The separation of polyethylene glycols and maleimide‐substituted polyethylene glycol derivatives based on the number of maleimide end‐groups under critical liquid chromatography conditions has been investigated on a reversed‐phase column. The critical solvent compositions for nonfunctional polyethylene glycols and bifunctional maleimide‐substituted polyethylene glycols were determined to be identical at about 40% acetonitrile in water on a reversed‐phase octadecyl carbon chain‐bonded silica column using mixtures of acetonitrile and water of varying composition as the mobile phase at 25°C. The maleimide‐functionalized polyethylene glycols were successfully separated according to maleimide functionality (with zero, one, two, or three maleimide end‐groups, respectively) under the critical isocratic elution conditions without obvious effect of molar mass. The separation was mainly due to the hydrophobic interaction between the maleimide end‐groups and the column packing. Off‐line matrix‐assisted laser desorption/ionization time of flight mass spectrometry was used to identify the repeating units and, especially, the end‐groups of the maleimide‐substituted polyethylene glycols. Liquid chromatography analysis at critical conditions could provide useful information to optimize the synthesis of functional polyethylene glycols. To our knowledge, this is the first report of the baseline separation of maleimide‐functionalized polyethylene glycols based on the functionality independent of the molar mass without derivatization by isocratic elution.  相似文献

Summary The concept of retention prediction for the separation of phenylthiohydatoin-amino acid derivatives in isocratic reversed-phase liquid chromatography is described. A novel retention-solubility parameter, R, is defined, for the retention prediction strategy and the performance of this R value is evaluated by comparing measured and predicted retention data. Excellent agreement between these values were observed. It is concluded that the R value has a very high potential in describing the retention of phenylthiohydantopinamino acid derivatives withdifferent types of separation systems consisting of C-18, C-8 and phenethyl bonded stationary phases and various mobile phases.  相似文献

The separation and characterization of fluorescein-labeled polyethylene glycols (PEG) is described. Firstly, the polyethylene glycols labeled with fluorescein isothiocyanate (FITC) were synthesized and separated using Sephadextrade mark LH-20 medium by a step gradient. Secondly, a TSK GEL G4000 PW XL column was developed for determining the FITC derivatives of PEG. The retention mechanism is based on the hydrophobic interaction between the FITC derivatives of PEG and the packing material of the TSK GEL G4000PW XL column. The retention time of the PEG compounds increased by adding of salts in the mobile phase and decreased by adding of organic modifier. In addition, various salts in the eluent can also change the chromatographic behavior of these compounds. Finally, the pH of the mobile phase can have an impact on the retention time of the PEG compounds.  相似文献

考察了烷基酚聚氧乙烯醚在反相色谱、正相键合色谱、硅胶吸附色谱、体积排阻色谱4种不同液相色谱分离模式中的分离效果,分别采用Kromasil C_(18)(250 mm× 4.6 mm,5 μm)、Agilent ZORBAX NH2(250 mm× 4.6 mm,5 μm)、Waters Spherisorb S3W(150 mm×2.0 mm,3 μm)和Shodex MSpak GF-310 2D(150 mm×2.0 mm,5 μm)色谱柱,以225 nm为紫外检测波长,对不同液相色谱分离模式的流动相组成、梯度洗脱条件、柱温、流速等进行了优化,并对烷基酚聚氧乙烯醚在不同液相色谱分离模式中的保留机理进行了初步探讨.结果表明,正相键合色谱实现了烷基酚聚氧乙烯醚的最佳分离;硅胶吸附色谱和体积排阻色谱的分离效果较正相键合色谱稍差.  相似文献

A mixed-mode chromatographic packing material, C18 and diol groups modified silica (C18-Diol), was prepared with controllable hydrophobicity and hydrophilicity. It demonstrated excellent aqueous compatibility and stability in aqueous mobile phase; compared to the traditional C18 column, improved peak shape of basic analytes was also obtained. Additionally, it exhibited both reversed-phase liquid chromatographic (RPLC) and hydrophilic interaction chromatographic (HILIC) performance; the analyte separation scope was thus enlarged, demonstrated by simultaneous separation of twenty acids, bases and neutrals. More interestingly, a novel on-line two-dimensional liquid chromatography on the single column (2D-LC-1C) was established by modifying the high performance liquid chromatographic instrument only with the addition of an extra six-port two-position valve. The early co-eluted components of the extract of Lonicera japonica on the 1st-dimension (RPLC) were collected for the online re-injection to the 2nd-dimension (HILIC) by conveniently varying the mobile phase components. Six more peaks were obtained. The established system was simple, easy operation and low cost, which had advantages in analyzing complicated samples.  相似文献

