This study was designed to examine the interaction of histamine H2-receptor antagonist drug ranitidine (RTN) with human serum albumin by multi-spectroscopic methods. The experimental results showed the involvement of dynamic quenching mechanism which was further confirmed by lifetime spectral studies. The binding constants (K a) at three temperatures (288, 298, and 308 K) were 2.058 ± 0.020, 4.160 ± 0.010 and 6.801 ± 0.011 × 104 dm3 mol?1, respectively, and the number of binding sites (m) were 1.169, respectively; thermodynamic parameters ΔH 0 (44.152 ± 0.047 kJ mol?1), ΔG 0 (?26.214 ± 0.040 kJ mol?1), and ΔS 0 (236.130 ± 0.025 J K?1 mol?1) were calculated. The distance r between donor and acceptor was obtained (r = 3.40 nm) according to the Förster theory of non-radiative energy transfer. Synchronous fluorescence, CD, AFM and 3D fluorescence spectral results revealed the changes in secondary structure of the protein upon interaction with RTN. A molecular modeling study further confirmed the binding mode obtained by the experimental studies.  相似文献

Dried duckweed (Landoltia punctata) powder is applied to remove IO3? from aqueous solutions under various conditions. The results indicate the Kd is 150 ml g?1 under general conditions. The sorption kinetics follow the pseudo-second-order equation, and the isotherm is well described by the Freundlich model. Hydroxyl and carbonyl groups contribute to IO3? sorption by ion-exchange, electrostatic attraction and redox reactions. Spectroscopic analyses prove that IO3? is reduced to I2 and I? by hydroxyl groups. These results demonstrate that duckweed (Landoltia punctata) is a promising biosorbent for environmental remediation of radioactive iodine pollution.  相似文献

The application of newly synthesized Fe3O4/TiO2–SiO2 that is modified with zinc (FTSZ) as a sorbent, for the removal of arsenic from contaminated water has been investigated in the present study. SEM, FTIR, XRD, BET, Zeta potential sizer (ξ) analyses are used to determine the sorbent characterization. The effect of the operational parameters such as initial pH, initial concentration, and the contact time were studied. In addition, the equilibrium behavior of FTSZ in As(III) removal was investigated in the temperature range of 20–40 °C. The results showed that the equilibrium data were fitted well with Langmuir than Freundlich isotherm model. The maximum monolayer adsorption capacity estimated by Langmuir isotherm was 24.010 mg g?1. Thermodynamic parameters, ?H°, ?S° and ?G° were also calculated from graphical interpretation of the experimental data. Standard heats of sorption (?H°) were found to be endothermic and ?S° values were calculated to be positive for the sorption of As(III) onto the adsorbent.  相似文献

In the presented study the binding of Cs, Co, and Tc with hardened cement paste (HCP) under hyperalkaline conditions, with respect to sorption reversibility, has been investigated using batch-type experiments. The Cs, Co, and Tc sorption kinetics were determined and the distribution coefficients K d , representing the experimental systems, were obtained. The reversibility of the element sorption has been discussed in accordance with the published data. It was shown that the sorption kinetics was rapid and the equilibrium was attained within 1–5 days. The K d values clearly demonstrated the sorption dependence on both the chemical composition of the HCP and the element speciation.  相似文献

Laser electrospray mass spectrometry (LEMS) measurement of the dissociation constant (Kd) for hen egg white lysozyme (HEWL) and N,N′,N″-triacetylchitotriose (NAG3) revealed an apparent Kd value of 313.2?±?25.9 μM for the ligand titration method. Similar measurements for N,N′,N″,N″’-tetraacetylchitotetraose (NAG4) revealed an apparent Kd of 249.3?±?13.6 μM. An electrospray ionization mass spectrometry (ESI-MS) experiment determined a Kd value of 9.8?±?0.6 μM. In a second LEMS approach, a calibrated measurement was used to determine a Kd value of 6.8?±?1.5 μM for NAG3. The capture efficiency of LEMS was measured to be 3.6?±?1.8% and is defined as the fraction of LEMS sample detected after merging with the ESI plume. When the dilution is factored into the ligand titration measurement, the adjusted Kd value was 11.3 μM for NAG3 and 9.0 μM for NAG4. The calibration method for measuring Kd developed in this study can be applied to solutions containing unknown analyte concentrations.

