The Pd0‐catalyzed C(sp3)‐H arylation of 2‐bromo‐N‐methylanilides leads to unstable benzazetidine intermediates that rearrange to benzoxazines through 4π electrocyclic ring‐opening and 6π electrocyclization. The introduction of a bulky, non‐activatable amide group on the nitrogen atom was key to favor the challenging reductive elimination step and disfavor undesired reaction pathways.  相似文献

Free to decide : Various 4‐(α‐styryl) γ‐lactams are synthesized in 61–88 % yield by a phosphine‐free palladium‐catalyzed carbopalladation/allylic alkylation domino sequence (see scheme). The cyclization is totally regio‐ and diastereoselective in favor of the 3,4‐trans‐disubstituted γ‐lactam. The process is successfully applied to the synthesis of a new aza analogue of the naturally occurring lignan (+)‐oxo‐parabenzlactone.

Diphenyl(3‐methyl‐2‐indolyl)phosphine (C9H8NPPh2, 1 ) gives stable dimeric palladium(II) complexes that contain the phosphine in P,N‐bridging coordination mode. On treating 1 with [Pd(O2CCH3)2], the new complexes [Pd(μ‐C9H7NPPh2)(NCCH3)]2 ( 2 ) or [Pd(μ‐C9H7NPPh2)(μ‐O2CCH3)]2 ( 3 ) were isolated, depending on the solvent used, acetonitrile or toluene, respectively. Further reaction of 3 with the ammonium salt of 1 led to the substitution of one carboxylate ligand to afford [Pd(μ‐C9H7NPPh2)3(μ‐O2CCH3)] ( 4 ), in which the bimetallic unit is bonded by three C9H7NPPh2? moieties and one carboxylate group. Using this methodology, [Pd2(μ‐C6H4PPh2)2(μ‐C9H7NPPh2)(μ‐O2CCX3)] (X=H ( 7 ); X=F ( 8 )) were synthesised from the ortho‐metalated compounds [Pd(C6H4PPh2)(μ‐O2CCX3)]2 (X=H ( 5 ); X=F ( 6 )). Complexes 3 , 4 , 7 , and 8 have been found to be active in the catalytic β‐boration of α,β‐unsaturated esters and ketones under mild reaction conditions. Hindrance of the carbonyl moiety has an influence on the reaction rate, but quantitative conversion was achieved in many cases. More remarkably, when aryl bromides were added to the reaction media, complex 7 induced a highly successful consecutive β‐boration/cross‐coupling reaction with dimethyl acrylamide as the substrate (99 % conversion, 89 % isolated yield).  相似文献

The first palladium‐catalyzed method for the arylation of α‐phosphonovinyl nonaflates is described. Using a catalyst comprised of Pd(OAc)2 and SPhos, terminal and internal α‐aryl vinylphosphonates could be efficiently accessed under mild conditions. The reaction features a broad coupling partner scope and tolerates many functional groups.  相似文献

A systematic study on the carbonylative transformation of 1‐bromo‐2‐fluorobenzenes with various nucleophiles has been performed. Different types of double nucleophiles, such as N?N, N?C, O?C, and N?S, can be effectively applied as coupling partners. The corresponding six‐membered heterocycles were isolated in moderate to good yields.  相似文献

The palladium‐catalyzed reaction of 2‐alkynylanilines with 2‐(2‐bromobenzylidene)cyclobutanone as an efficient route to 7,8‐dihydrobenzo[b]naphtho[2,3‐d]azocin‐6(5 H)‐ones was developed. The fused eight‐membered ring was constructed conveniently. During the reaction process, double carbometalation was involved, which resulted in excellent selectivity with the formation of three new bonds. This transformation is highly efficient and leads to fused polycycles in good to excellent yields with good functional group tolerance.  相似文献

Palladium complexes incorporating chiral N‐heterocyclic carbene (NHC) ligands catalyze the asymmetric intramolecular α‐arylation of amides producing 3,3‐disubstituted oxindoles. Comprehensive DFT studies have been performed to gain insight into the mechanism of this transformation. Oxidative addition is shown to be rate‐determining and reductive elimination to be enantioselectivity‐determining. The synthesis of seven new NHC ligands is detailed and their performance is compared. One of them, L8 , containing a tBu and a 1‐naphthyl group at the stereogenic centre, proved superior and was very efficient in the asymmetric synthesis of fifteen new spiro‐oxindoles and three azaspiro‐oxindoles often in high yields (up to 99 %) and enantioselectivities (up to 97 % ee; ee=enantiomeric excess). Three palladacycle intermediates resulting from the oxidative addition of [Pd(NHC)] into the aryl halide bond were isolated and structurally characterized (X‐ray). Using these intermediates as catalysts showed alkene additives to play an important role in increasing turnover number and frequency.  相似文献

