25 °C, [tri(tert-butoxy)silyl]methylmagnesium chloride exists in the equilibrium with bis{[tri(tert-butoxy)silyl]methyl}magnesium (in the ratio of 1: 1).  相似文献

将不同配比的吡唑与格氏试剂反应制得的吡唑基镁卤化物/四氢呋喃(THF)溶液用作可充镁电池电解液,采用循环伏安和恒电流充放电测试研究了该电解液的镁沉积-溶出性能和氧化分解电位;并通过X射线衍射(XRD)和扫描电镜(SEM)对沉积物的组分和形貌进行了分析. 结果表明,吡唑上的取代基、吡唑与格氏试剂的反应配比对电解液的电化学性能都有影响. 1 mol·L-1 1-甲基吡唑-PhMgCl(1:1摩尔比)/THF反应配制的电解液在不锈钢(SS)集流体的阳极氧化分解电位达到2.4 V(vs Mg/Mg2+),并具有镁沉积-溶出电位低、循环稳定性高、配制方便的特点,有希望应用于实际的可充镁电池体系中.  相似文献

The lithiation of 4-heterosubstituted dibenzothiins 1 (phenoxathiin, phenothiazine and thianthrene) with lithium and a catalytic amount of 4,4′-di-tert-butylbiphenyl (DTBB, 7.5% molar) in THF at temperatures ranging from −90 to −78°C gives the corresponding functionalised organolithium intermediate I, which by reaction with different electrophiles [H2O, D2O, ButCHO, PhCHO, Ph(CH2)2CHO, Me2CO, Et2CO, (CH2)5CO, (CH2)7CO] at the same temperature, followed by hydrolysis, gives the expected functionalised thiols 2. Cyclisation of some thiols 2 under acidic conditions leads to the corresponding seven-membered dibenzo heterocycles 5. In the case of thianthrene 1c, after addition of a carbonyl compound as the first electrophile [MeCHO, ButCHO, Me2CO, Et2CO, (CH2)5CO], the corresponding intermediate II can be lithiated again and react with a second electrophile. Diols 3 are obtained after hydrolysis when a carbonyl compound [ButCHO, PhCHO, Ph(CH2)2CHO, Me2CO, Et2CO, (CH2)5CO] is used as the second electrophile. Acidic cyclisation of diols 3 gives substituted phthalans 6 in almost quantitative yields. Finally, in the case of using carbon dioxide as the second electrophile, phthalides 4 are obtained after acidic hydrolysis.  相似文献

The reaction of enolates of α-unsubstituted carboxamides 3 with diphenyl phosphorazidate (DPPA) and di(tert-butyl) dicarbonate (‘Boc anhydride’) in THF at ?78° yielded 2-{[(tert-butoxy)carbonyl]amino}carboxamides 5 (Scheme 2) which are derivatives of α-amino acids. The lithiation of 1H,3H-benzo[de]isochromene (6) with lithium and a catalytic amount of 4,4′-di-tert-butylbiphenyl (DTBB, 5% molar) in THF at −50 °C gives dianionic intermediate 7, which by reaction with different electrophiles {H2O, D2O, tBuCHO, PhCHO, Me2CO, (CH3CH2)2CO, [CH3(CH2)4]2CO, (CH2)5CO, (CH2)7CO, (−)-menthone} at the same temperature followed by hydrolysis leads to functionalised alcohols 8. If after addition of a carbonyl compound as the first electrophile [tBuCHO, (CH2)5CO, (−)-menthone], the resulting dialcoholate 9 is allowed to react at 0 °C, a second lithiation takes place to give intermediate 10 which by reaction with a second electrophile [H2O, tBuCHO, (CH2)5CO, CO2], yields, after hydrolysis, 1,8-difunctionalised naphthalenes 11. Cyclization under acidic conditions of diols 8e-i gives oxygen-containing eight-membered heterocycles, which are homologous to the starting material 6.  相似文献

