Cereal preparation can be an excellent source of substances with proven health-promoting properties. Unfortunately, some types of bread, such as white flour bread, are devoid of many valuable nutrients. Therefore, it is necessary to look for ways to increase its density and nutritional value. The aim of the study was to investigate the effect of stabilized plant extracts on the quality of bread, its antioxidant activity and polyphenol content, and to evaluate the stability of bioactive compounds and antioxidant activity during in vitro digestion. The research material was the wheat bread baked with spray dried microcapsules of hawthorn bark, soybeans and onion husks in maltodextrin or inulin carriers. The addition of plant extracts resulted in the presence of phenolic compounds in the wheat bread, and its antioxidant activity significantly increased. There was no significant difference in antioxidant activity between breads containing microcapsules with different carriers. During in vitro digestion, procyanidins and isoflavones in bread were more resistant to the digestive processes than other compounds. The antioxidant activity during simulated digestion was the highest at the stage of gastric digestion, and its value depended on the extract used and the analytical method applied.  相似文献

This is the first study to examine the effects of in vitro digestion on biological activities of Sargassum spp., a broadly known brown seaweed for therapeutic potential. Three fractions (F1–F3) were obtained from hexane extract by column chromatography. Under in vitro simulated digestion, the anti-α-amylase capacity of F1 in oral and intestinal phases increases, while it significantly decreases in the gastric phase. The α-amylase inhibition of F2 promotes throughout all digestive stages while the activity of F3 significantly reduces. The cytotoxic activity of F1 against U266 cell-line accelerates over the oral, gastric, and intestinal stages. The fractions F2 and F3 exhibited the declined cytotoxic potentialities in oral and gastric phases, but they were strengthened under intestinal condition. Palmitic acid and fucosterol may play an active role in antidiabetic and cytotoxic activity against multiple myeloma U266 cell line of Sargassum spp. However, the involvement of other phytochemicals in the seaweed should be further investigated.  相似文献

A method was developed allowing the separation, detection and identification of Se species extracted from yeast supplements during simulated digestion processes. The in vitro gastric and intestinal digests were studied for their Se compounds by successive high-performance liquid chromatography–inductively coupled plasma mass spectrometry (HPLC-ICP-MS) and high-performance liquid chromatography–electrospray tandem mass spectrometry (HPLC-ES-MS-MS) analyses. The conditions for the separation were chosen as to be compatible with both ICP-MS and ES-MS-MS detection. HPLC-ICP-MS was used to screen the extracts for their Se content. By means of HPLC-ES-MS-MS, the compounds extracted were identified on-line according to their retention time, m/z of the molecular ion and the presence of typical product ions. From these results, it was clear that the main compound extracted by both gastric and intestinal fluid was Se-methionine, which was also the main Se compound extracted by proteolytic digestion from the yeast supplements. Two other minor compounds could be identified as Se-cystine and Se(O)-methionine, a degradation product of Se-methionine.  相似文献

Standardization of protein extracts for clinical purposes represents an important task in order to maintain adequate reactivity, presence of the relevant allergens, and safety among other factors. The main objective of this work was to explore the potential use of a chip‐based automated CE system commercially available to analyze several of the most common forms of allergenic extracts from olive pollen used in allergy clinics. These include experimental extracts prepared from olive pollens, in‐house reference extracts, extracts designed for skin prick test assays, and a panel of vaccine variants aimed to specific immunotherapy. As a major conclusion of the study, chip‐based CE allowed in all cases to determine accurate protein profiles with different degrees of sensitivity, where several allergens (particularly the major olive pollen allergen Ole e 1) were easily recognized. Moreover, several purified allergens were also analyzed by this method, and proposed as specific standards for different purposes. In the present condition, the method can only provide the protein profile of the extracts with respect to a preestablished standard extract, but not allergen identification. However, these and other future developments and applications are discussed.  相似文献

