Size-exclusion chromatography with multi-angle laser light scattering (SEC-MALLS) and acid–base titration were used to determine the molecular weight of a polyethylene glycol with low molecular weight. All potential uncertainty factors for each measurement were evaluated using cause-effect diagrams. Weight analysis was applied to harmonize the two different measurement results. It was found that the main uncertainty sources of SEC-MALLS measurement arose from the dn/dc value, the calibration constant of the differential refractive index (DRI) detector, and the intermediate precision of the instrument, while the sample mass, the titration volume and the concentration of titrant contributed to the uncertainty of titration measurement. The weighted mean value of the two measurement results was taken as the molecular weight of the polyethylene glycol.  相似文献

An analytical method based on matrix-assisted laser desorption ionization (MALDI) time-of-flight mass spectrometry has been developed to provide information on oligomer structure, average molecular weight, and molecular weight distributions of polydienes (e.g., polybutadiene and polyisoprene), an important class of industrial polymers. This MALDI method involves the use of all-trans-retinoic acid as the matrix, copper (II) nitrate as the cationization reagent, and tetrahydrofuran as the solvent. The incorporation of this copper salt generates Cu+ adducts with the polymer chains. It also improves the signal strength and extends the upper mass range when used with all-trans-retinoic acid, as compared to silver nitrate. With this formulation, it is demonstrated that polybutadienes of narrow polydispersity with masses up to 300,000 u and polyisoprenes of narrow polydispersity with masses up to 150,000 u can be analyzed. The upper molecular weight limit is set by the requirement of using higher matrix-to-polymer ratios with increasing polymer molecular weight, to the point where the instrument can no longer detect the small quantity of polymer present in the matrix host. It is also shown that this sample preparation generates previously unreported adduction behavior. The practical implications of this adduction behavior on polymer structural analysis, accuracy of molecular weight determination, and the upper molecular weight limit of oligomer resolution are discussed. It is illustrated that, in a linear time-lag focusing MALDI instrument, oligomer resolution can be obtained for polydienes with molecular weights up to 24,000, providing structural confirmation of the end-groups and the repeat unit. The average molecular weights of a number of polydienes of narrow polydispersity determined by MALDI are compared to those obtained by gel permeation chromatography, and discrepancies are noted.  相似文献

A simple, sensitive and fast method for the determination of melamine and its derivatives in milk powder using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) was developed. Neither time-consuming sample preparation, nor special target plates, or other extra equipment are necessary. The common matrix sinapinic acid (SA) was used with a dried-droplet preparation. Detection limits (signal-to-noise (S/N) ratio = 3) for standard solutions of melamine, ammeline and cyanuric acid were 10, 25 and 10 μg/L, respectively. The limit of quantification (LOQ) for melamine was 25 μg/L and excellent linearity (R(2): 0.9990) was maintained over the range of 10-2000 μg/L. Ammeline and cyanuric acid were analyzed with an LOQ of 50 μg/L and also excellent linearity (R(2): 0.9997 and R(2): 0.9998). Good accuracy and precision were obtained for all concentrations within the range of the standard curve. The developed method was successfully used for the determination of melamine, ammeline and cyanuric acid in milk powder samples with a simple sample preparation. The LOQ of melamine was 0.5 μg/g. Ammeline and cyanuric acid were detectable at 0.5 and 5 μg/g. This method showed excellent accuracy, precision and linearity and significantly reduces the needed analysis time, as only approximately 10 s/sample measuring time is required. To the authors' knowledge, this is the first published method to quantify melamine and derivatives by MALDI-TOF-MS.  相似文献

