A procedure for separation and quantitation of methylmercury by capillary electrophoresis using sample stacking as the injection technique is presented. The CE conditions have been optimized in order to separate the methylmercury from the excess cysteine peak and to concentrate large volumes of sample obtaining a low detection limit. Under the proposed operational conditions, the detection limit (S/N = 3) was 12 ng g and the limit of quantitation (S/N = 10) was 20 ng g−1 with a linear range of 20–100 ng g−1 (as methylmercury in samples). The method was tested using different reference materials with a certified methylmercury content.  相似文献

A large volume sample stacking procedure (LVSS) is developed here for metallothionein (MT) determinations in rabbit liver by using capillary electrophoresis with UV detection. A 10-time improvement in concentration-based LODs, when compared to the normal stacking mode (CE-UV analysis of samples solved in water), is achieved.The methodology is successfully applied to analysis of MTs in eel liver cytosolic extracts, preceded by an easy cleaning-up pre-treatment in order to eliminate the high salt content. Analysis of cytosol obtained from a group of eels previously exposed to Cd (induced group) resulted in several isoforms of MTs with differences in absorbance signal compared to a control group.  相似文献

Summary A systematic investigation of operational buffer systems, sample preparation and instrument parameters for achieving the best possible performance for determinating an homologous series of N-benzyl-N-alkyl-N,N-dimethylammonium chloride compounds by capillary zone electrophoresis with direct UV detection. The most effective separation was achieved within 3.5 min with the addition of acetonitrile (40%) in a phosphate buffer (20 mM pH 5.2) using a 40 cm fused-silica capillary operating at 25 KV and 20°C. Degassing of all electrolyte solutions and samples was very important. The linearity and repeatability for each compounds were satisfactory. To improve detection limits, on-column sample preconcentration, sample stacking, was investigated achieving a tenfold enrichment factor and quantitation limits about 10−7M.  相似文献

The condensation of low abundance zwitterion substance, such as protein and peptide, has great significance to the study on proteomics. This paper develops the theory on design of online stacking conditions of zwitterion by a moving reaction boundary (MRB) in capillary electrophoresis (CE). This concerns the choice of running and sample buffers, velocity design of MRB, and salt effect on the stacking. The theoretical results unveil that: (1) the velocity of MRB formed with weak acidic buffer and strong alkali should be set between zero and the velocity of zwitterion in the alkali phase, or no stacking occurs; (2) if a strong alkali is used to prepare the sample, a much long front plug of strong base must be injected before the alkaline sample plug for complete stacking, whereas no such front plug is needed if a weak alkali with enough high concentration and pH value is used to prepare the sample buffer; (3) the existence of salt in sample matrix has a weak effect on the stacking of zwitterion if sample is prepared with weak alkaline buffer, while has a dramatic effect on the same stacking if with a strong base buffer. In addition, the concentration of weak alkali used for preparation of sample should be set at the point, at which the velocity of MRB is as much as possible close to that of negative zwitterion. The developed theory and its computation are quantitatively proved by the experiments of zwitterion stacking by the MRB as shown in the previous and the accompanying papers. The proposed theoretic results hold obvious significances on-column stacking of low abundance zwitterions, such as amino acid, or peptides or proteins, in CE.  相似文献

To demonstrate the theoretic method on the stacking of zwitterion with moving reaction boundary (MRB) in the accompanying paper, the relevant experiments were performed. The experimental results quantitatively show that (1) MRB velocity, including the comparisons between MRB and zwitterionic velocities, possesses key importance to the design of MRB stacking; (2) a much long front alkaline plug without sample should be injected before the sample injection for a complete stacking of zwitterion if sample buffer is prepared with strong base, conversely no such plug is needed if using a weak base as the sample buffer with proper concentration and pH value; (3) the presence of salt in MRB system holds dramatic effect on the MRB stacking if sample solution is a strong base, but has no effect if a weak alkali is used as sample solution; (4) all of the experiments of this paper, including the previous work, quantitatively manifest the theory and predictions shown in the accompanying paper. In addition, the so-called derivative MRB-induced re-stacking and transient FASI-induced re-stacking were also observed during the experiments, and the relevant mechanisms were briefly demonstrated with the results. The theory and its calculation procedures developed in the accompanying paper can be well used for the predictions to the MRB stacking of zwitterion in CE.  相似文献

