The gas-phase H/D exchange reactions of arginine (R) and arginine-containing di- and tri-peptide (gly-arg (GR), arg-gly (RG), gly-gly-arg (GGR), gly-arg-gly (GRG) and arg-gly-gly (RGG)) [M+H]+ ions with deuterated ammonia (ND3) were investigated by using Fourier-transform ion cyclotron resonance mass spectrometry (FT-ICR), ion mobility-mass spectrometry (IM-MS), ab initio and density functional theory-based molecular orbital calculations and molecular modeling. Three exchanges are observed for arginine and arginine-containing tri-peptide [M+H]+ ions, whereas the di-peptide [M+H]+ ions undergo a single H/D exchange. In addition, C-terminal methylation blocks H/D exchange of arginine and the arginine-containing peptide [M+H]+ ions, and a single H/D exchange is observed for N-terminal acetylated arginine [M+H]+ ions. A general mechanism for H/D exchange involving a collision complex that is best described as a “solvated salt-bridge” structure is proposed.  相似文献

Analyses of a series of nitroaromatic compounds using fast atom bombardment (FAB) mass spectrometry are discussed. An interesting ion-molecule reaction leading to [M + O ? H]? ions is observed in the negative ion FAB spectra. Evidence from linked-scan and collision-induced dissociation spectra proved that [M + O ? H]? ions are produced by the following reaction: M + NO2? → [M + NO2]? → [M + O ? H]?. These experiments also showed that M ions are produced in part by the exchange of an electron between M and NO2? species. All samples showed M, [M ? H]? or both ions in their negative ion FAB spectra. Not all analytes studied showed either [M + H]+ and/or M+˙ in the positive ion FAB spectra. No M+˙ ions were observed for ions having ionization energies above ~9 eV.  相似文献

The observation that protonated molecules are present in solvents utilized for fast atom bombardment (FAB) mass spectrometric studies has been demonstrated using visible absorption spectrometry. Addition of porphyrins to thioglycerol, a solvent used for FAB analyses, results in partial protonation of the molecule. This reaction can be monitored by observing the shift in visible absorption maxima associated with the molecular transition from free base to protonated structure. A good correlation is observed between the degree of protonation indicated by the appropriate absorption bands and the abundance of the [M + H]+ ion in the FAB spectrum of the corresponding solution. Addition of certain non-polar porphyrin molecules to thioglycerol does not result in the protonation of the molecule in solution; in these cases, analyses of the corresponding solutions by FAB do not yield [M + H]+ ions. Subsequent addition of trifluoroacetic acid to the solvent has proved sufficient to protonate the analyte molecule, as indicated by the visible absorption spectrum; FAB analyses of these non-polar porphyins in acidified solvent result in the observation of [M + H]+ ions. These experiments demonstrate that analyses of these analyte molecules requires that they be present as ions in solution prior to analysis by FAB. This study provides experimental evidence for the presence of ions in solutions employed for FAB analysis, suggesting that these ions are essential for the generation of the protonated molecules observed during FAB mass spectrometric analyses.  相似文献

Positive and negative ion fast atom bombardment (FAB) mass spectra of some monosubstituted nitroaromatic isomers are reported. Generally ions carresponding to [M + H]+ and M+ are observed in the positive ion FAB spectra; ions such as [M ? H] ? and M?˙ are observed in the negative ion FAB spectra. The use of FAB mass spectra to distinguish the isomers is discussed. Comparisons of FAB, chemical ionization and electron impact mass spectra of the same isomers (wherever possible) are reported. The structural information obtained in the negative ion FAB spectra complement those obtained in the positive ion FAB spectra.  相似文献

Mass Spectra of unsubstituted, 2-methyl-, 3-methyl and 2,3-dimethylchromones were examined. These compounds showed [RDA]+˙ and [RDA + H]+ ions as characteristc ions, together with [M? H]+,[M? CO]+˙,[M? CHO]+ and [RDA? CO]+˙ ions. Based on deuterium labelling experiments and measurement of metastable peaks by the ion kinetic energy defocusing technique, the origin of transferred hydrogen in the [RDA + H]+ ion was clarified. The mechanism of the [RDA + H]+ ion formation is discussed.  相似文献

