This contribution describes a novel synthetic approach to very late antigen-4 (VLA-4) antagonist trans-4-[1-[[2,5-dichloro-4-(1-methyl-3-indolylcarboxyamide)phenyl]acetyl]-(4S)-methoxy-(2S)-pyrrolidinylmethoxy]cyclohexanecarboxylic acid (1) via tert-butyl trans-[(4S)-methoxy-(2S)-pyrrolidinylmethoxy]cyclohexanecarboxylate (2b) as a key intermediate. The synthesis, which includes n-Bu?NSO?H that catalyzed basic etherification of 12 and iodine-mediated cyclization to provide the 2,4-disubstituted pyrrolidine frame of 2b, is designed to utilize trans-4-hydroxycyclohexanecarboxylic acid (9) as a commercially available starting material. 相似文献

A series of novel 1-[(2,6-dichloro-4-trifluoromethyl)phenyl]-3-aryl-1H-pyrazole-4-carbaldehydes were synthesized using the Vilsmeier-Haack reagent. The structures of all the title compounds have been confirmed by elemental analysis, 1H-NMR and 13C-NMR and in addition, the structure of intermediate 5b was investigated by X-ray crystallography. 相似文献

Four homologous series of 4-[2-[4-alkoxy-2,3,5,6-tetrafluorophenyl)ethynyl] phenyl trans-4-alkylcyclohexyl-1-carboxylates have been prepared. Their liquid crystalline behaviour was investigated by optical polarizing microscopy and DSC. Series A, B and C exhibit the nematic phase. The phase transition behaviour of series D is anomalous. The compounds of series D with a short alkoxy chain exhibit enantiotropic smectic A and nematic phases, while those with a long alkoxy chain exhibit only the nematic phase. 相似文献

The properties of 4-methyl-4-(trichloromethyl)cyclohexa-2,5-dien-1-one which contains the trichloromethyl substituent at a quaternary carbon atom have been studied by cyclic voltammetry, rotating disk electrode voltammetry, and preparative electrolysis. Reductive dehalogenation involves initial two-electron reductive elimination of one of the geminal chlorine atoms followed by consecutive addition of proton, simultaneous elimination of the two residual halogen atoms to form carbene, and rearrangement of cyclohexadienone into the corresponding 4-methylcyclohepta-2,4,6-trien-1-one. 相似文献
