A simple flow injection (FI) conductometric system with gas diffusion separation was developed for the determination of Kjeldahl nitrogen (or proteins) in milk and chicken meat. The sample was digested according to the Kjeldahl standard method and the digest was diluted and directly injected into the donor stream consisting of 4 M NaOH. In alkaline medium, ammonium was converted to ammonia, which diffused through the PTFE membrane to dissolve in an acceptor stream (water). Dissociation of ammonia caused a change in conductance of the acceptor solution, which was linearly proportional to the concentration of ammonium originally present in the injected solution. A conductometric flow through cell and an amplifier circuit was fabricated, which helped improve sensitivity of the conductometric detection system. With using a plumbing Teflon tape as a gas diffusion membrane and without thermostating control of the system, a linear calibration graph in range of 10-100 mg L−1 N-NH4 was obtained, with detection limit of 1 mg L−1 and good precision (relative standard deviation of 0.3% for 11 replicate injections of 50 mg L−1 N-NH4). The developed method was validated by the standard Kjeldahl distillation/titration method for the analysis of milk and chicken meat samples. The proposed system had sample throughput of 35 h−1 and consumed much smaller amounts of chemical than the standard method (275 mg vs 17.5 g of NaOH per analysis, respectively).  相似文献

A flow injection method on the basis of gas phase molecular absorption is described for the sequential determination of ammonium and nitrate. Two hundred microliters of sample solution is injected into the flow line. For ammonium determination, the sample zone is directed to a line in which reacts with NaOH (13 M) and produces ammonia. But for nitrate determination, the sample zone is passed through the on-line copperized zinc (Zn/Cu) reduction column and produces ammonium ion and in the follows ammonia. The produced ammonia in both cases is purged into the stream of N2 carrier gas. The gaseous phase is separated from the liquid phase using a gas-liquid separator and then is swept into a flow through cell, which has been positioned in the cell compartment of an UV-Vis spectrophotometer. The absorbance of the gaseous phase is measured at 194 nm. Under selected conditions for sequential analysis of ammonium and nitrate, linear relations were found between the peak heights of absorption signals and concentrations of ammonium (10-650 μg ml−1) and nitrate (20-800 μg ml−1). The limit of detections for ammonium and nitrate analysis were 8 and 10 μg ml−1, respectively. The relative standard deviations of repeated measurements of 50 μg ml−1 of ammonium and nitrate were 2.0, 2.9%, respectively. Maximum sampling rate was about 40 samples/h. The method was applied to the determination of ammonium in pharmaceutical products and the sequential determination of ammonium and nitrate in spiked water samples.  相似文献

A simple flow injection gas/diffusion method for the determination of trimethylamine (TMA) in seafood with potentiometric detection using tungsten oxide electrode has been developed. The method is based on the diffusion of TMA through a PTFE membrane from a sodium hydroxide donor stream to a phosphate buffer acceptor stream. The TMA in the acceptor stream passes through an electrochemical flow cell containing a tungsten oxide wire and a silver/silver chloride electrode, where TMA was sensitively detected. The parameters affecting the sensitivity of the electrode such as sodium hydroxide concentration, buffer concentration, pH, flow rate and injected volume were studied in details. The electrode response was linear in the concentration range from 1 to 10 μg ml−1 TMA with a correlation coefficient (R2) of 0.991 and a detection limit of 0.05 μg ml−1 TMA. The intra- and inter-days precision (R.S.D.) was found to be, respectively, 1.20 and 1.6% (n=6). The method was applied to the determination of TMA in fish tissue and recoveries of 99-100% were obtained for fish extracts. Results were in close agreement with those obtained by the existing classical official method. Common interference from those species that can diffuse through the membrane were removed by the addition of formaldehyde to the seafood extract. The method is simple, feasible with satisfactory accuracy and precision and thus, could be used for monitoring seafood quality with a sampling rate of 20±2 sample h−1.  相似文献

This work presents new design of a gas diffusion unit, called ‘membraneless gas diffusion (MGD) unit’, which, unlike a conventional gas diffusion (GD) unit, allows selective detection of volatile compounds to be made without the need of a hydrophobic membrane. A flow injection method was developed employing the MGD unit to determine ethanol in alcoholic drinks based on the reduction of dichromate by ethanol vapor. Results clearly demonstrated that the MGD unit was suitable for determination of ethanol in beer, wine and distilled liquors. Detection limit (3S/N) of MGD unit was lower than the GD unit (GD: 0.68%, v/v; MGD: 0.27%, v/v). The MGD design makes the system more sensitive as mass transfer is more efficient than that of GD and thus, MGD can perfectly replace membrane-based designs.  相似文献

