模拟退火神经网络用于药物液相色谱梯度分离条件的优化。使用均匀设计法以乙腈在线性梯度展开时的初始浓度和线性梯度的斜率为优化参数,对六种药物混合体系进行优化。采用退火神经网络方法建立了有效的分离条件预测模型。对神经网络模型所预测的最佳分离条件进行试验,分离结果满意。模拟退火神经网络可有效地用于药物液相色谱分离条件的优化。 相似文献

五味子提取物高效液相色谱分析方法的优化 总被引:3,自引:1,他引:2
针对五味子乙醇提取物的复杂体系,借助于复杂样品分析系统软件(CSASS),根据组分在4次简单线性梯度下42个峰的保留时间,快速准确地计算出各组分的液相色谱保留参数a,c值和峰形参数σ,W1/2。借助这些参数,对五味子色谱的分离情况进行高精度仿真预测。在此基础上,应用移动重叠分辨分离图和谱图仿真技术,发展了计算机辅助的五味子提取物的高效液相色谱全局优化方法。在优化条件下,五味子提取物的高效液相色谱分析可在40 min内完成,且常量成分和部分低含量成分都能够得到较好的分离。所建立的方法已成功地用于五味子中化合物保留时间及峰形的预测,并在此基础上对其色谱分离条件进行优化。 相似文献

计算机辅助高效液相色谱(HPLC)分离条件优化可以低成本、快速地得到优化的分离条,因而已较为广泛地用于复杂样品的分离分析。基于移动重叠分离图方法,又发展了一种新型的多台阶梯度分离条件的优化方法可调移动重叠分离图法。该方法通过预测不同流动相条件下各组分的保留时间、峰宽和分离度,绘制出对于样品中各组分的重叠分离区域图。在对当前台阶流动相组成进行优化的同时,考虑其对后面一到两个台阶上流出组分保留的影响,实时地重新绘制对于后面台阶上流出组分的重叠分离区域图。通过观察当前台阶流动相条件对当前台阶和后面台阶上流出组分分离的影响,综合考虑样品中所有组分的分离情况,找到更接近全局最优的分离条件。通过扫描的方法对优化得到的分离条件进行微调,能够进一步提高分离效果。采用文献数据对可调移动重叠分离图法的应用加以说明,在二元流动相体系下,证明了该方法在HPLC方法建立方面的优越性。 相似文献

建立了梯度洗脱优化-离子色谱-脉冲安培检测技术分析婴幼儿配方乳粉中麦芽糖醇、半乳糖、葡萄糖、蔗糖、果糖、乳糖6种与代谢疾病紧密相关的糖和糖醇的方法。该方法对样品的前处理、色谱柱的选择、淋洗液的梯度等色谱条件进行了优化,重点研究了淋洗液梯度对各组分分离效果的影响。将梯度洗脱程序分解为弱保留组分分离、乳糖分离、基质消除、系统平衡4个子程序,并分别对其进行了条件优化,实现了6种组分的完全分离,并有效解决了弱保留组分色谱峰易重叠、乳糖色谱峰易拖尾等现象。6种组分的线性范围为0.5~100 mg/L,相关系数(r)>0.99,方法的检出限(S/N=3)为0.06~0.40 mg/L。该方法具有分离度好、灵敏度高等优点,可满足例行分析的要求,同时也为淋洗液的梯度优化提供了可借鉴的方法。 相似文献

Optimization procedure of gradient separations in ion-exchange chromatography using simplex optimization method in combination with the computer simulation program for ion-exchange chromatography is presented. The optimization of parameters describing gradient profile for the separation in ion chromatography is based on the optimization criterion obtained from calculated chromatograms. The optimization criterion depends on the parameters used for calculations and thus exhibits the quality of gradient conditions for the separation of the analytes. Simplex method is used to calculate new gradient profiles in order to reach optimum separations for the selected set of analytes. The Simplex algorithm works stepwise, for each new combination of parameters that describe the gradient profile a new calculation is performed and from the calculated chromatogram the optimization criterion is determined. The proposed method is efficient and may reduce the time and cost of analyses of complex samples with ion-exchange chromatography. 相似文献