Adsorption of the gadolinium from H2O and HCl solutions on the ion-exchange resin C100 is investigated. The experiments were carried out by varying the acidity of the liquid phase, the amount of sorbent, and the temperature. The maximal sorption of the ions Gd3+ is observed from the solution 0–0.2 M HCl under optimal conditions, the sorption reaches more than 99.5%. Sorption of Gd3+ on C100 from H2O solution occurs most intensively during the first 3 min then for 30 min the system smoothly comes to equilibrium. The maximal sorption capacity of the resin C100 amounted to 1.2 ± 0.1 mmol g?1. The thermodynamic parameters of sorption: ΔG = ? 24.20 kJ mol?1, ΔS = ? 90.27 J mol?1 K?1, ?H = ? 50.93 kJ mol?1 were evaluated. It is shown that the sorption of gadolinium on the ion-exchange resin C100 is described by models of kinetically pseudo-first and pseudo-second order. It is established that the Gd3+ sorption on the C100 resin is reversible second order chemical reaction.  相似文献

From three cell-associated β-xylosidases produced by Aureobasidium pullulans CBS 135684, the principal enzyme was enriched to apparent homogeneity and found to be active at high temperatures (60–70 °C) over a pH range of 5–9 with a specific activity of 163.3 units (U) mg?1. The enzyme was thermostable, retaining over 80% of its initial activity after a 12-h incubation at 60 °C, with half-lives of 38, 22, and 10 h at 60, 65, and 70 °C, respectively. Moreover, it was tolerant to xylose inhibition with a K i value of 18 mM. The K m and V max values against p-nitrophenyl-β-d-xylopyranoside were 5.57 ± 0.27 mM and 137.0 ± 4.8 μmol min?1 mg?1 protein, respectively. When combining this β-xylosidase with xylanase from the same A. pullulans strain, the rate of black liquor xylan hydrolysis was significantly improved by up to 1.6-fold. The maximum xylose yield (0.812 ± 0.015 g g?1 dry weight) was obtained from a reaction mixture containing 10% (w/v) black liquor xylan, 6 U g?1 β-xylosidase and 16 U g?1 xylanase after incubation for 4 h at 70 °C and pH 6.0.  相似文献

A novel peroxy group-containing silane coupling agent was synthesized and anchored on the surface of titanium dioxide nanoparticles (nano-TiO2) to form an immobilized-initiator-modified nano-TiO2 species. In this study, the kinetic parameters of the peroxy group-containing silane were tested and assessed using DSC. The pre-exponential factor (Ad) was 8.973?×?108 and the activation energy (Ea) was 80.736 kJ mol?1. Moreover, the empirical Arrhenius equation was determined to be ln Kd?=???80.736/RT?+?ln(8.973?×?108). To obtain continuous polymers, acrylonitrile (AN) and methyl methacrylate (MMA) were polymerized using the novel peroxy group-containing silane and FeSO4 as an initiator system. The number average molecular weights (Mn of PAN?=?3×104 and Mn of PMMA?=?1.4?×?105) and polydispersity indexes (PDI of PAN?=?2.76 and PDI of PMMA?=?1.65) were determined by GPC. It was suggested that the redox initiation system can generate highly reactive species on the surfaces of inorganic nanoparticles. The nano-TiO2-grafted polymers were successfully obtained.  相似文献

p-Phenylenediamine was oxidized with p-benzoquinone in the aqueous solutions of methanesulfonic acid (MSA). The conductivity of the products increased with increasing concentration of MSA from 1.5?×?10?12 S cm?1 in 0.1 M MSA up to 3.4?×?10?4 S cm?1 in 5 M MSA. The low-molecular-weight products are basically composed of one p-benzoquinone and two p-phenylenediamine molecules. Their molecular structure is discussed on the basis of mass, Fourier-transform infrared, Raman, NMR and electron paramagnetic resonance (EPR) spectroscopies. The formation of 2,5-di(p-phenylenediamine)-p-benzoquinone protonated with methanesulfonic acid best complies with the information provided by spectroscopic techniques. Its conversion to hydroquinone tautomer explains the formation of unpaired spins observed by EPR and their potential contribution to the conduction.  相似文献