An indirect batch spectrophotometric and direct flow-injection (FI) visible spectrophotometric methods have been developed for the determination of the novel anti-psychotic drug olanzapine (OLA). The batch method is based on the oxidation of olanzapine by a known excess of potassium hexacyanoferrate(III) in the presence of the mixture of sulphuric and phosphoric acids (1:1 (v/v)). The absorbance of unreacted oxidant is measured at 425 nm. The absorbance decreases linearly with increasing concentration of the assayed drug. The FI method with detection at 540 nm is based on the direct oxidation of olanzapine one of two oxidants, cerium(IV) sulphate or potassium hexacyanoferrate(III) in acidic medium. The calibration graph were linear over the range of 2.5-40 μg ml−1 in the batch method and 0.05-300 and 0.5-250 μg ml−1 in the FI methods, used cerium (IV) sulphate and potassium hexacyanoferrate (III) respectively. Both FI methods gave similar results in terms of precision and accuracy. The relative standard deviation (R.S.D.), was <1%. The accuracy, obtained from recovery experiments, was 97.9-99.4%. The batch method gave slightly higher R.S.D. values (up to 2.3%) and lower values of accuracy (the recovery was between 96.5 and 96.6%). The methods developed were applied to the determination of olanzapine in a pharmaceutical product.  相似文献

A simple procedure for the determination of ethanol in a liquor by near-infrared (NIR) spectrophotometry with flow injection (FI) is proposed. A liquor sample is equilibrated off-line with dried chloroform to extract ethanol into the organic phase. The extract is injected into a carrier stream of dried chloroform passing through a home-made flow through cell (1 mm path length) sitting in a NIR spectrophotometer for continuous monitoring of absorbance at 2305 or 2636 nm. The ethanol content can be evaluated from a calibration established by a plot of change in absorbance versus concentration of ethanol standard solutions. Optimization of the system will be discussed. A calibration is linear in the range of 20–50% (v/v) ethanol. A throughput of 240 injections h−1 can be obtained. The procedure is validated by comparing the results with an analysis using gas chromatography.  相似文献

In this work, a solid phase spectrophotometric method in association with flow injection analysis for formaldehyde determination has been developed with direct measurement of light-absorption in C18 material. The 3,5-diacetyl-1,4-dihydrolutidine produced from the reaction between formaldehyde and fluoral P was quantitatively retained on C18 support and the spectrophotometric detection was performed simultaneously at 412 nm. The retained complex was quickly eluted from C18 material with the eluent stream consisting of a 50% (v/v) ethanol solution. The results showed that the proposed method is simple, rapid and the analytical response is linear in the concentration range of 0.050-1.5 mg L−1. The limit of detection was estimated as 30 μg L−1 and the R.S.D. 2.2% using a sample volume of 625 μL. The system presented an analytical throughput of 20 determinations per hour. The method was successfully applied in the determination of formaldehyde in ethanol fuel.  相似文献

A new simple, sensitive, rapid and precise flow injection (FI) procedure based on the formation of copper complexes with some angiotensin converting enzyme (ACE) inhibitors has been developed and evaluated for the analysis of lisinopril (LN), enalapril maleate (EP), ramipril (RP) and perindopril tert-butylamine (PD). In this method, samples were injected into a flowing stream of distilled-deionized water, carried through the packed reactor of CuO for derivatization followed by ultraviolet (UV) detection. The flow rate was 1.5 ml min−1 and column temperature was ambient (25 °C). Lisinopril was injected directly into the flowing stream and the detector response was measured at 262 nm. The hydrolysis products of enalapril maleate, ramipril and perindopril tert-butylamine in 0.2N NaOH were injected after neutralization with 1N HCl and the detector response was measured at 272, 265 and 252 nm, respectively. The developed method was successfully applied to the determination of tested drugs in pharmaceutical preparations at a sampling rate of 60 samples h−1 and a recovery near 100% for all compounds.  相似文献

