The partition coefficients, P, in the n-octanol/water system and the acidity constants, Ka, of 15 chlorinated phenols and guaiacols at 20° were simultaneously determined by investigating the pH dependence of the apparent partition coefficients, Pa. For determining Pa, the relative responses of individual phenols and guaiacols in the aqueous phase after equilibration were measured by glass-capillary gas chromatography with electron-capture detection and compared with those in the octanol phase before partition. Curvefitting, linear regression, and non-linear regression were used for treating the partition data. The values of pKa and log P were compared with the values which could be found in the literature, and are discussed in relation to their affinity to methanol/water and acetonitrile/water mixtures.  相似文献

Summary A group of 17 β-carbolines was studied in HPLC and TLC systems in order to predict their partition coefficients (log P values). On account of the basic or acid character of some of these compounds, an ion pairing system gave the best results. Both HPLC and TLC data were comparable for log P prediction but severe pH conditions required the use of TLC plates. Retention data are quantitatively related to lipophilicity (expressed as the Hansch constant) and polarity (as the inductive constant) of the solute molecule.  相似文献

A rapid screening assay for the determination of octanol-water partition coefficients (log P(OW)) of pharmaceuticals was developed by using pressure-assisted microemulsion electrokinetic chromatography (MEEKC). The microemulsion system contains 50 mM sodium dodecyl sulfate, 0.87 M l-butanol, 82 mM heptane, and 50 mM borate-phosphate (2:3) at pH 10. Ten standard compounds with known log P(OW) values from -0.26 to 4.88 were used for constructing the calibration curve of log P(OW) against the MEEKC retention factor, log k. The log P(OW) values of the compounds were calculated based on the log k values measured by MEEKC and the slope and intercept of the calibration curve. For 13 literature and 32 Roche compounds, about 90% of the log P(OW) values measured by MEEKC are within 0.5 log units of the values from the literature and potentiometric titration. The throughput is about 2 samples/h using +20 kV voltage plus 5 mbar air pressure for separation. This MEEKC method is applicable for log P(OW) screening of weakly basic, weakly acidic, and neutral pharmaceuticals with log P(OW) = 0-5 and pKa < or = 10.  相似文献

The measured partition data of 26 steroids for six different compositions of ternary liquid-liquid systems consisting of 2,2,4-trimethylpentane, ethanol and water are used in calculations of partition data for new phase system compositions. It was possible to reproduce the original data and to verify new experimental data by this calculation with high accuracy. Calculations were done in three ways. In the first, non-linear regression of the partition coefficients as a function of the phase system composition was examined. With this classical approach it is possible to calculate the values of the partition coefficients, solute by solute, with good accuracy. In the second approach, principal component factor analysis was used. The phase system-specific factors for further phase system compositions were calculated with a non-linear regression of these factors on the phase system composition. In combination with the solute-specific factors, the values of the partition coefficients for all solutes in any phase system composition could be predicted with excellent accuracy. In the third approach, target transformation factor analysis was applied. A relationship between the partition coefficient and the phase system composition was established on the basis of either the phase system composition itself or a simple function of it. This method also gave very accurate predictions of known and unknown partition coefficients.  相似文献

This paper describes the determination of Henry's law coefficients by means of the EPICS (equilibrium partitioning in closed systems) technique in combination with SPME (solid-phase microextraction). The use of solid-phase microextraction-sampling allowed us to extend the possibilities of the equilibrium partitioning in closed systems technique with respect to the range of Henry's law coefficients which can be measured. Whereas the equilibrium partitioning in closed systems technique is limited to determine air–water equilibrium partitioning of volatile compounds with Henry's law coefficients of at least 0.06 (dimensionless), the current method allowed to measure coefficients between 0.0023 and 13.5. In this way Henry's law coefficients of 20 compounds, being in a range covering five orders of magnitude, were measured with relative standard deviations between 1.0 and 19.8% (mean standard deviation: 5.7%; median of standard deviations: 4.8%, n=99). Several types of compounds were examined i.e. aliphatic hydrocarbons, monocyclic and polycyclic aromatic hydrocarbons, chlorinated and fluorinated compounds, ethers and esters, biphenyl and N-containing compounds, including compounds for which availability of experimental Henry's law coefficients is limited. Measurement of the equilibrium partitioning in the 2 to 25°C range allowed to establish relations of Henry's law coefficient as a function of temperature.  相似文献

