The formation of covalently linked composites of multi–walled carbon nanotubes (MWCNT) and glucose oxidase (GOD) with high-function
density for use as a biosensing interface is described. The reaction intermediates and the final product were characterized
by using FT–IR spectroscopy, and the MWCNT-coated GOD nanocomposites were examined by atomic force microscopy (AFM) and transmission
electron microscopy (TEM). Interestingly, it was found that the GOD–MWCNT composites are highly water soluble. Electrochemical
characterization of the GOD–MWCNT composites that were modified on a glassy carbon electrode shows that the covalently linked
GOD retains its bioactivity and can specifically catalyze the oxidation of glucose. The oxidation current shows a linear dependence
on the glucose concentration in the solution in the range of 0.5–40 mM with a detection limit of 30 μM and a detection sensitivity
of 11.3 μA/mMcm2. The present method may provide a way to synthesize MWCNT related composites with other biomolecules and for the construction
of enzymatic reaction-based biofuel cells and biosensors.

In this work we present a strategy for the covalent immobilization of periodate oxidized glucose oxidase () to aminated silica nanoparticles (ASNPs) modified on gold electrodes. Silica nanoparticles greatly enhanced the catalytic
ability of GOx toward the oxidation of glucose and improved the electron transfer between the GOx and the electrode surface.
ASNPs of varying size—that is 100, 80, 60, and 30 nm—were prepared, and they were used to fabricate biosensors. Electrochemical
impedance spectroscopy (EIS) of ferrocyanide followed the assembly process and verified the successful immobilization of on ASNPs modified on gold electrodes. From the analysis of catalytic signals of biosensors using different sizes of ASNPs
under the same conditions, the surface concentration of electrically wired enzyme (Γ
ET) was estimated and was found to increase with decreasing ASNPs size. Therefore, the sensitivity of biosensors using smaller
ASNPs was higher than that using larger particles. Specifically, we utilized the ASNPs with optimal size (30 nm) to fabricate
the glucose biosensor. The resulting electrodes showed a wide linear response to glucose at least to 6 mM and reached 95%
of the steady-state current in less than 4 s with a sensitivity of 5.02 μA mM−1 cm−2 and a detection limit of 0.01 mM. The biosensor also showed excellent stability and good reproducibility.

Macroporous conducting polymer films were prepared by the electrochemical copolymerization of 3-methylthiophene and thiophene-3-acetic acid on the ITO-coated glass plates bearing different sizes of polystyrene template particles, and enzyme electrodes were fabricated by covalent immobilization of glucose oxidase on the macroporous copolymer films. It was found that the doping level and conductivity of the copolymer films was significantly affected by the treatment with solvent to remove the polystyrene particles, which was considered to result in deterioration in amperometric glucose-responding property of the enzyme electrodes fabricated with the copolymer films. Three-dimensionally ordered macroporous structure on the copolymer films led to enhancement of amperometric response of the enzyme electrodes, and this effect was attributed to the geometry of the interconnected channel structure formed by the linkage of macropores. It was suggested that the amperometric response of the enzyme electrodes was determined by whether the interconnected channel structure on the copolymer films had long distance regularity and a proper size to allow the enzyme and electron-mediator molecules to penetrate into the interior pores of the copolymer film. In particular, the interconnected channel structure seemed to play an important role in the electron-transfer reaction between the mediator molecules and the surface of electrodes. 相似文献

