Copper and silver N‐heterocyclic carbene (NHC) complexes were prepared through a simple, base‐free protocol involving the decomposition of corresponding imidazol(in)ium‐2‐carboxylates under thermolytic conditions and a subsequent reaction of the in situ generated carbenes with copper(I) or silver(I) chloride, respectively. The desired NHC metal complexes were isolated with good yields after simple crystallization.  相似文献

N-Heterocyclic carbene (NHC) ligand precursors, namely, HIm(A)Cl [1,3-bis(2-ethoxy-2-oxoethyl)-1H-imidazol-3-ium chloride] and HIm(B)Cl {1,3-bis[2-(diethylamino)-2-oxoethyl]-1H-imidazol-3-ium chloride}, functionalized with hydrophilic groups on the imidazole rings have been synthesized and were used in the synthesis of corresponding carbene complexes of silver(I) and copper(I), {[Im(A)]AgCl}, {[Im(A)]CuCl}, and {[Im(B)](2)Ag}Cl. Related Au(I)NHC complexes {[Im(A)]AuCl} and {[Im(B)]AuCl} have been obtained by transmetalation using the silver carbene precursor. These compounds were characterized by several spectroscopic techniques including NMR and mass spectroscopy. HIm(B)Cl and the gold(I) complexes {[Im(A)]AuCl} and {[Im(B)]AuCl} were also characterized by X-ray crystallography. The cytotoxic properties of the NHC complexes have been assessed in various human cancer cell lines, including cisplatin-sensitive and -resistant cells. The silver(I) complex {[Im(B)](2)Ag}Cl was found to be the most active, with IC(50) values about 2-fold lower than those achieved with cisplatin in C13*-resistant cells. Growth-inhibitory effects evaluated in human nontransformed cells revealed a preferential cytotoxicity of {[Im(B)](2)Ag}Cl versus neoplastic cells. Gold(I) and silver(I) carbene complexes were also evaluated for their ability to in vitro inhibit the enzyme thioredoxin reductase (TrxR). The results of this investigation showing that TrxR appeared markedly inhibited by both gold(I) and silver(I) derivatives at nanomolar concentrations clearly point out this selenoenzyme as a protein target for silver(I) in addition to gold(I) complexes.  相似文献

A series of novel imidazolium salts bearing hydrophilic tetraethylene glycol (TEG) and/or hydrophobic long-chain alkyl (n-C12) functionalities, which are precursors for desired N-heterocyclic carbene (NHC) ligands, were synthesized and characterized. Rh(I)-NHC complexes were prepared in good yields by the silver carbene transfer method with NHC-Ag species derived from the imidazolium salts. The molecular structure of the Rh(I)-NHC complex having n-C12 chains has been determined by a single-crystal X-ray diffraction study and the complex is found to possess extended alkyl chains with anti conformations in the solid state. Hydrosilylation with the Rh(I) complexes and Suzuki-Miyaura coupling reactions with the Pd(II) complexes with these NHC ligands were carried out. In the latter case, the TEG moiety enhances catalytic activity considerably.  相似文献

A silver‐catalyzed amidiniumation of N‐propiolic formamidines for the synthesis of novel enamine amido carbene precursors is reported. Isolation of a first silver intermediate in silver‐catalyzed amidiniumation of alkynes and other organogold intermediates supports our proposed mechanisms. Several control experiments reveal the unexpected effects of both HOTf and substrate substituents on the choice of either a π or σ,π silver activation mode and the cyclization fashion. Bis(hydroxyimidazol)ium salts were obtained through an unprecedented umpolung of propiolamides. The byproduct Ag2O as either an oxidant or silver source promotes the syntheses of N‐heterocyclic carbene (NHC) precursors or Ag/NHC complexes.  相似文献

A new imidazolium salt, 1,3-bis(2-diphenylphosphanylethyl)-3H-imidazol-1-ium chloride (2), for the phosphine/N-heterocyclic carbene-based pincer ligand, PC(NHC)P, and its palladium complexes were reported. The complex, [Pd(PC(NHC)P)Cl]Cl (4), was prepared by the common route of silver carbene transfer reaction and a novel direct reaction between the ligand precursor, PC(NHC)P.HCl and PdCl(2) without the need of a base. Metathesis reactions of 4 with AgBF(4) in acetonitrile produced [Pd(PC(NHC)P)(CH(3)CN)](BF(4))(2) (5). The same reaction in the presence of excess pyridine gave [Pd(PC(NHC)P)(py)](BF(4))(2) (6). The X-ray structure determination on 4-6 revealed the chiral twisting of the central imidazole rings from the metal coordination plane. In solution, fast interconversion between left- and right-twisted forms occurs. The twisting reflects the weak pi-accepting property of the central NHC in PC(NHC)P. The uneven extent of twisting among the three complexes further implies the low rotational barrier about the Pd-NHC bond. Related theoretical computations confirm the small rotational energy barrier about the Pd-NHC bond (ca. 4 kcal/mol). Catalytic applications of 4 and 5 have shown that the complexes are modest catalysts in Suzuki coupling. The complexes were active catalysts in Heck coupling reactions with the dicationic complex 5 being more effective than the monocationic complex 4.  相似文献