A sensitive LC method with pre-column fluorescence derivatization using 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate has been developed for the determination of defibrase from the venom of Agkistrodon acutus (also named defibrase in the Chinese Pharmacopoeia) in pharmaceutical formulation. By a denaturing procedure prior to derivatization, full and homogeneously derivatization of defibrase was obtained and demonstrated using MALDI-TOF-MS. The specificity of the method was validated by assessing interference from the placebo and by forced degradation. The method has demonstrated good linearity (r = 0.9998), accuracy (mean recovery 98.53%) and precision (RSD < 2.5%). Moreover, the method was found to be sensitive with a low limit of detection (3.35 ng mL?1) and limit of quantitation (10.16 ng mL?1). The method was successfully applied to the quantitative determination of defibrase in pharmaceutical formulation.  相似文献

To determine the accuracy to which hyperfine splittings can be measured using a cw dye laser with rf sideband tuning, a series of atomic beam experiments with59Co was performed. One hyperfine splitting of the first excited metastable atomic state was measured and compared with the results for the same splitting from magnetic rf resonance experiments. The uncertainty with the methods applied was found to be about 0.05 to 1% of the experimental linewidth in general; it is ±20 kHz or ±0.05% of the linewidth in the present Co experiment. The hyperfine splitting constants of the 3d 8 4p 2 F 7/2 state were found to beA=419.3(9) MHz,B=?77(17) MHz.  相似文献

报道以2,4,6-三羟基苯乙酮(2,4,6-THAP)为基体用基体辅助激光解吸/电离飞行时间质谱法(MALDI-TOF-MS)测定核酸分子的研究。2,4,6-THAP解吸和电离DNA分子的效率很高,测定DNA分子d(T)~1~0的[M-H]^-分辨率可达1130.0,信噪比为366.0,检出限达5×10^-^1^4mol,可以测定相对分子质量高达14800以上混合碱基组成的DNA分子。为提高基体对样品解吸/电离效率,对纯化DNA样品的方法进行了讨论。  相似文献

Software support for the Nordtest method of measurement uncertainty evaluation is described. According to the Nordtest approach, the combined measurement uncertainty is broken down into two main components??the within-laboratory reproducibility (intermediate precision) s Rw and the uncertainty due to possible laboratory bias u(bias). Both of these can be conveniently estimated from validation and quality control data, thus significantly reducing the need for performing dedicated experiments for estimating detailed uncertainty contributions and thereby making uncertainty estimation easier for routine laboratories. An additional merit of this uncertainty estimation approach is that it reduces the danger of underestimating the uncertainty, which continues to be a problem at routine laboratories. The described software tool??MUkit (measurement uncertainty kit)??fully reflects the versatility of the Nordtest approach: it enables estimating the uncertainty components from different types of data, and the data can be imported using a variety of means such as different laboratory data systems and a dedicated web service as well as manual input. Prior to the development of the MUkit software, a laboratory survey was carried out to identify the needs of laboratories related to uncertainty estimation and other quality assurance procedures, as well as their needs for a practical tool for the calculation of measurement uncertainty.  相似文献

Trichosporon jirovecii yeast cells are used for the first time as a source of l-cysteine desulfhydrase enzyme (EC 4.4.1.1) and incorporated in a biosensor for determining l-cysteine. The cells are grown under cadmium stress conditions to increase the expression level of the enzyme. The intact cells are immobilized on the membrane of a solid-state Ag2S electrode to provide a simple l-cysteine responsive biosensor. Upon immersion of the sensor in l-cysteine containing solutions, l-cysteine undergoes enzymatic hydrolysis into pyruvate, ammonia and sulfide ion. The rate of sulfide ion formation is potentiometrically measured as a function of l-cysteine concentration. Under optimized conditions (phosphate buffer pH 7, temperature 37 ± 1 °C and actual weight of immobilized yeast cells 100 mg), a linear relationship between l-cysteine concentration and the initial rate of sulfide liberation (dE/dt) is obtained. The sensor response covers the concentration range of 0.2-150 mg L−1 (1.7-1250 μmol L−1) l-cysteine. Validation of the assay method according to the quality control/quality assurance standards (precision, accuracy, between-day variability, within-day reproducibility, range of measurements and lower limit of detection) reveals remarkable performance characteristics of the proposed biosensor. The sensor is satisfactorily utilized for determination of l-cysteine in some pharmaceutical formulations. The lower limit of detection is ∼1 μmol L−1 and the accuracy and precision of the method are 97.5% and ±1.1%, respectively. Structurally similar sulfur containing compounds such as glutathione, cystine, methionine, and d-cysteine do no interfere.  相似文献