Sodium alginate (AlgNa) and poly(diallyldimethylammonium chloride) (PDDA) were mixed to obtain an interpenetrating polymer composite via electrostatic interaction and then cast on an Au electrode surface, followed by incorporation of metal ions (e.g. Fe3+ or Ca2+, to form AlgFe or AlgCa hydrogel) and glucose oxidase (GOx) (or lactate oxidase (LOx)), to prepare amperometric enzyme electrodes. The interactions of PDDA, Alg, and Fe3+ are studied by visual inspection as well as microscopic and electrochemical methods. Under optimized conditions, the PDDA-AlgFe-enzyme/Au and PDDA-AlgCa-enzyme/Au electrodes can give good analytical performance (e.g. nM-scale limit of detection of glucose or lactate, and sensitivities > 50 μA cm−2 mM−1) in the first-generation biosensing mode, which are better than the reported analogs using typical polysaccharide biopolymers as enzyme-immobilization matrices. The enzyme electrodes also worked well in the second-generation biosensing mode in the coexistence of p-benzoquione or ferrocene monocarboxylic acid artificial mediator. Biofuel cells (BFCs) with the enzyme electrodes as the bioanodes and glucose (or lactate) as the biofuel were also fabricated with satisfactory results. The proposed protocols for preparation of high performance Alg-based biocomposites may find wide applications in bioanalysis. 相似文献

A new approach to constructing an enzyme-containing film on the surface of a gold electrode for use as a biosensor is described.
A basic multilayer film (BMF) of (PDDA/GNPs)
n
/PDDA was first constructed on the gold electrode by electrostatic layer-by-layer self-assembly of poly(diallyldimethylammonium
chloride) (PDDA) and gold nanoparticles (GNPs). Glucose oxidase (GOx) was then sorbed into this BMF by dipping the BMF-modified
electrode into a GOx solution. The assembly of the BMF was monitored and tested via UV-vis spectroscopy and cyclic voltammetry
(CV). The ferrocenemethanol-mediated cyclic voltammograms obtained from the gold electrode modified with the (PDDA/GNPs)
n
/PDDA/GOx indicated that the assembled GOx remained electrocatalytically active for the oxidation of glucose. Analysis of
the voltammetric signals showed that the surface coverage of active enzyme was a linear function of the number of PDDA/GNPs
bilayers. This result confirmed the penetration of GOx into the BMF and suggests that the BMF-based enzyme film forms in a
uniform manner. Electrochemical impedance measurements revealed that the biosensor had a lower electron transfer resistance
(R
et) than that of a sensor prepared by layer-by-layer assembly of PDDA and GOx, due to the presence of gold nanoparticles. The
sensitivity of the biosensor for the determination of glucose, which could be controlled by adjusting the number of PDDA/GNPs

A glucose oxidase-based needle-type microsensor is described that is independent of the local oxygen concentration. The sensor consists of an enzyme-coated platinum microelectrode that is inserted in a glass capillary with a tip of 5–25 μm, and connected to the environment via an agar membrane. Oxygen is supplied to the enzyme coating from the shaft of the capillary. Depending on the electrode configuration, the sensor has a response time of 1–10 s, and the measuring range extends from 0.01 to 0.4 mmol l?1 or from 0.4 to 10 mmol l?1. A simple, rapid, reproducible and effective one-step procedure for biosensor construction, based on casting mixed poly(ester-sulfonic acid)-glucose oxidase solutions onto solid electrodes is described. The cation exchanger simultaneously performs the entrapment, charge permselectivity and antifouling functions. The discrimination against potential interferences is accompanied with a fast and sensitive response to glucose. With flow injection operation, 180 samples can be processed per hour, with a detection limit of 3 × 10?6 M. The effects of numerous experimental variables are explored. These properties of poly(ester-sulfonic acid) ionomers, coupled with their availability as aqueous dispersions, make them attractive and versatile for biosensor work. 相似文献

A mediator-free glucose biosensor, termed a “third-generation biosensor,” was fabricated by immobilizing glucose oxidase (GOD)
directly onto an oxidized boron-doped diamond (BDD) electrode. The surface of the oxidized BDD electrode possesses carboxyl
groups (as shown by Raman spectra) which covalently cross-link with GOD through glutaraldehyde. Glucose was determined in
the absence of a mediator used to transfer electrons between the electrode and enzyme. O2 has no effect on the electron transfer. The effects of experimental variables (applied potential, pH and cross-link time)
were investigated in order to optimize the analytical performance of the amperometric detection method. The resulting biosensor
exhibited fast amperometric response (less than 5 s) to glucose. The biosensor provided a linear response to glucose over
the range 6.67×10−5 to 2×10−3 mol/L, with a detection limit of 2.31×10−5 mol/L. The lifetime, reproducibility and measurement repeatability were evaluated and satisfactory results were obtained. 相似文献

