The palladium-catalyzed Suzuki-Miyaura reaction of 2,4-dichloropyrrolo[2,3-d]pyrimidine with aryl boronic acids has been studied. Pd(OAc)2/dicyclohexyl(2-biphenyl)phosphine/K3PO4 was found to be an efficient catalyst system to prepare 4-aryl-2-chloro- and 2,4-diarylpyrrolo[2,3-d]pyrimidines. Novel non-linear molecules consisting of a pyrrolo[2,3-d]pyrimidine core and aryl branches have been elucidated as blue light-emitters with fluorescence quantum yields ranging from 4% to 67% in THF solution. The impact of an electron-withdrawing t-BuOCO group attached to the pyrrole ring of pyrrolopyrimidine derivatives on optical properties is discussed.  相似文献

An improved methodology for the synthesis of substituted pyrido[2,3-d]pyrimidinediones from 6-aminouracils and the corresponding enaminone has been developed which provides high yields via an operationally simple process. The method has been extended to encompass a range of electron-rich enaminones, a substrate class that does not perform well under the standard conditions.  相似文献

N,N-Dimethyl-5-formylbarbituric acid 1 reacts with maleimide 2 and phenyl isocyanate/phenyl isothiocyanate 4 under microwave-assisted conditions in the solid phase to afford pyrano[2,3-d]pyrimidines 3 and oxazino[4,5-d]pyrimidines 5 in excellent yields. Under identical conditions, N,N-dimethyl-6-amino-5-formyluracil 6 reacts with 2 and 4 to give pyrido[2,3-d]pyrimidine derivative 7 and pyrimido[4,5-d]pyrimidines 8 in high yields.  相似文献

A convergent approach for a family of fluorescent nucleosides is described. It relies on thieno[3,2-d]pyrimidine-2,4(1H,3H)-dione that serves as a core heterocycle. This condensed pyrimidine is converted into an emissive pyrimidine nucleoside analogue by N-glycosylation and into an emissive purine nucleoside analogue by C-glycosidation at the thiophene's beta position. The design principles of this archetypical system are outlined together with the syntheses and photophysical properties of the resulting nucleosides.  相似文献

An effective approach to the new isoxazolo[3,4-d]thieno[2,3-b]pyridine system was provided by way of an intramolecular nitrone cycloaddition. The required nitrones were built in good yields starting from thiophene-2-carboxylic acids. The same skeleton was achieved in optically active form employing chiral nitrones derived from N-α-methylbenzyl- and the N-α-hydroxymethylbenzyl-hydroxylamines. The absolute configuration of the products was assigned by X-ray analysis.  相似文献

A novel I2-catalyzed one-pot multicomponent protocol for the synthesis of a variety of elusive furo[2,3-b]pyrrole and thieno[2,3-b]pyrrole libraries has been established. To date, cyclization among alkanone, hydrazine and 2-bromobenzofuran or 2-bromobenzo[b]thiophene has not been explored in one-pot. Thus, the proposed single step protocol provides a versatile alternative to existing routes for accessing useful furo[2,3-b]pyrrole and thieno[2,3-b]pyrrole libraries.  相似文献

The preparation of two pyrimido[4,5-c]isoquinoline-7,10-quinones from acylhydroquinones and 1,3-dimethyl-5-aminouracil and their cycloadditions with 1-trimethylsilyloxybutadiene and 1-dimethylamino-3-methyl-1-azabutadiene is described. The remarkable regiocontrol of these cycloadditions that yield stable 1:1 cycloadducts is discussed on the basis of steric interactions into the pyrimido[4,5-c]isoquinoline-7,10-quinones. The access to angucyclinone AB-pyridopyrimidine analogues from Diels-Alder adducts and preliminar evidences on their antitumour activities are also reported.  相似文献

We describe here a novel synthesis of benzo[b]naphtho[2,3-d]furan-6,11-diones based on the heteroannulation of 2-(2-bromophenyl)-3-hydroxy-1,4-naphthoquinones. The naphthoquinones were prepared from 3-(2-bromophenyl)naphthalen-2-ols, which were obtained by intramolecular aldol condensation of 2-[3-(2-bromophenyl)-2-oxo-propyl]benzaldehydes. Alternatively, benzo[b]naphtho[2,3-d]furan-6,11-diones were obtained more directly and efficiently by cyclization of 3-(2-bromophenyl)naphthalen-2-ols to benzo[b]naphtho[2,3-d]furans and oxidation of the resulting compounds. Furthermore, the first 6-oxabenzo[a]anthracen-5-one described was similarly obtained from 2-[3-(2-formylphenyl)-2-oxopropyl]benzoic acid and oxidized to 6-oxa-benzo[a]anthracene-5,7,12-trione.  相似文献

