Summary Molecularly imprinted polymers were prepared using 2-vinylpyridine and/or methacrylic acid as functional monomers in a self-assembly imprinting protocol. The resulting polymers were analyzed in aqueous media, and the effects from the pH of the mobile phase and the degree of added organic solvent were investigated. The results are indicative of the importance of ionic bonds in conjunction with hydrophobic interactions in the formation of the complexes between the analyte and the polymers.  相似文献

Molecular imprinting technology offers the unique opportunity to tailor chiral stationary phases with predefined chiral recognition properties by employing the enantiomers of interest as binding-site-forming templates. Added advantages, such as ease of preparation, chemical robustness, low-cost production, and the possibility of shaping molecularly imprinted polymers (MIPs) in various self-supporting formats, render them attractive materials for a broad range of chiral recognition applications. In this review a critical overview on recent developments in the field of MIP-based chiral recognition applications is given, focusing on separation techniques and molecular sensing. Inherent limitations associated with the use of enantioselective MIP materials in high-performance separation techniques are outlined, including binding site heterogeneity and slow mass transfer characteristics. The prospects of MIP materials as versatile recognition elements for the design of enantioselective sensor systems are highlighted.  相似文献

Molecularly imprinted polymers selective for phosphate were prepared with the two types of functional monomers, 1-allyl-2-thiourea and N-methyl-N′-(4-vinylphenyl)-thiourea, and the binding abilities of the polymers were evaluated. Phenylphosphonic acid or diphenyl phosphate were used as the template molecules and the imprinted polymers prepared with 1-allyl-2-thiourea as functional monomer showed high binding ability to phosphate in aqueous media and nearly 90% of phosphate could be recovered. Also, the imprinted polymer prepared with N-methyl-N′-(4-vinylphenyl)-thiourea as functional monomer had a high binding ability and specific interaction with phosphate in acetonitrile solution and over 90% of phosphate-derivatives could be recovered selectively.  相似文献

Three molecularly imprinted polymers (MIPs) were prepared corresponding to three structurally related template compounds 4-hydroxybenzoic acid (4-HBA), gentisic acid (GA) and salicylic acid (SA) that differ in intramolecular hydrogen bonding ability using acrylamide (AA) as a functional monomer. HPLC method was used to evaluate the binding performances of the MIPs to the templates and several analogues. The results showed that the difference in their molecular recognition ability was pronounced. The highest molecular recognition ability was observed for 4-HBA-imprinted polymer. It was proved that the hydrogen bond interaction between the functional monomer and the template (4-HBA) played a major role in the recognition process and Scatchard analysis showed that two classes of binding sites were formed in 4-HBA-imprinted polymer. Their dissociation constants were estimated to be 1.76×10−4 and 1.40×10−3 mol l−1, respectively. But for GA- or SA-imprinted polymer the molecular recognition ability was not improved compared to the blank polymer (BP). By comparison of the structures of the three templates, it was concluded that the molecular recognition ability will decrease when the template itself is able to form intramolecular hydrogen bond in the molecular imprinting process. This study will be helpful for us to understand the molecular recognition mechanism of MIPs and of instructive significance for the prediction of the selectivity of MIPs.  相似文献

This study aimed at elucidating the chiral recognition mechanism with molecularly imprinted polymers (MIPs) in aqueous environment. The system used ethylene glycol dimethacrylate (EGDMA), methacrylic acid (MAA), and 4-l-phenylalanylamino-pyridine (4-l-PheNHPy) as the cross-linking monomer, functional monomer and template, respectively, to assemble the imprinted polymer. A self-assembly mechanism, which includes the pre-organizing functional monomers around template before polymerization process, was proposed. This mechanism was supported by NMR titration test. Interactions between functional monomer and template were observed using UV-Vis spectroscopy of solutions of these components as well. These studies indicated a 1:2 molecular complex dominantly formed between 4-l-PheNHPy and MAA. Association constant was estimated to be 97,000 M−2. Based on these results, a model mainly involving two-spot interaction was proposed evolving from our reported concept of exact placement of functional group. Ionic interaction between the primary amino group of 4-l-PheNHPy and carboxylic acid group inside the microcavity on MIPs was believed to play a predominate role in the enantioselectivity as supported by the observation of the relationship between the retention factor of 4-l-PheNHPy and the pH of mobile phase. While thermodynamic study at different pH revealed that, the interaction between the pyridyl group of 4-l-PheNHPy and the carboxylic acid group on the MIPs is also strong, implying that it also plays a profound role in determining the highly chiral selectivity of MIPs.  相似文献

