beta-Crystallins (beta 1-, beta 2- and beta 3-crystallin) comprise nearly half the protein of the human lens. The effect of near-UV radiation, which is one of the possible risk factors in cataract formation, on the beta-crystallins is investigated in this study. Protein intersubunit crosslinking, change in charge of the protein subunits to more acidic species and changes in protein tertiary structure (conformation) by 300 nm irradiation are reported. The fluorescence yield of protein tryptophan residues decreases by 300 nm irradiation. There is an increase in nontryptophan fluorescence (lambda cx 340 nm, lambda cm 400-600 nm), and in protein absorption at 340 nm, due to the formation of tryptophan photooxidation products. Both tryptophan and its oxidation products can be photoexcited by 300 nm irradiation and the latter are known to be good photosensitizers. The results provide evidence for the generation of H2O2 in the irradiated human beta-crystallin solutions by the Type I photosensitizing action of the chromophores absorbing at 300 nm. The H2O2 is generated via the intermediate production of O2 anion; the latter spontaneously dismutates to H2O2, presumably via O2- protein interactions. The amount of H2O2 generated per absorbed photon is compared for various solutions of beta 1-, beta 2- and beta 3-crystallins from human lenses of different age.  相似文献

Abstract— The subcellular localization of protoporphyrin (PP) has been studied by microspectrofluo-rometric techniques in NCTC 2544 keratinocytes incubated with 5-aminolevulinic acid (ALA) for times up to 42 h. Whereas the plasma membrane shows strong staining, fluorescent spots are observed within the cytoplasm especially in the perinuclear region. Although the topographic pattern of the PP distribution does not change much with the incubation time with ALA, the fluorescence spectra suggest that the PP microenvironments are quite different at short and long incubation times. Addition of 18 uJW desferoxamine almost doubles the ALA-induced PP concentration. Colocalization experiments with rhodamine 123, a mitochondrial probe, and lucifer yellow (LY) or neutral red (NR), two lysosome probes, demonstrate that at least some of these spots are of lysosomal origin. Study of the time evolution of the NR fluorescence under irradiation with visible light in the presence and absence of ALA demonstrates that lysosomes are damaged in cells that have synthesized PP. No PP fluorescence can be detected in mitochondria after incubation with ALA. However, photosensitization of mitochondria occurs under irradiation with visible light. Very little formation of lipofuscins by photosensitization with exogenous PP or ALA-induced PP is observed with the NCTC 2544 keratinocytes, as compared to normal human fibroblasts.  相似文献

We have studied the hydrogenase-catalyzed production of H, when either its natural electron mediator cytochrome c3 (c3) or the artificial mediator methyl viologen (MV) was reduced by illumination of proflavin (PF) in the presence of ethylenediaminetetraacetate (EDTA). The reduction rates of MV and c3 were comparable at equivalent concentrations of PF, taking into account the four redox sites of c3. However, when the concentration of c3 exceeded that of PF, the reduction rate decreased. We explain this by light quenching. In the H2-producing system, MV was more efficient than c3 as electron mediator when the initial reaction rates were compared. However, under certain conditions with MV, the rate of H2 production decreased rapidly with time of illumination, whereas with c3 it consistently remained stable. Possible reasons for this difference are discussed.

Abstract— During the reaction HO2+ HO2 (or O2-) = H2O2+ O2 in aqueous solution, no luminescence in the region 620–720 nm, expected if the product O2 were formed in a singlet state, could be detected. If any singlet O2 is formed, its yield must be less than 10%. Faint luminescence, sometimes found at shorter wavelengths, was shown to arise from reaction of HO2 with impurities in the reagents present.  相似文献

Abstract— The rate constant for quenching of 1O2 by azide ion in water was determined to be (5.0 ± 0.4) × 108 M −1 s−1 using a variety of sensitizers (including humic acids) and 1O2 acceptors. The apparent second-order rate constant decreases with pH below pH 5.5 in accordance with the protonation of azide ion to form hydrazoic acid (p K a= 4.6). Quenching by hydrazoic acid is at least 2 orders of magnitude slower than by azide ion. Greater than 99% of all interactions between 1O2 and azide ion involve physical quenching rather than chemical reaction. Humic acid triplets are not significantly quenched by azide ion at concentrations less than 2 m M , allowing azide ion quenching to be used as a diagnostic test for the intermediacy of 1O2 in photosensitized oxidations in natural surface waters.  相似文献