Lithiation of 2,5-dimethylazaferrocene 1 with sec-BuLi/TMEDA in THF at −78 °C proceeds (as shown by quenching with D2O) to comparable extent on the methyl groups and the Cp ring. However, the outcome of the reaction of the lithiated 1 with an electrophile depends on the nature of this electrophile. In the reaction with 4-methoxybenzaldehyde only the product originated from the lateral lithiation is formed, whereas the reaction with 4-methoxyacetophenone and 4,4′-dimethoxybenzophenone afforded mixtures of the products resulting from lateral and ring-lithiation. Similar results were also obtained in the reaction of lithiated 1 with chlorodiphenylphosphine and diphenyl diselenide. On the other hand, the exclusive formation of the Cp-substituted product was observed in the reaction of lithiated 1 with N,N-dimethylformamide. The structures of selected products (oily compounds were transformed into the corresponding crystalline W(CO)5-complexes) were confirmed by X-ray diffraction. The presented reactions open a novel entry to specifically substituted azaferrocenes (especially those containing heteroatom substituents) with potential applications as ligands for the homogenous catalysis.  相似文献

A new approach to the synthesis of 4-amino-5-(tert-butyl-NNO-azoxy)-2-R-2H-1,2,3-triazole 1-oxides 1 was developed. Compounds 1 were obtained by reactions of 3-amino-4-(tert-butyl-NNO-azoxy)furoxan with aliphatic amines RNH2 (R = Me, Et, Pri, Bu, and But). 4-Amino-5-(tert-butyl-NNO-azoxy)-2-tert-butyl-2H-1,2,3-triazole 1-oxide was transformed under the action of acids into 4-amino-5-(tert-butyl-NNO-azoxy)-1-hydroxy-1H-1,2,3-triazole. Methylation of the latter with diazomethane mainly involves the O atom of the triazole oxide ring. Reduction of compounds 1 gave 4-amino-5-(tert-butyl-NNO-azoxy)-2-R-2H-1,2,3-triazoles and 4-amino-5-(tert-butyldiazenyl)-2-R-2H-1,2,3-triazoles (R = Me, Pri, and But). The structures of the compounds obtained were confirmed by 1H, 13C, and 14N NMR spectroscopy.  相似文献

Lithiation of a series of aryl benzyl ethers containing halogen substituents (-F, -Br, -I) was investigated. The resultant mono- and diorganolithium intermediates were converted into the corresponding aldehydes or diboronic acids in good yields. The dilithiation of aryl benzyl ethers containing a reactive hydrogen atom and halogen atom capable of halogen-lithium exchange proceeds quantitatively in THF at −50 °C. It was found that mono aryllithiums formed in the reaction can remove the reactive hydrogen atom from a molecule of aryl benzyl ether thus decreasing the yield of dilithiated species. This effect does not occur when t-BuLi is used instead of n-BuLi.  相似文献

The diastereoselective synthesis of 5,5-disubstituted 3,3-difluorotetrahydrofurans 2 was achieved using β,β-difluorohomoallylic alcohols 1 as the key starting material in the presence of ICl (3.0?equiv) and tert-BuOK (1.5?equiv) in THF. The corresponding iodocyclization products 2 were obtained in good to moderate yields with excellent diastereo- and regioselectivities (d.r.?=?>20/1, 5-endo-trig only).  相似文献

We report the synthesis of 1,1-dibromo-1-alkenes from partially and unprotected aldoses and the synthesis of glyco-1-ynitols from these dibromocompounds. The 1,1-dibromo-1-alkenes were obtained by the reaction of dibromomethyl-triphenylphosphonium bromide in the presence of zinc in refluxing 1,4-dioxane. As an example, when the reaction is performed on 2-deoxy-5-O-trityl-d-ribofuranose (1) the corresponding 1,1-dibromo-1-olefin, (2R,3S)-6,6-dibromo-1-O-trityl-hex-5-ene-1,2,3-triol (12), is obtained in 89% yield. These smooth reaction conditions led also to the achievement of the other olefins from other sugars with good yields (44-90%). The reaction of these olefins with n-butyllithium in THF at low temperature afforded the corresponding alkynes. So the reaction of (2R,3S)-6,6-dibromo-1-O-trityl-hex-5-ene-1,2,3-triol (12) with this alkyllithium reagent led to (2R,3S)-1-O-trityl-hex-5-yne-1,2,3-triol (23) in 87% yield. Other glyco-1-ynitols were obtained with satisfying yields (64-87%).  相似文献
