The palladium-catalyzed allylic alkylation reaction of 1,1-dimethylallyl acetate with dimethyl malonate is studied by a combination of isotope effects and theoretical calculations. A large 13C isotope effect of ≈1.037 is observed at the tertiary carbon, while small isotope effects are observed at the olefinic carbons. These results support rate-limiting ionization of a η2-Pd complex. The observed isotope effects are compared with predictions from calculational models employing either solvent models or ionization of an amidinium ion. The calculated transition structures are notably η2 in character, and the implications of this observation are discussed.  相似文献

A highly regioselective palladium-catalyzed allylic alkylation of fluorobis(phenylsulfonyl)methane has been studied. Using different allylic carbonates, a variety of terminal mono-fluoromethylated compounds were achieved in 85-99% yields with high regioselectivities.  相似文献

A novel Pd/1,2-diphosphite catalyzed asymmetric allylic alkylation of 1,3-diarylpropenyl acetate with malonates was developed. Catalyst optimization via a variation in the protecting groups at the 1,2- and/or 5,6-positions of d-mannitol skeleton and in biaryl moieties of the ligands led to a ‘lead’ catalyst, which efficiently mediated the allylic alkylations. The activities and enantioselectivities of the reaction clearly showed that the stereogenic centers of the skeleton and the axially chiral diaryl moieties of the ligands had a synergic effect. The ligand 1,2:5,6-di-O-isopropylidene-3,4-bis[(S)-1,1′-binaphthyl-2,2′-diyl]phosphite-d-mannitol afforded excellent yields (up to 99%) and high levels of enantioselectivies (ee up to 98%) in 1,4-dioxane/CH2Cl2 mixture (v/v, 1:1) using [Pd(π-allyl)Cl]2 as catalytic precursor and LiOAc as base. Dramatic changes in the sense and in the degree of the enantioselectivity depending on the configuration of the diaryl moieties of the ligands and reaction conditions were observed.  相似文献

Under catalysis of Pd(OAc)2-(P-n-Bu)3, Et2Zn promotes a variety of allyl alcohols to undergo nucleophilic allylation of aliphatic aldehydes and ketones at room temperature and provides homoallyl alcohols in 60-90 and ca. 60% isolated yield, respectively. The reaction is irreversible and kinetically controlled, and unique regio- and stereoselectivities observed for the allylation with unsymmetrically substituted allyl alcohols are discussed.  相似文献

The synthesis of a third generation phosphorus-containing dendrimer possessing 24 chiral iminophosphine end groups derived from (2S)-2-amino-1-(diphenylphosphinyl)-3-methylbutane is described. In situ complexation of this dendrimer by [Pd(η3-C3H5)Cl]2 affords a catalyst, which is used in asymmetric allylic alkylations of rac-(E)-diphenyl-2-propenyl acetate and pivalate. The percentage of conversion, the yield of isolated 2-(1,3-diphenylallyl)-malonic acid dimethyl ester, and its enantiomeric excess have been measured in each case, and were found to be good to very good (ee from 90% to 95%). Furthermore, the dendritic catalyst can be recovered and reused at least two times, with almost the same efficiency.  相似文献

Cooperative catalysis enables the direct enantioselective α-allylation of linear prochiral esters using Si-substituted allyl electrophiles. The Si-substituent directs the regioselectivity of enantioselective bond formation and provides products containing synthetically versatile pentafluorophenyl ester and vinylsilane moieties. Critical to the efficacy of this process was the recognition that the ancillary ligand on palladium could be altered to prevent formation of a deleterious ether by-product, whilst retaining enantioselectivity through the Lewis base catalyst. Flexibility such as this is unique to cooperative catalysis events and provides efficient access to an array of enantioenriched products that are orthogonally functionalized and easily modified.  相似文献

A series of readily available thioether-phosphinite ligands has been tested in the Pd-catalyzed allylic substitution reactions of several acyclic and cyclic allylic substrates (S1-S7). This series of ligands have been designed to uncover their important structural features and to determine the scope of the thioether-phosphinite ligands in these catalytic reactions. Systematic variation of the electronic and steric properties at the thioether moiety provide useful information about the ligand parameters that control the enantiodiscrimination. By carefully selecting the ligand parameters, good enantioselectivities with high activities were obtained for hindered linear substrates S1 and S2 (ee’s up to 95%) and for unhindered cyclic substrates S4 and S5 (ee’s up to 91%).  相似文献

A series of novel chiral phosphine–imine ligands have been prepared by a two-step transformation from chiral α-phenylethylamine. The resulting chiral ligands were found to be effective for the palladium-catalyzed asymmetric allylic alkylation of 1,3-diphenylprop-2-en-1-yl pivalate with dimethyl malonate, in which up to 94% ee and 99% conversions were obtained. The results demonstrate that the chirality resided on the chelate ring of P–Pd–N complex is more effective for the transfer of the stereochemical information by comparison with the result obtained by Hashimoto and coworkers’ phosphine–imine ligand, in which the chirality lay in the outside of P–Pd–N chelate ring. The effect of solvent, base and substitutent in phosphine–imine ligand on this catalytic reaction is also described.  相似文献