From the reaction of 1H‐imidazole ( 1a ), 4,5‐dichloro‐1H‐imidazole ( 1b ) and 1H‐benzimidazole ( 1c ) with p‐cyanobenzyl bromide ( 2 ), symmetrically substituted N‐heterocyclic carbene (NHC) [( 3a–c )] precursors, 1‐methylimidazole ( 5a ), 4,5‐dichloro‐1‐methylimidazole ( 5b ) and 1‐methylbenzimidazole ( 5c ) with benzyl bromide ( 6 ), non‐symmetrically substituted N‐heterocyclic carbene (NHC) [( 7a–c )] precursors were synthesized. These NHC? precursors were then reacted with silver(I) acetate to yield the NHC‐silver complexes [1,3‐bis(4‐cyanobenzyl)imidazole‐2‐ylidene] silver(I) acetate ( 4a ), [4,5‐dichloro‐1,3‐bis(4‐cyanobenzyl)imidazole‐2‐ylidene] silver(I) acetate ( 4b ), [1,3‐bis(4‐cyanobenzyl)benzimidazole‐2‐ylidene] silver(I) acetate ( 4c ), (1‐methyl‐3‐benzylimidazole‐2‐ylidene) silver(I) acetate ( 8a ), (4,5‐dichloro‐1‐methyl‐3‐benzylimidazole‐2‐ylidene) silver(I) acetate ( 8b ) and (1‐methyl‐3‐benzylbenzimidazole‐2‐ylidene) silver(I) acetate ( 8c ) respectively. The four NHC‐precursors 3a–c, 7c and four NHC–silver complexes 4a–c and 8c were characterized by single crystal X‐ray diffraction. The preliminary antibacterial activity of all the compounds was studied against Gram‐negative bacteria Escherichia coli, and Gram‐positive bacteria Staphylococcus aureus using the qualitative Kirby‐Bauer disc‐diffusion method. All NHC–silver complexes exhibited medium to high antibacterial activity with areas of clearance ranging from 4 to 12 mm at the highest amount used, while the NHC‐precursors showed significantly lower activity. In addition, all NHC–silver complexes underwent preliminary cytotoxicity tests on the human renal‐cancer cell line Caki‐1 and showed medium to high cytotoxicity with IC50 values ranging from 53 ( ± 8) to 3.2 ( ± 0.6) µM. A series of monomeric palladacycle complexes bearing n‐butyl‐substituted N‐heterocyclic carbenes, namely [Pd(NHC)X(dmba)] (dmba: dimethylbenzylamine and [Pd(NHC)X(ppy)]; NHC: 1‐n‐butyl‐3‐substituted benzylimidazol‐2‐ylidene; ppy: 2‐phenylpyridine), were prepared either by transmetallation from the corresponding silver carbene complexes or by the reaction of the corresponding acetate‐bridged palladacycle dimer with N‐heterocyclic carbene ligands in high yields. The palladium(II) complexes were characterized using elemental analyses, APCI‐MS, 1H NMR and 13C NMR spectroscopies. These complexes are efficient in the Suzuki–Miyaura coupling reaction between phenylboronic acid and aryl bromides.  相似文献

The synthesis, characterisation and biological activity of water‐soluble Ag(I)‐NHC complexes, general formula Na[(NHC)AgCl] where NHC is a sulfonated and sterically hindered N‐heterocyclic carbene, is reported. The Ag‐NHC complexes (2a–e) were synthesised by reacting the corresponding sulfonated NHC ligands with Ag2O in the presence of NaCl or NaBr in methanol/water (1:1) solution. Synthesised silver (I)‐N‐heterocyclic carbene complexes have been characterised by NMR, micro‐analysis and HRMS spectroscopic methods. The IC50 values of these complexes were determined by a proliferation BrdU enzyme‐linked immunosorbent assay (ELISA) against HeLa (human cervix carcinoma), HT29 (human adenocarcinoma) and L929 (mouse fibroblast) cell lines. These complexes have been highlighted as promising and original platforms for building new types of metalodrug. All new water‐soluble Ag(I) complexes demonstrated remarkable cytotoxic activity against HeLa, HT29 and L929 cell lines.  相似文献

Several silver(I) complexes with chiral amino-N-heterocyclic carbene (NHC) ligands, which are not diastereomerically pure, were prepared and used to generate in situ chelating NHC–amino palladium(II) complexes. The potential of these palladium(II) complexes in asymmetric catalysis was evaluated in the allylic alkylation reaction. The influence of the structure and of the diastereomeric purity of the ligands on enantioselectivity, as well as the role of the silver salts, were studied. Enantiomeric excesses of up to 80% were obtained with the best ligand.  相似文献

