 利用径向离子交换色谱法分离纯化了Nitschmann组分Ⅲ中的凝血酶原复合物 ( prothrombincomplexconcentrate ,PCC) ,流动相为pH 7.5的Tris HCl缓冲液 ,色谱柱为XK 1 6DEAEfastflowSepharose ( 0 .8cmi.d .× 5cm)及膜DEAE径向色谱柱 ( 3.0cmi.d .× 5.8cm)。通过改变不同上样流速及洗脱流速 ,研究了流速对所分离的凝血酶原复合物的蛋白质量浓度及凝固活性的影响 ,为今后在血浆蛋白分离纯化中进一步推广使用径向色谱技术和放大实验提供了依据。  相似文献

为了能从中华眼镜蛇蛇毒中简单快速地分离纯化神经生长因子(一种治疗各种神经性损伤和神经退行疾病的药物,简称NGF),采用不同的色谱柱联用的方式对NGF的纯化工艺进行了研究。结果表明,采用DEAE Sepharose F.F.和Sephadex G 50二步柱色谱工艺可以从蛇毒中快速分离得到神经生长因子。经十二烷基硫酸钠聚丙烯酰胺凝胶电泳和反相高效液相色谱分析, 证明所得到的NGF为单一组分,相对分子质量约为 29 000。对8 d龄鸡胚背根神经节采用体外培养法,结果证明,所得NGF具有明显的促进神经纤维  相似文献

A 0.46 cm x 10.0 cm silica monolith column was modified through the in situ covalent attachment of lysine (2,6-diaminohexanoic acid) groups. Due to the zwitterionic nature of the resultant stationary phase, the modified monolithic column contained both cation and anion exchange capacity. In the case of cation exchange, the capacity was found to be relatively low at between 5 and 6.5 micromoles Me2+ per column. However, as expected, the lysine monolith exhibited a higher anion exchange capacity at 12-13 micromoles A- per column (at pH 3.0), which was found to be dependent upon column pH, due to the dissociation of the weak acid carboxylic acid groups. High-performance separations of transition metal cations and inorganic anions were achieved using the modified monolith, with the effects of eluent concentration, pH and flow rate evaluated. Using elevated flow rates of up to 5 mL/min the separation of nitrite, bromate, bromide, nitrate, iodide and thiocyanate was possible in approximately 100 s with peak efficiencies of between 50 and 100,000 N/m and retention time %RSD of under 0.3%.  相似文献

采用乙醇分级沉淀、凝胶色谱纯化、阴离子交换色谱分离等步骤从灵芝深层发酵液中提取得到蛋白酶抑制剂GLPIA1 与GLPIA2。其中GLPIA2仅在215 nm处有紫外吸收,经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳鉴定为单一条带,相对分子质 量为15000。由其氨基酸组成分析谱图可看出,其酸性氨基酸含量较高,碱性氨基酸及芳香族氨基酸含量较低。GLPIA2抑 制剂的底物特异性研究表明,它对天冬氨酸族的胃蛋白酶和酵母蛋白酶A有相对较强的抑制作用。  相似文献

Auricularia polytricha is an edible mushroom, quite popular in China. Modern pharmacology research indicate that A. polytricha polysaccharides possess antitumor and immunomodulatory activities. In this paper, an antitumor Auricularia polytricha polysaccharide (APS)-2 was purified by radial flow chromatography (RFC) in a column of 500 mL bed-volume (40 cm i.d × 15 cm) packed with DEAE52-cellulose anion ion-exchange resin. The optimal separation conditions were: sample flow-velocity 15–20 mL min?1, sample volume 160–200 mL, and elution with distilled water and 0.2 M NaCl at a velocity of 35–45 mL min?1. The yield and content of APS-2 obtained under these conditions were 182 mg g?1 total polysaccharides and 98.35 %, respectively. The study provides guidelines for the separation and purification of polysaccharides using a radial flow chromatography column.  相似文献

