The reaction of 4-methyl-pyrrolidin-2-ones, chlorinated at the C(3) and C(6) positions, with n-propylamine constitutes a new method for the preparation of 5-propylimino-pyrrolidin-2-ones or 3-pyrrolin-2-ones in generally good yields. The transformation involves a series of eliminations, substitutions and double bond shifts. This constitutes a remarkable example of a functional rearrangement.  相似文献

A simple and efficient method for the synthesis of 4-hydroxy or 4-alkylamino isoxazolidin-3-ones and 4-hydroxy isoxazolidin-5-ones is described. The synthesis involved the ring opening of ethyl β-arylglycidates or ethyl N-alkylaziridine-2-carboxylates by free N-alkylhydroxylamines or hydroxylamine anions, followed by cyclization to give the title isoxazolidinones in moderate to good yields.  相似文献

Two new synthetic methodologies for 2,3-diarylacridin-9(10H)-ones were developed. The first one involves the Heck reaction of (E)-3-iodo-2-styrylquinolin-4(1H)-ones with styrenes, leading to (E,E)-2,3-distyrylquinolin-4(1H)-ones, which when heated at high temperatures cyclize in two different ways. Electrocyclization and further in situ oxidation leads to 2,3-diarylacridin-9(10H)-ones, while tautomerization, cyclization by nucleophilic addition and further in situ oxidation produces (E)-2-aryl-4-styrylfuro[3,2-c]quinolines as the main compound. The second method gives only 2,3-diaryl-10-methylacridin-9(10H)-ones and involves the Heck reaction of (E)-3-iodo-1-methyl-2-styrylquinolin-4(1H)-ones and styrenes, leading to (E,E)-1-methyl-2,3-distyrylquinolin-4(1H)-ones, which when heated at high temperatures cyclize through electrocyclization and oxidation processes affording the expected compounds. The structures of all new compounds were established by extensive NMR studies.  相似文献

4-Arylphthalaz-1-ones (2a–e) are prepared by the hitherto unknown reaction of N-aminophthalimide (1) with aromatic hydrocarbons under Friedel-Crafts conditions. The reaction of N-aminophthalimide with Grignard reagents presents another convenient method for the synthesis of 4-alkyl or 4-arylphthalaz-1-ones.  相似文献

A simple method for the synthesis of acetylenic β-diketones is described. They can be cyclized to the corresponding 4H-pyran-4-ones with acids. Their reaction with hydrazine hydrate gave pyrazole derivatives isomeric with those obtained from 4H-pyran-4-ones. With hydroxyl-amine hydrochloride in ethanol, 1-hydroxy-4-pyridones are formed.  相似文献

Summary. An efficient one-pot method for synthesis of an array of quinazolin-4(3H)-ones from anthranilic acid, triethyl orthoformate, and amines using ZrOCl2 is described. This method offers improvements for the synthesis of quinazolin-4(3H)-ones with regard to short reaction time, yield of products, and simplicity in operation.  相似文献

A versatile synthetic method for preparing 1-substituted 3-nitroquinolin-4(1H)-ones from corresponding 2-fluoro-α-nitroacetophenones is demonstrated by the synthesis of 6,7-difluoro derivatives 7a-c . The method involves sequential treatment of the starting nitroacetophenone with triethyl orthoformate and the appropriate amine, followed by a nucleophilic cyclization reaction under mild conditions. The C-7 fluorine atom of 7 can be displaced by cyclic amines. Substituted 6-fluoro-7-(4-methyl-1-piperazinyl)-3-nitroquinolin-4(1H)-ones 8a-c were prepared in this way.  相似文献

4-Arylmethylidene-3-isopropylisoxazol-5-ones and 4-arylmethylidene-1-phenyl-3-isopropylpyrazol-5-ones have been prepared from the reactions of hydroxylamine and phenylhydrazine with methyl 4-methyl-3-oxovalerate and aromatic aldehydes in an aqueous medium at room temperature in the presence of pyridine under ultrasonic irradiation. The simple and facile method produced products in good yields.

