Racemic 3-methoxyphenyl(1-naphthyl)phenylphosphine 1 was effectively resolved via an oxidative resolution procedure utilizing l-menthyl bromoacetate as the resolving agent to give enantiopure 3-methoxyphenyl(1-naphthyl)phenylphosphine oxide (R)-2 in 41% yield. Reduction of the resolved (R)-4 with HSiCl3/NEt3 provided the corresponding phosphine (R)-1 in >97% ee. Ortho-iodination of the enantiopure (R)-4 followed by Ullmann coupling of the resulting iodoarylphosphine oxide gave a P-stereogenic and atropoisomeric biphenyl diphosphine dioxide as a single diastereoisomer. The latter transformation constitutes the first example of an effective transfer of a P-centered chirality to an axial chirality of the atropoisomeric biaryl system. The absolute configurations of the resolved phosphine and the atropoisomeric biaryl system have also been established.  相似文献

Enantiopure O-phenyl ethylphosphonothioic acid 1 was easily obtained by the enantioseparation of racemic 1, which was prepared from commercially available phosphorothioic trichloride in four steps. Enantiopure 1 was found to show an excellent chiral recognition ability for various 1-arylethylamine derivatives during the diastereomeric salt formation. In particular, enantiopure 1 was able to recognize the chirality of o- and m-substituted 1-arylethylamine derivatives, of which the chirality is generally difficult to establish by conventional resolving agents. X-ray crystallographic analyses of the less-soluble diastereomeric salts revealed that the conformation of the phenoxy group in enantiopure 1 could change in the diastereomeric salt crystals and that the excellent chiral recognition ability of enantiopure 1 resulted from the effective CH/π interaction of the phenoxy phenyl group.  相似文献

Pd(0)-catalyzed allylic amination of 3-bromocyclohexene with (1S,2S)-2-amino-1,2-diphenylethanol and subsequent intramolecular oxyselenenylation of the resulting allylic amine 6 followed by oxidation–elimination afforded the valuable cis-fused bicyclic olefin 10. Oxyselenenylation of cyclohexene with (1S,2S)-N-(benzyloxycarbonyl)-2-amino-1,2-diphenylethanol and subsequent oxidation–elimination gave the allylic ether 18. Intramolecular aminoselenenylation of 18 followed by oxidation–elimination provided the cis-fused bicyclic olefin 21, which is the regioisomer of 10. Further stereoselective transformation of 10 afforded enantiopure aminocyclohexitols.  相似文献

The synthesis of enantiomerically pure (4aS,5R)-hexahydro-4a,5-dimethyl-2(3H)-naphthalenone (−)-1 is described for the first time. The synthesis starts from (R)-3-methylcyclohexanone and involves the preparation of Piers enol lactone 6 in its enantiopure form as the key intermediate. Treatment of (+)-6 with methyl lithium followed by an intramolecular aldol reaction gives the bicyclic enone (−)-1.  相似文献

Starting from the Staudinger [2+2] cycloaddition between the 1-azadiene 1 and acetoxyacetylketene, generated in situ from acetoxyacetyl chloride, we developed a seven-step synthesis of the novel azeto[3′,4′:2,3]pyrano[4,5-c]pyrazole skeleton 9 in both racemic and enantiopure forms. Silver carbonate treatment of the functionalised hydrazonoyl chloride 7 promoted in situ generation of the corresponding nitrilimine 8, which underwent a clean, stereoselective cycloaddition to the target tricyclic β-lactam 9. The key step of the synthetic pathway leading to enantiopure 9 was the production of the enantiopure azetidinone (3R,4S)-3 by enzymatic resolution with Amano Lypase PS of the racemic precursor (3S1,4R1)-2.  相似文献

Enantiopure O-ethyl (2-naphthyl)phosphonothioic acid 1 was designed on the model of O-ethyl phenylphosphonothioic acid, based on the fact that the chiral recognition abilities of enantiopure mandelic acid and cis-1-aminoindan-2-ol were improved upon by replacing their phenyl/phenylene groups with naphthyl/naphthylene groups, respectively. Enantiopure 1 was easily obtained by the enantioseparation of racemic 1, which was easily synthesized from commercially available 2-bromonaphthalene and diethoxyphosphonothioic chloride, with enantiopure 1-phenylethylamine. Enantiopure 1 showed an excellent chiral recognition ability for several 1-arylethylamine derivatives during the diastereomeric salt formation. The effect of the enlarged aromatic part of 1 is discussed on the basis of X-ray crystallographic analyses.  相似文献

The enantiopure (ee >99%) antiepileptic (R)-(+)-Stiripentol has been stereoselectively synthesized via cross metathesis of 5-vinylbenzo[d][1,3]dioxole 1 and (R)-(+)-4,4-dimethylpent-1-en-3-ol (R)-(+)-2. A novel one-pot two-step pathway for the synthesis of 5-vinylbenzo[d][1,3]dioxole 1 starting from 3,4-dihydroxycinnamic acid has been introduced. A lipase catalyzed kinetic resolution access to enantiopure (R)-(+)-4,4-dimethylpent-1-en-3-ol (R)-(+)-2 (ee >99%) has also been developed.  相似文献