A commercially available 4.6 mm id x 50 mm polymethacrylate-based monolithic strong anion exchange column (ProSwift SAX-1S) designed for the separation of proteins has been successfully used to separate small inorganic anions in the presence of a seawater sample matrix. Using a hydroxide eluent with suppressed conductivity detection the ion exchange capacity of this column declined over time; however, using KCl as the eluent, the column performance was stable with a capacity of 530 microequiv. for nitrate. The optimum conditions for the separation of iodate, bromate, nitrite, bromide and nitrate were assessed by constructing van Deemter plots using 1.00 and 0.100 M KCl. Efficiencies of up to 26 700 plates/m were recorded using 1.00 M KCl, at a flow rate of 0.20 mL/min but iodate was not baseline resolved from the void peak. By reducing the concentration of the eluent to 0.100 M, efficiencies of up to 39 900 plates/m could be obtained at 0.35 mL/min. By employing a linear gradient ranging from 0.05 to 1.00 M KCl the ions dissolved in distilled water or a salt water matrix could be baseline separated in less than 3 min at a flow rate of 2.50 mL/min.  相似文献

A method for determining cis-diamminedichloroplatinum(II) (CDDP), an anticancer drug, in plasma and urine by HPLC with UV detection and column-switching has been developed. Typical conditions were as follows. An apparatus was composed of two columns, two pumps, a UV detector, a sample injector with a 100 microL loop, a switching valve, a column oven and a recorder. A Rheodyne model 7125 sample injector was used as the switching valve. A precolumn (4.6 mm ID x 25 cm) was packed with MCI GEL CK10S (a strong cation exchanger), and an analytical column (4.6 mm ID x 5 cm) was packed with MCI GEL CDR10 (a strong anion exchanger). Both columns were connected in series via the switching valve. The CDDP-containing fraction of the effluent from the precolumn was loaded to the analytical column by column-switching and the effluent from the analytical column was monitored at 210 nm. An eluent of 0.3 M sodium dihydrogen phosphate was pumped at a flow rate of 1 mL/min and the columns were maintained at 40 degrees C. CDDP was eluted at about 11 min and the identity of the peak of CDDP on the chromatogram was confirmed by its 3-dimensional chromatogram and analysis of platinum in the column effluent. Under the conditions described above, a linear relationship was obtained between peak height and concentration of CDDP up to 100 microM. Correlation efficients were 0.998 for plasma and 0.999 for urine. The detection limit was 0.1 microM for CDDP in both plasma and urine (S/N = 3,0.005 AUFS). A specific and sensitive direct-injection high performance liquid chromatography electrospray ionization tandem mass spectrometry (HPLC/ESI-MS/MS) method has been developed for the rapid identification and quantitative determination of olanzapine, clozapine, and N-desmethylclozapine in human plasma. After the addition of the internal standard dibenzepin and dilution with 0.1% formic acid, plasma samples were injected into the LC/MS/MS system. Proteins and other large biomolecules were removed during an online sample cleanup using an extraction column (1 x 50 mm i.d., 30 microm) with a 100% aqueous mobile phase at a flow rate of 4 mL/min. The extraction column was subsequently brought inline with the analytical column by automatic valve switching. Analytes were separated on a 5 microm Symmetry C18 (Waters) analytical column (3.0 x 150 mm) with a mobile phase of acetonitrile/0.1% formic acid (20:80, v/v) at a flow rate of 0.5 mL/min. The total analysis time was 6 min per sample. The inter- and intra-assay coefficients of variation for all compounds were <11%. By eliminating the need for extensive sample preparation, the proposed method offers very large savings in total analysis time.  相似文献