A dynamic continuous-flow dialysis (CFD) method with on-line electrothermal atomic absorption spectrometric (ETAAS) and pH measurements for study of simulated gastrointestinal digestion has been developed for prediction of iron bioavailability. The method used to estimate mineral bioavailability was based on gastric digestion in a batch system then dynamic continuous-flow intestinal digestion. The intestinal digestion was performed in a dialysis bag placed inside a chamber containing a flowing stream of dialyzing solution. Mineral concentration and dialysate pH were monitored by ETAAS and use of a pH meter, respectively. The amount of dialyzed minerals in the intestinal digestion stage was used to evaluate the dialyzability. The dialysis profile and pH change can be used to understand or examine differences between the dialyzability of different food samples. To test the proposed system it was used to estimate the iron dialyzability of different kinds of milk. Iron dialyzability for powdered cow milk, cereal milk, and two brands of soymilk was found to be 1.7, 20.4, 24.9, and 37.7%, respectively. The developed CFD–ETAAS–pH system is a simple, rapid, and inexpensive tool for bioavailability studies, especially for minerals at ultratrace levels.  相似文献

An in vitro gastrointestinal digestion approach in combination with species-specific isotope dilution analysis has been employed for the first time to study the transformation reactions as well as the solubilisation of butyltin species throughout a simulated human digestion. Different sample preparation procedures were assayed in order to avoid problems derived from lack of isotope equilibration between the endogenous and the isotopically-enriched added species. A triple spike approach, which can be used to calculate the corrected concentrations of mono-, di-, and tributyltin (MBT, DBT and TBT, respectively), as well as six interconversions, was employed throughout this work. In order to calculate and compare the species degradation factors, a triple spike solution containing each butyltin species enriched in a different isotope was added to the simulated gastric and intestinal fluids before the digestion procedures in the presence and in the absence of a solid biological matrix (commercial mussel tissue). Additionally, by analysing the soluble and insoluble fractions resulting from the simulated digestion of a commercial mussel tissue (gastric and gastric plus intestinal digestion), total mass balances for each butyltin compound could be derived. For this purpose, the isotopically-enriched species were added after the enzymatic digestions in order to avoid problems derived from lack of isotope equilibration. The mass balances provided information not only about the solubilisation but also about the degradation of the butyltin species during the digestion procedures. Good agreement between the degradation factors calculated under all experiments performed in this work and between those reported in previous works were obtained. The most serious degradation observed was that of DBT to produce MBT, whereas slight degradations of TBT and MBT were detected. Moreover, a worrying 61% of the original total butyltin content present in a commercial mussel tissue was found to be solubilised after complete simulated gastrointestinal digestion, with minimal degradation of TBT.  相似文献

Hydrocolloids in the form of polymeric ingredients as well as natural biopolymer assemblies provide much of the macroscopic structure of foods. The controlled disassembly of hydrocolloid-structured foods in the digestive tract determines numerous nutritional properties driven by the rates of passage, digestion, absorption, and fermentation. Despite convincing evidence for health benefits of hydrocolloids (particularly dietary fibre) from epidemiology, and numerous in vitro model system studies, the detailed underlying mechanisms operating in the digestive tract are currently understood to only a limited extent. Distinct hydrocolloid-based processes occur in each of the gastric, small intestinal and large intestinal environments, with significant biological cross-talk between the sites. Hydrocolloids offer a major opportunity to tailor nutritional value and provide potential health benefits through control of gastric emptying and ileal brake mechanisms (satiety and potentially obesity), glycemic response (diabetes), plasma cholesterol levels (cardiovascular disease), and carbohydrate fermentation throughout the large intestine (colon cancer). There is often a parallel between the functionality of the plant-based foods which the human digestive tract evolved to digest and the use of extracted hydrocolloids in modern food structuring technology.  相似文献

This study was carried out to evaluate the application of food irradiation technology as a method for reducing food allergy. Milk β-lactoglobulin, chicken egg albumin, and shrimp tropomyosin were used as model food allergens for experiments on allergenic and molecular properties by gamma irradiation. The amount of intact allergens in an irradiated solution was reduced by gamma irradiation depending upon the dose. These results showed that epitopes on the allergens were structurally altered by radiation treatment and that the irradiation technology can be applied to reduce allergenicity of allergic foods.  相似文献

