Abstract

A method for the thin layer chromatographic (TLC) separation of enantiomers and diastereomers involving the use of chiral ion interaction agents is described. Several aromatic amino alcohols were resolved by TLC on diol and/or high performance silica gel plates using a mobile phase containing (1R)-(-)- ammonium-10-camphorsulfonate or N-benzoxycarbonyl-glycyl-L-proline (ZGP). Many of these chiral aromatic amino alcohols are of pharmacological importance as α- and β-adrenergic blockers, adrenergic compounds, and anti-glaucoma agents. A comparison was made between various N-CBZ-amino acid derivatives as chiral counter ions/chiral mobile phase additives (CMAs). These separations could not be achieved on other normal phase TLC stationary phases including microcrystalline cellulose, alumina and ordinary silica gel plates.  相似文献

采用包夹聚合法,将硅小球同硅烷化试剂反应制得乙烯基硅胶,然后将该乙烯基硅胶同经十一烯酰氯、4-甲基苯甲酰氯衍生的纤维素共聚,制备出含不同官能团的聚合物包夹硅基的键合型纤维素(4-甲基苯甲酸酯)类手性固定相。分别以正己烷异丙醇、正己烷四氢呋喃为流动相,对此键合型手性固定相的手性识别能力进行了评价。为了与同类型的涂敷型纤维素(4-甲基苯甲酸酯)手性固定相作比较,合成了涂敷型纤维素(4-甲基苯甲酸酯)手性固定相。结果表明,键合型纤维素(4-甲基苯甲酸酯)手性固定相具有一定的手性识别能力,可以拆分所研究的6种手性化合物中的4种。  相似文献

将邻乙酰水杨酸纤维素酯通过间隔臂--2-(3,4-环氧环己基)乙基三甲氧基硅烷键合于硅胶之上,制得键合型手性固定相CSP1,考察了其与同类纤维素衍生物制得的涂敷型手性固定相在拆分效果方面的异同,并探讨了其差异的原因与机理。为了印证间隔臂中环己基对于手性拆分的作用,本文又将CSP1与间隔臂中不具有环己基的键合型手性固定相CSP2进行了拆分对比,进一步证实了间隔臂中环己基的作用。结果表明,键合型手性固定相相比涂敷型固定相在拆分咪唑类药物方面具有优势,这种优势一方面来自于流动相范围的扩大,另一方面是由于间隔臂中环己基对纤维素表面结构的修饰作用。  相似文献

研究了3, 5-二甲基苯基异氰酸酯对羟丙基-β-环糊精固定相的手性识别影响.通过异氰酸丙基三乙氧基硅烷作偶联剂,将羟丙基-β-环糊精键合到3-氨丙基硅烷化硅胶上,再用3, 5-二甲基苯基异氰酸酯对β-环糊精和硅胶其余羟基进行衍生化,制得一种新型的3, 5-二甲基苯基氨基甲酸酯全衍生化羟丙基-β-环糊精键合硅胶手性固定相.在反相色谱条件下,对9种手性药物进行了拆分,结果表明,3, 5-二甲基苯基氨基全衍生化固定相较之羟丙基-β-环糊精固定相有更好的分离效果.  相似文献

合成了海藻糖、龙胆二糖、蜜二糖三种二糖类键合硅胶高效液相色谱手性固定相,采用湿法装柱制备了色谱柱.在高效液相色谱正相条件下,该类固定相对醇类、胺类、氨基酸类对映异构体以及一些手性药物表现出了一定的拆分效果.特别是海藻糖固定相在所拆分的9种手性化合物中,有6种手性化合物能得到较好的分离,表现出较好的手性分离性能.并且手性固定相之间具有较好的互补性.  相似文献