Summary The feasibility of using columns prepared by mixing together ion exchange and reversed-phase packing materials, for the simultaneous separation of ionised and unionised molecules, has been examined using a range of test solutes. Columns prepared in this way showed chromatographic properties which were intermediate between those of the individual phases. The dual nature of the retention mechanism allowed the retention of ionisable molecules to be adjusted using pH whilst that of uncharged compounds was unaffected. The simultaneous chromatography of model compounds and their glucuronide, sulphate and glycine conjugates, under a variety of conditions, was demonstrated.  相似文献

The present study described the preparation and application of a reversed-phase/zwitterionic/hydrophilic interaction liquid chromatography stationary phase, named as SIL-PS. The SIL-PS was prepared through a four-step reaction, chemical bonding, nucleophilic addition, SN1 substitution, and sulfonation on the silica matrix. It was featured with C12 alkyl chain, quaternary ammonium, tertiary amine, and sulfonate groups. After SIL-PS was packed into the stainless steel column (150?× 2.1 mm i.d.), chromatographic parameters, including acetonitrile content, pH, and ionic strength of the mobile phase, and the column temperature, were systematically investigated to study the retention mechanism. Electrostatic adsorptive/repulsive, partition, and hydrogen-bonding interactions were demonstrated to contribute to the retention. The stability of the SIL-PS was satisfactory, with relative standard deviations of retention factors of 1.93, 2.08, and 1.90% for loxoprofen, adenosine, and liquiritin, respectively. Additionally, to investigate the separation selectivity, the non-steroidal anti-inflammatory drugs, nucleobases/nucleotides, and alkaloids/glycosides were separated; the HPLC fingerprinting of the Cortex phellodendri extract was also conducted, and the separation performance was superior to that of the C18 column in terms of peak shape, resolution, and analytical time. The results revealed that the prepared SIL-PS possessed multifunctionalities for multiretention and could be promising for complicated samples.  相似文献

将硅烷偶联剂γ-(2,3-环氧丙氧)丙基三甲氧基硅烷与十八酸反应,再键合到硅胶整体柱上,得到了酯型十八烷基键合固定相,并用红外光谱、元素分析对其进行了表征。在以甲醇-水为流动相的反相色谱条件下分离了苯、联苯和蒽的混合样品,评价了该整体柱的色谱性能,考察了该整体柱适用的pH范围,以及柱压降、柱效与流速的关系。结果表明,该硅胶整体柱键合效果良好,具有较好的反相色谱性能,且在pH=2~8时稳定性好,柱压降、柱效受流速影响较小,可有效地用于化合物的快速分离分析。  相似文献

Summary The present research studies the possibility of using the correlation dependence between molecular parameters of unsubstituted polyaromatic hydrocarbons (PAH) and their retention in reversed-phase liquid chromatography to optimize the conditions for the separation and identification of unknown peaks on the chromatograms of multicomponent mixtures. A linear correlation equation, that takes the number and environment of the carbon atom in the PAH molecule into account as well as the differences in the specific interactions of isomeric molecules with polar eluent, has been proposed. The adequacy of the proposed PAH retention model was verified by comparing the calculated retention values with the experimental data. The possibility of identifying unsubstituted PAH according to the number of carbon atoms of various types and according to the values of the molecules lengths (calculated on the basis of the retention of these substances under different eluent compositions) was exemplified by various chromatographic systems (reversed phase-eluent-PAH molecules).  相似文献

Summary An optimization strategy for the separation of a small number of peptides from a complex biological sample by two-dimensional liquid chromatography is presented. Ion-exchange chromatography is followed by reversed-phase separation. The ion-exchange separation is performed with a step gradient which admits a high sample load and simplifies instrumentation. The reversed-phase separation complements the first dimension with a different retention mechanism and higher resolution by linear gradient elution.Chromatographic theory is combined with experimental design to find separation conditions, for both dimensions, that allow the fastest gradient in the second dimension, giving short separation time, low detection limits and high load capacity. This is illustrated by the separation of a peptide from rat brain tissue, with a simple off-line arrangement. The strategy presented is useful in both analytical and preparative applications, and is widely applicable as it does not rely on special instrumentation or extensive knowledge of the sample.Dedicated to Professor Leslie S. The chromatographic behavior of 8 ionic liquids - 7 homologues of 1-alkyl-3-methylimidazolium and 4-methyl-N-butylpyridinium - has been investigated with a strong cation exchange adsorbent. In particular, the dependence of the retention properties of these solutes on mobile phase composition, pH, and buffer concentration was evaluated with the aim of optimizing and improving the selectivity and retention of solute separation. While using the SCX stationary phase, several interactions occurred with varying strengths, depending on the mobile phase composition. Cation exchange, nonspecific hydrophobic interactions, and adsorption chromatography behavior were observed. Reversed phase chromatography occurred at low concentrations of acetonitrile, electrostatic and adsorption interactions at higher organic modifier concentrations. Elevated buffer concentrations lowered the retention factors without affecting the selectivity of ionic liquids. Obtained results were further compared to the chromatographic behaviour of ionic liquids in the reversed phase system. All analyzed ionic liquids follow reversed-phase behavior while being separated. Much lower selectivity in the range of highly hydrophilic compounds is obtained. This suggests preferred use of ion chromatography for separation and analysis of compounds below 4 carbon atoms in the alkyl side chain.  相似文献