A series of [1,2,4]triazolo[4,3-a]pyridine derivatives bearing a sulfide substructure was designed, synthesized and characterized via 1H·NMR, 13C·NMR, IR and elemental analyses. Bioassay Results indicated some of the derivatives displayed good fungicidal activity on Rhizoctonia cerealis, moderated insecticidal activity against Plutella xylostella and good insecticidal activity on Helicoverpa armigera. The inhibitory effects of compounds 4g and 4u against Rhizotonia cerealis were 70.9% at 50 μg mL?1; the IC50 values of compounds 4d and 4s against Plutella xylostella were 43.87 and 50.75 μg mL?1, respectively. And the IC50 values of compounds 4d, 4q, and 4s on Helicoverpa armigera were 58.3, 77.14 and 65.31 μg mL?1, respectively, which were better than that of commercial chlorpyrifos (103.77 μg mL?1).  相似文献

Two cis-dioxomolybdenum(VI) complexes [MoO2L] (L: L 1, 2 and L: L 2, 3) in a phenol-based sterically encumbered N2O2 ligand environment have been synthesized, and their crystallographic characterizations are reported. The orange crystals of 2 are monoclinic, space group P21/a with unit cell dimensions as a=16.2407(17) Å, b=7.2857(8) Å, c=18.400(2) Å, β=98.002(9)°, Z=4, and d cal=1.486 g cm?3. The light orange crystals of 3, however, are orthorhombic, space group, Pbcn, with unit cell dimensions a=8.3110(12) Å, b=12.637(3) Å, c=34.673(5) Å, Z=4, and d cal=1.187 g cm?3. The structures were refined by a full-matrix least-squares procedure on F 2 to a final R=0.046 (0.055 for 3) using 4944 (3677) all independent data. In both the cases, the Mo atom exists in a distorted octahedral geometry defined by a N2O4 donor set, which features a cis-Mo(–O)2 and a trans-Mo(OPh)2 arrangement. Compound 2 undergoes a quasireversible one-electron reduction at ?1.3 V vs Ag/AgCl reference due to MoVIO2/MoVO2 electron transfer and thus providing a rare example of steric solution to the comproportionation–dimerization problem encountered frequently in the development of valid biomimetic models for the active sites of oxomolybdenum enzymes.  相似文献

The interaction between 2,4-dichlorophenol (DCP) and bovine serum albumin (BSA) was investigated by fluorescence spectroscopy combined with UV-vis absorption and circular dichroism (CD) spectroscopy under simulative physiological conditions. The experiment results show that the fluorescence intensity of BSA is dramatically decreased owing to the formation of a DCP–BSA complex. The corresponding effective quenching constants (K a) between DCP and BSA at four different temperatures (292, 298, 304 and 310 K) were determined to be 10.08×104, 9.082×104, 8.177×104, and 7.260×104 L?mol?1, respectively. The thermodynamics parameters enthalpy change (ΔH) and entropy change (ΔS) were calculated to be ?13.64 kJ?mol?1 and 49.08 J?mol?1?K?1, which suggested that hydrophobic interaction was the predominant intermolecular force. Site marker competitive experiments indicated that the binding of DCP to BSA primarily takes place in subdomain IIA. The binding distance (r) between DCP and the tryptophan residue of BSA ias 4.09 nm according to Förster’s theory of non-radioactive energy transfer. The conformational investigation demonstrated that the presence of DCP decreased the α-helical content of BSA and induced a slight unfolding of the polypeptides of protein, which confirmed the occurrence some micro environmental and conformational changes of BSA molecules.  相似文献