An automated method for deconvolution of overloaded band profiles in gradient elution is described. The instrumental set-up consists of a pseudo-bidimensional HPLC system, where overloaded band profiles generated in the first direction are sampled on the second one. The method, previously employed under isocratic conditions, has been now extended to gradient elution, where one has to face the problem of band compression (and possibly band interference) during gradient, which decreases the time-window available along the first direction for sampling the overloaded profiles. The effect of the gradient steepness on the problem of defining a minimum number of sampling points to reconstruct single component bands from overloaded profiles is investigated. This approach is particularly useful in the framework of using inverse method (IM) to determine adsorption isotherm parameters for preparative gradient elution. In fact, it allows for the gathering of the information necessary to run IM calculations with minimum efford, also in cases where the individual component forming the mixture have different UV spectra. 相似文献

A gradient kinetic plot method is used for theoretical characterisation of the performance of polymeric particulate anion exchange columns for gradient separations of small inorganic anions. The method employed requires only information obtained from a series of isocratic column performance measurements and in silico predictions of retention time and peak width under gradient conditions. Results obtained under practically constrained conditions provide parameters for the generation of high peak capacities and rapid peak production for fast analysis to be determined. Using this prediction method, a maximum theoretical peak capacity of 84 could be used to achieve separation of 26 components using a 120 min gradient (Rs > 1). This approach provides a highly convenient tool for development of both mono- and multidimensional ion chromatography (IC) methodologies as it yields comprehensive understanding of the influence of gradient slope, analysis time, column length and temperature upon kinetically optimised gradient performance. 相似文献

The selectivity of size-exclusion chromatography (SEC) can be modified by adding non-ionic micelles to the mobile phase. Surfactant-aided size-exclusion chromatography (SASEC) can therefore very well be performed in a gradient mode on an SMB, as is reported in this paper. A method has been developed for correctly positioning a micellar gradient over an SMB. The method is applied for size-exclusion chromatography with the non-ionic surfactant C12E23 as gradient forming solute, and demonstrated by applying it to a relevant chromatographic protein separation problem. 相似文献

Analytical equilibrium gradient methods are non-linear separation methods in which the separation mechanism involves a force gradient along the separation channel. These methods can be classified into two categories: those in which the gradient is a field gradient applied along the separation channel (i.e., field gradient), and those in which the channel is subjected to a constant field with a gradient formed in some other property (i.e., constant field). Standard deviation of peak width, resolution and peak capacity are important parameters in characterizing equilibrium gradient methods, and general expressions can be obtained from considering both the point of force acting on the analyte and the basic flux equation. Several successful examples, such as density gradient sedimentation, isoelectric focusing and electromobility focusing are discussed. Based on equilibrium gradient methods in the field gradient category, a method to dynamically improve peak capacity is described. Summary For any point on a gradient extremal path, the gradient is an eigenvector of the hessian. Two new methods for following the gradient extremal path are presented. The first greatly reduces the number of second derivative calculations needed by using a modified updating scheme for the hessian. The second method follows the gradient extremal using only the gradient, avoiding the hessian evaluation entirely. A computer-assisted method is presented for optimization of mobile phase compositions and development distance in gradient two-step development HPTLC. The method is based on a system which can predict the final Rf values for gradient two-step development from values measured using five preliminary runs. This technical note describes a new bench-top method for producing anisotropic hydrogels composed of gradient layers of soluble factors, particles, polymer concentrations or material properties. Each gradient layer was produced by a previous gradient method in which a droplet of one precursor solution was added to a thin layer of a second solution. The ensuing rapid capillary flow along the open channel generated a gradient precursor solution, which was then crosslinked to form a gradient gel. Repeating these steps allowed a layered gel to be iteratively constructed with as many gradient layers as desired. This technique renders the synthesis of multi-layered gradient gels accessible to virtually any researcher and should help simplify the production of more biologically relevant cellular microenvironments. 相似文献