An efficient, convenient, and general synthetic method for indeno[1,2-c]pyrazole-4-ones through oxidation of indenopyrazoles by treatment with a base and molecular oxygen is described, and a possible mechanism for the oxidation is proposed. The product indeno[1,2-c]pyrazole-4-ones were elaborated further by Suzuki coupling and Mitsunobu reaction.  相似文献

The condensation of 2-aminobenzamide with aldehydes or ketones has been achieved using cellulosesulfonic acid under mild reaction conditions to furnish 2,3-dihydroquinazolin-4(1H)-ones in good yields with a high selectivity. The use of biodegradable solid acid catalyst, cellulosesulfonic acid makes this method quite simple, more convenient, and practical. This catalyst was also found to be very active for the synthesis of hydroxyalkylquinazolin-4-ones from cyclic enol ethers.  相似文献

To develop a method of synthesis of the potentially physiologically active compounds, 1′-substituted 6-amino-spiro-4-(piperidine-4′)-2H,4H-pyrano[2,3-c]pyrazoles, we studied the three-component condensation of substituted piperidin-4-ones, malononitrile and pyrazolin-5-ones. It was found that the electrochemical method of synthesis is more regioselective, the products of the reaction are analytically pure and do not require further recrystallization.  相似文献

2,7-Diaryl-3-cyano-4-methylpyrano[4,3-b]pyridin-5-ones were synthesized by Ni(acac)2-catalyzed condensation of aroylacetonitriles with acetoacetates. The competitive Knoevenagel reaction gave 6-aryl-5-cyano-4-methylpyran-2-ones as by-products. A preparative method for the synthesis of the latter compounds in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) as a catalyst was proposed.  相似文献

The reaction between 1,3-cyclohexanediones and chalcones (or their vinilogs) in the presence of 2.5 equiv of cerium(IV) ammonium nitrate afforded trans-2-arylcarbonyl-3-aryl (or styryl)-2,3,6,7-tetrahydrobenzofuran-4(5H)-ones in good to excellent yields and in high diastereoselectivities. The method was also extended to the preparation of derivatives of the 5,6-dihydro-2H-cyclopenta[b]furan-4(3H)-one system. The fused 2,3-dihydrofuran derivatives were transformed into 1-alkyl-2-acyl-3-aryl-6,7-dihydroindole-4(5H)-ones by 2,3-dehydrogenation followed by reaction with primary amines. The direct reaction of the tetrahydrobenzofuran-4(5H)-one compounds derived from dimedone with amines gave 1-alkyl-2-alkylimino-3-aryl-6,7-dihydroindole-4(5H)-ones, while starting materials derived from 1,3-cyclohexanedione underwent an unprecedented 2-deacylation reaction and gave 1-alkyl-3-aryl-6,7-dihydroindole-4(5H)-ones.  相似文献

1,2-Benzisothiazolin-3-ones were synthesized from 1,3-benzoxathiin-4-one 1-oxides by means of a nonhazardous and inexpensive method. The 1,3-benzoxathiin-4-one 1-oxides were prepared by oxidation of 1,3-benzoxathiin-4-ones with hydrogen peroxide in the presence of a catalyst. Attack of amines on the carbonyl groups of the 1,3-benzoxathiin-4-one 1-oxides and subsequent elimination of carbonyl compounds likely produced sulfenic acids, which then underwent ring closure to afford the 1,2-benzisothiazolin-3-ones.  相似文献

A new simple method has been used to prepare 6-substituted 4-(subst. amino)-5-aryldiazenyl-1-arylpyridazinium salts from N-methyl- or N-aryl-3-amino-1-phenylbut-2-en-1-ones and 4-aminopent-3-en-2-ones and substituted benzenediazonium tetrafluoroborates or hexafluorophosphates. We examined a method for the preparation of 4-hydroxypyran-2-ones in two steps from aryl ketones and ethyl malonyl chloride (or ethyl 2-methylmalonyl chloride). These 4-hydroxypyran-2-ones are useful precursors as they may be transformed in two steps to the 2-methoxypyran-4-ones, which is found in phenoxan—a naturally occurring compound discovered to have anti-HIV activity.  相似文献

Pyrrolin-4-ones/pyrrolin-3-ones and its derivatives are important heterocyclic systems which are observed in vast variety of natural products, pharmaceuticals, and biologically important compounds. The Buchwald-Hartwig Pd-catalyzed C-N coupling reaction of 5-substituted 3-chloro-4H-1,2,6-thiadiazin-4-ones with primary (het)arylamines is described, affording twenty new 3,5-disubstituted 4H-1,2,6-thiadiazin-4-ones in 56–99% yield. The protocol enables the mild preparation of difficult to access 3,5-dianilino-1,2,6-4H-thiadiazin-4-ones. The reaction scope and limitations are discussed.  相似文献

Methylation of 3-methyl-4-arylhydrazonoisoxazol-5-ones with methyl iodide affords both 2,3-dimethyl-4-arylazoisoxazol-5-ones and 3-methyl-4-(N-methylarylhydrazono)isoxazol-5-ones but with dimethyl sulfate only the former products are formed. 3-Phenyl-4-arylhydrazonoisoxazol-5-ones behave in a similar way on methylation with methyl iodide and dimethyl sulfate.  相似文献