Preliminary testing of dissolved air flotation (DAF) for wastewater treatment is presented. A combined coagulation-flocculation/DAF column system is used to remove oil and 60Co from nuclear industry wastewater. In this work, operational conditions and coagulant/flocculant concentrations are optimized by varying pH. Determinations of air-solids ratio (G/S), retention time (θ), pressure (P), volume of depressurized air–water mixture (V), turbidity and 60Co concentrations are reported. The effect of the treatment on the efficiency of separation of oily residues is also discussed. The results establish that the coagulant/flocculant system, formed by a modified polyamine (25 mgL?1) and a slightly cationic polyacrylamide (1.5 mgL?1), under specific operational conditions (pH = 7, mixing intensity Im1 = 300 s?1 and Im2 = 30 s?1), allowed the destabilization of colloidal matter, resulting in resistant flocs. It was concluded that by using G/S = 0.3, θ = 15 min, P = 620 kPa and V = 0.0012 m3, the greatest percentage removals of oil, turbidity, total cobalt and 60Co were obtained. These preliminary results then show that dissolved air flotation represents a good alternative for treatment of nuclear industry wastewater contaminated with radionuclides.  相似文献

Adenylate deaminase (AMPD, EC 3.5.4.6) is an aminohydrolase that widely used in the food and medicine industries. In this study, the gene encoding Aspergillus oryzae AMPD was cloned and expressed in Escherichia coli. Induction with 0.75 mM isopropyl β-d-l-thiogalactopyranoside resulted in an enzyme activity of 1773.9 U/mL. Recombinant AMPD was purified to electrophoretic homogeneity using nickel affinity chromatography, and its molecular weight was calculated as 78.6 kDa. Purified AMPD exhibited maximal activity at 35 °C, pH 6.0 and 30 mM K+, with apparent K m and V max values of 2.7 × 10?4 M and 77.5 μmol/mg/min under these conditions. HPLC revealed that recombinant AMPD could effectively catalyse the synthesis of inosine-5′-monophosphate (IMP) with minimal by-products, indicating high specificity and suggesting that it could prove useful for IMP production.  相似文献

Molecular properties are computed as responses to perturbations (energy derivatives) in coupled-cluster (CC)/many-body perturbation theory (MBPT) models. Here, the CC/MBPT energy derivative with respect to a general two-electron (2-e) perturbation is assembled from gradient theory for 2-e property evaluation, including the electron repulsion energy. The correlation energy (?E) is shown to be the sum of response kinetic (?T), electron–nuclear attraction (?V), and electron repulsion (?V ee ) energies. Thus, evaluation of total V ee for energy component analysis is simple: For total energy (E), total 1-e responses T and V, and nuclear–nuclear repulsion energy (V NN ), V ee  = E ? V NN  ? T ? V is the true 2-e response value. Component energy analysis is illustrated in an assessment of steric repulsion in ethane’s rotational barrier. Earlier SCF-based results (Bader et al. in J Am Chem Soc 112:6530, 1990) are corroborated: The higher-energy eclipsed geometry is favored versus staggered in the two repulsion energies (V NN and V ee ), while decisively disfavored in electron–nuclear attraction energy (V). Our best quality calculations (CCSD/cc-pVQZ) attain practical Virial Theorem compliance (i.e., agreement among the kinetic energy, potential energy, and total energy representations) in assigning 2.70 ± 0.06 to the barrier height; ?195.80 kcal/mol is assigned to the drop in “steric” repulsion upon going to the eclipsed geometry. Steric repulsion is not responsible for any fraction of the ~3 kcal/mol barrier.  相似文献

The apparent protonation constant of an amino butanol (AB) and its carbamate formation constant in the CO2–H2O–AB system were determined at T = 298.15–328.15 K, and at various ionic strengths up to 0.2 mol·L?1 by potentiometric titration. The Debye–Hückel equation was used to extrapolate the protonation constants to zero ionic strength. The variation of the thermodynamic equilibrium constant for carbamate formation with temperature was modeled according to the relationship of log10 K1 = 280.91/T ? 0.1105, while the temperature dependency of the amine protonation constant was correlated by log10 K2 = 1926.53/T + 2.9482. Van’t Hoff type plots of the pK values showed linear relationships indicating that the standard enthalpy changes of reaction are constant over this range of temperatures. Hence, our current findings are crucial for designing efficient unit operations involving separation of CO2 from natural or flue gases.  相似文献

