建立了高效液相色谱柱前衍生化法测定大鼠腹腔注射(i.p.)给药盐酸氟西汀前后脑渗析液中5-羟色胺(5-HT)的浓度变化情况。在大鼠给药盐酸氟西汀前和给药后不同时间点取其脑渗析液,加入衍生化试剂反应后,以乙腈-20 mmol/L醋酸盐缓冲液(pH 5.0)(体积比为45∶55)(含20 mmol/L辛烷磺酸钠)作为流动相,在Hypersil C18色谱柱 (250 mm×2.0 mm,5 μm)上进行分离,荧光检测波长为λex=330 nm,λem=455 nm。5-HT在浓度为0.25~5.0 nmol/L 范围内线性关系良好(r=0.9991);5-HT的最低定量限为0.25 nmol/L。本法准确可靠、简便,适用于生物样品中5-HT的浓度测定。  相似文献

建立了在线衍生、双波长高效液相色谱-荧光检测器同时检测血清中犬尿氨酸(kynurenine, Kyn)和犬尿喹啉酸(kynurenic acid, KYNA)含量的方法。血清标本经5%高氯酸溶液去除蛋白质后,上清液直接进样分析测定。采用的色谱柱为Hypersil C8柱;流动相为0.25 mol/L醋酸锌-50 mmol/L醋酸溶液(含3%乙腈),流速为1.5 mL/min。在0~10 min时间段,在激发波长和发射波长分别为365 nm和480 nm时检测Kyn;10 min后,在激发波长和发射波长分别变换为344 nm和404 nm时检测KYNA。Kyn的保留时间约为8.1 min,线性范围为98~19600 nmol/L,最低检出浓度为50 nmol/L,平均回收率为94.88%,日内、日间测定值的相对标准偏差(RSD)均低于4%。KYNA的保留时间约为13.0 min,线性范围为2.62~1047 nmol/L,最低检出浓度为0.11 nmol/L,平均回收率为102.72%,日内、日间测定的RSD均低于4%。苯丙氨酸、酪氨酸、色氨酸和5-羟色胺等物质对目标物的检测无干扰。71例健康成人血清中,Kyn和KYNA含量分别为(1.40±0.34) μmol/L和(24.22±8.67) nmol/L。该方法简便、快速、灵敏、特异,适于临床和科研应用。  相似文献

建立了醋酸锌在线衍生高效液相色谱法同时测定血浆中色氨酸(Trp)、犬尿氨酸(Kyn)、5-羟吲哚乙酸(5-Hiaa)和犬尿喹啉酸(Kyna)的方法。以3-硝基酪氨酸为内标(IS),采用Hypersil C-18柱(250 mm×4.0 mm, 5 μ m),以250 mmol/L醋酸锌溶液(pH 5.5)-乙腈(95:5, v/v)为流动相,流速为0.8 mL/min,柱温30℃。荧光检测波长设定:5-Hiaa为278 nm(λex)/343 nm(λem), Kyna为244 nm(λex)/400 nm(λem);紫外检测波长设定:Kyn和IS为360 nm, Trp为302 nm。4种物质的回收率在91.62%~114.17%之间;线性范围分别为2.50~320.00 μ mol/L(Trp), 0.32~15.36 μ mol/L(Kyn), 3.27~104.60 nmol/L(5-Hiaa), 14.00~464.80 nmol/L(Kyna);检出限分别为0.078 μ mol/L(Trp), 0.056 μ mol/L(Kyn), 0.690 nmol/L(5-Hiaa), 1.290 nmol/L(Kyna)。利用该方法对30例正常孕妇和28例女性健康志愿者的血浆进行测定,结果表明两组间Trp, Kyn和Kyna含量有显著性差异。该方法操作简便,重复性好,灵敏度高,适合于临床检测。  相似文献

建立了一种高效液相色谱-程序波长紫外检测法同时测定血浆中色氨酸(Trp)及其主要代谢产物犬尿氨酸(Kyn)和5-羟色胺(5-HT)。以茶碱为内标(IS),采用BDS-Hypersil-C8柱(150 mm×4.6 mm, 5 μm)分离。流动相为10 mmol/L醋酸钠缓冲液(pH 4.5)-乙腈(94:6, v/v),流速为0.6 mL/min;柱温为25 ℃;紫外检测波长设定: Kyn和IS为360 nm, 5-HT为220 nm, Trp为302 nm。3种物质的平均回收率为87%~113%;线性范围分别为3.97~400 μmol/L(Trp), 0.421~20.2 μmol/L(Kyn), 4.36~980 nmol/L(5-HT);检出限分别为0.134 μmol/L(Trp), 0.0160 μmol/L(Kyn), 2.03 nmol/L(5-HT)。利用该方法对15例抑郁症患者和15例健康志愿者的血浆进行测定,结果表明两组间Trp的代谢存在显著的差异。  相似文献