Capillary zone electrophoresis (CZE) with UV detection was used to determine vanadium species. Nitrilotriacetic acid (NTA), hydroxyethylethylenediaminetriacetic acid (HEDTA), ethylenediaminetetraacetic acid (EDTA), diethylenetriaminepentaacetic acid (DTPA), ethylene glycol-bis(2-aminoethylether)-tetraacetic acid (EGTA) and 2,6-pyridinedicarboxylic acid (PDCA) were investigated to determine whether these ligands formed stable anionic complexes with vanadium. Of all the ligands studied HEDTA was the most suitable ligand because it gave the largest UV response with reasonable migration time. Electrospray mass spectrometry (ES-MS) was used to confirm the formation of [VO2(HEDTA)]2− and [VO(HEDTA)]1− in solution. An electrolyte containing 25 mM phosphate, 0.25 mM tetradecyltrimethylammonium bromide (TTAB) at pH 5.5 was optimum for the separation of these anionic vanadium complexes. Sample stacking techniques, including large-volume sample stacking (LVSS) and field-amplified sample injection (FASI), were tested to improve the sensitivity. Best sensitivity was obtained using FASI, with detection limits of 0.001 μM, equivalent to 0.4 μg L−1, for [VO2(HEDTA)]2− and 0.01 μM, equivalent to 3.4 μg L−1 for [VO(HEDTA)]1−. The utility of the method for the speciation of V(IV) and V(V) was demonstrated using ground water samples.  相似文献

Large volume sample stacking using electrosmotic flow pump (LVSEP) in capillary electrophoresis is a useful tool for on-line concentration of low-abundant protein samples for proteomics research. While the stacking of only small anions with high pKa values has been reported at low pH buffers (pH<4), protein samples (trypsin inhibitor and α-lactalbumin) with high pI values (pI>4) were stacked successfully in our system using LVSEP. Initially, the whole column of the SIL, or LPA-coated capillary was filled with protein samples (1 μg/ml), then a high voltage was applied without polarity switching with high pH buffer (pH 8.5). An enhancement factor of more than 100 times was achieved compared to the conventional pressure injection method.  相似文献

The reason why a moving reaction boundary (MRB) can stack analyte in highly saline sample in capillary electrophoresis [C.X. Cao, Y.Z. He, M. Li, Y.T. Qian, S.L. Zhou, L. Yang, Q.S. Qu, Anal. Chem. 74 (2002) 4167] is still unclear. To illuminate the mechanism of such stacking, three MRBs formed by formic acid-NaOH buffer and sodium formate as well as 40, 80 and 120 mmol/L sodium chloride in matrixes were studied. The computation with MRB theory shows that sodium chloride in matrix has weak effect on the stacking efficiency, whether the concentration of sodium chloride is set at 40, or 80, or 120 mmol/L. The conclusion has been highly manifested by numerous experiments. Furthermore, the computer simulation and theoretical analyses depict that this kind of stacking is induced by the mechanism of MRB, rather than that of electrostacking or isotachophoresis (ITP) under the given electrolytic system. Finally, the application of the sample condensation was achieved for the stacking of analyte(s) in highly saline biological sample of skeletonema costarum’ culture with up to 527 mmol/L total salt and health human urine with 150-320 mmol/L inorganic ions (Cl, Na+, K+, PO43−, etc.). The results herein have a clear significance to the design on stacking of analyte in highly saline biological sample.  相似文献