The clinical effectiveness of succinylated gelatin as a plasma substitute depends strongly on its molar mass, determined conventionally by size exclusion chromatography (SEC). This study evaluates different SEC calibration standards in comparison with two independent "absolute" methods for determining the weight average molar mass (M(w)) of a succinylated gelatin sample. SEC calibrated using succinylated gelatin fractions correlated well with size exclusion chromatography-multi-angle laser light scattering (SEC-MALLS) and sedimentation equilibrium whereas SEC calibrated with unmodified gelatin, sodium polystyrene sulfonates or pullulans overestimated M(w) by over 20%. Universal calibration was equivocal. The problems associated with the preparation of succinylated gelatin fractions suggest that an absolute method such as SEC-MALLS may be a more suitable choice for determining the M(w) in succinylated gelatins.  相似文献

The separation of long-chain polyprenols was successfully achieved using supercritical fluid chromatography (SFC). Each 100-mer greater component was separated using tetrahydrofuran as a mobile phase modifier. The molecular mass distributions derived from SFC analyses agreed with the results of matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) analyses. The number-average molecular mass calculated by MALDI-TOF-MS data were also in accord with the results of quantitative 1H-NMR analysis of terminal groups. A combination of SFC and MALDI-TOF-MS analyses is a powerful tool for the elucidation of the complicated structures of natural polyprenols.  相似文献

Size-exclusion chromatography (SEC) coupled with online laser light-scattering (LS) and refractive index (RI) detection provides an excellent approach to determine the molecular weights (Mw) of proteins by the “two-detector” approach. Mw is determined only at the maximum of a peak, using either peak heights or area ratio from the two detectors. However, proper calibration of the SEC/LS/RI system is critical to obtain high precision.Today, an essential part of any analysis is to evaluate the uncertainty associated with the method. Basically, it is possible to distinguish between factors related to signal nature, precision and those due to signal processing. Given the signal of interest is the peak height or area ratio from two detectors, the signal ratio uncertainty was calculated using the random propagation of error formula. In this case, the effect of signal correlation was evaluated to avoid the uncertainty overestimation. In the second case, the sources of uncertainty affecting analytical measurement were estimated with the information from the precision assessment. For this, two designs with two-factor fully nested were followed for each method. Finally, the contributions from various uncertainty sources related with calibration are also analysed in detail. There are in fact only three main sources of measurement uncertainty: intermediate precision, calibration and repeatability. Of these, method precision is always the greatest, regardless of approach.For all proteins and peptides studied, the Mw calculated using both methods are close to the theoretical results, independently of the design, but the contributions of individual terms to combined uncertainty depend on both the design and method used. For example, the combined uncertainty varied between 223 and 813.2 Da for carbonic anhydrase, although higher values were found for human insulin and ovalbumin dimer. Other considerations that can have a significant impact on the results are discussed.The reproducibility of the two methods versus that based on ASTRA software used as reference method was performed using the concordance correlation coefficient. The methods’ reproducibility depends on the permitted losses in precision and accuracy.  相似文献

Realistic internal validation of a method implies the performance validation experiments under intermediate precision conditions. The validation results can be organized in an X Nr×Ns (replicates×runs) data matrix, analysis of which enables assessment of the accuracy of the method. By means of Monte Carlo simulation, uncertainty in the estimates of bias and precision can be assessed. A bivariate plot is presented for assessing whether the uncertainty intervals for the bias (E ± U(E)) and intermediate precision (RSDi ± U(RSDi) are included in prefixed limits (requirements for the method). As a case study, a method for determining the concentration of nitrate in drinking water at the official level set by 98/83/EC Directive is assessed by use of the proposed plot.  相似文献