An ion-pair reversed phase high performance liquid chromatographic method was developed for the quantitative determination of lipoperoxide (as malondialdehyde, MDA) in animal tissue using a ODS column and a mobile phase of methanol + phosphate buffer (53:47 v/v, pH 6.5). Tetrabutylammonium bromide was used as an ion-pair reagent and the detection wavelength was set at 532 nm. The method was sensitive, accurate and simple. A linear relationship was obtained between the peak height and the amount of MDA from 50 nmol/L to 1200 nmol/L and the lower limit of detection of MDA was 15 nmol/L (signal-to-noise ratio greater than 2), and the coefficient of variation at the 200 nmol/L level was 3.7% (n = 7). The results correlated well with those from the 2-thiobarbituric acid colorimetric method. The method has been used in the pathogenic studies of cerebral ischemia and acute renal failure for the determination of trace lipoperoxide.  相似文献

A highly sensitive method for the determination of cholesterol in biological fluids is described. Unsaponifiable lipids from rat serum and thoracic duct lymph chylomicron samples were treated with cholesterol oxidase. The product of the enzymatic reaction, delta 4-cholestenone, was analysed by normal-phase high-performance liquid chromatography (HPLC) using hexane-isopropanol (95:5, v/v) as a mobile phase and detected with a UV spectrophotometer at 240 nm. When the standard samples containing varying amounts of cholesterol (0.15-3 nmol) were treated with cholesterol oxidase and analysed by HPLC (injected amounts 0.09-1.8 nmol of cholesterol), the peak areas increased proportionally with the amounts of authentic cholesterol with a correlation coefficient of 0.996. The values in these biological fluids determined by the HPLC method were identical to those obtained by enzymatic-colorimetric or gas chromatographic methods. Moreover, the detection limit (0.09 nmol) of the present method (0.15 nmol are required for the sample preparation) is lower than those of conventional methods (approximately 30 nmol). Because of the excellent sensitivity and reproducibility, this method is well suited for the determination of cholesterol in biological fluids where cholesterol concentration is low.  相似文献

利用细胞色素c与Nafion,通过自组装方式,构建了纳米簇模拟过氧化物酶;利用电子透射显微镜、圆二色谱仪、紫外可见光分光光度仪及电化学工作站等设备测量模拟酶结构变化、酶促动力学参数以及电化学特性;在50 mmol/L Na2HPO4-NaH2PO4缓冲溶液(pH 10.0)中,测得模拟酶米氏常数Km、催化速率及催化效率分别为2.5 μmol/L、0.069 s-1、0.028±0.005(μmol/L)-1s-1,其催化效率为天然辣根过氧化物酶的39%±5%,该模拟酶具有很好的稳定性和活性,能够替代辣根过氧化物酶.该模拟酶可以固定在功能化多壁纳米管与纳米金复合材料修饰玻碳电极上,在50 mmol/L Na2HPO4-NaH2PO4缓冲溶液(pH 7.0)中测得模拟酶与电极之间电子迁移速度常数k8为0.65 s-1.利用循环伏安法测量模拟酶催化浓度为0.02~10 nmol/L的H2O2,其阴极峰电流随H2O2浓度增加而增大,并且得出模拟酶催化H2O2检出限为0.05 nmol/L,电化学表观米氏常数Kmapp为2.3×10-10 mol/L.  相似文献

L-谷氨酸消旋化反应对于制备光学纯D-谷氨酸具有重要的意义。本文利用532 nm激光诱导荧光共聚焦检测式毛细管阵列电泳仪研究了L-谷氨酸基于席夫碱中间物的消旋化反应,采用羧基四甲基罗丹明琥珀酰亚胺酯(TAMRA)荧光探针标记消旋化产物后,以含有2 mmol/L环糊精(β-CD)的100 mmol/L Tris-硼酸缓冲液(pH 10)为分离电解质进行毛细管阵列电泳分析,在该条件下TAMRA标记的谷氨酸对映体可以得到基线拆分。详细考察了醛的种类及用量、羧酸溶剂的种类、反应体系的含水量对消旋化的影响,结果表明L-谷氨酸在含20%(体积分数)水的乙酸溶剂中,以水杨醛作为催化剂(水杨醛与L-谷氨酸的物质的量比为0.2)的优化条件下可以快速地被消旋化。  相似文献