Two highly labile silver carbene cluster complexes are described, which are unique in that they mark the transition point at which the carbene center transmutes from a fairly common NHC‐like nucleophilic behavior to an electrophilic character befitting reactive silver carbene intermediates of relevance in various catalytic transformations. This amphoteric character is the distinguishing attribute of a μ‐bridged donor/donor carbene entity that connects two silver atoms of triangular or tetrahedral metallic core units.  相似文献

A new type of quinoline-functionalized palladium N-heterocyclic carbene (NHC) complexes has been synthesized via silver transmetallation. The quinoline moiety was either directly attached to the imidazole ring or linked to it by a methylene group. NHCs with a methylene linker tend to form trans biscarbene complexes in the reaction of Pd(COD)Cl2, while NHCs without any linker form chelating NHC-quinoline (NHC-N) complexes. These two types of carbenes also react with [Pd(allyl)Cl]2 to give monodentate NHC palladium eta(3)-allyl chlorides [Pd(NHC)(allyl)Cl]. Fluxionality in the NMR time scale was observed for most complexes, and the origin of their dynamic behaviors was discussed for each type of structure. For [Pd(NHC)(allyl)Cl] with a relatively small wing tip group of the NHC, the fluxionality (selective line-broadening of (1)H NMR signals) is caused by selective eta(3)-eta(1)-eta(3) allyl isomerization. For NHC with a bulkier (t)Bu group, a different line-broadening pattern was observed and was ascribed to partially hindered Pd-C(carbene) bond rotation. For cationic chelating complexes [Pd(NHC-N)(allyl)]BF4, the dynamic exchange process likely originates from a dissociative boat-to-boat inversion of 7-membered palladacycles. Activation parameters were measured for this process. Crystal structures were reported for representative complexes in each category.  相似文献

A series of unsymmetrically substituted N‐heterocyclic carbene (NHC) precursors ( 1a , 1b , 1c , 1d , 1e ) were synthesized from the reaction of N‐phenylbenzimidazole with various alkyl halides. These compounds were used to synthesize NHC–silver(I) complexes ( 2a , 2b , 2c , 2d , 2e ). The five new 1‐phenyl‐3‐alkylbenzimidazolium salts ( 1a , 1b , 1c , 1d , 1e ) and their NHC–silver complexes ( 2a , 2b , 2c , 2d , 2e ) were characterized by the 1H NMR, 13C NMR and FT‐IR spectroscopic methods and elemental analysis techniques. Also, the two NHC–silver complexes 2b and 2c were characterized by single‐crystal X‐ray crystallography, which confirmed the linear C―Ag―Cl arrangements. The antibacterial activities of the NHC precursor and NHC–silver complexes were tested against three Gram‐positive bacterial strains (Bacillus subtilis, Listeria monocytogenes and Staphylococcus aureus) and three Gram‐negative bacterial strains (Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa) using the microdilution broth method. The NHC–silver complexes showed higher antibacterial activity than the NHC precursors. In addition, silver complexes 2a , 2b , 2c , 2d showed high antibacterial activity against the Gram‐positive bacteria L. monocytogenes and S. aureus compared to the standard, tetracycline. This paper reports the synthesis and characterization of a variety of ruthenium complexes coordinated with phosphine and N-heterocyclic carbene (NHC) ligands. These complexes include several alkylidene derivatives of the general formula (NHC)(PR(3))(Cl)(2)Ru=CHR', which are highly active olefin metathesis catalysts. Although these catalysts can be prepared adequately by the reaction of bis(phosphine) ruthenium alkylidene precursors with free NHCs, we have developed an alternative route that employs NHC-alcohol or -chloroform adducts as "protected" forms of the NHC ligands. This route is advantageous because NHC adducts are easier to handle than their free carbene counterparts. We also demonstrate that sterically bulky bis(NHC) complexes can be made by reaction of the pyridine-coordinated precursor (NHC)(py)(2)(Cl)(2)Ru=CHPh with free NHCs or NHC adducts. Two crystal structures are presented, one of the mixed bis(NHC) derivative (H(2)IMes)(IMes)(Cl)(2)Ru=CHPh, and the other of (PCy(3))(Cl)(CO)Ru[eta(2)-(CH(2)-C(6)H(2)Me(2))(N(2)C(3)H(4))(C(6)H(2)Me(3))], the product of ortho methyl C-H bond activation. Other side reactions encountered during the synthesis of new ruthenium alkylidene complexes include the formation of hydrido-carbonyl-chloride derivatives in the presence of primary alcohols and the deprotonation of ruthenium vinylcarbene ligands by KOBu(t). We also evaluate the olefin metathesis activity of NHC-coordinated complexes in representative RCM and ROMP reactions.  相似文献

[(NHC)AuCl] complexes (NHC = N-heterocyclic carbene), in conjunction with a silver salt, were found to efficiently catalyze the rearrangement of allylic acetates under both conventional and microwave-assisted heating. The optimization of several reaction parameters (solvent, silver salt, and ligand) as well as a study of the reaction scope are reported. The steric hindrance of the ligand bound to gold was found crucial for the outcome of the reaction as only extremely bulky ligands permitted the isomerization.  相似文献