In the present study, humic acids (HAs) were applied as both a matrix for matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) and an adsorbent of magnetic solid phase extraction (MSPE) for the first time. As natural macromolecule compounds, HAs are inherently highly functionalized and contain laser energy absorbing–transferring aromatic structures. This special molecular structure made HAs a good candidate for use as a MALDI matrix in small molecule analysis. At the same time, due to its good adsorption ability, HAs was prepared as MSPE adsorbent via a simple co-mixing method, in which the commercially available HAs were directly mixed with Fe3O4 magnetic nanoparticles (MNPs) in a mortar and grinded evenly and completely. In this process, MNPs were physically wrapped and adhered to tiny HAs leading to the formation of magnetic HAs (MHAs). To verify the bi-function of the MHAs, Rhodamine B (RdB) was chosen as model compound. Our results show that the combination of MHAs-based MSPE and MALDI-TOF-MS can provide a rapid and sensitive method for the determination of RdB in chili oil. The whole analytical procedure could be completed within 30 min for simultaneous determination of more than 20 samples, and the limit of quantitation for RdB was found to be 0.02 μg/g. The recoveries in chili oil were in the range 73.8–81.5% with the RSDs less than 21.3% (intraday) and 20.3% (interday). The proposed strategy has potential applications for high-throughput analysis of small molecules in complex samples.  相似文献

Matrix-assisted laser desorption ionization was performed on an external ion source Fourier transform ion cyclotron resonance mass spectrometer equipped with a 7-T superconducting magnet to analyze end groups of synthetic polymers in the mass range from 500 to 5000 u. Native, perdeutero methylated, propylated, and acetylated polyethylene glycol and polyvinyl pyrrolidone with unknown end-group elemental composition were investigated in the mass range up to 5000 u by using a 2,5-dihydroxybenzoic acid matrix. A small electrospray setup was used for the deposition of the samples. Two methods to process data were evaluated for the determination of end groups from the measured masses of the component molecules in the molecular weight ranges: a regression method and an averaging method. The averaging method is demonstrated to allow end-group mass determinations with an accuracy within 3 mu for the molecular weight range from 500 to 1400 and within 20 mu for the molecular weight range from 3400 to 5000. This is sufficient to identify the elemental composition of end groups in unknown polymer samples.  相似文献

In this study, complete purification and biochemical characterization of protein is presented. The protein was purified by using Sephadex G-75 gel filtration column followed by reverse-phase high-performance liquid chromatography in a C18 column. The molecular weight of the protein was determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis, mass spectrum matrix-assisted laser desorption/ionization-time-of-flight-mass spectrometry (MALDI-TOF-MS) and liquid chromatography-electrospray ionization tandem mass spectrometry. Protein was fragmented by trypsin based on the m/z values obtained by MALDI-TOF-MS analysis. The peptide fragments sequence showed homology with DEAD-box-ATP-dependent RNA helicase 45, present in a public domain, National Centre for Biotechnology Information. The protein exhibited antibacterial activity against selected Gram +/? bacteria. The analgesic activity was determined by conducting acetic-acid-induced writhing test in mice.  相似文献