建立了固相萃取-离子色谱(SPE-IC)测定饮用水中痕量卤代乙酸(HAAs)(包括一氯乙酸、二氯乙酸、三氯乙酸、一溴乙酸和二溴乙酸)的方法。固相萃取采用LiChrolut EN SPE柱来进行痕量待测物的预浓缩(25倍)和基体杂质的消除,用NaOH(10 mmol/L)洗脱;色谱分离采用亲水性、高容量、氢氧化物选择型阴离子交换柱Dionex IonPac AS16(250 mm×4 mm i.d.),以NaOH为流动相进行浓度梯度淋洗,淋洗速度为0.8 mL/min,电导检测,进样量为500 μL。结果表明,用SPE-IC法测定HAAs,一溴乙酸的检测限为12.5 μg/L,其余4种HAAs的检测限为0.38~1.69 μg/L。该法可实现对饮用水中痕量卤代乙酸的测定。  相似文献

Pulsed flow modulation (PFM) is based on higher flow rate time compression of the first GC column effluent, which prior to the injection into the second column is stored for a few seconds in a standard fused silica wide bore transfer line. We constructed the PFM device with two standard 1/16 in. brass compression fittings with the insertion of the two columns inside the wide bore 0.53 mm i.d. fused silica storage transfer line for the elimination of dead volumes. This simple arrangement provides a combination of flexibility in the length of the sample storage transfer line hence comprehensive two-dimensional gas chromatography (GC x GC) cycle time, inert sample path and full elimination of cooling gas consumption. A record short second column injection time of 20 ms is demonstrated. Practical injection times are the sample collection time (such as 4s) divided by the second to first column flow rate ratio (such as 20/0.7), which is typically around 150 ms. Due to the low cost of the device it can also be considered for use with non comprehensive time segmented GC x GC to remove a few accidental coelutions. PFM-GCxGC excels with high second column capacity due to the use of 0.32 mm i.d. columns with high flow rates as the second dimension GC x GC column. As a result, PFM-GCxGC can have up to two orders of magnitude higher second column sample capacity and linear dynamic range for improved reduction of adverse matrix interference effects due to column overloading.  相似文献

Several configurations using 6- and 10-port switching valves were studied for high flow, on-line extraction of rat plasma coupled to an electrospray triple quadrupole mass spectrometer. Each plasma sample was diluted 1:1 with an aqueous internal standard solution. The sample was injected into a 2.1 x 20 mm cartridge column packed with 25 microm divinylbenzene/N-vinylpyrrolidone packing using 100% aqueous mobile phase at 4 mL/min. After sample loading and sample cleanup, the analytes were eluted from the extraction column with a 1.0-min gradient at 0.4 mL/min. The samples were either analyzed directly after elution from the extraction column or after additional separation using a short high performance liquid chromatography (HPLC) column. The different configurations were tested using an acidic drug (diflunisal) and a basic drug (clemastine) in rat plasma. On-line analysis was performed by injecting 200 microL of diluted plasma. The mass spectrometer was operated in the multiple reaction monitoring (MRM) mode. All calibration standards gave relative standard deviations (RSDs) below 5%. A column switching LC method is presented for the analysis of fluoxetine (FLU) and norfluoxetine (NFLU) by direct injection of human plasma using a lab-made restricted access media (RAM) column. A RAM-BSA-octadecyl silica (C-18) column (40 mm x 4.6 mm, 10 microm) is evaluated in both backflush and foreflush elution modes and coupled with a C-18 lab-made (50 mm x 4.6 mm, 3 microm) analytical column in order to perform online sample preparation. Direct injection of 100 microL of plasma samples is possible with the developed approach. In addition, reduction of sample handling is obtained when compared with traditional liquid-liquid extraction (LLE) and SPE. The total analysis time is around 20 min. A LOQ of 15 ng/mL is achieved in a concentration range of 15-500 ng/mL, allowing the therapeutic drug monitoring of clinical samples. The precision values achieved are lower than 15% for all the evaluated points with adequate recovery and accuracy. Furthermore, no matrix interferences are found in the analysis and the proposed method shows to be an adequate alternative for analysis of FLU in plasma.  相似文献

