Novel Synthesis of 5-Amino-3-bromo-1-(tert-butyl)-1H-pyrazole-4-carbonitrile: A Versatile Intermediate for the Preparation of 5-Amino-3-aryl-1-(tert-butyl)-1H-pyrazole-4-carboxamides

A simple, novel, and efficient route for the synthesis of 5-amino-3-aryl-1-(tert-butyl)-1H-pyrazole-4-carboxamides 1 has been devised. Preparation of pyrazole bromide 3 from potassium tricyanomethanide can be accomplished in only two steps in good yield and features a selective Sandmeyer reaction on the corresponding diaminopyrazole. This allows for a more versatile synthesis of 5-amino-3-aryl-1-(tert-butyl)-1H-pyrazole-4-carboxamides 1 than was previously possible.

Synthesis and reactions of 2, 5-dimethyl-4-(3′-hydroxy-3′-methyl-1′-butynyl)-4-piperidinol

The addition of dimethyl ethynyl carbinol to the carbonyl group of 2, 5-dimethyl-4-piperidinone under the conditions of the Favorskii reaction has been studied. It has been shown that the addition takes place stereo-selectively and leads to the formation of a single isomeric acetylenic glycol. The hydration of this glycol has given a heterocyclic tetrahydrofuran with a spiran structure. The exhaustive hydrogenation of the acetylenic glycol has yielded the corresponding saturated glycol. By its cyclization a tetrahydrofuran derivative has been synthesized.     

Radical addition reaction of alcohols to 1-(4-nitrophenyl)-5H-pyrrolin-2-one

Synthesis and Crystal Structure of 4-Bromo-5-ethoxy-3-methyl-5- （naphthalen- 1-yl）- 1-tosyl- 1H-pyrrol-2（5H）- one

The title compound 4-bromo-5-ethoxy-3-methyl-5-(naphthalen-1-yl)-1-tosyl-1H- pyrrol-2(5H)-one 1 (C24H22BrNO4S, Mr = 500.40) has been synthesized and its crystal structure was determined by single-crystal X-ray diffraction analysis. It crystallizes in monoclinic, space group P21/n with a = 8.8562(15), b = 18.118(3), c = 14.055(2) , β = 99.855(3)o, V = 2221.9(6) 3, Z = 4, Dc = 1.496 g/cm3, μ = 1.975 mm-1, λ = 0.71073 , F(000) = 1024, R = 0.0607 and wR = 0.1371.

The title compound 1-（（5-（4-（4-chlorophenoxy）-2-chlorophenyl）-2,2,3-trimethyl-oxazolidin- 5-yl）methyl）-lH-1,2,4-triazole （C21H22Cl2N4O2）has been synthesized and characterized by elemental analysis, IR, 1H NMR, MS and single-crystal X-ray diffraction. The crystal belongs to the triclinic system, space group Pi with a = 6.891（2）, b = 9.074（2）, c = 18.258（4）AA, α = 99.292（4）, β = 95.105（4）, γ = 108.068（3）°, C21H22Cl2N4O2, Mr = 433.33, V= 1059.3（4） Aa, Z = 2, Dc = 1.359 g/cm3, F（000） = 452,μ = 0.331 mm-1, the final R = 0.0448 and wR = 0.0994 for 3727 unique reflections. The dihedral angle between the oxazolidine ring taking an envelope conformation with a local pseudo-mirror and the triazole ring is 27.7（9）°. Weak intermolecular C-H...N hydrogen bonds and π-π interactions exist between the triazole rings of neighboring molecules, forming a three-dimensional network, which stabilizes the crystal structure. The primary biological test shows the target compound has certain fungicidal activity.     

Crystal and molecular structure of 1-ferrocenyl-r-3,t-5-diphenyl-c-4-(1-ferrocenylvinyl)-1-cyclohexene

X-ray diffraction has been used to establish the crystal and molecular structure of one diastereoisomer of 1-ferrocenyl-3,5-diphenyl-4-(1-ferrocenylvinyl)-1-cyclohexene. Synthesis, IR spectra, crystal structure and DFT studies on 1-acetyl-3-(4-chlorophenyl)-5-(4-methylphenyl)-2-pyrazoline

1-Acetyl-3-(4-chlorophenyl)-5-(4-methylphenyl)-2-pyrazoline has been synthesized and characterized by elemental analysis, IR and X-ray single crystal diffraction. Density functional (DFT) calculations have been carried out for the title compound by using the B3LYP method at the 6-311G** basis set level. The calculated results show that the predicted geometry can reproduce well the structural parameters. Predicted vibrational frequencies have been assigned and compared with experimental IR spectra and they are supported each other. On the basis of vibrational analyses, the thermodynamic properties of the title compound at different temperatures have been calculated, revealing the correlations between C(0)(p, m), S(0)(m), H(0)(m) and temperatures.

The title compound ethyl 1-(2-bromoethyl)-3-(4-methoxyphenyl)-1H-pyrazole-5-carboxylate 1 has been synthesized and structurally characterized by single-crystal X-ray diffraction.The crystal is of monoclinic(C15H17BrN2O3,Mr = 353.22),space group C21 with a = 24.691(7),b = 6.7678(17),c = 17.884(5) ,β = 97.184(5)o,V = 2965.1(13) 3,Z = 8,Dc = 1.583 g.cm-3,F(000) = 1440,μ = 2.784 mm-1,the final R = 0.0260 and wR = 0.0596 for 2684 observed reflections with I > 2σ(I).All the carbon atoms in the molecule are nearly coplanar except C(15),with a large conjugated system among the carbonyl group,pyrazole ring and the benzene ring.Three non-classical intermolecular hydrogen bonds help to stabilize the crystal lattice.The regioselectivity was rationalized based on the coordination of potassium ion with the N-anion and the carbonyl oxygen atom.     

1,3-Dipolar cycloaddition of diarylnitrones to 1-(4-nitrophenyl)-5H-pyrrolin-2-one

The reaction of diarylnitrones with 1-(4-nitrophenyl)-5H-pyrrolin-2-one to give 2-oxa-6-oxo-3-phenyl-4-(R-phenyl)-7-(4-nitrophenyl)-3,7-diazabicyclo(3,3,0)octanes has been examined. The structures of the products were established by NMR and IR spectroscopy, and x-ray diffraction studies. The stereoselectivity of the reaction is due to exo-approach of the reactants with cis-stereospecific addition of the trans-forms of the C,N-diarylnitrones to the pyrrolinone double bond.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 12, pp. 1675–1678, December, 1990.

Stereospecificity of the reaction of (+)-(4S, 5S)-4-(4-nitrophenyl)-1-aza-3,7-dioxabicyclo[3.3.0]octane with methyl iodide

The MINDO3, MNDO, AM1, and PM3 methods have been used to optimize the geometries of (+)-(4S,5S)-4-(4-nitrophenyl)-1-aza-3,7-dioxabicyclo[3.3.0]octane and (+)-(4S,5S)-4-phenyl-1-aza-3,7-dioxabicyclo-[3.3.0]octane. It has been shown that the stereospecificity, of the reaction of the former with methyl iodide is due to the high stability of one of the possible conformers.
