Synthesis of novel 2,3,4,5-tetrasubstituted isoxazolidine and 2,3,5-trisubstituted isoxazolidine by 1,3-dipolar cycloaddition of nitrones with electron-deficient and electron-rich olefins is described. 1,3-Dipolar cycloaddition of nitrones with β-nitrostyrene, gives exclusively endo-diastereoisomer of isoxazolidine and with ethyl vinyl ether, gives exo-diastereoisomer of isoxazolidine with de >95%. The stereochemistry of the products were assigned using extensive NMR studies. 相似文献

The 1,3-dipolar cycloaddition reactions of α-2-methoxyaryl nitrones with nitrostyrenes and chalcones have been investigated. The regiochemistry and stereochemistry of the resultant cycloadducts have been determined with the help of NMR spectroscopy and X-ray analysis. β-Nitrostyrenes add to nitrones to give two geometrical isomers, while the β-methyl-β-nitrostyrene gives a single isomer in relatively low yield. The chalcones give a single cycloadduct upon 1,3-dipolar addition. 相似文献

2-O- and 4-O-Allyl and -propargyl glucose and the corresponding oxime derivatives were prepared from readily available glucose dithioacetals. Intramolecular 1,3-dipolar cycloaddition of the N-benzyl and N-methyl nitrones of the above acyclic 2-O-allyl glucose derivatives led to the diastereoselective formation of chiral isoxazolidines incorporating the tetrahydrofuran ring. The EI mass spectra revealed a characteristic cleavage of the C-alkyl group adjacent to the furan oxygen atom. An enantiopure trisubstituted tetrahydrofuran was obtained by the reductive cleavage of the isoxazolidine ring of one of the cycloadducts. In contrast, the nitrile oxide cycloaddition of the 2-O-allyl derivatives afforded diastereomeric mixtures of the corresponding dihydroisoxazolines, the stereochemistry of which was tentatively assigned on the basis of the principle of optical superposition. The exclusive formation of a tetrahydrofuran ring from pentaallyl nitrone or nitrile oxide demonstrated the preferred formation of a five-membered ring to that of six or seven-membered rings. The nitrile oxide generated from a 3,4,5,6,7-pentaallyloxy-1-nitroheptane derivative obtained from pentaallylglucose underwent diastereoselective cycloaddition to give an isoxazoline fused to a pyran ring. Enantiopure isoxazoles containing tetrahydrofuran and oxepane rings were also prepared in good yields by the nitrile oxide cycloaddition of the 2-O- and 4-O-propargyl derivatives. 相似文献

Bicyclic isooxazolidinyl enediynes 1 and 2 have been prepared by intramolecular nitrone cycloaddition between the two arms of an acyclic enediyne precursor 13. A stereoselective intramolecular 1,3-dipolar nitrone cycloaddition useful in the synthesis of chromanes is described. The reaction relies on the use of a chiral auxiliary on the nitrone partner. Key to the success of the reaction is the choice of auxiliary and the choice of Lewis acid catalyst. Utilizing an auxiliary with a pendant coordinating group, and Zn(OTf)(2) as the Lewis acid, diastereoselectivities up to 22:1 could be achieved. 相似文献

By the Cu(II)-catalyzed reaction of 2-(4-diazo-3-oxoalkyl)pyridines (2), 4-alkoxycarbonyl (or 4-acyl)-3-oxo-1,2,3,4-tetrahydroquinolizinium ylides (3) were obtained in high yields. AbstractA facile, atom-economic synthesis of isoxazilidino withaferin, a novel hybrid of withaferin A, has been accomplished via two-step reaction of nitrone synthesis followed by nitrone 1,3-dipolar cycloaddition. The reaction is highly chemoselective (preferential reaction only on one of the two double bonds present on withaferin A) and diastereoselective affording exclusively the cis-fused products. The molecular mechanism for the 1,3-dipolar cycloaddition of nitrone with sulfonylethene chlorides has been studied using ab initio and DFT methods at the HF, MP2 and B3LYP levels together with the 6-31G* basis set. Relative rates, stereo and regioselectivity, have been analysed and discussed. For this cycloaddition four reactive channels associated with the formation of two pairs of diastereoisomeric regioisomers have been characterized. Analysis of the geometries of the corresponding transition structures shows that the cycloaddition takes place along a concerted but asynchronous mechanism. Activation energies as asynchronicity are dependent on the computation level. Thus, while HF calculations gave large barriers, MP2 calculations tend to underestimate them. DFT calculations gave reasonable values. These 1,3-dipolar cycloadditions present an endo stereoselectivity while the meta regioselectivity depends on the computational level. Thus, while HF and DFT calculations predict meta path, in agreement with the experimental results, MP2 calculation predict ortho regioselectivity. The frontier molecular orbitals analysis shows that the reaction is controlled by the (HOMOdipole–LUMOdipolarophile) interaction in agreement with the charge transfer analysis carried out at the transition structures. Inclusion of diffuse functions at the B3LYP/6-31+G* level increases the energy barriers about 4 kcal/mol, giving a similar endo/meta selectivity. The review considers the main results of the cycloaddition reactions involving cyclopropenes and methylenecyclopropanes, the compounds bearing strained three-membered rings and, respectively, endo- and exocyclic double bonds. The main attention is focused on the reactions of these compounds with 1,3-dipoles (nitrones, azomethine imines, azomethine ylides, carbonyl ylides, etc.), which gave complex heterocyclic systems with high regio- and stereoselectivity.

We report the synthesis of a broad variety of functionalized molecules for assembly on gold, allowing the formation of biologically relevant SAMs by a modular approach: either utilizing 1,3-dipolar cycloaddition of alkynes and azides in solution or by 'click on SAM'.