将层层自组装技术与仿生矿化技术相结合,由聚苯乙烯磺酸钠、聚二甲基二烯丙基氯化铵和二氧化硅成功制备(聚苯乙烯磺酸钠-聚二甲基二烯丙基氯化铵)2-二氧化硅复合微囊.采用扫描电子显微镜、红外光谱和热重对微囊的形貌和化学结构进行了表征.以该复合微囊作为理想载体固定化葡萄糖氧化酶.结果表明,固定于复合微囊中的葡萄糖氧化酶的热稳定性、pH稳定性、操作稳定性得到了提高;在最适条件下,复合微囊固定化葡萄糖氧化酶的酶活回收率为72.85%,米氏常数是游离葡萄糖氧化酶的2.21倍.复合微囊在化学/生物催化、药物/基因传递系统和生物传感器应用方面具有一定的潜能. 相似文献

We are proposing for the first time the use of a Nafion/multi-walled carbon nanotubes dispersion deposited on glassy carbon electrodes (GCE) as a new platform for developing enzymatic biosensors based on the self-assembling of a chitosan derivative and different oxidases. The electrodes are obtained by deposition of a layer of Nafion/multi-wall carbon nanotubes dispersion on glassy carbon electrodes, followed by the adsorption of a chitosan derivative as polycation and glucose oxidase, l-aminoacid oxidase or polyphenol oxidase, as polyanions and biorecognition elements. The optimum configuration for glucose biosensors has allowed a highly sensitive (sensitivity = (0.28 ± 0.02) μA mM−1, r = 0.997), fast (4 s in reaching the maximum response), and highly selective (0% interference of ascorbic acid and uric acid at maximum physiological levels) glucose quantification at 0.700 V with detection and quantification limits of 0.035 and 0.107 mM, respectively. The repetitivity for 10 measurements was 5.5%, while the reproducibility was 8.4% for eight electrodes. The potentiality of the new platform was clearly demonstrated by using the carbon nanotubes/Nafion layer as a platform for the self-assembling of l-aminoacid oxidase and polyphenol oxidase. Therefore, the platform we are proposing here, that combines the advantages of nanostructured materials with those of the layer-by-layer self-assembling of polyelectrolytes, opens the doors to new and exciting possibilities for the development of enzymatic and affinity biosensors using different transdution modes. 相似文献

The direct electrochemistry of glucose oxidase (GOD) immobilized on a hexagonal mesoporous silica modified glassy carbon electrode was investigated. The adsorbed GOD displayed a pair of redox peaks with a formal potential of -417 mV in 0.1 M pH 6.1 phosphate buffer solution (PBS). The response showed a diffusion-controlled electrode process with a two-electron transfer coupled with a two-proton transfer reaction process. GOD immobilized on a hexagonal mesoporous silica retained its bioactivity and stability. In addition, the immobilized GOD could electrocatalyze the oxidation of glucose to gluconlactone by taking ferrocene monocarboxylic acid (FMCA) as a mediator in N(2) saturated solutions, indicating that the electrode may have the potential application in biosensors to analyze glucose. The sensor could exclude the interference of commonly coexisted uric acid, p-acetaminophenol and ascorbic acid and diagnose diabetes very fast and sensitively. This work demonstrated that the mesoporous silica provided a novel matrix for protein immobilization and the construction of biosensors. 相似文献