A new chiral stationary phase of 3,5-dimethylphenylcarbamates of cellulose chemically bonded to 3-aminopropyl silica gel was prepared, which may be used with a wide range of solvents including standard and non-standard ones. Several racemic indole derivatives have been resolved using standard and non-standard solvents on the immobilized chiral column (15 cm × 0.46 cm) at a flow rate of 1.0 mL/min or 0.5 mL/min with a UV detection at 230 nm. Separation of indole derivatives on immobilized and coated chiral stationary phases (CSP) in HPLC using a mixture of hexane/2-propanol as mobile phase was compared. The resolution factors for immobilized and coated chiral column were 0.57–2.02 and 0.61–4.03, respectively. It was found that both coated and immbolized chiral stationary phases were suitable for the separation of indole derivatives; however, the coated CSP possesses a higher resolving power than the immobilized one. The chromatographic behaviour of nitrophenols on thin layers of silica gel and cellulose was compared, both without impregnation and impregnated with non-aqueous polar stationary phases (formamide, dimethylformamide) and less polar stationary phases (liquid paraffin, octan-1-ol, 1-bromonaphthalene). Cellulose is preferred when using formamide or dimethylformamide if a pure partition process is required. For each particular analyte a certain amount of the stationary phase is always required to suppress the adsorption activity of silica gel. Separation by reversed-phase thin-layer chromatography is strongly affected by the type of stationary phase (the possibility of forming charge-transfer complexes with 1-bromonaphthalene) and its support (the acidic properties of silica gel and its adsorption activity), the mobile phase (content of organic modifier, pH, presence of salts) and the properties of the solutes (polarity, ionizability).  相似文献

Summary To elucidate the mechanism of chiral recognition of cholic acid-based stationary phases, four new cholic acid derivatives, with differently substituted carbamate or three acetoxy groups, were bonded to a hydrosilyl-modified silica gel. Their capacity to discriminate between enantiomers was evaluated in normal-phase high-performance liquid chromatography. The results were compared with those from equivalent separations on trihydroxy- and 3α-phenylcarbamate-substituted cholic acid-based bonded phases. The influence of mobile phase composition of the separation of the enantiomers of amino alcohols was shown. Different mechanisms of chiral discrimination are discussed, highlighting the influence of the nature of the carbamate on enantioselectivity.  相似文献

Chiral stationary phases (CSPs) for high-performance liquid chromatographic (HPLC) have been prepared by coating silica gel with cellulose tribenzoate or cellulose trisphenylcarbamate. The effect of chiral additives on preparation of the CSPs was studied with (+)-l-mandelic acid, (−)-2-phenyl-1-propanol, (+)-1-phenyl-1,2-ethanediol and (−)-1-(1-naphthyl)ethanol as chiral additives for cellulose tribenzoate and (−)-2-phenyl-1-propanol and (+)-phenylsuccinic acid as chiral additives for cellulose trisphenylcarbamate. The results showed that chiral recognition by these stationary phases was increased in comparison with the original CSPs, especially the resolution (R S) obtained. The method can be used to improve the efficiency of enantiomer separation by silica gel stationary phases coated with polymers.  相似文献

纤维素三(3,5-二甲基苯基氨基甲酸酯)是液相色谱中使用最广泛的手性柱。该文详细地研究了不同程度衍生的纤维素(3,5-二甲基苯基氨基甲酸酯)以及不同硅胶(粗制硅胶、氨丙基粗制硅胶、精制硅胶、氨丙基精制硅胶、大孔硅胶、氨丙基大孔硅胶)作为支撑体对该柱手性分离能力的影响。自制了13根手性色谱柱,分别考察了其对16种外消旋体的拆分,分离结果显示:三取代纤维素柱 > 二取代纤维素柱 > 纤维素柱;精制硅胶和大孔硅胶优于粗制硅胶,大孔硅胶的柱压更低;硅胶的氨丙基化对手性选择性有一定的影响;这些手性柱之间具有一定的互补性,尤其是纤维素柱。该文有助于人们更深刻地理解和更好地把握高效液相色谱手性柱的制备。  相似文献

采用六亚甲基二异氰酸酯作为键合试剂,制备了键合型纤维素3,5-二甲基苯基氨基甲酸酯手性固定相。分别往正己烷/乙醇或正己烷/异丙醇中添加四氢呋喃和二氯甲烷,系统地考察了四氢呋喃和二氯甲烷的含量对安息香外消旋体拆分的影响。实验结果表明,安息香两对映体的容量因子随二者含量的增加而降低,但分离效果变化不大,分离因子均在1.2以上。所制备的CSP在采用含有高浓度四氢呋喃(23%)、二氯甲烷的流动相进行手性分离时仍具有良好的稳定性。与涂敷型固定相相比,流动相选择范围更广。  相似文献