The indium trichloride‐catalyzed reaction between aromatic imines and α,β‐unsaturated N,N‐dimethylhydrazones in acetonitrile afforded 1,2,3,4‐tetrahydroquinolines bearing a hydrazone function at C4 through a one‐pot diastereoselective domino process that involves the formation of two C? C bonds and the controlled generation of two stereocenters, one of which is quaternary. This reaction constitutes the first example of an α,β‐unsaturated dimethylhydrazone that behaves as a dienophile in a hetero Diels–Alder reaction. The related reaction between anilines, aromatic aldehydes, and methacrolein dimethylhydrazone in CHCl3 with BF3?Et2O as catalyst afforded polysubstituted 1,2,3,3a,4,8b‐hexahydropyrrolo[3,2‐b]indoles as major products through a fully diastereoselective ABB′C four‐component domino process that generates two cycles, three stereocenters, two C? C bonds, and two C? N bonds in a single operation.  相似文献

Enantioenriched N‐allyl tetrahydro‐β‐carbolines were prepared by chiral phosphoric acid‐catalyzed Pictet–Spengler reactions. The compounds undergo Pd0‐catalyzed cyclizations through a tandem deprotection/cyclization process. The regioselectivity of the attack is controlled by the chain length and by the substitution pattern of the allene function. Products resulting from 5‐exo‐ or 6‐exo‐attack were obtained with diastereoisomeric ratio up to 95:5. Azepinopyrrido[3,4‐b]indoles were obtained by 7‐endo‐cyclizations.  相似文献

A general catalytic protocol for the α‐arylation of aryl ketones has been developed. It involves the use of a preformed, bench‐stable Pd–N‐heterocyclic carbene pre‐catalyst bearing IHept as an ancillary ligand, and allows the coupling of various functionalized coupling partners at very low catalyst loading. Careful choice of the solvent/base system was crucial to obtain optimum catalyst performance. The pre‐catalyst was also successfully tested in the synthesis of an industrially relevant compound.  相似文献

A mild, palladium(II)‐catalyzed reaction of α‐allenols with α‐allenic esters in a heterocyclization/cross‐coupling sequence, applicable to a wide range of substitution patterns, has been developed for the preparation of 2,3,4‐trifunctionalized 2,5‐dihydrofurans. Our studies indicate high levels of chemo‐ and regiocontrol. The possibility of using optically active substrates as well as substrates of increased steric demand, such as tertiary α‐allenols, makes this novel sequence of heterocyclization/cross‐coupling an attractive method in organic synthesis. The current mechanistic hypothesis invokes a regiocontrolled palladium(II)‐mediated intramolecular oxypalladation of the free allenol component, that then undergoes a cross‐coupling reaction with the allenic ester partner, followed by a trans‐β‐deacyloxypalladation with concomitant regeneration of the PdII species.  相似文献

Treatment of 1,4‐disubstituted 1,2,3‐triazoles with aryl chlorides in the presence of potassium carbonate under palladium catalysis and microwave irradiation at 250 °C for 15 min leads to arylation of the triazole at the 5‐position. A variety of functional groups, including ester and hydroxy groups, are compatible. The procedure is suitable for the regioselective preparation of trisubstituted triazoles. Microwave irradiation accelerates the reaction, thus allowing the rapid synthesis of trisubstituted triazoles, which are difficult to synthesize selectively.  相似文献

A novel and efficient palladium‐catalyzed C2 arylation of N‐substituted indoles with 1‐aryltriazenes for the synthesis of 2‐arylindoles was developed. In the presence of BF3 ? OEt2 and palladium(II) acetate (Pd(OAc)2), N‐substituted indoles reacted with 1‐aryltriazenes in N,N‐dimethylacetamide (DMAC) to afford the corresponding aryl–indole‐type products in good to excellent yields.  相似文献

Sterically hindered substrates can be employed in an enantioselective palladium‐catalyzed α‐arylation with the chiral monophosphorus ligand BI‐DIME. This process enabled an efficient synthesis of the antidepressant (S)‐nafenodone, a four‐step enantioselective synthesis of the Sceletium alkaloid (+)‐sceletium A‐4, a concise five‐step enantioselective synthesis of (?)‐corynoline, as well as a three‐step preparation of (?)‐DeN‐corynoline.  相似文献