Compound-specific carbon isotope ratio analysis is a promising tool to assess the origins and fates of organic contaminants in many fields. The present study aims to investigate the isotope effects of dichloro-diphenyl-trichloroethane (DDT) and its degradates in carrots during digestion processes simulating the human gastrointestinal tract. To accurately interpret isotopic data obtained for unreleased DDTs (p,p-DDT, p,p-DDD and p,p-DDE) from carrots during digestion, spiked carrots with known delta(13)C values of DDTs were incubated in simulated gastric and intestinal solutions using a static in vitro model of the human gut. The delta(13)C values and concentrations of DDTs remaining in the carrot and in the digestive solutions were measured. The difference in the delta(13)C values of the added DDTs and the matrix-bound DDTs after digestion was always <0.50 per thousand, which is within the technical specification of the analytical system, although the released DDTs increased or decreased under the digestion conditions. The study demonstrated that there were no significant carbon isotopic fractionations during digestion. To our knowledge, this is the first report demonstrating the isotope effect of organic compounds during stomach and small intestine digestion, and the first reported isotope analyses of DDTs. 运用体积排阻高效液相色谱-电感耦合等离子体质谱联用技术(SEC-HPLC-ICP-MS)分析了高镉积累扇贝和低镉积累菲律宾蛤仔中镉的存在形态,并结合体外全仿生消化技术,研究了在唾液、胃、肠无机物和有机物(含消化酶)作用下,扇贝和菲律宾蛤仔中镉的主要存在形态.结果发现:扇贝中Cd总量约为菲律宾蛤仔的10倍;在扇贝中检测到3种Cd形态:金属硫蛋白(MT)-Cd、谷胱甘肽(GSH) Cd和半胱氨酸(Cys)-Cd;在菲律宾蛤仔中检测到2种Cd形态:MT-Cd和GSH-Cd;以峰面积作参考进行比较,扇贝中MT-Cd和GSH-Cd含量分别约为菲律宾蛤仔的5 6和2.0倍.结合体外全仿生模型发现,在扇贝胃全仿生提取液中,检测到1种未知小分子有机镉形态(Cd-X),在扇贝肠全仿生提取液中检测到4种Cd形态,其中MT-Cd是主要形态;而在菲律宾蛤仔胃、肠全仿生提取液中均仅检测到1种未知小分子有机态镉(Cd-X).本实验证明贝类中的MT-Cd,GSH-Cd,Cys-Cd中络合的Cd在生物体胃肠消化液作用下会发生解离.  相似文献

The relationship between gastric acidity and the bioavailability of two kinds of sustained-release indomethacin (IM) formulations was investigated in gastric acidity-controlled beagle dogs, and compared with that of rapid-release IM formulations. All test dosage forms were more rapidly dissolved in simulated intestinal fluid than in simulated gastric fluid. Gastric acidity did not affect the bioavailability of IM from the rapid-release formulation. However, the bioavailability of IM from the two kinds of sustained-release formulations were markedly influenced by gastric acidity. The rates of IM bioavailability from both of the sustained-release formulations were faster under low acidity conditions than under high acidity conditions (p less than 0.01). In addition, Tmax and mean residence time (MRT) were approximately the same for the rapid-release and sustained-release formulations under low acidity conditions. These results suggest that the IM sustained-release formulations showed a rapid-release profile under low acidity conditions.  相似文献