Mechanisms are proposed for the formation of M+, [M + 2H]+ and [M + 3H]+ ions in the fast atom bombardment (FAB) mass spectra of 4-(2,2,6,6-tetramethyl-1-oxyl)-piperidol and its carboxylates. Free radical quenching induced by the fast atom beam has been observed. The effects of temperature on the radical quenching and of acid on the FAB mass spectra are discussed. The experiment showed that the volatile liquid samples with vapour pressures higher than that for glycerol produced M+ even-electron molecular ions, and the FAB mass spectra were similar to the corresponding electron ionization mass spectra. For the solid samples, it was found that the free radicals were quenched during the FAB process so that the mononitroxide and dinitroxide compounds produced [M + 2H]+ and [M + 3H]+ ions, respectively. Further experiments showed that the intensities and stabilities of [M + 2H]+ and [M + 3H]+ ions could be improved by addition of acids.  相似文献

The structures of the major adduct ions formed in ammonia chemical ionization of thirteen aliphatic and aromatic ketones have been studied by mass analysed ion kinetic energy spectrometry. The [M+NH3+H]+ ion is shown to have a protonated carbinolamine structure, [M+2NH3+H]+ to be a protonated carbinolamine with hydrogen-bonded ammonia and [2M+NH3+H]+ to be, at least in part, a protonated carbinolamine with hydrogen-bonded ketone. These structures may imply a nucleophilic addition of ammonia at the carbonyl of the ketone-ammonium ion complex. An unusual hydroxy migration is seen in the internal rearrangement of the [2M+NH3+H]+ ion leading to the formation of a protonated imine.  相似文献

Some ion-formation processes during fast atom bombardment (FAB) are discussed, especially the possibility of reactions in the gas phase. Divided (two halves) FAB probe tips were used for introducing two different samples into the source at the same time. Our results showed [M + A]+ ions (where M = crown ethers and A = alkali metal ions), can be produced, at least in part, in the gas phase when crown ethers and sources of alkali metal ion are placed on two halves of the FAB probe tip. The extent of this ion formation depends on the volatility of the crown ether and on steric factors. Cluster ions such as (M + LiCl)Li+, (2M + LiCl)Li+, [2M + K]+ and [2M + Na]+ are also observed to form in the gas phase. Unimolecular decompositions contribute to some ions detected in FAB. When the alkali ion salt and the crown ether are mixed together the probability of [M + A]+ ion formation increases significantly, regardless of the volatility of the crown ether.  相似文献

A series of new synthetic tetrabenzyl N-glucosidic, N-mannosidic and N-galactosidic isomers were investigated by fast atom bombardment (FAB)/mass-analysed ion kinetic energy (MIKE) spectrometry. The [M + H]+ ions were obtained with high abundance in the FAB spectra when using 3-nitrobenzyl alcohol as the matrix. The FAB/MIKE spectra provide characteristic daughter ions fragmented from selected molecular parent ions, allowing these isomers to be differentiated. In addition, an interesting rearrangement was found from the MIKE spectra, indicating that the benzyl (Bzl) group on the sugar ring is rearranged on to the N atom of the base (R) group to form [R + Bzl + H]+ and [R+ 2Bzl]+ ions.  相似文献

The gas-phase H/D exchange of bradykinin [M + H]+, [M + Na]+, [M + 2H]2+, and [M + H + Na]2+ ions; des-Arg1-bradykinin, des-Arg9-bradykinin, and bradykinin fragment 2-7 [M + H]+ ions; and O-methylbradykinin [M + H]+ and [M + 2H]2+ ions with D2O have been examined by electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry at 9.4 T. The different peptides vary widely in exchange rate and extent of deuterium incorporation. H/D exchange was slowest and deuterium incorporation was least for bradykinin [M + H]+, [M + H + Na]2+ and bradykinin methyl ester [M + 2H]2+ ions. In contrast, H/D exchange and extent of deuteration are higher for des-Arg1-bradykinin, des-Arg9-bradykinin, and bradykinin fragment 2-7 [M + H]+ ions; and highest for bradykinin [M + Na]+ and [M + 2H]2+, and O-methylbradykinin [M + H]+. Because the most likely site of protonation is the guanidino group of arginine, the above reactivity pattern strongly supports a zwitterion form for protonated gas-phase bradykinin.  相似文献

The fast atom bombardment (FAB) mass spectra of telluronium salts were studied. The spectra exhibit the intact cation (C+) and cluster ions ([M + C]+). The principal fragment ions in the FAB mass spectra of telluronium salts are [RTe]+, [R2Te]+˙, [R2Te − H]+, [RTeR′]+˙, and [RTeR′ + H]+. When the anion was [BPh4], interesting cluster ions such as [M + C − BPh3]+ appeared.  相似文献

