Ethyleneglycol derivatives of 2-methyl (and ethyl)-3-hydroxy-4-pyridinone were synthesized by a modified Hilbert-Johnson reaction. The synthesis proceedes by reaction of O-protected 2-alkyl-3-hydroxy-4-pyridinone with hexamethyldisilazane in the presence of chlorotrimethylsilane followed by trimethylsilyl trifluoromethanesulfonate catalized alkylation with benzyloxy-ethoxymethylchloride in dichloroethane and deprotection by hydrogenation. The overall yield was 87%. This method provides a useful way to produce oligo- or polyethyleneglycol substituted hydroxypyridinones.  相似文献

A series of novel dihydro-alkylthio-benzyloxopyrimidine (S-DABO) and 1-[(2-hydroxyethoxy) methyl]-6-(phenylthio)thymine (HEPT) analogs bearing a (benzo[d]thiazol-2-yl)methyl moiety at the C6 position of the pyrimidine core have been synthesized. 5-Allyl-6-{(benzo[d]thiazol-2-yl)methyl}-2-thiouracil and 5-allyl-6-{(benzo-[d]thiazol-2-yl)methyl}uracil were alkylated to give, respectively, S2- and N1- ethoxymethyl and -methylthiomethyl uracil derivatives. 5-Allyl-6-[(benzo[d]thiazol-2-yl)methyl]-2-thiouracil was also alkylated by S2 with methyl bromoacetate and hydrolyzed to the corresponding acid.  相似文献

A simple quantitative structure-activity relationship (QSAR) method of analysis used to predict biological activity for congeneric series of compounds is reported. This method is based on the application of bilinear or multilinear partial least squares regression to a data set, which is a binary matrix representing the substituents of a framework. It is appraised here to a series of (S)-N-[(1-ethyl-2-pyrrolidinyl)methyl]-6-methoxybenzamides, compounds with affinity towards the dopamine D2 receptor subtype and showed high predictive ability, even when compared to a refined three-dimensional (3D) approach.  相似文献

2-[(2-Hydroxyphenylimino)methyl]phenol (H2L1) and 1-[(2-hydroxyphenylimino)methyl]naphthalen-2-ol (H2L2) reacted with copper(II) acetate hydrate and sulfanilamide (Sf1), sulfathiazole (Sf2), sulfaethidole (Sf3), sulfadiazine (Sf4), and sulfadimidine (Sf5) in ethanol to give mixed-ligand copper chelates with the composition Cu(Sf1–5)(L1–2) · n H2O (n = 1, 2). All these complexes are monomeric. Salicylaldehyde imines (H2L1 and H2L2) behave as doubly deprotonated tridentate O,N,O ligands, whereas sulfanilamides (Sf1–5) are unidentate ligands. Thermolysis of the synthesized complexes includes dehydration at 70–90°C, followed by complete thermal decomposition (290–380°C). The complexes [Cu(Sf1)(L1)] · 2H2O and [Cu(Sf3)(L1)] · H2O at a concentration of 10−4 M inhibited growth and reproduction of 100% of human myeloid leukemia cells (HL-60). The inhibitory effect was 90 and 75%, respectively, at a concentration of 10−5 M, whereas no antitumor activity was observed at a concentration of 10−6 M.  相似文献

The crystal structures of (μ-4,4-bipyridine)-di(nitrato-1-[(2-hydroxyethylimino)methyl]naphthalen-2-olocopper (I) and catena-di(μ-4,4’-bipyridine)di(μ-4,4’-bipyridine)-di(nitrato-2-[2-(hydroxyethylimino) methyl]phenolocopper)diaquacopper(II) nitrate (II) were determined. In the crystal of I, each of the two copper atoms coordinates a singly deprotonated tridentate azomethine molecule, a nitrate ion, and bipyridine, which functions as a bridge between the central atoms. The copper coordination polyhedron is a slightly distorted tetragonal pyramid with the base formed by the imine and bipyridine nitrogen atoms and the phenol and alcohol oxygen atoms. The axial site in the pyramid is occupied by the oxygen atom of the monodentate nitrate groups. In the trinuclear structure II with C2 crystal chemical symmetry, the terminal coordination unit is composed through copper coordination of monodeprotonated 2-[2-(hydroxyethylimino)methyl]phenol, bipyridine, and the nitrate anion. In the crystal, the trinuclear molecules form infinite ribbons along the z axis in which the pyridine molecules perform the bridging function. The central copper atom has an octahedral configuration formed by the nitrogen atoms of four 4,4’-bipyridine molecules and oxygen of two water molecules.  相似文献

研究了以2-羟基-4-甲氧基苯甲醛和2-氨基-6-甲基吡啶为原料合成新型席夫碱化合物5-甲氧基-2-[(E)-(6-甲基吡啶-2-亚氨基)甲基]苯酚的方法。当2-羟基-4-甲氧基苯甲醛与2-氨基-6-甲基吡啶摩尔比为1∶1.6,反应时间为6 h,反应温度为75℃时,反应产率最高。采用元素分析、UV-Vis、IR、1H NMR、X-射线单晶衍射等方法进行结构表征。该化合物为单斜晶系,空间群P21/c,a=1.1886(3)nm,b=0.65948(16)nm,c=1.6897(4)nm,β=108.505(3)°,V=1.2560(5)nm3,Dc=1.281 g·cm-3,Z=4,F(000)=512,μ=0.087 mm-1,R1=0.0477,wR2=0.1342。通过π···π堆积和分子内氢键O2-H2···N1、C8-H8A···N2形成较稳定的晶体结构,并具有蓝色荧光。  相似文献

The solid phase FT-IR and FT-Raman spectra of 1-[6-(4-chlorophenyl)-1-[(6-chloropyridin-3-yl)methyl]-2-[(6-chloropyridin-3-yl)methylsulfanyl]-4-methyl-1,6-dihydropyrimidin-5-yl]ethanone (C25H21Cl3N4OS) were recorded in the region 4000-400 and 3500-100cm(-1), respectively. The vibrational spectra have been computed using density functional theory (B3LYP) and ab initio molecular orbital calculation (HF) with 6-31G(d, p) basis sets. A close agreement was achieved between the observed and calculated frequency by employing normal coordinate calculations. The observed and simulated spectra were found to be well comparable.  相似文献

1-[(Dimethylamino)methyl]pyrrole from Trimethyl (1-pyrrolyl)ammonium Ion Trimethyl (1-pyrrolyl)ammonium iodide ( 5a ) and the corresponding p-toluene-sulfonate 5b are transformed by strong bases into 1-[(dimethylamino)methyl]-pyrrole ( 9 ), i.e. into a N-Mannich base, a type of compound novel in the pyrrole series. In this reaction, which is very fast in DMSO, the cation of compounds 5 is deprotonated to form the nitrogen ylide 6 . The latter undergoes a Stevens-type rearrangement to 9 . Several facts, namely the negative outcome of a cross-reaction experiment with 3,4-dimethylpyrrole and of an attempt to obtain 9 from pyrrole and dimethyl (methylidene)ammonium iodide in the presence of one equivalent of sodium methoxide, as well as unsuccessful CIDNP studies point to a rearrangement mechanism via the contact ion pair 12 .  相似文献