A large number of food allergens able to induce allergic symptoms in predisposed individuals, including severe, even life-threatening reactions, have been identified and characterized. However, proteins able to cause such IgE-mediated reactions can be assigned to only a limited number of protein families. Detailed knowledge about the characteristics of food allergens, their 3D structures, biological activity and stability, will help to improve diagnosis of food allergy, avoid unnecessary exclusion diets and assess the risk of cross-reactive allergies to other food sources. This review is dedicated to summarizing current knowledge about the most important food allergen protein families and to presenting data from the EuroPrevall allergen library, a proof-of-concept collection of highly purified, characterized and authenticated food allergens from animal and plant food sources to facilitate improved diagnosis of food allergies. β-Cyclodextrin containing polymer brushes were grafted to magnetic nanoparticles following free radical copolymerization of N,N-dimethylacrylamide and allyl glycidyl ether on the surface. The products were characterized by Fourier transform infrared spectroscopy, thermogravimetric analysis, elemental analysis, and transmission electron microscopy and employed for solid phase extraction of naproxen in human urine. The profile of the naproxen adsorbed by the nanoparticles demonstrated high accessibility of the β-cyclodextrin. Scatchard analysis indicated that the capacity of the nanoparticles was 112 µmol g?1 at 25°C and a pH of 5. The adsorption data of naproxen were considered by Langmuir, Freundlich, Redlich–Peterson, and Temkin isotherms. Approximately 55% of naproxen was released in simulated gastric fluid in 30 min and 94% in simulated intestinal fluid in 30 h. These data have indicated the utility of the naproxen loaded- β-cyclodextrin containing polymer brushes grafted to magnetic nanoparticles for enteric drug delivery.  相似文献

The aim of this work was identifying and selecting hazelnut marker peptides and subsequently developing a complementary method of common immunoassay for the detection of hazelnut. For this purpose, at first, an in silico digestion of three major hazelnut allergens (Cor a 8, Cor a 9 and Cor a 11) was performed to get information about expected peptides. After extraction and trypsin digestion of hazelnut proteins, the samples were measured with tandem mass spectrometry (MS/MS) by direct infusion, which led to identification of 14 peptides. Eight of them with the highest MS signal were synthesized and used as standards for developing a liquid chromatography (LC)–MS/MS method in selected reaction monitoring (SRM) mode. Since almost all food allergens derived from nuts belong to the seed storage protein family and have homologue structure, a Basic Local Alignment Search Tool (BLAST) search was performed to identify the hazelnut specificity of the developed method. According to BLAST, only one peptide occurs in three other nuts, and the remaining seven selected peptides are hazelnut specific. Additionally to hazelnut, the eight other listed nuts in Directive 2003/89/EC as allergen were extracted, digested and measured with the developed method to prove the BLAST results. The analytical data confirmed that six peptides are hazelnut specific, on the contrary to anti-hazelnut antibodies, which showed cross-reactivities to all other nut extracts. Comparing these results, it could be shown that with this LC–MS/MS method in SRM mode, the specific detection of hazelnut is possible.  相似文献

Sequential extraction (water, Driselase, protease XIV) and extraction with simulated gastric and intestinal fluids were proposed to characterize the binding and the bioaccessibility of chromium in two commercial food supplements obtained by incorporation of this element into yeast. Chromium in Cr-enriched yeast was found to be hardly extractable with water, Driselase, or simulated gastric fluid (recoveries of approximately 10–20%), but proteolysis or gastrointestinal fluid digestion released more than half of the chromium present. Fractionation with size-exclusion chromatography with Cr-specific detection by inductively coupled plasma mass spectrometry (ICP MS) allowed the distinction of two fractions: one below approximately 1 kDa and one 1–5 kDa; they contained the entirety of the released Cr with proportions varying as a function of the extracting solution and the origin of sample. When collected and investigated by reversed-phase high-performance liquid chromatography–ICP MS, the low molecular mass fraction was found to release Cr(III), whereas the heavier one showed most of Cr bound in fairly stable hydrophobic complexes. However, an attempt of their identification by electrospray ionization MS/MS and matrix-assisted laser desorption ionization MS was not successful.   相似文献

In this study, dark chocolates (DCh) containing zinc lactate (ZnL) were enriched with extracts from elderberries (EFrE), elderflowers (EFlE), and chokeberries (ChFrE) to improve their functional properties. Both dried plant extracts and chocolates were analyzed for antioxidant capacity (AC) using four different analytical methods: 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), cupric ion-reducing antioxidant capacity (CUPRAC), and ferric-reducing antioxidant power (FRAP), while total phenolic content (TPC) was determined by Folin–Ciocalteu (F–C) assay. An increase in antioxidant properties of fortified chocolates was found, and the bioaccessibility of their antioxidants was evaluated. The highest AC and TPC were found in ChFrE and chocolate with chokeberries (DCh + ChFrE) before and after simulated in vitro digestion. Bioaccessibility studies indicated that during the simulated digestion the AC of all chocolates reduced significantly, whereas insignificant differences in TPC results were observed between chemical and physiological extracts. Moreover, the influence of plant extracts on physicochemical parameters such as moisture content (MC), fat content (FC), and viscosity of chocolates was estimated. Furthermore, scanning electron microscopy with dispersive energy spectroscopy (SEM-EDS) was used to analyze surface properties and differences in the chemical composition of chocolates without and with additives.  相似文献