Abstract— Photolysis of naphthalene on the surface of SiO2 under an atmosphere of air produces phthalic acid as the only major photoproduct, accounting for 49%o of the consumed naphthalene. Photolysis on Al2O3 also produces phthalic acid, in 31% yield. Photolysis of 1 -methylnaphthalene on SiO2 proceeds under similar conditions to produce 2-acetylbenzoic acid (35%) as the major photoproduct with the production of a small amount of I-naphthaldchyde (6%). 1-Cyanonaphthalene does not photooxidize under similar conditions. The presence of oxygen is necessary for the photodecomposition of naphthalene and 1-methylnaphthalene to proceed. Superoxide formed from the photolysis of naphthalene at the SiO2/air interface is readily observed by electron paramagnetic resonance spectroscopy. In the absence of naphthalene no superoxide is observed. A mechanism involving electron transfer from the S1 state of the naphthalene to O2 is proposed on the basis of these observations and related literature precedent.  相似文献

Ultraviolet-A radiation has weak effects on the release of inflammatory mediators by skin cells due to the poor overlap between UVA wavelengths and the absorption spectra of the relevant chromophores of key biomole-cules. However, this situation could be very different in the presence of a photosensitizing drug. To investigate this issue, we have irradiated human skin cells (keratinocytes and fibroblasts) in the presence of fenofibric acid (the active phototoxic metabolite of fenofibrate). The results of this research show a dual effect on the production/release of inflammatory mediators: the synthesis of the proinflammatory cytokine interleukin-6 becomes strongly inhibited at photosensitizer concentrations that clearly stimulate the production of prostaglandins (PGE2) by skin cells. We have found evidences showing that the de novo synthesis of cytokines is inhibited in photosensitized cells due to the fact that cellular mRNA is degraded. Interestingly, when the medium taken from irradiated cultures is added to nonexposed cells, a significant stimulation of cytokine synthesis is observed that can be inhibited by anti-PGE2 antibodies. These observations may be relevant in vivo, where prostaglandins released by photosensitized skin cells could stimulate cytokine synthesis by underlying, nonirradiated cells.  相似文献

Abstract— Chlorophyll a and I2 form a 1:1 addition compound, which exhibits a strong absorption maximum at about 360 nm. When dissolved in anaerobic methanol, this compound can be reversibly bleached by illumination with red light. Prolonged illumination at low temperatures (˜– 75°C) or intense illumination at room temperature reduces the height of the red absorp tion maximum to about half of its normal value. Since the thermal back reaction is negligibly slow at – 75°C, presumably the chlorophyll-I2 complex is converted completely to its bleached form, when illuminated under those conditions. This assumption leads to a simple mechanism which is consistent with the experimental data.  相似文献

Abstract— The bioluminescent oxidation of reduced flavin mononucleotide by bacterial luciferase involves a long-lived flavoenzyme intermediate whose chromophore has been postulated to be the 4a-sub-stituted peroxy anion of reduced flavin. Reaction of long chain aldehyde with this intermediate results in light emission and formation of the corresponding acid. These experiments show that the typical aldehyde-dependent, luciferase-catalyzed bioluminescence can also be obtained starting with FMN and H2O2 instead of FMNH2 and O2. We postulate that the 4a-peroxy anion intermediate is formed directly by attack of H2O2 on FMN. The latter may be bound to luciferase. An enzyme bound intermediate is formed which by kinetic analysis, flavin specificity for luminescence, aldehyde dependence, and bioluminescent emission spectrum appears to be identical with the species generated by reaction of FMNH, and O2 with luciferase. The quantum yield of the H2O2-- and FMN-initiated biolumlnescence is low but can be enhanced by certain metal ions, which also stimulate a chemiluminescent reaction of oxidized flavin with H2O2. The peak of this chemiluminescence. however, appears to be at a shorter wavelength than that (490 nm) of the bioluminescence.  相似文献

Neutrophils stimulated with formyl-methionyl-leucyl-phenylalanine (fMLP) or leukotriene B4 (LTB4) generated kinetically distinctive luminol augmented chemiluminescence (LCL). Inhibitors of .O2- [superoxide-dismutase (SOD) or tiron], H2O2 (catalase), myeloperoxidase, MPO, (NaN3), HOCl (taurine) and .OH (mannitol) hampered LCL dose-dependently with similar characteristics for both stimuli. In cell free systems it was found that .O2- (generated in the xanthine/xanthine-oxidase reaction) or H2O2 produced LCL. Superoxide dismutase inhibited .O2- -induced LCL dose dependently. The MPO + H2O2 system, which generated more pronounced LCL than either component alone, was inhibited by catalase and taurine but not by SOD. When neutrophils, treated with luminol, but where extracellular luminol had been removed, were stimulated with fMLP or LTB4, they produced less than 2% of the LCL where luminol was present in the medium. When neutrophil LCL and superoxide formation by the cytochrome C method were assessed in parallel experiments, in all instances the peak LCL response coincided with the linear phase in that response. Thus, LCL, induced by LTB4 and the corresponding fMLP peak, are extracellular events with similar chemical backgrounds, closely related to generation of reactive oxygen species. Consequently, the kinetical differences in LCL between fMLP and LTB4 suggest that LTB4, by yet unknown mechanisms, activates the NADPH oxidase more rapidly than fMLP.  相似文献