Cellulose trisphenylcarbamate is regioselectively bonded to 3-aminopropyl silica gel and underivatized silica gel, respectively, at the 6-position of the primary hydroxyl group on the glucose unit of cellulose with 4,4'-diphenylmethane diisocyanate (DPDI) as a spacer. Enantioseparations are evaluated on these prepared chiral stationary phases (CSPs) with several organic acids as the modifiers in the mobile phase by high-performance liquid chromatography. The influence of the amount of DPDI used on chiral resolution is investigated. Also, the corresponding coated-type phase is also prepared for the aim of comparison. It is observed that the bonded-type phase shows a lower chiral recognition power but a better column efficiency than the coated-type phase under the liquid chromatographic mobile phase with hexane-alcohol. However, the bonded-type CSPs are compatible with a wider number of solvents such as tetrahydrofuran (THF) or chloroform, which generally result in the solubility or swelling of the cellulose derivatives on the coated-type CSPs. The results obtained from this study indicate that the bonded-type CSP may provide complementary enantioselectivity over the coated-type phase by adopting THF as a component in the mobile phase.  相似文献

Seven new stationary phases with different number of proline units and/or different linkage to silica gel were prepared and evaluated in order to improve the performance of proline chiral stationary phases. The average separation factor achieved with the 53 analytes increases with the number of proline units in the stationary phases. When the proline peptides are directly attached to the 3-methylaminopropyl silica gel without using the 6-methylaminohexanoic acid linker, the stationary phases perform better overall. For decaproline chiral stationary phase 8, the separation also depends on the mobile phase system used. For this stationary phase, the CH2Cl2/hexanes/2-propanol system significantly outperforms the 2-propanol/hexanes system. For the 53 analytes tested, the separation factors achieved with this stationary phase compare well with those for three commercial columns.  相似文献

Abstract

In this work, we present the first separation of enantiomers in gas chromatography (GC) using a fused‐silica capillary column containing cellulose triacetate, cellulose triphenylcarbamate, or cellulose tris(3,5‐dimethylphenylcarbamate) as the new chiral stationary phase. The separated solutes included alcohols, amine, ketone, ether, ester, and amino acid. Their column efficiency, polarity, and chiral selectivity were studied. The retention mechanism was discussed. The results showed that those derivatives had relatively high chiral recognition abilities and can be used as the chiral stationary phases in GC.  相似文献

Thin layer chromatography (TLC) is normally carried out on single-phase (adsorbent) plates. However, it may be necessary to use nore than one adsorbent to achieve the separation of complex mixtures. Literature reports indicate that the use of mixed phases in TLC is not very common, possibly because the plates are difficult to prepare and precoated single-phase plates of different absorbents are readily available (silica gel, reversed phase, cellulose. alumina, etc.).  相似文献

Abstract

A method for the direct separation of racemates by HPLC is described. A chiral stationary phase is synthesized, suitable for ligand exchange chromatography. L-proline is chemically bonded to silica gel via 3-glycidoxypropyltrimethoxysilane. The bonded support is loaded with Cu(II) ions as a complexing agent. Complete resolution of amino acid racemates can be obtained in less than ten minutes. α-values up to 3.5 are observed.  相似文献

A sensitive, selective, and robust high-performance TLC (HPTLC) method using chiral TLC plates for qualitative and quantitative analysis of phyllanthin (A), hypophyllanthin (B), niranthin (C), and nirtetralin (D), the active lignans of Phyllanthus species, was developed and validated. The effectiveness and role of various stationary phases viz TLC silica gel 60F(254), HPTLC silica gel 60F(254), and chiral TLC plates in the quantitation were evaluated. A precoated chiral TLC plate was found suitable for the simultaneous analysis of four pharmacologically active lignans. For achieving good separation, the optimized mobile phase of n-hexane/acetone/1,4-dioxane (9:1:0.5 by volume) was used (R(f) = 0.30, 0.36, 0.41, and 0.48 for compounds A, B, C, and D, respectively). A densitometric determination of the above compounds was carried out in reflection/absorption mode at 620 nm. Optimized chromatographic conditions provide well-separated compact bands for the tested lignans. The calibration curves were found linear in the concentration range of 100-500 ng/band. Recoveries of A-D were 99.98, 100.51, 99.22, and 98.74%, respectively. The method was validated according to ICH guidelines. The method reported here is reproducible and applied for the quantitative analysis of the above lignans in the leaves of four Phyllanthus species, i. e., P. amarus, P. maderaspatensis, P. urinaria, and P. virgatus.  相似文献