With the aim of obtaining compounds with strong antitumor activity, a quantitative structure-activity relationship (QSAR) of antitumor phenolic compounds (long-chain phenols) was derived using the Hansch-Fujita equation. The ED50 values against Chinese hamster V-79 cells were analyzed in terms of log P as the hydrophobic parameter and the energy of the lowest unoccupied molecular orbital (ELUMO) calculated by using the modified neglect of differential overlap (MNDO) method as the electronic parameter, by means of multiple regression analysis. It was found that the activities mainly depended on log P (an optimum log P of 8.3) and a low-lying ELUMO value. 4-Undecylcatechol, selected on the basis of the above results, exhibited strong antitumor activity against Sarcoma 180 ascites and P-388 lymphocytic leukemia.  相似文献

Fluid Phase Equilibria, 8: 25–35.A method has been developed for measuring the partition coefficients of volatile compounds between the gas and liquid phases of aqueous systems, based on the direct analysis of both phases. A gas mixture containing a known proportion of a volatile compound is drawn through the liquid (water) until equilibrium is established. A defined volume of the liquid phase is then withdrawn through a porous-polymer trap while maintaining the system at equilibrium. The residual water in the trap is then expelled by a stream of nitrogen gas, and the deposit remaining is thermally desorbed and analyzed by gas chromatography. This approach, together with an experimental technique for producing gas mixtures containing an accurately known concentration of hydrocarbon at low values, makes it possible to determine accurately the partition coefficients of low-solubility compounds, such as for hydrocarbons in aqueous systems, at very low solute concentrations in the system. The method has been verified by measuring the partition coefficient of hexane between the gas and liquid phases of an aqueous system at various concentrations and temperatures.  相似文献

构建了定量结构-保留关系(QSRR)测定马兜铃酸A、马兜铃酸B、马兜铃内酰胺及白黎芦醇的正辛醇-水分配系数(K_(ow))。采用反相高效液相色谱(RP-HPLC)法,以甲醇-水为流动相,以16种已知K_(ow)值的酸性和中性苯系物为模型化合物,以保留时间两点校正法(DP-RTC)校正保留时间,并由Snyder-Soczewinski方程得100%水相保留因子k_w,建立了表观正辛醇-水分配系数K_(ow)″与k_w的定量关系(QSRR模型),并对模型进行了内、外部验证。结果显示,不同pH下的QSRR模型线性相关性良好(相关系数R~2为0.980~0.987),内部验证(交叉验证相关系数R_(cv)~2为0.982~0.988)和外部验证结果(6种验证化合物的相对误差RE为0.6%~10.9%)令人满意。将建立的QSRR模型应用于中药中4种潜在肝肾毒性化合物的K_(ow)测定,并与软件计算值、摇瓶法(SFM)实验值比较,结果显示该方法准确性更高,且简单快捷。该文提出的采用中性及酸性苯系物建立QSRR模型,通过对结构与性质相似的中药材组分进行RP-HPLC分析,得到各待测组分的保留时间即可获得其K_(ow)值的简便策略,解决了中药组分复杂且难以分离、无法通过SFM测定其K_(ow)值的问题,为通过定量-构效关系(QSAR)模型实现快速预测中药组分的肝肾毒性提供了可靠的K_(ow)数据。  相似文献