A protocol for the preparation of polymeric samples for time-of-flight matrix-assisted laser desorption ionization mass spectrometry (TOF-MALDI-MS) analysis was developed. Dithranol was identified as a good matrix for polystyrene (PS), and the addition of silver for cationization of molecules was determined to be necessary. Based on this preparative method, low molecular weight samples of other polymers [polyisoprene, polybutadiene, poly(ethylene oxide), poly(methyl methacrylate), and polydimethylsiloxane] were analyzed with molecular weights up to 49 ku. The effects of laser intensity were determined to influence the molecular weight distribution of intact oligomers, most significantly for low molecular weight polymers. Linear and reflectron modes of analysis were evaluated; better signal intensity and resolution were obtained in the reflectron mode. The TOF-MALDI-MS measurements are compared with time-of-flight secondary ion mass spectrometry (TOF-SIMS) and gel permeation chromatography (GPC) for the same polymers. The M n values calculated by TOF-MALDI-MS consistently are higher than values calculated by TOF-SIMS for all classes of polymers with molecular weights up to 8 ku. The molecular weights of the PS calculated from TOF-MALDI-MS are in good agreement with GPC (±10%). The composition of the terminal group on a polymer chain may affect the ion yields. The ion yields of intact oligomers were evaluated as a function of end group composition for both TOF-MALDI-MS and TOF-SIMS. The slight disparity of results between TOF-SIMS and TOF-MALDI-MS for the perfluoroalkyl-terminated PS suggests that the oligomers are desorbed preferentially from the surface in the TOF-SIMS analysis, rather than having an increased ionization probability.  相似文献

Continuous flow hydride generation coupled with laser induced fluorescence spectrometry (HG-LIF) has been used for the determination of Pb in aqueous samples. Lead hydride is generated in K3Fe(CN)6-HCl medium using NaBH4 as the reducing agent. Parameters such as acidity and the concentrations of oxidising and reducing agents have been studied in order to obtain the highest sensitivity. Laser excitation of Pb has been performed at 283.306 nm and fluorescence has been detected at 365 nm and 405.8 nm. The limit of detection calculated for the optimised conditions (1% K3Fe(CN)6 in 0.3% Oxalic Acid, 0.055 mol L?1 HCl, 1% NaBH4 in 0.1% NaOH) is 0.3 ng mL?1. However, a significant Pb background was observed for blank measurements, which degraded the limit of detection. The limit of detection that is estimated for a Pb free blank was determined by detuning the laser to a wavelength of 282.806 nm and was found to be 0.45 pg mL?1. Method repeatability is 3.5% RSD at the 10 ng mL?1 level. The accuracy of this method has been evaluated by analysis of a water reference material. The results obtained for a multielement standard determined using the HG-LIF approach have been found to be in agreement with those obtained by using a comparison ICP-AES approach. The HG-LIF approach has been applied to the measurement of Pb in a contaminated sediment sample material. The results of this study demonstrate that a small tunable laser system combined with hydride generation sample introduction can be used to measure Pb with high sensitivity, precision and selectivity in different sample matrices.  相似文献

Two cellouronic acids [sodium (1 → 4)-β-polyglucuronates, CUAs] and one 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO)-oxidized wood cellulose (TOC) became soluble in 8 % lithium chloride/N,N-dimethylacetamide (LiCl/DMAc) after the methylation of C6 carboxyl groups in these samples using trimethylsilyldiazomethane (TMSD). The obtained solutions were diluted to 1 % LiCl/DMAc and subjected to size-exclusion chromatography combined with multi-angle laser-light scattering (SEC-MALLS). Neither depolymerization nor side reactions took place during methylation; this was confirmed by SEC-MALLS and nuclear magnetic resonance analyses, using CUAs as models. The SEC-MALLS analysis of the original wood cellulose and the carboxyl-methylated TOC prepared from it, using 1 % LiCl/N,N-dimethyl-2-imidazolidinone and 1 % LiCl/DMAc, respectively, as eluents, showed that the weight-average degree of polymerization of the original wood cellulose decreased from 3,100 to 2,210 through TEMPO-mediated oxidation. The molecular-mass distributions of the original wood cellulose and the TOC both consisted of one large peak with a small shoulder, indicating that some of the oxidized hemicelluloses remained in the TOC. The combination of methylation of carboxyl groups in polysaccharides using TMSD and subsequent SEC-MALLS analysis using 1 % LiCl/DMAc as an eluent may be applicable not only to TOCs, but also to other polysaccharides with carboxyl groups, for evaluation of their molecular-mass parameters.  相似文献