Leachate samples from a sanitary landfill of Araraquara city and composting usine of Vila Leopoldina, São Paulo, Brazil were lyophilized to remove the water content. TG/DTG curves at different heating rates were recorded. The second step of the thermal decomposition of leachate from the Araraquara landfill (CB1), from the composting usine from Vila Leopoldina (CB2) from the organic phase extracted (FO) and aqueous phase (FA) were all kinetically evaluated using the non-isothermal method.By Flynn-Wall isoconversional method the following values were obtained: E=234±3.65 kJ mol?1 and logA=29.7±0.58 min?1 for CB1; E=129±1.66 kJ mol?1 and logA=11.8±0.10 min?1 for CB2; E=51.6±1.35 kJ mol?1 and logA=6.09±0.09 min?1 for FO and E=76.91±6.33 kJ mol?1 and logA=8.88±0.7 min?1 for FA with 95% confidence level. Applying the procedures of Málek and Koga, SB kinetic model (?esták-Berggren) is the most appropriate to describe the decomposition of CB1, CB2, FO and FA.  相似文献

The heat capacities of 1-butyl-3-methylimidazolium lactate ionic liquids ([C4mim][Lact]) were measured with a highly accurate automatic adiabatic calorimeter over the temperature range from 79 to 406 K. And the experimental values of molar heat capacities were fitted to a polynomial equation using least square method in the appropriate temperature ranges. The standard molar heat capacity was determined to be 1734.46?±?5.12 J K?1 mol?1 at 298.15 K. The molar enthalpy and molar entropy of the transition were determined to be 15.575?±?0.045 and 64.44?±?0.14 J K?1 mol?1. Other thermodynamic properties, such as (HT???H298.15) and (ST???S298.15), were also calculated. Furthermore, when the temperature reaches 241.87 K, the strongest peaks appeared by analysis of the heat capacity curve. This phenomenon could be explained from the interionic interaction, which is the hydrogen bond between the anions and cations.  相似文献

LnBaCuCoO5 + δ (Ln = Y, Dy) cuprocobaltites were prepared. Their unit cell parameters were determined and their thermal expansion, electrical conductivity (σ), and Seebeck coefficient (S) were studied in air in the range 300–1100 K. The compounds have tetragonal structures (space group P4/mmm). Their unit cell parameters are a = 0.3867(2) nm, c = 0.7550(7) nm, V = 112.9(2) × 10?3 nm3 for YBaCuCoO4.98; and a = 0.3872(2) nm, c = 0.7562(7) nm, V = 113.4(2) × 10?3 nm3 for DyBaCuCoO5.01. They are p-type semiconductors. The electrical conductivity of DyBaCuCoO5 + δ is slightly lower and its Seebeck coefficient is 1.5–2 times higher than the respective values for YBaCuCoO5 + δ apparently because of different electronic configurations of the rare-earth cations in LnBaCuCoO5 + δ (4d 0 for Y3+ and 4f 9 for Dy3+). Dilatometric measurements show that the LnBaCuCoO5 + δ phases in the range 300–1100 K do not experience structural phase transitions, and their linear thermal expansion coefficients (LTEC) are 14.3 × 10?6 K?1 for Ln = Y and 14.7 × 10?6 K?1 for Ln = Dy.  相似文献

Interaction of ammonia with H-MCM-22 zeolite (Si/Al = 24.5) was investigated by temperature-programmed desorption technique in order to obtain information on thermodynamics of the process. Average activation energy for desorption of ammonia from Brønsted acid sites of H-MCM-22 zeolite was estimated from the data obtained under conditions varying in heating rate and also flow rate of carrier gas. It resulted in value of E d = 127 kJ mol?1 for heat rate variation method, whereas flow rate variation led to E d value of 111 kJ mol?1. Obtained E d values are compared with those reported in the literature for other zeolitic materials and discussed in the broader context of zeolite acidity. Comparison of E d values estimated here for H-MCM-22 zeolite with corresponding data for other protonic zeolites shows that H-MCM-22 displays mediocre/lower activation energy for ammonia compared with other high-silica zeolites.  相似文献
