Three new sesquiterpenes (1-3), along with five known (5-9), were isolated from the organic extract of the red alga Laurencia obtusa, collected from the coastal rocks of Serifos in the Aegean Sea. A new dimeric sesquiterpene of the cyclolaurane-type (4) along with four previously reported (7, 10-12) metabolites, were isolated from the extract of Laurencia microcladia, collected at Chios island in the North Aegean Sea. The structures and the relative stereochemistry of the compounds are proposed on the basis of their spectral data. The cytotoxicity of the isolated metabolites was evaluated against several cell lines including human tumor cell lines.  相似文献

Chromatographic separation of a crude extract obtained from the fungus Aspergillus sp., isolated from the Mediterranean sponge Tethya aurantium, yielded a new tryptophan derived alkaloid, 3-((1-hydroxy-3-(2-methylbut-3-en-2-yl)-2-oxoindolin-3-yl)methyl)-1-methyl-3,4-dihydrobenzo[e][1,4]diazepine-2,5-dione (1), and a new meroterpenoid, austalide R (2), together with three known compounds (35). The structures of the new compounds were unambiguously elucidated on the basis of extensive one and two-dimensional NMR (1H, 13C, COSY, HMBC, and ROESY) and mass spectral analysis. Interestingly, the compounds exhibited antibacterial activity when tested against a panel of marine bacteria, with 1 selectively inhibiting Vibrio species and 2 showing a broad spectrum of activity. In contrast, no significant activity was observed against terrestrial bacterial strains and the murine cancer cell line L5178Y.  相似文献

Novel cytotoxic rearranged kaurene- and ent-kaurene-type diterpenoids against HL-60 cells have been isolated from the unidentified New Zealand liverwort Jungermannia species together with previously known ent-kaurane-type diterpenoids. Chromatographic fractionation of a hexane extract from stem-bark of Premna latifolia yielded three new icetexane diterpenes (1-3). Their structures were elucidated on the basis of spectroscopic data, mainly NMR and MS, data. Compounds 1-3 were also evaluated for the cytotoxicity against the cancer cell lines (HT-29, A-431, MCF-7, Hep-G2, PC-3, A-549, B-16 F10 and ACHN). Compounds 1 and 2 displayed potent cytotoxic activity against HT-29 and Hep-G2 cell lines with IC50 values of 0.04 and 0.18 μg/mL, respectively.  相似文献

Seven new prenylated indole diketopiperazine alkaloids, including compound 1, 3 spirotryprostatins C-E (2-4), 2 derivatives of fumitremorgin B (5 and 6), and 13-oxoverruculogen (7), have been isolated from the holothurian-derived fungus Aspergillus fumigatus, along with 12 known ones (8-19). The structures of the new compounds were determined on the basis of extensive spectroscopic data and amino acid analysis. All new compounds were evaluated for their cytotoxic activities on MOLT-4, A549, HL-60, and BEL-7420 cell lines by the MTT and SRB methods.  相似文献

Two novel cyclized meroditerpenoids atomarianones A, and B (1 and 2), were isolated from the organic extract of the brown alga Taonia atomaria collected at Serifos island in the Central Aegean Sea. This is only the second report on metabolites having a functionalized indane moiety instead of a benzofurano- or a benzopyrano-ring connecting their aromatic and diterpenoid parts. Atomarianone A contains an unprecedented cis B-C ring fusion while atomarianone B is the epimer of A at C-7. Both metabolites were found to exhibit significant cytotoxic activity against two lung cancer cell lines.  相似文献

Three new furanocoumarins, named feroniellins A (1), B (2), and C (3), were isolated from the roots of Feroniella lucida. Compounds 1-3 are novel structures having an oxolane, oxane, and oxepane moiety. Their overall structures and configurations were determined by spectral and chemical methods. Kehokorins A (1)-C (3), three novel dibenzofurans, have been isolated from field-collected fruit bodies of the myxomycete, Trichia favoginea var. persimilis, and their structures were elucidated by spectral data. Kehokorin A (1) was a α-l-rhamnopyranoside of kehokorin B (2), while kehokorin C (3) was a 1-demethoxy analog of 2. Eleven new dihydronaphthalenones 1-11, together with five known compounds; 5-hydroxydihydrofusarubin C (12), javanicin, bostrycoidin, anhydrofusarubin, and 3-O-methylfusarubin, were isolated from the endophytic fungus Fusarium sp. BCC14842. Structures were elucidated by analyses of the NMR spectroscopic and mass spectrometry data. Javanicin, 3-O-methylfusarubin, compounds 3 and 7 exhibited antimycobacterial and cytotoxic activities. Javanicin also displayed antifungal activity with IC50 of 6.16 μg/mL, while compounds 2, 4, 5, 8, and 12 showed only cytotoxic activity.  相似文献