The molecular recognition properties of molecular imprinted polymers which bind the carbamate function were studied. Functional monomers potentially able to form non-covalent interactions with the model molecule N,O-dibenzylcarbamate were selected on the basis of a computational approach describing possible interactions between the template and a small library of vinylic monomers. These results were in accordance with N,O-dibenzylcarbamate batch-rebinding measurements performed on several miniMIPs prepared with the same library. From these preliminary results, four polymers were prepared by thermoinduced radical polymerization, using ethylene dimethacrylate as a cross-linker, chloroform (MIP1, MIP3) or acetonitrile (MIP2, MIP4) as porogens and methacrylic acid (MIP1, MIP2) or acrylamide (MIP3, MIP4) as functional monomers. Molecular recognition features of these materials were studied by high-performance liquid chromatography. In this manner selectivity was evaluated by considering the column retention of a library of sixteen structural analogues of dibenzylcarbamate, characterized by the transformation of the carbamate into a related function, or by the alteration of the molecular structure.The experimental results show that methacrylic acid is more efficient than acrylamide as a functional monomer (imprinting factors: MIP1 = 24.1, MIP2 = 25.6, MIP3 = 13.3, MIP4 = 2.44), and that chloroform enhances polymer selectivity. As regards structural motifs which conditionate the selectivity, the carbamate function strictly controls the presence/absence of molecular recognition, while shape and dimension of the substituents modulate the recognition itself. In particular, a marked recognition for analogs which were slightly bigger than the template was observed (N-benzyl-O-phenethylcarbamate: MIP1 α = 1.13, MIP2 α = 1.41, MIP3 α = 0.97; N-phenethyl-N-benzylcarbamate: MIP1 α = 1.61, MIP2 α = 1.17, MIP3 α = 0.81; N,O-diphenethylcarbamate: MIP1 α = 0.89, MIP2 α = 1.20, MIP3 α = 0.55).  相似文献

Micro-contact imprinting has been used to form thin-film molecular imprints of ovalbumin (OVA) in polymers supported on glass slides. Thermocalorimetric data was used to optimise the choice of functional monomer and cross-linker to maximise selectivity and minimise non-specific recognition.A polymer comprising polyethyleneglycol 400 dimethacrylate (95 vol.%) and methacrylic acid (5 vol.%) showed both maximum recognition for OVA when made as a molecularly imprinted polymer (MIP), and minimal recognition when made as a non-imprinted, i.e. control polymer. OVA rebinding to the molecularly imprinted polymer, from a buffered 2 µM OVA solution, was 1.55 × 10− 11 mol cm− 2, while the control polymer showed 10-fold less re-binding, i.e. 0.154 × 10− 11 mol cm− 2.Experiments in which human serum albumin (HSA), conalbumin, ovomucoid or lysozyme, were re-bound to the polymers, either as single proteins or in competition with OVA, showed them to have low affinity for the polymer formulation used. Of the competing proteins examined, in non-competitive binding experiments, HSA showed the greatest affinity 0.45 × 10− 11 mol cm− 2 for the OVA imprinted polymer. In two protein competition experiments, i.e. with OVA and a competing protein present at equal concentrations (2 µM), OVA binding to the OVA imprinted polymer was in all cases significantly greater than that of the competitor.  相似文献

Molecular imprinting technology can be used to generate specific artificial polymeric receptors, i.e., high affinity stationary phases, as already shown for peptides and many other food ingredients. In this work, polymers have been molecularly imprinted with three exemplary antioxidants, butylated hydroxyanisole (BHA), butylated hydroxytoluene (BHT) and propyl gallate (PG). Results of chromatographic evaluations are presented, demonstrating the specificity of the imprinted polymers (MIP) compared to non-specific control polymers (CP). For a BHA imprinted polymer and the respective analyte BHA a separation factor of α=kBHA-MIP′/kCP′=1.12 was found, whereas for the BHT-MIP a higher selectivity was determined with α=kBHT-MIP′/kCP′=1.47. Using the PG selective MIP for PG as analyte a value of α=kPG-MIP′/kCP′=1.24 could be achieved. Furthermore, results of extraction procedures based on MIP and CP phases are presented, demonstrating for instance a higher selectivity of the BHA-MIP in comparison with its CP when using methanol or acetonitrile as analyte solvent, and of the PG-MIP when performing the extraction in acetonitrile.  相似文献