Selected hydrophilic interaction chromatography (HILIC) columns packed with bare silica, bridge-ethyl hybrid silica, or an amide sorbent chemistry were utilized for an investigation of chromatographic behavior and separation selectivity of tryptic peptides. Retention model was proposed allowing for retention prediction of peptides with correlation coefficient R(2)~0.92-0.97 for various columns. The values of optimized amino acid retention coefficients were compared to those obtained for reversed-phase liquid chromatography (Gilar et al., Anal. Chem. 2010, 82, 265-275) and used to elucidate the impact of different amino acid on peptide HILIC retention. In contrast to reversed-phase chromatography, where presence of Phe, Trp, Ile, and Leu amino acid residues in sequence strongly promoted, and presence of hydrophilic His, Lys and Arg residues strongly reduced peptide retention, the effects of these amino acid residues in HILIC were opposite (His, Lys and Arg promote, Phe, Trp, Ile and Leu demote peptide retention in HILIC). Retention coefficient optimized for pH experiments illustrated the impact of silanols on HILIC retention.  相似文献

Abstract

The retention of ethoxylated nonylhylphenyl surfactants was determined in reversed-phase high-performance liquid chromatography (RP-HPLC) using various supports (C1, C2, C6, C8, C18, polyethylene-coated silica, and polyethylene-coated alumina). The retention data matrix was evaluated both by principal component analysis and cluster analysis. The retention characteristics of both polyethylene-coated supports were similar to that of C1, the retention capacity of RP columns for surfactants increased with the increasing length of the covalently bonded hydrocarbon chain. The retention of ethoxylated surfactants depended nonlinearly on the length of ehtyleneoxide chain suggesting that the polar ethyleneoxide chains are in folded state under the chromatographic separation.  相似文献

用超临界流体色谱法进行了黄酮醇异构体的分离研究。考察了温度、压力、流动相组成、柱条件等对分离的影响。在实验的温度范围40~60℃和压力范围15~30MPa内,这组化合物都能得到很好的分离;流动相组成是影响色谱分离的最显著的因素,磷酸的加入大大改变了各物质的保留行为;考察了三种硅胶基质键合相对分离的影响,发现苯基柱用于这组异构体的分离最为合适。  相似文献

Summary The relationship between the logarithmic capacity factor measured in reversed-phase liquid chromatography and the operating conditions including the mobile phase composition and the column temperature is investigated. The strategy described herein can offer the possibility to predict the retention of polycyclic aromatic hydrocarbons without any experiments and standard materials, by utilizing equations describing the relationships between retention, temperature, mobile phase composition and physicochemical properties of the solutes previously stored in the program of the microcomputer-assisted retention prediction system.This concept is one of the most promising techniques for the optimization of the separation conditions in reversed-phase liquid chromatography.  相似文献

在反相和正相色谱模式下,研究了几种嘌呤衍生物在葫芦[6]脲单轮烷键合硅胶固定相上的高效液相色谱行为,并在反相模式下与ODS固定相进行了比较,考察了流动相中甲醇含量、流动相pH值和离子强度对嘌呤化合物保留的影响。研究结果表明:在反相模式下,嘌呤化合物与葫芦[6]脲单轮烷键合相之间存在多种相互作用,除疏水作用外,分离过程中还存在与ODS不同的色谱分离机制。在正相条件下,多作用力的色谱分离机制同样存在。葫芦[6]脲单轮烷键合相与溶质之间存在疏水、氢键、π-π和偶极-偶极等多种作用力,协同作用提高了固定相对嘌呤化合物的分离选择性。  相似文献