Three unique propeller-shaped helicenyl amines compounds: N,N-diphenyl-N-naphtho[2,1-b]thieno[2,3-b:3′,2′-d]dithiophene-5-yl-amine (1), N-phenyl-N,N-di(naphtho[2,1-b]thieno[2,3-b:3′,2′-d]dithiophene-5-yl)amine (2), and N,N,N-tri(naphtho[2,1-b]thieno[2,3-b:3′,2′-d]dithiophene-5-yl)amine (3) were efficiently synthesized by Wittig reaction and oxidative photocyclization. The crystal structures of 1, 2 and molecular configuration optimization (DFT-B3LYP/6-31+G(d)) of 3 reveal that the steric hindrance from the moiety of trithia[5]helicene effectively forces the nitrogen atom and the three bonded carbon atoms to coplanar and the interplanar angles of the facing terminal thiophene ring and benzene ring becoming larger when the helical arm increased from 1 to 3. Electrochemical properties and UV–vis absorption behaviors of 1, 2, 3 were primarily determined by the moiety of trithia[5]helicene.  相似文献

Sharpless epoxidation of 4-amino-3-methyl-5-styrylisoxazoles 1 resulted in the formation of 3-methyl-5-aryl-4H-pyrrolo[2,3-d]-isoxazoles 4 in a one-step reaction. The reaction initially involves epoxide formation, followed by ring-opening and cyclization. Finally dehydration leads to the title compounds. The pyrrolo[2,3-d]-isoxazoles 4 were also synthesized via an alternative procedure.  相似文献

The Suzuki-Miyaura reaction of methyl-5-bromo-8-(tosyloxy)-1,6-naphthyridine-7-carboxylate(5),with 2 equiv. of arylboronic acids gave diarylated product, 5,8–diaryl-1,6-naphthyridine-7-carboxylate(7), whereas 1 equiv. of arylboronic acid resulted in site-selective formation of 5-aryl-8-(tosyloxy)-1,6-naphthyridine-7-carboxylate(8). The reactions proceeded with excellent chemo-selectivity in favor of the bromide group. Likewise, one-pot reaction with completely different boronic acids by sequential addition produced 1,6-naphthyridine-7-carboxylates,(10) containing two different aryl groups at 5 and 8 positions.  相似文献

Indole-fused cyclic anhydrides earlier deemed unreactive in the Castagnoli-Cushman reaction with imines have been rendered valid participant in this process. The new reaction format involves the use of respective indole-based dicarboxylic acids and in situ cyclodehydration of the latter by acetic anhydride. This finding validates a fundamentally new approach to synthesizing compounds based on the privileged 1,2,3,4-tetrahydropyrazino[1,2-a]indole core characterized by hitherto undescribed substitution pattern.  相似文献

A mild and efficient method for the synthesis of 1H-chromeno[2,3-d]pyrimidine-5-carboxamide derivatives via a one-pot, three-component reaction of an isocyanide, barbituric acid, and a salicylaldehyde in the presence of acetic acid in ethanol/water mixture at 75 °C is reported. The reaction of readily accessible 3-(thien-2-yl)allylamines with maleic anhydride, followed by a domino sequence involving successive acylation/[4+2] cycloaddition steps, leads to the formation of the thieno[2,3-f]isoindole core. The key step, the intramolecular Diels-Alder vinylaren (IMDAV) reaction, proceeds with high level of diastereoselectivity and with formation of a single diastereoisomer of the target product 4,4a,5,6,7,7a-hexahydro-3aH-thieno[2,3-f]isoindole-4-carboxylic acids in excellent yields. If the reaction is carried out at room temperature, it occurs in 2–3 days and the proton migration (H-shift) does not take place at the last stage. In boiling benzene, the reaction is complete after three hours, but in this case a slight impurity of byproducts bearing aromatic thiophene ring – 4a,5,6,7,7a,8-hexahydro-4H-thieno[2,3-f]isoindole-4-carboxylic acids is formed.  相似文献

Regioselective reaction of C60Cl6 with thioamides via a radical annulation to form fullereno thiazole derivatives is reported. The reaction is promoted by K2CO3, which might deprotonate thioamide to initiate a single electron transfer from thioamide anion to C60Cl6. The experiments with various thioamides establish the proposed base-promoted reaction as a facile route for synthesis of fullereno fused thiazole derivatives starting from C60Cl6, a prevalent synthon in fullerene chemistry. In addition, the tunable electrochemical properties of the fullereno thiazole products have been investigated for their potential photovoltaic application.  相似文献

Post-multicomponent reaction modifications of Cushman reaction-derived 1,2,3,4-tetrahydroisoquinolin-4-carboxylic acids turned out to be a surprisingly underdeveloped strategy in scaffold-oriented synthesis. In this Letter, we describe a concise synthesis of hitherto unreported pyrazino- and diazepino-fused isoquinolones in two chemical operations. The synthesis involves the use, for the first time, of aryl glyoxals as carbonyl components for the Cushman multicomponent reaction.  相似文献