This paper describes a method for the determination of lysine based on a flow injection (FI) differential potentiometry system. The flow injection manifold is composed of a support electrolyte solution channel and a water channel acting as a carrier into which the sample solution is injected. The lysine biosensor was consisted of lysine oxidase chemically immobilized on a nylon membrane and attached to an all-solid-state ammonium electrode. A circular ammonium electrode was used as a reference. Hence, the possible interference of endogenous ammonium can be partly corrected by differential potentiometry. In order to increase the sensitivity of the response, the reaction was kinetically developed following a stopped-flow method. As a result, the sensitivity increased from 20 to 40 mV per decade when comparing the FI and the stopped-flow values. Furthermore, the peak-to-peak stopped-flow signals generated can be used as a more selective analytical response for lysine. The quantification of lysine in mixture samples containing small amounts of ammonium can be achieved with an acceptable accuracy, with prediction errors lower than 4%. However, when the ammonium concentration exceeded the lysine concentration, multivariate calibration with non-linear partial least squares (PLS) regression was needed to improve the lysine quantification, with an overall prediction error around 10%.  相似文献

A gas diffusion-flow injection system was developed for the determination of carbon dioxide in gaseous samples. The calibration was based on the use of either gaseous carbon dioxide or aqueous sodium carbonate standards. Gaseous carbon dioxide samples and gaseous or aqueous standards were injected directly into a donor stream of 1.0×10−4 M H2SO4. In the gas diffusion unit, carbon dioxide diffused through a PTFE membrane into an acceptor stream containing a mixed acid/base indicator. The absorbance of the acceptor stream was monitored spectrophotometrically at 554 nm. The calibration plot was linear over the range of 5.00×102 to 1.27×104 μl l−1 with a sample throughput of 28 h−1 and 3.2% R.S.D. ([CO2]=2.37×103 μl l−1, n=12). The detection limit was determined as 2.50×102 μl l−1. The flow system was successfully applied to the analysis of several natural gaseous samples and the headspace of milk containers during storage. The flow injection results were found to be statistically indistinguishable at the 95% confidence level from those obtained by gas chromatography using thermal conductivity detection.  相似文献

This work reports a novel flow injection (FI) method for the determination of captopril, 1-[(2S)-3-mercapto-2-methylpropionyl]-l-proline (CPL), based on the enhancement CPL affords on the chemiluminescence (CL) reaction between luminol and hydrogen peroxide. For this purpose alkaline luminol and hydrogen peroxide solutions were mixed online, the sample containing CPL was injected into an aqueous carrier stream, mixed with the luminol-hydrogen peroxide stream and pumped into a glass flow cell positioned in front of a photomultiplier tube (PMT). The increase in the CL intensity was recorded in the form of FI peaks, the height of which was related to the CPL mass concentration in the sample. Different chemical and instrumental parameters affecting the CL response were investigated. Under the selected conditions, the log-log calibration curve was linear in the range 5-5000 μg l−1 of CPL, the limit of detection was 2 μg l−1 (at the 3σ level), the R.S.D., sr was 3.1% at the 100 μg l−1 level (n=8) and the sampling rate was 180 injections h−1. The method was applied to the determination of CPL in pharmaceutical formulations with recoveries in the range 100±3%.  相似文献

A novel gas diffusion-flow injection method has been developed for the rapid and sensitive determination of total inorganic carbon (TIC) in water. The method is based on the diffusion of CO2 across gas permeable membrane from a donor stream containing 0.1 M HCl to an acceptor stream of sodium acetate (10−5 mol l−1 and pH 10). The CO2 trapped in the acceptor stream passes through an electrochemical flow cell contains a tungsten oxide wire and a silver/silver chloride electrode, where it was sensitively detected. The parameters affecting the sensitivity of the electrode such as buffer concentration, pH, flow rate and injected volume were studied in detail. The electrode response was linear in the concentration range from 5 to 100 μg ml−1 CO32− with a correlation coefficient (R2) of 0.998. Precision (R.S.D.) was 1.42% for 20 μg ml−1 standard solution of CO32− (n=10). The detection limit was 0.20 μg ml−1 CO32−. The method was evaluated by the injection of real natural water samples and an average recovery of 100.1% was obtained. The sampling rate was 30 samples h−1. The method is simple, feasible with satisfactory accuracy and precision and thus could be used for monitoring TIC in water.  相似文献