Glucose oxidase electrodes of a terpolymer poly(aniline-co-o-anisidine-co-o-toluidine) as biosensors
A simple technique is described for constructing a glucose sensor by the entrapment of glucose oxidase (GOD) in a poly (aniline-co-o-anisidine-co-o-toluidine) [P(A-co-o-A-co-o-T)] thin films, which were electrochemically deposited on a platinum plate in phosphate and acetate buffer. The maximum current response was observed for the said electrodes at pH 5.5 and potential 0.60 V (versus Ag/AgCl). The phosphate buffer gives high stability and fast response as compared to acetate buffer in amperometric measurements. 相似文献

A monolithic silica gel matrix with entrapped glucose oxidase was constructed as a bioactive element in an optical biosensor
for glucose determination. Physicochemical and biochemical characterizations of the catalytic matrix were performed, and the
intrinsic fluorescence of immobilised glucose oxidase (GOD) was investigated in the UV and visible range by performing steady
state and time course measurements. In all cases, the silica gel matrix proved to be a suitable support for optical biosensing
owing to its superior optical properties (e.g., high transmittance and reliable fluorescence and GOD absorption spectra after
immobilisation). From steady state measurements, calibration curves were obtained as a function of glucose concentration.
When time course measurements were performed, the silica gel support displayed a larger linear calibration range and higher
sensitivity than other immobilisation systems. In addition, a glucose optical biosensor was developed and characterised using

A highly sensitive amperometric nanobiosensor has been developed by integration of glucose oxidase (GOx) with a gold nanowires array (AuNWA) by cross-linking with a mixture of glutaraldehyde (GLA) and bovine serum albumin (BSA). An initial investigation of the morphology of the synthesized AuNWA by field emission scanning electron microscopy (FESEM) and field emission transmission electron microscopy (FETEM) revealed that the nanowires array was highly ordered with rough surface, and the electrochemical features of the AuNWA with/without modification were also investigated. The integrated AuNWA–BSA–GLA–GOx nanobiosensor with Nafion membrane gave a very high sensitivity of 298.2 μA cm−2 mM−1 for amperometric detection of glucose, while also achieving a low detection limit of 0.1 μM, and a wide linear range of 5–6000 μM. Furthermore, the nanobiosensor exhibited excellent anti-interference ability towards uric acid (UA) and ascorbic acid (AA) with the aid of Nafion membrane, and the results obtained for the analysis of human blood serum indicated that the device is capable of glucose detection in real samples. 相似文献

Direct electron transfer between active glucose oxidase (GOD) and a gold electrode was obtained when GOD was immobilized in poly(N-methylpyrrole) electrochemically prepared on the gold electrode. When electropolymerization was accomplished at 50 °C, after glucose addition, the cyclic voltammograms showed an increased oxidation peak at ca. ?0.45 V vs. Ag/AgCl. The application of an inexpensive, easy-to-make and disposable biosensor, based on a chemically modified graphite electrode with an adsorbed and cross-linked layer of glucose dehydrogenase, as a detector in a flow-injection system for on-line monitoring of glucose in fermenters is described. The performance of the sensor as a function of time is investigated, and used for assaying glucose in a wine fermentation process. It is concluded that the sensor operates satisfactorily during at least 3 days of continuous use. No interference from ethanol is encountered. With regard to practical applications, special emphasis is placed on the interfacing of the fermenter and the analytical system. 相似文献

A glucose biosensor using an enzyme-immobilized eggshell membrane and oxygen electrode for glucose determination has been fabricated. Glucose oxidase was covalently immobilized on an eggshell membrane with glutaraldehyde as a cross-linking agent. The glucose biosensor was fabricated by positioning the enzyme-immobilized eggshell membrane on the surface of a dissolved oxygen sensor. The detection scheme was based on the depletion of dissolved oxygen content upon exposure to glucose solution and the decrease in the oxygen level was monitored and related to the glucose concentration. The effect of glutaraldehyde concentration, pH, phosphate buffer concentration and temperature on the response of the glucose biosensor has been studied in detail. Common matrix interferents such as ethanol, d-fructose, citric acid, sodium benzoate, sucrose and l-ascorbic acid did not give significant interference. The resulting sensor exhibited a fast response (100 s), high sensitivity (8.3409 mg L−1 oxygen depletion/mmol L−1 glucose) and good storage stability (85.2% of its initial sensitivity after 4 months). The linear response is 1.0×10−5 to 1.3×10−3 mol L−1 glucose. The glucose content in real samples such as commercial glucose injection preparations and wines was determined, and the results were comparable to the values obtained from a commercial glucose assay kit based on a spectrophotometric method. 相似文献