Abstract— A mutant of Chlamydomonas reinhardtii (NL–11) isolated from a wild type (137c+) was inactivated in the light in the presence of methionine at concentrations where the wild type was not inactivated. The inactivation was suppressed by either catalase or superoxide dismutase (SOD). Light-induced H2O2 formation and nitroblue tetrazolium (NBT) reduction inNL–11 were greater than those in the wild type. Methionine stimulated both the H2O2 formation and the NBT reduction inNL–11 as well as the wild type. The light-induced NBT reduction inNL–11 in the presence of methionine was partially suppressed by externally added SOD suggesting the participation of O-2. These results suggest that the hypersensitivity ofNL–11 to methionine in the light is due to stimulated formation of H2O2 and O-2.  相似文献

Abstract— The role of chloride in photosynthetic oxygen evolution was investigated by means of thermoluminescence measurements. It was found that chloride depletion in isolated chloroplasts almost completely abolished the B1 thermoluminescence band (S3QB recombination) but diminished only slightly the amplitude of the B2 band (S2QB recombination). The B2 band could be excited to full intensity by the first flash of a flash series and subsequent flashes caused no further change in the amplitude of the band. These observations suggest a block in the S2→S3 transition of the water-splitting system in chloride-depleted chloroplasts. Readdition of chloride provided evidence that the inhibitory effect of chloride removal is reversible.  相似文献

Abstract— Although the mechanism of bioluminescent reactions in various species, such as fireflies, ostracod crustaceans ( Cypridina ), sea pansies ( Renilla ), and the deep-sea shrimp Oplophorus , are thought to involve dioxetanone intermediates, studies reported in the past from different laboratories have included widely different experimental results, most likely due to various factors including the effects of contaminating CO2. With the improved technique employed in the present study, bioluminescent reactions of the firefly and Cypridina in 18O2 gas resulted in an incorporation of over 75% of 18O into one oxygen of the product CO2. with a reproducibility within a few per cent. When 13CO2. instead of the product CO2 of the bioluminescent reaction, was studied in an H218O medium, the exchange of one oxygen of 13CO2 with H2O was 64%. and the effect of contaminant CO2 amounted to 1418% of the total CO2. These results suggest that every molecule of CO2 formed in the bioluminescent reactions of the firefly and Cypridina had intially contained 1 oxygen atom derived from O2.  相似文献

Abstract— The quantum yield and the kinetics of O -induced luminol chemiluminescence was investigated in a broad pH interval at varying luminol and concentrations. It is suggested that the weak chemiluminescence observed is mediated via a luminol-superoxide-adduct proposed to be an a-hydroxyperoxyl radical. At pH 7 the maximum quantum yield of chemiluminescence per initial percent was determined to be 4 times 10-8. The degree of involvement in phagocytosis and related processes should be viewed against this maximum limit.  相似文献

Abstract— We have described the covalent binding of 124-kDa oat phytochrome to large unilamellar liposomes composed of either dioleoyl phosphatidylcholine or dipalmkoyl phosphatidylcholine or soybean lecithin, without affecting the photochromic properties of the protein. These phytochrome-liposome systems have now been studied by laser flash photolysis. The liposomes, independent of their membrane rigidity (liquid-crystal vs gel-like phase), do not influence the ratio and reactivity of the two primary photoproducts, Ii700- of the red absorbing form of phytochrome, Pl Thus, the lifetimes of the Ii700 intermediates and the activation parameters associated with Ii700Iibl are the same as those measured for nonbound phytochrome in buffer solution. The temperature increase from about 273 K. to 297 K lowers the population of the shorter-lived Ii700 intermediate to the same extent both in the liposome-Pl and in nonbound Pl, whereas it does not affect the relative population of the Ii700 intermediates from non-bound Pl in the presence of 25% ethylene glycol added to the buffer solution (ionic strength 0.17).  相似文献