An electrophoretic stacking method has been developed for monitoring the therapeutic level of the antibiotic ceftazidime in blood plasma and microdialysates taken from peripheral soft tissues of the lower limbs of patients with diabetic foot syndrome. The biological samples are treated by addition of acetonitrile in an amount of 75% v/v and injected into a capillary in a large volume; after turning on the separation voltage, the residual acetonitrile is forced out of the capillary by the application of hydrodynamic pressure. The clinical samples were separated in an optimised background electrolyte composed of 50 mM chloroacetic acid +20% v/v methanol +0.5% v/v INST coating solution. The attained LOD for ceftazidime equalled 0.42 μg mL−1 (0.8 μM) and the migration time equalled 3.75 min when using a 25 μm capillary with minimum length of 31.5 cm. The separation was controlled by a maximum voltage of +30 kV and the movement of the analyte was accelerated by a pressure of 50 mbar. The RSD values for intra-day repeatability of the migration time and peak area are 0.14% and 3.8%, respectively; the inter-day values equalled 0.25% for the migration time and 7.3% for peak area, respectively. Pharmacological studies revealed that ceftazidime passes from the blood circulation to the peripheral tissues of the lower limbs with an efficiency of 20%. The introduction of CE control of ceftazidime level in diabetic foot represents a very important improvement in achieving the targeted therapeutic effect.  相似文献

mAbs are highly complex proteins that present a wide range of microheterogeneity that requires multiple analytical methods for full structure assessment and quality control. As a consequence, the characterization of mAbs on different levels is particularly product‐ and time‐consuming. CE‐MS couplings, especially to MALDI, appear really attractive methods for the characterization of biological samples. In this work, we report the last instrumental development and performance of the first totally automated off‐line CE‐UV/MALDI‐MS/MS. This interface is based on the removal of the original UV cell of the CE apparatus, modification of the spotting device geometry, and creation of an integrated delivery matrix system. The performance of the method was evaluated with separation of five intact proteins and a tryptic digest mixture of nine proteins. Intact protein application shows the acquisition of electropherograms with high resolution and high repeatability. In the peptide mapping approach, a total number of 154 unique identified peptides were characterized using MS/MS spectra corresponding to average sequence coverage of 64.1%. Comparison with NanoLC/MALDI‐MS/MS showed complementarity at the peptide level with an increase of 42% when using CE/MALDI‐MS coupling. Finally, this work represents the first analysis of intact mAb charge variants by CZE using an MS detection. Moreover, using a peptide mapping approach CE‐UV/MALDI‐MS/MS fragmentation allowed 100% sequence coverage of the light chain and 92% of the heavy chain, and the separation of four major glycosylated peptides and their structural characterization.  相似文献

An on-line sample stacking method, namely field-amplified sample injection, has been developed for the separation and determination of carnosine, anserine, and homocarnosine by capillary electrophoresis. Using electrokinetic injection, about 130- to 160-fold improvement of sensitivity was achieved without loss of separation efficiency when compared to conventional sample injection. For conventional injection, the samples were dissolved in running buffer and then hydrodynamically injected for 10 s (3.45 kPa). Various parameters affecting separation and sample stacking were optimized. Under optimum conditions, linear responses were obtained over two orders of magnitude and the detection limits (defined as S/N = 3) of carnosine, anserine, and homocarnosine were 1.5 x 10(-8) to 1.6 x 10(-8) mol/L.  相似文献

Ceria‐zirconia composites at different molar ratios were synthesized. Several methods were used to characterize these composites, including X‐ray photoelectron spectroscopy, surface area and surface acid‐base property detection. A one‐step method for isolation and identification of phosphopeptides from peptide mixture was created using these ceria‐zirconia composites. Using tryptic digest of standard phosphorylated protein, we have shown that these enrichment and dephosphorylation activities are effective. The adsorption capacity and catalytic property of ceria‐zirconia composites at different molar ratios and calcinated temperatures were studied. In combination with MALDI‐TOF‐based peptide mass finger printing technique, we have established a method to utilize the enrichment/dephosphorylation dual properties of these ceria‐zirconia composites for the analysis of phosphoprotein in nonfat milk successfully.  相似文献

