An efficient procedure for the stereoselective synthesis of (E)- and (Z)-2-alkene-4-ynoates and -nitriles by a simple reaction of vic-diiodo-(E)-alkenes with acrylic esters and nitriles catalyzed by in situ prepared Pd(0) nanoparticles in water has been developed. Addition of acrylic esters leads to (E)-isomers exclusively, whereas (Z)-isomers are obtained in high stereoselectivity from reactions of acrylonitrile. The aqueous slurry of Pd nanoparticles is recycled. A probable mechanism has been suggested.  相似文献

The addition of pinacol allylboronic esters to aromatic and aliphatic aldehydes smoothly occurred at -78 degrees C in toluene in the presence of a catalytic amount of AlCl3 or Sc(OTf)3 (10 mol %) to give the corresponding homoallyl alcohols in high yields. The reactions proceeded regio- and diastereospecifically, yielding the isomerically pure syn- and anti-homoallyl alcohols from (Z)- and (E)-allylboronic esters, respectively. The protocol was also applied to enantioselective reactions by using a chiral Lewis acid catalyst.  相似文献

The highly stereoselective synthesis of (E)- and (Z)-alpha-fluoro-alpha,beta-unsaturated esters and (E)- and (Z)-alpha-fluoro-alpha,beta-unsaturated amides is described. 1-Bromo-1-fluoroalkenes (E/Z approximately 1:1), which are readily available starting materials, have been found to isomerize to high E/Z ratios after storage at -20 degrees C for 1 week or by photolysis at 254 nm. Since the (E)-isomers have been found to react faster than the corresponding (Z)-isomers at room temperature in Pd(0)-catalyzed reactions, the palladium-catalyzed carboalkoxylation of high E/Z 1-bromo-1-fluoroalkenes lead to a high Z/E (Z/E >/= 98:2) ratio of the alpha-fluoro-alpha,beta-unsaturated esters. When 1-bromo-1-fluoroalkenes (E/Z approximately 1:1) were reacted with HCOOH/NBu(3)/Pd(II)/DMF, the (E)-isomer was selectively reduced, and the remaining (Z)-1-bromo-1-fluoroalkenes were recovered in essentially pure isomeric form. The resulting mixture of (Z)-1-bromo-1-fluoroalkenes and the reduced products underwent similar palladium-catalyzed carboalkoxylation reactions at 70 degrees C, and the (E)-alpha-fluoro-alpha,beta-unsaturated esters were stereospecifically obtained. This methodology was also successfully applied for the stereospecific synthesis of (Z)- and (E)-alpha-fluoro-alpha,beta-unsaturated amides: the palladium-catalyzed carboamidation reaction of high E/Z and (Z)-1-bromo-1-fluoroalkenes lead to pure (Z)- and (E)-alpha-fluoro-alpha,beta-unsaturated amides, respectively.  相似文献

(E)- and (Z)-(fluoroalkenyl)boronates were prepared stereospecifically by the reaction of 2-fluoroalkylideneiodonium ylide generated from (E)- or (Z)-(2-fluoroalkenyl)iodonium salts with di(p-fluorophenoxy)alkylboranes, followed by transesterification to pinacol esters. The resulting pinacol esters of (fluoroalkenyl)boranes were used for the stereoselective synthesis of trisubstituted fluoroalkenes by cross-coupling reactions.  相似文献

With the aim of enhancing selectively the beneficial biological effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) a number of polyunsaturated fatty acids containing sulfur or oxygen atoms in the chain has been synthesized starting from EPA and DHA, respectively. Oxidative degradation of these acids led to the corresponding aldehydes all-(Z)-3,6,9,12-pentadecatetraenal and all-(Z)-3,6,9,12,15-octadecapentaenal. Reactions with DBU converted these aldehydes quantitatively into the conjugated isomers (2E,6Z,9Z,12Z)-pentadecatetraenal and (2E,6Z,9Z,12Z,15Z)-octadecapentaenal, respectively. The four aldehydes were transformed by a sequence of reactions comprising reduction to the alcohols, halogenation and substitution with mercapto esters into the corresponding sulfur containing polyunsaturated fatty acid esters. The oxygen containing esters were prepared from the respective alcohol by boron trifluoride catalysed reaction with ethyl diazoacetate.  相似文献

OsO4-catalysed dihydroxylation reactions of the aryl ketimine derivatives of (E)-gamma-amino-alpha,beta-unsaturated esters gave anti selectivity ranging from 6.7 : 1 to 19 : 1, whereas the opposite syn selectivity was consistently observed with those of (Z)-gamma-amino-alpha,beta-unsaturated esters (5.4 : 1 to > 100 : 1).  相似文献