A novel fragmentation of metastable peptide [M + H]+ ions is described. Loss of the C-terminal amino acid residue is accomqanied by retention of one of the carboxyl oxygens, as judged by 18O-labeling. The retained 8O label is located at the new C-terminus. Sequential mass spectrometric analyses indicate that the structure of the first-generation product ion is indistinguishable from that of the [M + H]+ ion of the peptide with one fewer amino acid residues. Thus, for example, the metastable decompositions of ions of m/z 904 are similar whether they correspond to des-Arg9-bradykinin [M + H]+ ions or to fragments derived from bradykinin [M + H]+ ions. No corresponding rearrangements have been observed for peptides with C-terminal amide or ester functions. The mechanism of this fragmentation may be considered to be analogous to that previously suggested for fragmentations of [M + alkali metal cation]+ ions. For the examples of bradykinin and related peptides, the rearrangement is strongly promoted when arginine is the amino acid residue lost. The same fragmentation is also favored by the presence of an arginine residue at or near the N-terminus. The strong influence of peptide amino acid composition, including residues remote from the C-terminus, on the prevalence of this fragmentation suggests mechanistic complexities that require further elucidation.

The competitive formation of molecular ions M and protonated molecules [M + H]+ under fast atom bombardment (FAB) conditions was examined using various kinds of organic compounds. The use of protic/hydrophilic matrices such as thioglycerol and glycerol resulted in relatively large values of the peak intensity ratio I([M + H]+)/I(M) compared with the use of relatively aprotic/hydrophobic matrices such as m-nitrobenzyl alcohol and o-nitrophenyl octyl ether. The change of matrix from thiol-containing such as thioglycerol and dithiothreitol to alcoholic such as glycerol and pentamethylene glycol increased the I([M + H]+)/I(M) ratio. Furthermore, the change of matrix increased the peak intensity ratio of the doubly charged ion [M + 2H]2+ to [M + H]+ in the FAB mass spectra of angiotensin I and gramicidin S. The addition of acids to the matrix solution increased the I([M + H]+)/I(M) ratio, although such an effect did not always occur. The acetylation of simple aniline compounds markedly increased the I([M + H]+)/I(M) ratio. It was concluded from these results that the hydrogen bonding interaction between hydroxyl groups(s) of the matrix and basic site(s) of analyte molecules in solution acts advantageously as a quasi-preformed state for [M + H]+ formation, and that the presence of significant proton acceptor(s) such as carbonyl group in analytes hinder the M formation which may generally occur under FAB conditions. The formation of M and [M + H]+ ions seemed to occur competitively, reflecting or according to the interaction or solvation states between the analyte and matrix molecules in solution and the structural characteristics of the analytes.  相似文献

The unimolecular fragmentations of [M + H]+ and [M – H]? ions from four 2-aryl-2-methyl-1,3-dithianes are described and clarified with the aid of deuterated derivatives. Comparison of the MIKE spectra of [M + H]+ species obtained under chemical ionization and fast atom bombardment (FAB) conditions reveals differences which are attributed to the different energetics involved in the two ionization processes. It is suggested that FAB is a ‘softer’ ionization technique but, at the same time, it provides, for the possibility of solvation, reaction sites not available in gas-phase protonation. [M – H]? species and anionic fragments thereof were generally not obtained under FAB(?) conditions. [M – H]? ions are readily produced in gas-phase reactions with OH? via proton abstraction from C(4) or C(5), and from the 2-methyl substituent; and they fragment according to several reaction pathways.  相似文献

When mass-analysed ion kinetic energy (MIKE) spectra are required to discriminate between isomeric ions formed under conditons of fast atom bombardment (FAB) in the ion source, severe interference may be observed. The interfering peaks in MIKE spectra obtained with a reversed-geometry instrument can arise from different sources. Moreover, the intensity distribution of the true ions from the selected precursor ion may depend strongly on the instrument being used. This means that the FAB–MIKE or collisionally induced dissociation (CID) spectrum is not an absolute characteristic of a particular ion. The [M + H ? HOAc]+ ion in the spectrum of peracetylated ribopyranose is used as an example to illustrate this and to trace and discuss the origin of the phenomena observed.  相似文献