The common SDS microemulsion (i.e. 3.3% SDS, 0.8% octane, and 6.6% butanol) and organic solvents were investigated for the stacking of cationic drugs in capillary zone electrophoresis using a low pH separation electrolyte. The sample was prepared in the acidic microemulsion and a high percentage of organic solvent was included in the electrolyte at anodic end of capillary. The stacking mechanism was similar to micelle to solvent stacking where the micelles were replaced by the microemulsion for the transport of analytes to the organic solvent rich boundary. This boundary is found between the microemulsion and anodic electrolyte. The effective electrophoretic mobility of the cations reversed from the direction of the anode in the microemulsion to the cathode in the boundary. Microemulsion to solvent stacking was successfully achieved with 40% ACN in the anodic electrolyte and hydrodynamic sample injection of 21 s at 1000 mbar (equivalent to 30% of the effective length). The sensitivity enhancement factors in terms of peak height and corrected peak area were 15 to 35 and 21 to 47, respectively. The linearity R2 in terms of corrected peak area were >0.999. Interday precisions (%RSD, n = 6) were 3.3–4.0% for corrected peak area and 2.0–3.0% for migration time. Application to spiked real sample is also presented.  相似文献

Acupuncture sample injection is a simple method to deliver well‐defined nanoliter‐scale sample plugs in PDMS microfluidic channels. This acupuncture injection method in microchip CE has several advantages, including minimization of sample consumption, the capability of serial injections of different sample solutions into the same microchannel, and the capability of injecting sample plugs into any desired position of a microchannel. Herein, we demonstrate that the simple and cost‐effective acupuncture sample injection method can be used for PDMS microchip‐based field amplified sample stacking in the most simplified straight channel by applying a single potential. We achieved the increase in electropherogram signals for the case of sample stacking. Furthermore, we present that microchip CGE of ΦX174 DNA‐HaeⅢ digest can be performed with the acupuncture injection method on a glass microchip while minimizing sample loss and voltage control hardware.  相似文献

An EOF counter-balanced ITP boundary has been used to stack anions from high conductivity samples during continuous electrokinetic injection of the sample. In a polystyrenesulfonate/poly(diallyldimethylammonium chloride) polyelectrolyte coated capillary, the time at which the ITP boundary exited the capillary could be prolonged by balancing the movement of the boundary with the EOF. Using a bis-tris-propane electrolyte, the ITP boundary was removed from the capillary within 7 min, while when using triethanolamine the ITP boundary was still at 30% of the capillary after 2 h of injection. Using these systems, the sensitivity of a mixture of simple organic acids in 100 mM Cl was improved by 700–800-fold using bis-tris-propane with a whole-capillary injection of the sample and 5 min of electrokinetic injection at +28 kV, and 1100–1300-fold using triethanolamine and 60 min of electrokinetic injection under the same conditions. The potential of the method to be applicable to high conductivity samples was demonstrated by stacking a whole capillary filled with urine spiked with naphthalenedisulfonic acid, with limits of detection 450 times lower than those achievable with a normal hydrodynamic injection.  相似文献

Simultaneous electrokinetic and hydrodynamic injection (SEHI) of organic cations (tricyclic antidepressant and beta blocker drugs) with on-line sample concentration using micelle to solvent stacking (MSS) was studied in micellar electrokinetic chromatography. Compared to conventional injection, >300-fold improvements in signals were obtained by hydrodynamic injection. However, with SEHI the amount of sample ions introduced into the capillary was increased which afforded a higher gain of up to 4000-fold without compromise to separation efficiency. The electrokinetic injection at negative polarity (anode at the detector end) introduced the micelle bound analytes. The hydrodynamic injection also maintained the MSS boundary inside the capillary. The stability of the MSS boundary affected SEHI where mild conditions that were low voltage as well as pressure injection were desired. The limits of detection were in the range from 0.6–4.2 ng mL−1. A strategy for optimization was described and the method was applied to the ng mL−1 analysis of spiked wastewater after simple dilution and centrifugation.  相似文献