Two series of molecularly imprinted polymers (MIPs) for the class-selective recognition of glucuronides have been prepared by using lipophilic substructures of the target analyte as template molecule and potent host monomers against oxyanions, that are expected to establish a strong stoichiometric interaction with the single carboxylic group of the template. The polymers were tested as stationary phases in liquid chromatography for specific recognition. A preliminary investigation of the imprinting properties of eleven MIPs was carried out, by comparing the retention time of the template and of structurally related compounds on the MIP column with that on the corresponding non-imprinted polymer (NIP). The two polymers showing the best performance were selected to further test cotinine, mycophenolic acid, testosterone and their respective glucuronides as model compounds. The high specificity obtained against glucuronides and the different chemical structure of the parent drug make the two MIPs class-selective imprinted receptors, also suitable for SPE application.  相似文献

The application of molecularly imprinted polymers in the selective adsorption of macromolecules such as proteins by monolithic protein‐imprinted columns requires a macroporous structure, which can be provided by cryogelation at low temperature in which the formation of ice crystals gives a porous structure to the molecularly imprinted polymer. In this study, we applied this technique to synthesize lysozyme‐imprinted polyacrylamide cryogels containing 8% w/v of total monomers and 0.3% w/v of lysozyme. The synthesized cryogel was sponge‐like and elastic with very fast swelling and reshaping properties, showing a swelling ratio of 24.5 ± 3 and gel fraction yield of about 72%. It showed an imprinting effect of 1.58 and a separation factor of 1.37 for cytochrome c as the competing protein. Adsorption studies on the cryogel revealed that it follows the Langmuir isotherm, with a maximum theoretical adsorption capacity of 36.3 mg lysozyme per gram of cryogel. Additionally, it was shown that a salt‐free rebinding solution at low flow rate and pH = 7.0 is favorable for lysozyme rebinding. This kind of monolithic column promises a wide range of application in separation of various biomolecules due to its preparation simplicity, good rebinding characteristics, and macroporosity.  相似文献

A rational design approach was taken to the planning and synthesis of a molecularly imprinted polymer capable of extracting caffeine (the template molecule) from a standard solution of caffeine and further from a food sample containing caffeine. Data from NMR titration experiments in conjunction with a molecular modelling approach was used in predicting the relative ratios of template to functional monomer and furthermore determined both the choice of solvent (porogen) and the amount used for the study. In addition the molecular modelling program yielded information regarding the thermodynamic stability of the pre-polymerisation complex. Post-polymerisation analysis of the polymer itself by analysis of the pore size distribution by BET yielded significant information regarding the nature of the size and distribution of the pores within the polymer matrix. Here is proposed a stepwise procedure for the development and testing of a molecularly imprinted polymer using a well-studied compound—caffeine as a model system. It is shown that both the physical characteristics of a molecularly imprinted polymer (MIP) and the analysis of the pre-polymerisation complex can yield vital information, which can predict how well a given MIP will perform.  相似文献

Dibenzofuran (DBF) recognition was studied by a molecularly imprinted polymer (MIP) made of commercially available polymers: polyvinyl chloride (PVC), polysulfone, polystyrene, and polyacrylonitrile. Preparation of MIPs was carried out by phase inversion of the polymer. Respective polymer solutions were coagulated with DBF template in water (non-solvent). In addition, reference polymers without the template were prepared in same manner as unimprinted polymers. After the DBF template was extracted from the solidified polymers by washing with methanol (MeOH), recognition experiments of DBF by the imprinted polymers were carried out for DBF in MeOH solutions. Equilibrium constants for DBF were calculated from saturation binding kinetics. Equilibrium experiment results reveal that recognition was effective for DBF binding to the DBF-imprinted polymer, especially for the imprinted PVC.  相似文献

Surface imprinting over nanosized support materials is particularly suitable for protein templates, considering the problems with mass transfer limitation and low binding capacity. Previously we have demonstrated a strategy for surface protein imprinting over vinyl-modified silica nanopartiles with lysozyme as a model template by polymerization in high-dilution monomer solution to prevent macrogelation. Herein, the synthesis process was further studied toward enhancement of the imprinting performance by examining the effect of several synthesis conditions. Interestingly, the feed crosslinking degree was found to have a great impact on the thickness of the formed imprinting polymer layers and the recognition properties of the resulting imprinted materials. The imprinted particles with a crosslinking degree up to 50% showed the best imprinting effect. The imprinting factor achieved 2.89 and the specific binding reached 23.3 mg g−1, which are greatly increased compared to those of the lowly crosslinked imprinted materials reported previously. Moreover, the relatively high crosslinking degree led to no significant retarding of the binding kinetics to the imprinted particles, and the saturated adsorption was reached within 10 min. Therefore, this may be a promising method for protein imprinting.  相似文献