The acid fruit of the "xoconostle" cactus belongs to the genus Opuntia family of cacti. It is used as a functional food for its bioactive compounds. Several studies reported that xoconostle fruits have a high amount of ascorbic acid, betalains, phenols, tannins, and flavonoids. These compounds confer antioxidant, antibacterial, anti-inflammatory, and hepatoprotective gastroprotective activity. Xoconostle fruit extracts were tested by in vitro assays where the digestion conditions were simulated to measure their stability. At the same time, the extracts were protected by encapsulation (microencapsulation, multiple emulsions, and nanoemulsions). Applications of encapsulated extracts were probed in various food matrices (edible films, meat products, dairy, and fruit coatings). The xoconostle is a natural source of nutraceutical compounds, and the use of this fruit in the new food could help improve consumers’ health.  相似文献

The aim of this study was to develop pellet formulations that could be used to improve the dissolution and bioavailability of a poorly water-soluble model drug, cisapride. Six different types of pellets were prepared by coating sugar spheres in a fluidized bed coater. When the sugar spheres were single layered containing cisapride and solubilizer such as polysorbate 80, the resulting pellets provided an instant release of cisapride in the simulated gastric fluid. Dissolution tests carried out in the simulated intestinal fluid showed that there were negligible amounts of cisapride released, regardless of the pellet formulation. To succeed in attaining dissolution and the sustained release of cisapride at a neural pH, the single layered pellets were coated again with a coating suspension containing Eudragit RS 30D and L 30D. Scanning electron microscopy revealed that the dual layered pellets had a crack-free and spherical surface. Interestingly, the dual layered pellets provided the sustained release of cisapride in both the simulated gastric and intestinal fluids. The composition and components of the dual layers were found to be key parameters affecting the pattern of cisapride dissolution. Significant improvement in the bioavailability of cisapride was achieved when the dual layered pellets were administered orally to dogs. Overall, these results suggest that the dual layered pellets have potential as a sustained release dosage form for poorly water-soluble drugs.  相似文献

A method for measuring trace concentration of Fe, Co, Ni, Cu, Zn and Pb in plant materials was developed, optimized, validated and compared using reference standards and fresh samples. The method is based on rapid material digestion by radicals produced with laser-assisted pulsed UV (PUV) photolysis of aqueous solutions containing H2O2. Metals in the resulting solution were pre-concentrated as carbamates. Samples were analyzed with particle induced X-ray emission (PIXE). Combined PUV digestion and pre-concentration techniques resulted in enhanced detection levels of selected trace elements in foods and non-food commodities. The method was applied to measure trace elements in wastewater irrigated fruits, vegetables and crops.  相似文献

Two types of mesoporous silica nanospheres (MSNs) were synthesized for use as controlled-release agents. One was prepared by grafting with 5,6-dihydroxyhexylsilane (DH-MSN) and the other one by further coating with cholic acid-crosslinked poly(lactic acid) (CA-PLA-MSN). We studied the release of the antidepressant venlafaxine from each of the materials in simulated gastric fluid (SGF), in simulated gastric acid solution (SGA), and in simulated intestinal fluid without pancreatin (SIF). The CA-PLA-MSN material was able to significantly delay the release of the drug in intestinal condition compared with gastric acid surrounding due to the fast decomposition rate of PLA in gastric acid. Moreover, it successfully avoided the initial burst to a certain extent in SGF. The enzyme pepsin played a favorable obstruct role in both DH-MSN and CA-PLA-MSN systems to reduce release rate. A model based on Weibull model was built to fit the release results, and based on it, the mechanisms about release processes were brought out tentatively.  相似文献