New chiral C1-symmetric dipyridylmethane ligands have been prepared from naturally occurring monoterpenes according to a method based on two consecutive constructions of the pyridine rings. These ligands have been assessed in the enantioselective palladium catalyzed allylic alkylation of 1,3-diphenylprop-2-enyl acetate with dimethylmalonate. New chiral 2-alkyl-8-quinolinyl-oxazolines were synthesized from 2-alkyl-8-quinolinecarboxylic acids and enantiomerically pure amino alcohols using a convenient procedure. Enantioselective palladium-catalyzed allylic alkylation of 1,3-diphenyl-2-propenyl acetate with dimethyl malonate in the presence of 2-alkyl-8-quinolinyl-oxazolines provided an alkylation product with an opposite configuration compared to those obtained from unsubstituted quinolinyl-oxazoline ligands.  相似文献

Rapid access to enantioenriched spirocycles possessing a 1,4-dicarbonyl moiety spanning an all-carbon quaternary stereogenic spirocenter was achieved using a masked bromomethyl vinyl ketone reagent. The developed protocol entails an enantioselective palladium-catalyzed allylic alkylation reaction followed by a one-pot unmasking/RCM sequence that provides access to the spirocyclic compounds in good yields and selectivities.  相似文献

The synthesis and characterization of two new ferrocenyl Schiff bases: [Fc-CHN-(CH2)n-(C4H3S)] (2) {Fc represents (η5-C5H5)Fe(η5-C5H4)- and n = 1(2a) or 2(2b)} containing the thienyl (C4H3S) group are reported. NMR studies indicate that 2 have the anti-(E) conformation in solution and the X-ray crystal structure of 2a confirms that it also adopts the anti-(E) form in the solid state. Ligands 2 have been tested in the palladium catalyzed allylic alkylation of (E)-3-phenyl-2-propen-1-yl (cinnamyl) acetate using sodium diethyl 2-methylmalonate as nucleophile. The reaction of 2 with [Pd(η3-1-Ph-C3H4)(μ-Cl)]2 in the presence of a slight excess K[PF6] produced [Pd(η3-1-Ph-C3H4){Fc-CHN-(CH2)n-(C4H3S)}][PF6] {n = 1(5a) or 2(5b)}, which are the intermediates of this catalytic process. NMR studies of 5 reveal the coexistence of several isomers in solution. The stoichiometric reactions of 5 with the nucleophile are also reported. The comparison of the results obtained for 2, [Fc-CHN-(C6H4-2SMe)] (1a) and [(2,4,6-Me3-C6H2)-CHN-(C6H4-2SMe)] (1b) has allowed to establish the importance of the nature of the substituents on the imine group on the regioselectivity of the process.  相似文献

The “trans rule” in Pd-catalyzed allylic substitutions predicts trans to phosphorus additions of nucleophiles to Pd-allyl intermediates, e.g., with P,N-ligands. This computational study reveals that not only the intrinsic electronic differentiation between P- (i.e., PH3) and N-ligands (i.e., para-X-substituted pyridines), but also the “late” or “early” nature of the transition structures is crucial for strong cis vs. trans discriminations and hence for selectivity. Although para-nitro pyridine exhibits less intrinsic electronic differentiation than para-dimethylamino pyridine, the higher reactivity of the Pd-allyl-intermediate and the earlier nature of the transition structure yield a higher sensitivity for electronic differentiation for XNO2 than XNMe2.  相似文献

Asymmetric synthesis of 6,7-dimethoxy-1-vinyltetrahydroisoquinolines through Pd-catalyzed intramolecular allylic amination of 3-(amidoethylphenyl)prop-2-enyl carbonates was studied, using a library of fine-tunable monodentate phosphoramidite ligands. Under optimized conditions, excellent enantiopurity (up to 96% ee) and 100% product selectivity were achieved. 1-Vinyltetrahydroquinoline thus obtained is a highly versatile intermediate for the synthesis of various biologically active alkaloids of medicinal interest.  相似文献

The Pd-catalyzed allylation of 3-acetyl-2-oxindoles with allyl alcohol is performed using 3 mol% of Pd(dba)2, rac-BINAP and BINOL phosphoric acid as catalytic mixture. This procedure allows the in situ synthesis of 3-allyl-2-oxindole by adding Triton B to the reaction mixture. The deacylative allylation of 3-acetyl-3-methyl-2-oxindoles with allylic alcohols is carried out with 3 mol% of Pd(OAc)2, dppp and 1.5 equiv. of LiOtBu as base affording the corresponding 3,3-disubstituted 2-oxindoles in good yields. Both methodologies can be combined for the preparation of unsymmetrical 3,3-diallylated 2-oxindoles such as compound 7. The DaA must be carried out in the absence of oxygen in order to avoid the competitive formation of 3-alkyl-3-hydroxy-2-indoles. The later compounds can be easily obtained by deacylative oxidation of 3-alkylated 3-acetyl-2-oxindoles with LiOEt at rt under air.  相似文献

A new route to racemic diethyl 2-methyl-bicyclo[3.1.1]hept-2-ene-6,6-dicarboxylate (±)-1a by means of a palladium-catalyzed intramolecular allylic alkylation exercised on malonate-ester derivatives has been developed from R-(−)-carvone. The stereocontrolled synthesis of the enantiomerically pure (−)-1b by application of a six-step reactions sequence with a 28% overall yield from acetal 8, has been accomplished.  相似文献