A new approach, combining metal coordination with the molecular imprinting technique, was developed to prepare affinity materials. Magnetic poly(glycidyl methacrylate) microspheres in monosize form were used for specific recognition toward the target protein. The magnetic poly(glycidyl methacrylate) microspheres were prepared by dispersion polymerization in the presence of magnetite nanopowder. Surface imprinted magnetic poly(glycidyl methacrylate) microspheres based on metal coordination were prepared and used for the selective recognition of human serum albumin. Iminodiacetic acid was used as the metal coordinating agent and human serum albumin was anchored by Cu2+ ions on the surface of magnetic poly(glycidyl methacrylate) microspheres by metal coordination. The magnetic poly(glycidyl methacrylate) microspheres were coated with a polymer formed by condensation of tetraethyl orthosilicate and 3‐aminopropyltrimethoxysilane. The human serum albumin imprinted magnetic poly(glycidyl methacrylate) microspheres were characterized by scanning electron microscopy, attenuated total reflectance Fourier transform infrared spectroscopy and particle size analysis. The maximum adsorption capacity of human serum albumin imprinted magnetic poly(glycidyl methacrylate) microspheres was 37.7 mg/g polymer at pH 6.0. The selectivity experiments of human serum albumin imprinted magnetic poly(glycidyl methacrylate) microspheres prepared with different concentrations in the presence of lysozyme, bovine serum albumin and cytochrome C were performed in order to determine the relative selectivity coefficients.  相似文献

A phosphate-selective molecularly imprinted polymer was prepared using 1-allyl-2-thiourea as a functional monomer, and the binding ability and selectivity of the polymer were evaluated. The imprinted polymer showed high binding ability to and selectivity for phosphate in aqueous media. The recoverability of phosphate from the imprinted polymer was also examined, and nearly 70% of highly concentrated phosphate could be recovered.  相似文献

Molecular imprinted polymers (MIPs) prepared using combination of acrylamide (ACM) and 4-vinylpyridine (4-Vpy) as co-functional monomers exhibited efficient recognition properties in both organic and aqueous media as HPLC stationary phase. The results indicate that amide and pyridine groups in functional monomers formed strong hydrogen-bonding interaction with the template molecule, and specific recognition sites were created within the polymer matrix during the imprinting process. When sulfamethoxazole (SMO) was used as template, a MIP prepared in a polar organic solvent (acetonitrile) using the combination of ACM and 4-Vpy showed better recognition of template than the polymer prepared in the same solvent using the combination of acidic monomer (methacrylic acid) and basic monomer 4-Vpy. On the contrary, when sulfamethazine (SMZ) was used as template, a MIP prepared using the combination of methacrylic acid (MAA) and 4-Vpy showed better recognition of template than the polymer prepared using the combination of ACM and 4-Vpy. Our results indicate that in organic media the degree of retention of the sample molecules on the imprinted polymers was controlled by the hydrogen-bonding interaction between the sample molecules and the polymer, while in aqueous media it was determined to a considerable extent by hydrophobic interactions. In both media the shape, size and the electronic structure of the template molecule were all-important factors in the recognition process.  相似文献