The ion-molecule reactions of dimethyl ether ions CH3OCH3 + and (CH3OCH3)H+, and four- to seven-membered ring lactams with methyl substituents in various positions were characterized by using a quadrupole ion trap mass spectrometer and a triple-quadrupole mass spectrometer. In both instruments, the lactams were protonated by dimethyl ether ions and formed various combinations of [M + 13] +, [M + 15] +, and [M + 45] + adduct ions, as well as unusual [M + 3] + and [M + 16] + adduct ions. An additional [M + 47] + adduct ion was formed in the conventional chemical ionization source of the triple-quadrupole mass spectrometer. The product ions were isolated and collisionally activated in the quadrupole ion trap to understand formation pathways, structures, and characteristic dissociation pathways. Sequential activation experiments were performed to elucidate fragment ion structures and stepwise dissociation sequences. Protonated lactams dissociate by loss of water, ammonia, or methylamine; ammonia and carbon monoxide; and water and ammonia or methylamine. The [M + 16] + products, which are identified as protonated lactone structures, are only formed by those lactams that do not have an N-methyl substituent. The ion-molecule reactions of dimethyl ether ions with lactams were compared with those of analogous amides and lactones.  相似文献

We have developed a combined EI/FI source for gas chromatography/orthogonal acceleration time‐of‐flight mass spectrometry (GC/oaTOFMS). In general, EI (electron ionization) and FI (field ionization) mass spectra are complementary: the EI mass spectrum contains information about fragment ions, while the FI mass spectrum contains information about molecular ions. Thus, the comparative study of EI and FI mass spectra is useful for GC/MS analyses. Unlike the conventional ion sources for FI and EI measurements, the newly developed source can be used for both measurements without breaking the ion source vacuum or changing the ion source. Therefore, the combined EI/FI source is more preferable than the conventional EI or FI ion source from the viewpoint of the reliability of measurements and facility of operation. Using the combined EI/FI source, the complementarity between EI and FI mass spectra is demonstrated experimentally with n‐hexadecane (100 pg): characteristic fragment ions for the n‐alkane such as m/z 43, 57, 71, and 85 are obtained in the EI mass spectrum, while only the parent peak of m/z 226 (M+) without any fragment ions is observed in the FI mass spectrum. Moreover, the field desorption (FD) measurement is also demonstrated with poly(ethylene glycol)s M600 (10 ng) and M1000 (15 ng). The loss of 60 u from protonated peptide ions containing an arginine residue at the C-terminus has been investigated by means of low energy tandem mass spectrometry. The lowest energy conformation of singly charged bradykinin is thought to involve a salt-bridge structure, which may lead to the formation of two isomeric forms. It is thought that one isomer retains the ionizing proton at the C-terminal end of the peptide, leading to the formation of the [b n?1 + H + OH]+ fragment ion, and the other isomer retains the charge at the N-terminus, leading to the formation of the [M + H ? 60]+ fragment ion. It was found that the formation of the [M + H ? 60]+ ion occurs only from singly charged precursor ions. In addition, the loss of 60 u occurs from peptides in which the charge is localized at the N-terminus. These results indicate that the mechanism of formation of the [M + H ? 60]+ ion may be driven by a charge-remote process.  相似文献

The behaviour towards low collision energy processes (eV range) of [M  H] prepared under negative ion chemical ionization (NICI) ammonia conditions from 17β-estradiol-17-fatty acid esters has been investigated. From such bifunctional compounds containing two acidic sites (i.e. phenol and ester groups), two isomeric forms (i.e. phenoxide and enolate forms) characterize the [M  H] ion structures, whose distribution depends on the ion preparation mode. Here NICI (ammonia) provides both phenoxide and enolate forms as the [M  H] species. This behaviour contrasts with the regioselectivity observed for proton abstraction from phenol under NICI (N2O) and fast atom bombardment conditions. Production of both phenoxide and enolate forms in NICI (ammonia) is demonstrated under NICI (ND3) conditions in which DO-labelled [Md  H] enolate ions are produced in a similar yield to unlabelled [Md  D] phenoxide ions. Collisionally activated dissociation (CAD) spectra of both isomeric deprotonated molecules differ strongly by the presence of two different pairs of complementary daughter ions, suggesting that these ionic species are unconvertible. This is due to a steric hindrance effect on the long-distance proton transfer. A mechanistic investigation on the formation of fragment ion pairs produced under CAD was performed with various deuterium-labelled molecules. From these experiments, evidence is provided for molecular isomerizations into ion-dipole complexes (prior to dissociation) which are structurally dependent on the initial charge location. Direct dissociation of these intermediates competes with the occurrence of exothermic proton transfer(s) yielding the formation of other isomeric intermediate forms. The orientation of these proton transfers is dictated by the relative acidities of both moieties of the complex.  相似文献