Reaction of 2-(diphenylphosphino)phenylphosphonous acid tetramethyldiamide 1 with (+)-menthol, (1S,2S,3S,5R)-isopinocampheol and (1R,2R)-trans-cyclohexanediol affords enantiopure phosphino-phosphonite ligands 35. The X-ray structures of 1 (space group P21/n) and 3 (space group P21) have been determined. The reaction of 1 with (1R,2R,3S,5R)-(−)-pinanediol proceeds diastereoselectively to afford a novel type of enantiopure phosphino-phosphonite ligand 6 with an asymmetric substituted P atom. On reaction of (+)-cedryl alcohol with 1 the adduct 7 of the phosphonous acid 2-Ph2PC6H4P(O)(H)OH 9 and its dimethylammonium salt is formed through elimination of water and subsequent hydrolysis. The structure of 7 (space group P1̄) was elucidated by X-ray structural analysis. Reduction of the chlorophosphine 8 with LiAlH4 yields the novel primary–tertiary phosphine 10, which is a valuable starting material for the synthesis of the enantiopure C1 symmetric bidentate phospholane ligands 11 and 12.  相似文献

Isoplagiochin C 1 was prepared for the first time in enantiopure form from synthetic racemic material, using a novel stereochemical concept. This macrocyclic bisbibenzyl contains two biaryl axes. Of these, axis A is configurationally stable by its fixation within the macrocyclic framework, while axis B is stereochemically unstable. By diesterification of rac-1 with enantiopure (P)-1,1′-binaphthyl-2,2′-dicarboxylic acid, axis B is locked in its P-configuration, thus allowing the resolution and separate saponification of dilactones (PA,PB,P)-3 and (MA,PB,P)-3 to give the pure enantiomers of 1. This concept permits recycling of any undesired enantiomer of 1 by thermal equilibration of the respective diastereomer of 3.  相似文献

Kinetic resolution of racemic 1-(benzofuran-2-yl)ethanols rac-1ad was performed by lipase-catalyzed enantiomer selective acylation (E≫100) yielding (1R)-1-acetoxy-1-(benzofuran-2-yl)ethanes (R)-2ad and (1S)-1-(benzofuran-2-yl)ethanols (S)-1ad in highly enantiopure form. The degree of enantiomer selectivity for enzymatic alcoholysis/hydrolysis processes starting from racemic 1-acetoxy-1-(benzofuran-2-yl)ethane rac-2 was also tested under various conditions including supercritical CO2 medium. Racemization-free lipase-catalyzed ethanolysis of the (1R)-1-acetoxy-1-(benzofuran-2-yl)ethanes (R)-2ad yielded almost quantitatively the enantiopure (1R)-1-(benzofuran-2-yl)ethanols (R)-1ad.  相似文献

The kinetic bioresolution of 2-nitrocyclohexanol 1 was investigated by screening a range of hydrolases both for enantioselective transesterification and for enantioselective hydrolysis of the corresponding acetate. Racemic ethyl 3-hydroxybutanoate rac-1 was transformed into ethyl (R)-acetoxybutanoate (ee = 92%) with 85–90% chemical yields using enantioselective acylation with isopropenyl acetate in the presence of Candida antarctica lipase B (CAL-B, Novozym 435) under solvent-free conditions, followed by mesylation of the unreacted (S)-alcohol in the reaction mixture and inversion of configuration with cesium acetate in DMF in one pot. When the (R)-acetoxybutanoate was subjected to alcoholysis with ethanol and CAL-B, enantiopure (R)-1 (ee >99%) was produced.  相似文献

Basic hydrolysis of the ester group of title compounds 3ac gave the corresponding, readily functionalisable carboxylic acids. A concise enantiopure synthesis of six monoterpenic alkaloids of the actinidine series possessing a cyclopenta[c]pyridine skeleton, (+)-deoxyrhexifoline (4), (+)-boschniakinic acid (5), (+)-boschniakine (6), (−)-plantagonine (7), (−)-indicaine (8) and (−)-tecostidine (9) is reported starting with the chiral precursor 3-bromo-5-((4R)-phenyloxazolin-2-yl)pyridine (10). It involves a C-4 regioselective connection of a butene appendice and an intramolecular 5-exo-trig Heck annulation sequence followed by hydrogenation of the exocyclic alkene. Mixture of (3R)- and (3S)-7-((4R)-phenyloxazolin-2-yl)cyclopenta[c]pyridines was separated by HPLC before being transformed into enantiopure natural products (4-9) by modification of the oxazoline group.  相似文献

The novel, chiral glycinamides (S,S)-3 and (S,S)-4 were prepared in good yields from C2-symmetric chiral amines (S,S)-1 and (S,S)-2, respectively. Enolate formation and addition to methyl iodide and benzyl bromide proceeded in good yield and high diastereoselectivity, especially in the presence of LiCl or DMPU. β2-(3,4-Dihydroxybenzyl)-β-alanine [β2-Homo-Dopa, 1] is a novel β-amino acid homologue of Dopa, the most successful therapeutic agent in the treatment of Parkinson's disease. Enantioenriched (R)-1 and (S)-1 were obtained via the diastereoselective alkylation of enantiopure pyrimidinone (R)- and (S)-3, chiral derivatives of β-alanine, with veratryl iodide. The major diastereomeric products (2S,5R)-4 and (2R,5S)-4 were hydrolyzed with 57% HBr, and the desired β-amino acids were purified by silica gel chromatography. Alternatively, enantioenriched (R)- and (S)-1 were prepared by means of the highly diastereoselective alkylation (3,4-dimethoxybenzyl iodide) of open-chain β-aminopropionic acid derivatives (R,R,S)-8 and (S,S,R)-8 containing the chiral auxiliary α-phenylethylamine. Finally, nearly enantiopure (R)- and (S)-1 were obtained by resolution of racemic N-benzyloxycarbonyl-2-(3,4-dibenzyloxybenzyl)-3-aminopropionic acid, rac-12, with (R)- or (S)-α-phenylethylamine, followed by catalytic hydrogenolysis.  相似文献