Water-soluble polymers (poly-N-vinylpyrrolidone, poly-N-vinylcaprolactam, polyethylene glycol) were used for the first time for extraction of phenols from aqueous solutions. The influence exerted by pH of the aqueous solution and kind of the polymer on the degree of recovery of phenol and some of its methyl and nitro derivatives was elucidated. Sorption of phenol, 3-cresol, 2-, 3-, 4-nitrophenols, 2,4-, 2,6-dinitrophenol, 2,4,6-trinitrophenol, and 1-naphthol by polyether- and polyester-type polyurethane foams (PUF) was investigated. The effects of sorption time, pH, phenol concentration and the structure of tested phenols and PUF were studied. The mechanism of sorption of tested compounds on foams is discussed. It is shown that the hydrophobicity (logP, octanol-water distribution constant) and pK(a) values of the compounds play an important role in the sorption process. A regression equation connecting distribution coefficient of phenols by PUF with their hydrophobicity parameter and pK(a) values were derived. Good correlation between logD and values logP and pK(a) was observed.  相似文献

The elution behaviour of a selection of sequence variant and modified insulins has been compared on a C18 reversed-phase column. Observed elution times were compared with those expected from the nature of the differences from bovine insulin. In some cases prediction rules established for peptides are adequate to explain the observed elution and in others detailed knowledge of the structure of the protein is of considerable importance in understanding elution behaviour.  相似文献

Microemulsion electrokinetic chromatography (MEEKC) was assessed as a tool for determination of octanol-water partition coefficients using 34 solutes encompassing 8 carbonate esters. It was confirmed that microemulsions containing 1.44-2.88% w/w SDS, 6.49% w/w 1-butanol, and 0.82% w/w n-heptane constitute a good model of octanol-water partitioning in the pH range of 1.4-7.4. Use of the migration index concept led to improved repeatability of the MEEKC method compared to the use of retention factors. Using a dynamical coating, a high electroosmotic flow at pH 1.4 and 4.75 was achieved expanding the practical pH working range of the MEEKC system. The correlation obtained between the migration index and log P was unaffected by pH indicating that the properties of the microemulsion droplets and, thus, partitioning are independent of pH. No evidence for congeneric behavior was found for the sample set comprising solutes with different hydrogen bonding properties suggesting that simple reference compounds can be used as calibrators. Lipophilicity estimates for the series of carbonate esters were obtained. The increase in lipophilicity with chain length was smaller than expected from the Hansch substituent constant, pi.  相似文献

Carbon tetrabromide (CBr4) catalyzes O-tert-butoxycarbonylation of functionalized phenols without any side reactions (bromination, addition of CBr3 to a double bond, and formation of symmetrical diaryl carbonates, cyclic carbonates, or carbonic-carbonic anhydrides). The parent phenols are regenerated from the O-t-Boc derivatives by the catalyst system CBr4-PPh3 without affecting other protecting groups (aryl alkyl ether, alkyl ester, and thioacetal) or competitive side reaction such as bromination, nitrene (from NO2) and alpha,alpha-dibromoolefine (with CHO/COMe) formation, and transesterification (with CO2Me/Et) taking place.  相似文献

The kinetic method is based on catalysis, by thiobenzamide and/or its derivatives, of the oxidation of azide by iodine at 25°C in 0.1 M acetate buffer (pH 4.7) containing iodate, iodide and azide. Thiobenzamide and its 4-methoxy-, 4-methyl-, 4-chloro- and 4-nitro derivatives (0.5–3 μM) are determined by measuring the decrease in the absorbance of iodine at 350 nm at a fixed time (2 min). The detection limits are 0.05–0.1 μM and the relative standard deviations are <2%. The proposed method is used to determine the partition coefficients of thiobenzamide and its derivatives in water/1-octanol; the equilibrated aqueous phase was analysed.  相似文献

The retention factors (k) of 104 hydrophobic organic chemicals (HOCs) were measured in soil column chromatography (SCC) over columns filled with three naturally occurring reference soils and eluted with Milli-Q water. A novel method for the estimation of soil organic partition coefficient (Koc) was developed based on correlations with k in soil/water systems. Strong log Koc versus log k correlations (r>0.96) were found. The estimated Koc values were in accordance with the literature values with a maximum deviation of less than 0.4 log units. All estimated Koc values from three soils were consistent with each other. The SCC approach is promising for fast screening of a large number of chemicals in their environmental applications.  相似文献