A new polyhydroxylated macrolide, named mangiferaelactone (1) was isolated from a solid culture of the endophytic fungus Pestalotiopsis manguiferae, together with ten known compounds [(6S,1′S)-LL-P880α; (6S,1′S,2′R)-LL-P880β; (1′S,2′R)-LL-880γ; (1′R)-dehydropestalotin; (−)-5-carboxylmellein; (−)-5-methylmellein; (−)-5-hydroxylmethylmellein; arabenoic acid; 5,6-dihydro-4-methoxy-2H-pyran-2-one; and the (−)-2-hexylidene-3-methylsuccinic acid]. P. manguiferae was isolated from Hyptis dilatata, a small shrub common in the central region of Panama. The structure of compound 1 was elucidated by a combination of spectroscopic methods (IR, MS, optical rotation, 1D and 2D NMR spectroscopy). The absolute configuration of 1 was established as 4R,7R,8R,9S by application of vibrational circular dichroism (VCD). Compound 1 showed a minimum inhibitory concentration (MIC) of 1.6863 mg/mL against Listeria monocytogenes, and 0.5529 mg/mL against Bacillus cereus. No activity was observed for compound 1 against Plasmodium falciparum or Trypanosoma cruzi; likewise, no cytotoxic activity was observed against A2058 and H522-T1 cells.  相似文献

Zygosporamide (1), a new cyclic pentadepsipeptide, was isolated from the seawater-based fermentation broth of a marine-derived fungus identified as Zygosporium masonii. The structure of 1, which is composed of α-hydroxyleucic acid and both d- and l-amino acids, was determined by combined spectral and chemical methods. Despite a simple structure, zygosporamide illustrated significant cytotoxicity in the NCI’s 60 cell line panel (median GI50 = 9.1 μM), with highly enhanced selectivity against the CNS cancer cell line SF-268 (GI50 = 6.5 nM) and the renal cancer cell line RXF 393 (GI50 ? 5.0 nM).  相似文献

From an antimicrobial gram-positive actinomycete strain of Saccharomonospora azurea (MTCC11714) isolated from high altitude soil of Kargil (J&K, India), a new isocoumarin saccharonol B (2) along with two known compounds viz. saccharonol A (1) and piericidin A3 (3) was isolated and characterized. The structure of the new compound was established based on extensive 2D-NMR data. Saccharonol B (2) exhibited mild antimicrobial activity against a standard panel of microorganisms Staphylococcus aureus ATCC 29213, Candida albicans ATCC 90028, and Aspergillus fumigatus MTCC 1811 with MIC values in the range of 128–248 μg/mL. Saccharonol B (2) and piericidin A3 (3) showed selective cytotoxic activity against human pancreatic carcinoma cell line (MIAPaCa-2) with IC50 values of 9 and 8 μM, respectively. Mechanistic studies indicated that saccharonol B (2) arrests S-phase of the cell cycle and causes dose-dependent loss of mitochondrial potential in MIAPaCa-2 cells.  相似文献

Chemical investigation of the endophytic fungus Diaporthe melonis, isolated from Annona squamosa, yielded two new dihydroanthracenone atropodiastereomers, diaporthemins A (1) and B (2), together with the known flavomannin-6,6′-di-O-methyl ether (3). The structures of the new compounds were established on the basis of extensive 1D and 2D NMR spectroscopy, as well as by high resolution mass spectrometry and by CD spectroscopy. Compounds 13 were tested for their antimicrobial activity against a multi-resistant clinical isolate of Staphylococcus aureus 25697, a susceptible reference strain of S. aureus ATCC 29213 and against Streptococcus pneumoniae ATCC 49619. Compound 3 strongly inhibited S. pneumonia growth with a MIC value of 2 μg/mL, and showed moderate activity against the S. aureus multi-resistant clinical isolate and susceptible reference strain (MIC 32 μg/mL), whereas 1 and 2 were not active against the tested strains.  相似文献

A new sesquiterpene lactone, rufescenolide C (1), the first furanoheliangolide dimer, was isolated from the leaves of Piptocoma rufescens, collected in the Dominican Republic. Its structure was determined by interpretation of its spectroscopic data, with the absolute configuration being established by analysis of the CD spectrum. A plausible biogenesis of this dimer is proposed. This compound showed potent cytotoxicity with an IC50 value of 150 nM, when tested against HT-29 human colon cancer cells.  相似文献