Experimental data on the stereoselectivity of base-catalyzed 1,2-elimination reactions that produce conjugated carbonyl compounds are scarce in spite of the importance of these reactions in organic and biochemistry. As part of a comprehensive study in this area, we have synthesized stereospecifically-deuterated beta-tosyloxybutanoate esters and thioesters and studied the stereoselectivity of their elimination reactions under non-ion pairing conditions. With the availability of both the (2R*,3R*) and (2R*,3S*) diastereomers the innate stereoselectivity could be determined unambiguously. (1)H and (2)H NMR data show that these substrates produce 5-6% syn elimination, the usual amount for acyclic substrates undergoing E2 reactions. Contrary to earlier suggestions, activation by a carbonyl group has virtually no influence upon the stereoselectivity. Elimination of the (2R*,3R*) diastereomer of the beta-tosyloxyester and thioester produces 21-25% of the (Z)-alkene, much more than observed with a poorer beta-nucleofuge. A relatively large amount of (Z)-alkene product seems to be a good marker for an E2 pathway, in which the transition state is E1cB-like, rather than an E1cB(irrev) mechanism. Syn KIE values were higher than those for anti elimination for the esters as well as the thioesters. Experimental challenges to the synthesis of stereospecifically-deuterated beta-tosyloxyesters are discussed.  相似文献

[reaction: see text] The (Z)-2,3-difluoro-3-stannylacrylic ester is readily prepared from (Z)-1,2-difluorovinyltriethylsilane via stereospecific stannyl/silyl exchange with KF/(Bu3Sn)2O or Bu3SnCl in DMF at 70 degrees C. The corresponding (E)-2,3-difluoro-3-stannylacrylate is prepared by stereospecific carbonylation of (E)-1,2-difluorovinyl iodide followed by low temperature/in situ stannylation of the resultant (Z)-2,3-difluoroacrylic ester. With Cu(I) iodide and Pd(PPh3)4 catalysis, the (Z)- and (E)-stannylacrylate esters readily couple with aryl iodides and vinyl bromides, as well as 2-iodothiophene, at room temperature to stereospecifically produce the respective (E)- and (Z)-2,3-difluoro-3-aryl substituted acrylic esters or conjugated dienes in high yields.  相似文献

Many addition reactions on Si-P or Ge-P bonds of 1,2-dimethyl 1-diethylphosphinosila (or germa) cyclopentanes to unsaturated dipolar reagents (aldehydes, ketones, heterocumulenes, ethylenic esters and nitriles) have been carried out. Abstract

Studies were performed on the reaction between alkyl- and arylselenyl chlorides with esters of allenic phosphonic acids, variously substituted at their terminal C-atom. With the esters of the C-3 diasubstituted acids the reaction is highly regioselective and only oxaphosphol heterocyclization occurs to 2,5-dihydro-1,2-oxaphosphols 21-e. With the esters of the propadiene phosphonic acid the reaction is regio- and (Z)-stereoselective: only the 2,3-adducts 3a-b are formed, where the (Z)-isomer and the allenic phosphonates 4a-b are prevalent, as a result of 1,3-sigmatropic isomerization of 3a-b. With the esters of the C-3 monosubstituted acids, complex reaction mixtures are formed. From them were isolated: (E)- and (Z)-isomers of 2,5-dihydro-1,2-oxaphosphols 2f-j; (E)-and (Z)-isomers of the 2,3-adducts 3c-h; (E)-and (Z)-isomers of 4c-f; and (E)-and (Z)-isomers of the 1,2-adducts 5a-d. We report the preparation of the deuterated tetradecanoic acids [2,2,3,3-(2)H(4)]-, [2,2,3,3,10,10-(2)H(6)]-, and [2,2,3,3,13,13-(2)H(6)]-tetradecanoic acids (1, 2, and 3, respectively) and their use to investigate the mechanism of the enzymatic transformation of (Z)-11-tetradecenoic acid into (E,E)-10,12-tetradecadienoic acid. Probes 2 and 3 were prepared from intermediate ketones 7 and 10, which were transformed into the labeled bromides 17 and 18 by reduction with NaBD(4), tosylation of the resulting alcohol, replacement of the tosyloxy group by deuteride with LiAlD(4), hydrolysis, and reaction with N-bromosuccinimide. The resulting bromides were converted into the alpha-acetylenic esters 21 and 22, respectively, and the additional deuterium labels were introduced by reduction of the conjugated triple bond with Mg in deuterated methanol. The same sequence of reactions starting with 11-bromoundecane afforded 27. Saponification of the labeled esters 23, 24, and 27 gave the deuterated acids 2, 3, and 1, respectively. The results of the biochemical experiments showed that C10-H removal, but not elimination of C13-H, was sensitive to deuterium substitution in the transformation of (Z)-11-tetradecenoic acid into (E,E)-10,12-tetradecadienoic acid, which is consistent with the hypothesis that this desaturase reaction involves a first slow, C10-H bond cleavage, with probable formation of an unstable allylic intermediate, followed by a second fast C13-H bond removal and concomitant rearrangement.  相似文献