建立了一种快速、高效测定天冬酰胺合成酶B(AS-B)酶活性的反相高效液相色谱法(RP-HPLC)。酶反应体系中的氨基酸经2,4-二硝基氟苯(DNFB)柱前衍生,通过RP-HPLC测定酶反应体系前后底物及产物的变化来分析酶的活性。采用的色谱柱为Agilent C18柱(250 mm×4.6 mm,5 μm),以50 mmol/L醋酸钠缓冲液(pH 6.2)-乙腈(体积比为15:85)为流动相,流速为1.0 mL/min,柱温为30 ℃,检测波长365 nm,于6 min内实现了各组分的基线分离。通过该方法测定反应动力学参数进行AS-B的抑制定量分析。将已知AS-B抑制剂L-谷氨酸-γ-甲酯作用于酶反应体系,测得的抑制剂的抑制常数与文献值相接近,证明该方法可用于AS-B抑制剂的筛选。  相似文献

A CZE with near‐infrared (NIR) LIF detection method has been developed for the analysis of six low molecular weight thiols including glutathione, homocysteine, cysteine, γ‐glutamylcysteine, cysteinylglycine, and N‐acetylcysteine. For this purpose, a new NIR fluorescent probe, 1,7‐dimethyl‐3,5‐distyryl‐8‐phenyl‐(4'‐iodoacetamido)difluoroboradiaza‐s‐indacene was utilized as the labeling reagent, whose excitation wavelength matches the commercially available NIR laser line of 635 nm. The optimum procedure included a derivatization step of the free thiols at 45°C for 25 min and CZE analysis conducted within 14 min in the running buffer containing 16 mmol/L pH 7.0 sodium citrate and 60% v/v ACN. The LODs (S/N = 3) ranged from 0.11 nmol/L for N‐acetylcysteine to 0.31 nmol/L for γ‐glutamylcysteine, which are better than or comparable to those reported with other derivatization‐based CE‐LIF methods. As the first trial of NIR CE‐LIF method for thiol determination, the practical application of the proposed method has been validated by detecting thiols in cucumber and tomato samples with recoveries of 96.5–104.3%.  相似文献

A rapid and robust CE method using a long wavelength fluorescent reagent 1,7‐dimethyl‐3,5‐distyryl‐8‐phenyl‐(2‐maleimide)difluoroboradiaza‐s‐indacene as the labeling reagent has been developed for the simultaneous determination of thiols, including glutathione, cysteine, homocysteine, N‐acetylcysteine, cysteinylglycine, and penicillamine. The derivatization reaction was carried out in 14 mmol/L pH 8.5 borate buffer at 30°C for 6 min and the labeled thiols derivatives were separated with the running buffer containing 30 mmol/L pH 7.4 phosphate, 30% v/v acetonitrile and 8 mmol/L SDS within 12 min. Detection limits ranged from 0.4 to 2.4 nmol/L. To demonstrate the capability of this method, it was applied to the analysis of thiols in human urine with recoveries of 92.4–105.6%. The derivatization reaction was much faster at milder conditions, and the analysis was rapider. Moreover, with excitation wavelength at long wavelength region, background interference from samples was reduced effectively. The present method seems to be a potential choice for quantifying thiols in human urine.  相似文献

建立了亲水作用色谱(HILIC)测定组织中全基因组DNA甲基化水平的方法。采用苯酚-氯仿提取组织中的DNA,提取的DNA用88%甲酸在140 ℃下裂解,经N2吹干后,加乙腈-水(9:1, v/v)溶解,用Waters BEH HILIC柱进行分离,在277 nm波长下检测胞嘧啶(Cyt)及5-甲基胞嘧啶(5-mCyt)含量。结果表明,以乙腈-10 mmol/L甲酸铵溶液(94:6, v/v)为流动相,流速为0.5 mL/min, Cyt与5-mCyt分离较好,保留时间分别为2.6与3.1 min。胞嘧啶的线性范围为1~900 μmol/L,相关系数为0.9999; 5-甲基胞嘧啶的线性范围为1~64 μmol/L,相关系数为0.9998。胞嘧啶和5-甲基胞嘧啶的检出限为54 nmol/L(柱中为0.54 pmol),定量限为250 nmol/L(柱中为2.5 pmol);在5~900 μmol/L的添加水平下,胞嘧啶和5-甲基胞嘧啶的平均加标回收率为94.7%~100.5%,相对标准偏差小于1.48%。用该方法检测了结肠癌组织中DNA甲基化水平,结果显示该癌组织中全基因组的DNA甲基化均值为4.0%。该方法快速、简单,稳定性好,灵敏度较高,能满足全基因组DNA甲基化的检测要求。  相似文献