A highly selective molecularly imprinted polymer (MIP) for the recognition of the pesticide carbaryl in water has been synthesized using halogenated bisphenol A compounds as one of the polymeric precursors and carbaryl as the template molecule. On the basis of the heavy-atom effect, both the brominated and the iodinated MIPs allowed analyte detection by room-temperature-phosphorescence measurements. In the presence of an oxygen scavenger (sodium sulphite) the halide, included in the polymeric structure, induced efficient room-temperature phosphorescence of the analyte (once it had been selectively retained by the MIP). The MIP cavity can be easily regenerated for subsequent sample injections with 2 mL methanol. The optosensing system developed has demonstrated high selectivity for carbaryl, even in the presence of other luminophores that could be unspecifically adsorbed onto the MIP surface. Under optimal experimental conditions, the detection limit for the target molecule was 4 μg/L (3-mL sample injection volume), and the linear range extended up to 1 mg/L of the analyte. Good reproducibility was achieved (a relative standard deviation of 3% was obtained for ten replicates of 150 μg/L carbaryl). The synthesized sensing material showed good stability for at least 3 months after preparation. Finally, the applicability to carbaryl determination in real samples was evaluated through the successful determination of the pesticide in spiked mineral and tap water samples. The aim of this study was the evaluation of the binding performances and selectivity of molecularly imprinted beads prepared toward several penicillins (i) by hierarchical bulk polymerization in the pores of template‐grafted silica microbeads (hMIPs) and (ii) by Pickering emulsion polymerization in the presence of template‐decorated silica nanobeads (pMIPs). 6‐Aminopenicillanic acid was chosen as the common fragmental mimic template. Both approaches produced micron‐sized polymeric beads with good recognition properties toward the target ligands whereas the selectivity pattern appeared quite different. The polymer prepared by the Pickering emulsion approach showed binding properties similar to imprinted beads prepared by hierarchical approach. Equilibrium binding constants changed their values from 0.1–0.2 × 106 (hMIPs) to 0.2–0.6 × 106 M?1 (pMIPs), while the binding site densities changed from 3.7–4.8 (hMIPs) to 0.3–0.55 μmol/g (pMIPs). Compared to the hierarchical polymerization, Pickering emulsion polymerization represents a more practical approach when a template mimic needs to be used.  相似文献

Uniformly sized molecularly imprinted polymers (MIPs) for Boc-l-Trp were prepared using ethylene glycol dimethacrylate (EDMA) as the cross-linker, and methacylic acid (MAA) and/or 4-vinylpyridine (4-VPY) as the functional monomers or without use of a functional monomer. The MIPs prepared were evaluated using acetonitrile or a mixture of phosphate buffer and acetonitrile as the mobile phase. The Boc-l-Trp-imprinted EDMA polymers can recognize Boc-l-Trp by its molecular shape, and can thus afford the enantioseparation of Boc-Trp. Besides the molecular shape recognition, the hydrophobic interactions with the polymer backbones as well as the hydrogen-bonding interactions of Boc-l-Trp with carboxyl and pyridyl groups in the polymers should work for the retention and recognition of Boc-l-Trp on the imprinted MAA-co-EDMA and 4-VPY-co-EDMA polymers, respectively, in the hydro-organic mobile phase. The hydrogen-bonding interactions seem to become dominant when only acetonitrile is used as the mobile phase. The Boc-l-Trp-imprinted 4-VPY-co-EDMA polymers gave the highest retentivity and enantioselectivity for Boc-Trp among the MIPs prepared. However, the simultaneous use of MAA and 4-VPY was not effective for the enantioseparation of Boc-Trp in a hydro-organic mobile phase. Furthermore, the baseline separation of Boc-Trp enantiomers was attained within 10 min on the Boc-l-Trp-imprinted 4-VPY-co-EDMA polymers under the optimized HPLC conditions.  相似文献

Three kinds of molecularly imprinted polymers (MIPs) were obtained with surface molecular imprinting technique on functionalized potassium tetratitanate whisker (F-PTW). The results of adsorption experiments indicated that MIP prepared using PTW modified with N-(2-aminoethyl)-3-(trimethoxysilyl)propylamine (AAPTS) (F-PTW A) as support [MIP(1)] was superior to the other two polymers, then MIP(1) was selected to analyze the 4-nitrophenol (4-NP) adsorption process from aqueous solution in this study. AAPTS offered hydrophilic exterior that allowed to self-assemble with the template 4-NP through intermolecular interaction rather than based on the interactions between the functional monomers and template. Equilibrium adsorption data were analyzed by the Langmuir and Freundlich isotherm models at various temperatures. Kinetic properties were successfully investigated by pseudo-first-order model, pseudo-second-order model, intraparticle diffusion equation, initial adsorption rate, half-adsorption time. A diffusion-controlled process as the essential adsorption rate-controlling step was also proposed. The performance of such imprinted polymer was further demonstrated by high-performance liquid chromatography, and the results showed that the selectivity of MIP(1) exhibited higher affinity for template 4-NP over competitive phenolic compounds than that of non-imprinted polymer NIP(1). MIP(1) could be reused four times without significant loss in the adsorption capacity.  相似文献