The consumption of ethanol is known to increase the likelihood of oral cancer. In addition, there has been a growing concern about possible association between long term use of ethanol-containing mouthwashes and oral cancer. Acetaldehyde, known to be a carcinogen, is the first metabolite of ethanol and it can be produced in the oral cavity after consumption or exposure to ethanol. This paper reports on the development of a gas-diffusion flow injection method for the online determination of salivary acetaldehyde by its colour reaction with 3-methyl-2-benzothiazolinone hydrazone (MBTH) and ferric chloride. Acetaldehyde samples and standards (80 μL) were injected into the donor stream containing NaCl from which acetaldehyde diffused through the hydrophobic Teflon membrane of the gas-diffusion cell into the acceptor stream containing the two reagents mentioned above. The resultant intense green coloured dye was monitored spectrophotometrically at 600 nm. Under the optimum working conditions the method is characterized by a sampling rate of 9 h−1, a linear calibration range of 0.5–15 mg L−1 (absorbance = 5.40 × 10−2 [acetaldehyde, mg L−1], R2 = 0.998), a relative standard deviation (RSD) of 1.90% (n = 10, acetaldehyde concentration of 2.5 mg L−1), and a limit of detection (LOD) of 12.3 μg L−1. The LOD and sampling rate of the proposed method are superior to those of the conventional gas chromatographic (GC) method (LOD = 93.0 μg L−1 and sampling rate = 4 h−1). The reliability of the proposed method was illustrated by the fact that spiked with acetaldehyde saliva samples yielded excellent recoveries (96.6–101.9%), comparable to those obtained by GC (96.4–102.3%) and there was no statistically significant difference at the 95% confidence level between the two methods when non-spiked saliva samples were analysed.  相似文献

A slurry sampling inductively coupled plasma mass spectrometry (ICP-MS) method has been developed for the determination of Ge, As, Cd, Sb, Hg and Bi in cosmetic lotions using flow injection (FI) vapor generation (VG) as the sample introduction system. A slurry containing 2% m/v lotion, 2% m/v thiourea, 0.05% m/v l-cysteine, 0.5 μg mL−1 Co(II), 0.1% m/v Triton X-100 and 1.2% v/v HCl was injected into a VG-ICP-MS system for the determination of Ge, As, Cd, Sb, Hg and Bi without dissolution and mineralization. Because the sensitivities of the analytes in the slurry and that of aqueous solution were quite different, an isotope dilution method and a standard addition method were used for the determination. This method has been validated by the determination of Ge, As, Cd, Sb, Hg and Bi in GBW09305 Cosmetic (Cream) reference material. The method was also applied for the determination of Ge, As, Cd, Sb, Hg and Bi in three cosmetic lotion samples obtained locally. The analysis results of the reference material agreed with the certified value and/or ETV-ICP-MS results. The detection limit estimated from the standard addition curve was 0.025, 0.1, 0.2, 0.1, 0.15, and 0.03 ng  g−1 for Ge, As, Cd, Sb, Hg and Bi, respectively, in original cosmetic lotion sample.  相似文献

A flow injection (FI) method with flame atomic absorption spectrometry (FAAS) detection was developed for the determination and speciation of nitrite and nitrate in foodstuffs and wastewaters. The method is based on the oxidation of nitrite to nitrate using a manganese(IV) dioxide oxidant microcolumn where the flow of the sample through the microcolumn reduces the MnO2 solid phase reagent to Mn(II), which is measured by FAAS. The absorbance of Mn(II) are proportional to the concentration of nitrite in the samples. The injected sample volume was 400 μL with a sampling rate of analyses was 90 h−1 with a relative standard deviation better than 1.0% in a repeatability study. Nitrate is reduced to nitrite in proposed FI-FAAS system using a copperized cadmium microcolumn and analyzed as nitrite. The calibration curves were linear up to 20 mg L−1 and 30 mg L−1 with a detection limit of 0.07 mg L−1 and 0.14 mg L−1 for nitrite and nitrate, respectively. The results exhibit no interference from the presence of large amounts of ions. The method was successfully applied to the speciation of nitrite and nitrate in spiked natural water, wastewater and foodstuff samples. The precision and accuracy of the proposed method were comparable to those of the reference spectrophotometric method.  相似文献