A novel near-infrared (NIR) cyanine 1-(epsilon-succinimidyl-hexanoate)-1'-methyl-3,3,3',3'-tetramethyl-indocarbocyanine-5,5'-disulfonate potassium (MeCy5-OSu) has been developed in our laboratory. Simultaneous determination of MeCy5-OSu-derivatized polyamines spermine (Spm), spermidine (Spd), cadaverine (Cad), and putrescine (Put) based on the separation by CE combined with diode LIF detection has been accomplished. The highest derivatization efficiency was achieved in 0.2 mol/L borate buffer (pH 8.8) for 20 min at 25 degrees C. Polyamine derivatives were separated within 14 min in the phosphate running buffer (pH 3) containing 50 mmol/L phosphoric acid, 40 mmol/L SDS, and 35% methanol v/v. Linearity of response was obtained in the range of 10-200 nmol/L. The detection limits (S/N = 3) for Spm, Spd, Cad, and Put were 0.8, 1, 3, and 2 nmol/L, respectively. The proposed method has been successfully applied to the analysis of polyamines in erythrocytes of two healthy persons and one cancer patient. Average recoveries for erythrocyte samples were 93.6-106% and coefficients of variation ranged from 1.8 to 5.4%. The analysis of polyamines in erythrocytes can be used for studying the relationship between their changes and the carcinogenesis process involved in erythrocytes.  相似文献

建立了柏子养心丸(Baizi Yangxin Wan, BZYXW)毛细管区带电泳指纹图谱(CEFP)。采用三棱柱优化法优化背景电解质(BGE),以色谱指纹图谱分离量指数(RF)为实验条件优化的目标函数,最终确定BGE为50 mmol/L硼砂-50 mmol/L磷酸氢二钠-200 mmol/L硼酸-150 mmol/L碳酸氢钠(体积比为7:7:1:1,含4%乙腈,pH 9.70)。在其他优化的毛细管电泳条件为紫外检测波长228 nm,运行电压12 kV,重力进样25 s (高度14 cm)条件下,采用未涂层石英毛细管(70 cm(有效分离长度57 cm)×75 μm)分离,以阿魏酸峰为参照,确定了17个共有指纹峰。对样品聚类分析后用其中9批生成对照CEFP(RCEFP),以其为标准,用系统指纹定量法鉴别出12批BZYXW质量为: 3批好,1批良好,3批中,1批一般,4批劣。三棱柱优化法为BGE选择提供了重要参考,建立的BZYXW-CEFP精密度好、重现性高,可用于BZYXW的质量控制。  相似文献

A simple, rapid, and accurate method for the separation and determination of physcion, chrysophanol, aloe-emodin, and emodin in Rhubarb, Juemingzi, and Chinese herbal preparations was developed by combination of flow injection-capillary zone electrophoresis for the first time. The analysis was carried out using an unmodified fused-silica capillary (75 mm x 50 microm ID x 375 microm OD, effective separation length of 48 mm) and direct ultraviolet detection at 254 nm. By a series of optimization, the sample solvent consisted of NaOH (100 mmol/L) and ACN (1:1 v/v), and a running buffer composed of 15 mmol/L sodium borate - 12.5 mmol/L sodium dihydrogen phosphate - 42% v/v ACN (pH 10.1) was applied for the separation of the four anthraquinones. The separation was rapid and highly reproducible, with complete resolution of all four compounds within 6 min. The sample throughput rate could reach up to 12 per h. The repeatability (defined as relative standard deviation) was 4.45, 4.44, 4.34, 0.61% with peak height evaluation and 1.62, 0.89, 2.49, 2.19% with peak area evaluation for physcion, chrysophanol, aloe-emodin, and emodin, respectively.  相似文献

建立了体外直接测定血管紧张素转化酶抑制剂活性的高效液相色谱分析方法。以马尿酰-组氨酰-亮氨酸为反应底物,血管紧张素转化酶为催化剂,反应所生成的马尿酸为测定指标,未加酶抑制剂的反应为空白对照。使用ZORBAX SB-C18色谱柱(4.6 mm i.d.×150 mm,填料粒径5 μm),柱温25 ℃,流动相为乙腈-超纯水(体积比为25∶75,各含0.05%(体积分数)三氟乙酸及0.1%(体积分数)三乙胺),流速0.5 mL/min,检测波长228 nm。在马尿酸浓度为0.005~1.000 mmol/L时,马尿酸浓度与其峰面积呈良好的线性关系(r=0.9999),最小检测限为0.50 μmol/L;该方法对马尿酸的回收率为99.48%~105.64%,相对标准偏差(RSD)为2.20%(n=6)。该方法可适用于血管紧张素转化酶抑制剂活性的体外测定,具有操作简便、精密度和准确性高的特点,为降血压药物的研制提供了方便可靠的检测手段。  相似文献