We have constructed a glucose biosensor by immobilizing glucose oxidase (GOD) on titanium-containing MCM-41 (Ti-MCM-41) modified screen-printed electrodes. The strategy of the sensing method is to monitor the extent of the decrease of the reduction current of O2 upon adding glucose at a selected potential. The detection can be done at the applied potential of −0.50 V and can efficiently exclude the interference from commonly coexisted substances. The constructed sensor has a high sensitivity to glucose (5.4 mAM−1 cm−2) and a linear response range of 0.10-10.0 mM. The detection limit is 0.04 mM at a signal-to-noise ratio of 3. The sensor also shows high stability and remains its catalytic activity up to 60 °C. The biocompatibility of Ti-MCM-41 means that this immobilization matrix not only can be used for immobilizing GOD but also can be extended to other enzymes and bioactive molecules, thus providing a promising platform for the development of biosensors. 相似文献

An amperometric glucose biosensor was developed using an anionic clay matrix of hydrotalcitic nature (Ni/Al-NO3 HT) as enzyme support, which was electrochemically synthesized at −0.90 V versus SCE, using a rotating disk Pt electrode to assure homogeneity of the electrodeposition suspension. The biorecognition element was glucose oxidase (GOx) immobilized on HT during the electrosynthesis, which was followed by cross-linking with glutaraldehyde vapours to avoid the enzyme release.The performances of the biosensor, in terms of sensitivity to glucose calculated from the slope of the calibration curve, are dependent on parameters related to the electrodeposition.An experimental design was applied to detect the optimal conditions of electrosynthesis in order to optimize the glucose biosensor performance. The factors taken into account were enzyme concentration and Ni/Al molar ratio. A full factorial design was performed to study linear interactions between factors and their quadratic effects and the optimal setup was evaluated by the isoresponse curves. The significant factors were enzyme concentration (linear and quadratic terms) and the interaction between enzyme concentration and Ni/Al molar ratio. Under the optimized electrodeposition conditions, the reproducibility of the biosensor fabrication was very good, being the RSD of the sensitivity about 5%. 相似文献

A one-step procedure for the immobilization of glucose oxidase in fused-silica capillaries and in glass microchips was developed based on enzyme entrapment in a polyacrylamide-based monolithic column. The inner capillary surface was silanized with gamma-methacryloxypropyltrimethoxysilane (gamma-MAPS) to allow covalent binding of the gel to the surface. The composition of the polymer was optimized to prevent the formation of bubbles, allow liquid transportation by electroosmotic flow and to maintain the enzymatic activity. These requirements resulted in the addition of polyethylene glycol and poly(acrylic acid) to the acrylamide mixture. The gel containing the enzyme was formed in situ in the capillaries, respectively, in one channel of the microchip. In the microchip, it was limited to the sample injection channel by accordingly controlled silanization of the inner capillary surface. Glucose was detected via the amperometric determination of hydrogen peroxide. A linear correlation between signals and glucose concentration was observed from 0.05 to 1.1 mM glucose with a correlation coefficient of 0.999. The enzymatic monolithic microreactor showed no loss of activity during 8 h of continuous use and during storage in the running buffer at 4 degrees C for about 2 months. Interferents, such as ascorbic acid, were separated from the analyte electrophoretically, so that glucose could be quantified in diluted juices. 相似文献