In this work, it was developed a method for the determination of nitrite and nitrate in groundwater by high-resolution continuum source electrothermal molecular absorption spectrometry of NO produced by thermal decomposition of nitrate in a graphite furnace. The NO line at 215.360 nm was used for all analytical measurements and the signal obtained by integrated absorbance of three pixels. A volume of 20 μL of standard solution or groundwater sample was injected into graphite furnace and 5 μL of a 1% (m/v) Ca solution was co-injected as chemical modifier. The pyrolisis and vaporization temperatures established were of 150 and 1300 °C, respectively. Under these conditions, it was observed a difference of thermal stability among the two nitrogen species in the presence of hydrochloric acid co-injected. While that the nitrite signal was totally suppressed, nitrate signal remained nearly stable. This way, nitrogen can be quantified only as nitrate. The addition of hydrogen peroxide provided the oxidation of nitrite to nitrate, which allowed the total quantification of the species and nitrite obtained by difference. A volume of 5 μL of 0.3% (v/v) hydrochloric acid was co-injected for the elimination of nitrite, whereas that hydrogen peroxide in the concentration of 0.75% (v/v) was added to samples or standards for the oxidation of nitrite to nitrate. Analytical curve was established using standard solution of nitrate. The method described has limits of detection and quantification of 0.10 and 0.33 μg mL−1 of nitrogen, respectively. The precision, estimated as relative standard deviation (RSD), was of 7.5 and 3.8% (n = 10) for groundwater samples containing nitrate–N concentrations of 1.9 and 15.2 μg mL−1, respectively. The proposed method was applied to the analysis of 10 groundwater samples and the results were compared with those obtained by ion chromatography method. In all samples analyzed, the concentration of nitrite–N was always below of the limit of quantification of both the methods. The concentrations of nitrate–N varied from 0.58 to 15.5 μg mL−1. No significant difference it was observed between the results obtained by both methods for nitrate–N, at the 95% confidence level.  相似文献

Two simple, rapid and selective spectrophotometric methods have been described for the determination of promethazine hydrochloride (PMH) either in pure or pharmaceutical formulations. The methods are based on the formation of a green colored product with sulphanilic acid (SPA) in presence of N-bromosuccinimide (NBS) or a red colored, chloroform soluble product with p-nitroaniline (PNA) in presence of ceric ammonium sulphate (CAS). The chromogen formed has maximum absorption at 600 nm and at 510 nm with SPA and PNA, respectively. The optimum reaction conditions and other analytical parameters are evaluated. Beer's law is valid in the concentration range of 0.5–25 g/mL (R= 0.9983) with SPA and 2–75 g/mL (R = 0.9981) with PNA. Molar absorptivity values as calculated from the Beer's law data are 5.54 × 103 L/mol/cm and 3.05 × 103 L/mol/cm for SPA and PNA, respectively. The results are in good agreement with those of the official method. No interference was observed from common pharmaceutical excipients.  相似文献

Xanthones are well known for their interesting phytochemical properties, which make them attractive to the pharmaceutical and medicinal industry. We have therefore developed a method to analyse the major xanthones in Garcina mangostana. The xanthones were extracted by pressurized liquid extraction with ethanol and separated at the semi-preparative scale by centrifugal partition chromatography (CPC) with a biphasic solvent system consisting of heptane/ethyl acetate/methanol/water (2:1:2:1, v/v/v/v). A CPC-electrospray ionisation MS coupling was performed and used to simultaneously separate and identify the compounds. Thanks to a variable flow splitter and an additional stream of ethanol/1 mol L−1 ammonium acetate (95:5, v/v), all the compounds were ionised, detected and monitored whatever the solvents used in mobile phase for the CPC separation. The dual mode or elution–extrusion which are less solvent-consuming and faster than the elution mode were used without loss of ionisation and detection.  相似文献

A novel method for preconcentration of methylmercury and inorganic mercury from water samples was developed involving the determination of ng l−1 levels of analytes retained on the silica C18 solid sorbent, previous complexation with ammonium pyrrolidine dithiocarbamate (APDC), by slurry sampling cold vapor atomic absorption spectrometry (SS-CVAAS) in a flow injection (FI) system. Several variables were optimized affecting either the retention of both mercury species, such as APDC concentration, silica C18 amount, agitation times, or their determination, including hydrochloric acid concentration in the suspension medium, peristaltic pump speed and argon flow-rate. A Plackett-Burman saturated factorial design permitted to differentiate the influential parameters on the preconcentration efficiency, which were after optimized by the sequential simplex method. The contact time between mercury containing solution and APDC, required to reach an efficient sorption, was decreased from 26 to 3 min by the use of sonication stirring instead of magnetic stirring. The use of 1 mol dm−3 hydrochloric acid suspension medium and 0.75% (m/v) sodium borohydride reducing agent permitted the selective determination of methylmercury. The combination of 5 mol dm−3 hydrochloric acid and 10−4% (m/v) sodium borohydride was used for the selective determination of inorganic mercury. The detection limits achieved for methylmercury and inorganic mercury determination under optimum conditions were 0.96 and 0.25 ng l−1, respectively. The reliability of the proposed method for the determination of both mercury species in waters was checked by the analysis of samples spiked with known concentrations of methylmercury and inorganic mercury; quantitative recoveries were obtained.  相似文献