This study describes approaches for stacking a large volume of sample solutions containing a mixture of mercaptopurine monohydrate, 6-methylmercaptopurine, thioguanine, thioguanosine, and thioxanthine in capillary electrophoresis (CE). After filling the run buffer (60 mM borate buffer, pH 8.5), a large sample volume was loaded by hydrodynamic injection (2.5 psi, 99.9 s), followed by the removal of the large plug of sample matrix from the capillary using polarity switching (-15 kV). Monitoring the current and reversing the polarity when 95% of current recovered, the separation of anionic analytes was performed in a run buffer < 20 kV. Around 44- to 90-fold improvement of sensitivity for five analytes was achieved by large-volume stacking with polarity switching when compared with CE without stacking. This method was feasible for determination of the analytes spiked in plasma. Removing most of electrolytes from plasma is a key step for performing large-volume sample stacking. Solid-phase extraction was used for pretreatment of biological samples. To our knowledge, this study is one of few applications showing the possibilities of this stacking procedure to analyze biological samples by large-volume sample stacking with polarity switching (LVSSPS) in CE.  相似文献

The main aim of this work was to demonstrate the applicability of capillary zone electrophoresis in combination with field enhanced sample stacking in targeted metabolome analyses of adenine nucleotides – AMP, ADP, ATP, coenzymes NAD+, NADP+ and their reduced forms in Paracoccus denitrificans. Sodium carbonate/hydrogencarbonate buffer (100 mM, pH 9.6) with the addition of β‐CD at a concentration of 10 mM was found to be an effective BGE for their separation within 20 min. Besides this, special attention was paid to the development of the procedure for the extraction of specific metabolites from the bacterium P. denitrificans. This procedure was not only optimised to achieve the highest metabolite yields but also to obtain a sample that was fully compatible with the online preconcetration strategy used. The developed methodology was finally applied in a study of the bacterium P. denitrificans at various stages of the active respiratory chain.  相似文献

Different sample handling methods for hydrophobic proteins and peptides were evaluated in association with the utilization of a structured matrix-assisted laser/desorption ionization (MALDI) target for increased sensitivity. The fluorinated organic solvent hexafluoroisopropanol (HFIP) was used for the solubilization of both the full-length protein bacteriorhodopsin (BR) and a cyanogen bromide digest thereof, and compared to the performance of the non-ionic detergents octyl--d-glucopyranoside (OG), dodecyl--d-maltoside (DM), and Triton X-100. A concentrating effect was seen when using the structured MALDI plate for BR dissolved in all the different detergents, of which OG generated the best-quality spectra for the full-length integral membrane protein as well as for the hydrophobic peptides. However, the uneven analyte distribution obtained with the detergent preparations required selective and thus time-consuming acquisition of spectra. When instead HFIP was used as sample solvent, a tenfold increase in sensitivity was achieved for full-length BR. Addition of acids to the HFIP-solubilized sample, or to the MALDI matrix solution, improved the signals for a few of the peptides, while degrading the spectra of others. Consequently, the addition of acid could be used as a complementary sample preparation method for hydrophobic peptides. On-target washing to remove contaminants (e.g., salt) was performed, and a recrystallization protocol for signal improvement specifically suited for hydrophobic peptides is described. Results from digestion and solubilization in different micro centrifuge tubes were examined to determine the influence of different materials on the possible sample loss due to wall adhesion. Studies of sample solution storage times suggest immediate analysis after solubilization to obtain best results.  相似文献

In this work, we demonstrate the applicability of electroextraction (EE) to urine metabolites. To investigate which urine metabolite classes are susceptible to EE, off‐line EE experiments were carried out with a prototype device, in which urine metabolites were electroextracted from ethyl acetate into water. The obtained extracts were examined with direct infusion MS and the results demonstrated that several compound classes could be extracted, amongst which amino acids and acylcarnitines. Acylcarnitines were selected for evaluation of the performance of EE. For this, the EE setup was adapted to capillary EE (cEE) to be able to analyze large urine sample series, and it was coupled online to LC‐MS. cEE‐LC‐MS of acylcarnitines was optimized and characterized. The recovery, linearity, repeatability, and detection limit of the cEE‐LC‐MS method was good to excellent. To demonstrate the versatility of EE for sample preparation in analytical procedures, extracts were injected into a CZE‐MS system, resulting in detection of the acylcarnitines along with more than 100 presumed metabolite peaks. The results presented here indicate that EE can be used as a fast sample preconcentration technique of low abundant urine metabolites, in combination with both LC and CZE.  相似文献