Lewis acid catalyzed intramolecular Diels-Alder reactions of trienes (E,E,Z)-1a-d, (E,E,Z)-4a-d, and (E,Z,Z)-7a,b are described. Trienes containing enal or enone dienophiles cyclize in excellent yield under mild conditions using substoichiometric amounts of MeAlCl(2), in most cases with high levels of diastereoselectivity. The thermal IMDA reactions of 1a, 4a, and 7a require forcing conditions and proceed in low yield with reversed stereoselectivity in the cases of 1a and 4a.  相似文献

The E/Z ratio of lactone was high regardless of the R group (auxiliary) or the initial alkene stereochemistry. [reaction: see text] A study of the effect of Michael acceptor stereochemistry on the efficiency of intramolecular Morita-Baylis-Hillman (MBH) reactions has been performed. The reactions were catalyzed by a phosphine, and the reaction substrates studied were enones containing a pendant aldehyde moiety attached at the beta-position of the alkene group. In all cases examined with PPh3 as the catalyst, cyclization substrates possessing (Z)-alkene stereochemistry afforded a much higher yield of the desired product than did the E isomeric substrates under identical reaction conditions. This was also true when a polymer-supported phosphine catalyst was used. While both alkene isomers afforded the same product, in parallel reactions, the Z isomer afforded 2.5-8.5 times higher yield than did the corresponding E isomer. It is proposed that steric effects are a possible source of this dramatic difference in reactivity. Substrates where the beta-substituent is cis to the electron-withdrawing substituent are relatively more accessible to react with the nucleophile catalyst than are their trans counterparts. These findings are expected to be useful in the design of synthetic intermediates, as intramolecular MBH reactions are being increasingly used in the preparation of complex synthetic targets.  相似文献

New thermo-crosslinking reactions of poly(glycidyl methacrylate), copolymers of glycidyl methacrylate with methyl methacrylate, styrene or ethyl acrlate with various active esters such as di[S-(2-benzothiazoly)] thioadipate (BTAD), di(S-phenyl) thioadipate (PTAD), di(4-nitrophenyl) adipate (NPAD), diphenyl adipate (PAD), and di(S-phenyl) thioisophthalate (PTIP), and other polyfunctional esters were carried out in the film state using various catalysts such as quarternary ammonium or phosphonium salts, tert amines, or the crown ether 18-crown-6 = potassium salts system. Addition reactions of pendant epoxide groups in the polymer with the active esters such as NPAD and PTAD proceeded selectively to give gel compounds without other side reactions. The rates of reaction with the thioesters such as BTAD and PTAD were relatively faster than those with the phenyl esters such as PAD and NPAD at 70°C. The rates of reactions with the esters having flexible segments such as PTAD were also faster than those with the esters having rigid skeletons such as PTIP. Furthermore, it was found that the rate of reaction was affected strongly by reaction temperature, catalyst concentration, length of alkyl chain in the catalyst, kind of counterion of quarternary ammonium salts as a catalyst, content of pendant epoxide groups in the polymer, and kind of copolymer unit in the polymer, respectively.  相似文献

The treatment of alpha-bromo-alpha,beta-unsaturated esters 2 with FSO(2)CF(2)CO(2)Me and CuI in DMF/HMPA constitutes a new synthetic scheme for the preparation of alpha-trifluoromethyl-alpha, beta-unsaturated esters 3. The trifluoromethylation of (Z)/(E)-ethyl 3-[(S)-2,2-dimethyl-1,3-dioxolan-4-yl]-2-bromo-2-propenoate (2e), which is derived from 1-(R)-glyceraldehyde acetonide, yields the key intermediate alpha-trifluoromethyl-alpha,beta-unsaturated esters 3e. This is transformed into anomeric acetates 8a and 8b and is used for the synthesis of a number of 2', 3'-dideoxy-2'-trifluoromethylnucleosides.  相似文献

Alkenylalumination of substituted styrene oxides with [alpha-(ethoxycarbonyl)alkenyl]diisobutylaluminum, in the presence of BF(3).Et(2)O, affords the corresponding (Z)-alpha-alkylidene-gamma-aryl-gamma-hydroxy esters in 81-100% Z-selectivity. Chromatographic separation of isomers, followed by lactonization with trifluoroacetic acid, provides isomerically pure (Z)-alpha-alkylidene-gamma-aryl-gamma-butyrolactones in 53-78% overall yield. Isomerization of the (Z)-alkylidene hydroxyl esters using LDA, followed by protonation using a bulky proton source, such as BHT, provides a simple route to the corresponding alpha-(E)-alkylidene-gamma-phenyl-gamma-hydroxy esters in 72-78% yield, which were cyclized to obtain the corresponding (E)-butyrolactones in 78-85% yield.  相似文献