The aim of this work was to develop a fast method using capillary electrophoresis for the determination of creatinine in human urine samples. The pH and constituents of the background electrolyte were selected by inspection of effective mobility of creatinine and candidate urine interferents versus pH curves. The tendency of the analyte to undergo electromigration dispersion and the buffer capacity were evaluated by the Peakmaster software and considered in the optimization of the background electrolyte, composed by 10 mmol L(-1) tris(hydroxymethyl)aminomethane and 20 mmol L(-1) 2-hydroxyisobutyric acid (HIBA) at pH 3.93. Separation was conducted in a fused-silica capillary (32 cm total length and 8.5 cm effective length, 50 microm I.D.), with short-end injection configuration and direct UV detection at 215 nm. The migration time of creatinine was only 22s. A few figures of merit of the method are as follows: good linearity in the concentration interval of 5-70 mg L(-1) (R(2)>0.99), limit of detection of 0.5 mg L(-1), inter-day precision better than 2.7% (n=9) and recovery in the range 99.0-103.7% at three concentration levels (50, 100 and 150 mg L(-1)). Urine samples were prepared by deproteination with acetonitrile (1:3 sample:acetonitrile, v/v), centrifugation and dilution of a deproteinated aliquot with 12.5 mmol L(-1) HIBA (1:4, v/v). Creatinine concentrations between 489 and 1063 mg L(-1) were obtained in the urine of four healthy volunteers.  相似文献

采用毛细管区带电泳法建立了逍遥丸(Xiaoyao Pill, XYP)的毛细管电泳指纹图谱(CEFP)。运用正方形优化法,以色谱指纹图谱分离量指数(RF)为优化的目标函数,对建立指纹图谱的实验条件进行了优化,确定了最佳背景电解质(BGE)溶液50 mmol/L硼砂-50 mmol/L磷酸氢二钠-150 mmol/L磷酸二氢钠-50 mmol/L碳酸氢钠(1:1:1:5, v/v/v/v; pH 7.40)、紫外检测波长228 nm、运行电压12 kV、重力进样25 s (高度14 cm)的分离检测条件。采用未涂层石英毛细管(70 cm×75 μm,有效分离长度57 cm)分离,以咖啡酸色谱峰为参照,确定13批逍遥丸样品的21个共有指纹峰。通过聚类分析确定用其中10批样品生成对照CEFP,以此为标准用系统指纹定量法鉴别13批逍遥丸的质量,结果显示: S3号样品的化学成分数量和分布比例不合格,S10和S12号样品含量明显偏高,其余批次质量均合格。所建立的正方形优化法操作简便,适用于中药的毛细管区带电泳BGE的选择;所建立的逍遥丸CEFP具有较好的精密度和重现性,可以为逍遥丸的质量控制提供新的参考。  相似文献

采用改良Stöber法制备420 nm亚微米单分散二氧化硅微球,采用C18硅烷化修饰后装填成毛细管色谱柱。采用该色谱柱,在加压毛细管电色谱平台上成功地实现了3对手性三唑类农药烯效唑、烯唑醇和丙环唑的同时拆分和分离。考察了各因素对手性分离效果的影响,优化后的色谱条件为:流动相为乙腈-20 mmol/L磷酸盐缓冲液(pH=6.8)(45:55,v/v),其中缓冲液中含20 mmol/L羟丙基-γ-环糊精(HP-γ-CD);泵流速为0.04 mL/min;施加电压-9.4 kV;检测波长220 nm。在上述条件下,烯效唑、烯唑醇和丙环唑3种对映体同时得到拆分和分离,相邻两峰之间的分离度依次为4.20、12.9、4.41、4.09、1.70,分离时间仅为12 min,柱效最高达到310000 plates/m。该研究为手性三唑类农药的同时分离提供了新的分离分析思路。  相似文献

A simple, reliable, and reproducible method for separation and determination of five enkephalin-related peptides based on CE with amperometric detection (AD) is described in this paper. A potential of 1.0 V was applied to the carbon disk electrode, which was used as a working electrode in this system. At 15 kV of applied voltage, the five compounds were separated within 18 min in a 20 mmol/L phosphate buffer solution (PBS, pH 7.6) including 2.5% methanol v/v. LOD for five enkephalins were ranged from 6.31 to 54.3 nmol/L. The method was applied successfully to determine the five compounds added in the human cerebrospinal fluid (CSF) and the recoveries were in the range of 95.8-98.2%.  相似文献