A high performance liquid chromatographic method for the analysis of chlortetracycline (CTC) using postcolumn fluorescence detection has been developed. After chromatographic separation of CTC on a polystyrene-divinylbenzene copolymer column, a highly fluorescent derivative isochlortetracycline (iso-CTC) was formed postcolumn in an on-line reaction coil with the addition of 25% NaOH (w/v). Chromatographic separation was achieved on a PRP-1 column, 15 cm x 4.6 mm, with 27:73 acetonitrile:0.2% perchloric acid (v/v), at 1.0 mL/min. Fluorescence derivatization was achieved by the on-line addition of 25% NaOH (w/v), at a flow rate of 0.2 mL/min, into the column eluant in a post-column reaction coil. The reaction coil was 9 m of teflon (1/16 in o.d., 0.3 mm i.d.) knitted into a six-sided coil. The fluorescent derivative was detected at lambda ex 355 nm and lambda em > 389 nm. Using this method after a simple sample cleanup, CTC can be detected in milk at 0.04 micrograms/mL, which is comparable to that obtained by microbiological assays. The detection method was linear between 0.02 micrograms/mL and 4 micrograms/mL. Because of the chromatographic separation, the method is more selective than microbiological assays and more sensitive than ultraviolet detection. With the chromatographic system described, the keto tautomeric forms of CTC and 4-epi-CTC are separated in a system which minimizes their formation on-column. In acidic aqueous organic solutions, the keto tautomer of CTC is the only product formed to any significant amount.  相似文献

In this study, a 38 mL monolith with homogeneous porous structure was produced by a single polymerization from glycidyl methacrylate (GMA) and ethylene dimethacrylate (EDMA) in the presence of porogens and an initiator. The uniform temperature distribution within the reaction system was achieved by adding reactant mixture continuously and enhancing the heat transfer ability of the polymerization system. Homogeneous porous structure in the monolith was proved by SEM and the pore size distribution profiles measured by mercury intrusion porosimetry. Experimental results from proteins separation indicated that the dynamic capacity and resolution of radial flow monolithic column were independent of flow rates. Furthermore, the pressure drop on the column was linearly dependent on the flow rate and did not exceed 1.7 MPa even at a flow rate of 50 mL/min, which proved that the prepared monolith could be used in the quick separation and preparation of biopolymers.  相似文献

建立了一种灵敏度高、操作简单的定量分析水产品中残留的微量吡喹酮的高效液相色谱方法。样品经乙酸乙酯提取、极性硅胶柱净化后进行高效液相色谱分析,以乙腈-水(体积比为50∶50)为流动相,流速为0.9 mL/min,ZORBAX C18色谱柱分离,紫外检测波长为214 nm。在此条件下吡喹酮在0.02~20 mg/L范围内呈线性关系,相关系数为0.99998;吡喹酮的加标回收率在85%以上,检出限为10 μg/kg。  相似文献

结合连续加料降低聚合体系的放热量,及强化体系的换热能力等措施,实现了无搅拌模式聚合体系中温度分布的均一化。通过单次聚合过程构造了体积为38mL的大体积整体固定相。SEM及孔隙率分布测试证明了整体固定相具有均匀的孔结构,动态容量测定及分离蛋白质的实验,表明所制备的径向流动模式整体柱的容量及分辨率不依赖于流速,同时在高流速下具有低背压的特征,可用于生物大分子的分离制备。  相似文献

A method has been developed for the analysis of a broad spectrum of pharmaceuticals using packed column supercritical fluid chromatography (pSFC) on a cyanopropyl silicagel stationary phase. Five 25 cm x 4.6 mm I.D., 5.0 microm columns were coupled to generate ca. 100000 plates. The selectivity was tuned by varying the nature and concentration of various modifiers and additives in the carbon dioxide mobile phase. It was noted that pressure influences both efficiency and selectivity of the chromatographic process. Final method conditions are: outlet pressure 100 bar, flow 2.0 mL/min, temperature 40 degrees C, organic modifier program from 5% (1 min) to 40% at 2.0%/min, organic modifier composition methanol:acetonitrile in a ratio of 3:1 (variable according to sample composition) with peak symmetry additives trifluoroacetic acid and diisopropylamine both at levels of 0.5%.  相似文献
