A general, stereoselective synthesis of 4,5-disubstituted imidazolidines-2-ones from alpha-aminoacids has been developed: the key steps are a Blaise reaction of bromoacetate on alpha-aminonitriles and further reduction. Although reduction with sodium cyanoborohydride afforded a mixture of cis and trans isomers 6a-e with moderate to good stereoselectivity, reduction with sodium in liquid ammonia gave the trans isomers 8a-e with complete stereoselectivity. Acidic hydrolysis of the urea gave 4-amino-pyrrolidinones, which can be precursors to beta,gamma-diaminoacids or 3-aminopyrrolidines.  相似文献

Cross coupling of 2-arylpyrrole with benzoylbromoacetylene over aluminum oxide at room temperature gave 45–94% of 2-(benzoylethynyl)-5-arylpyrroles. Intermediate 2-(2-benzoyl-1-bromoethenyl)-5-arylpyrroles were isolated in up to 19% yield. The reaction was accompanied by formation of less than 5% of adducts of the initial pyrroles with the cross-coupling products, 2-benzoyl-1,1-bis(5-arylpyrrol-2-yl)ethenes.  相似文献

It has been shown that the most efficient catalysts for the synthesis of 4,5,7-trimethyl-4,5,6,7-tetrahydropyrrolo[3,2-c]pyridine from 1,2,5-trimethylpyridin-4-one and acetylene under Trofimov conditions are rubidium and potassium hydroxides. Use of Triton B or a mixture of trimethylbenzylammonium chloride with rubidium hydroxide as catalyst gives O-alkylated oxime. Their configurations and conformations were established through separation of the individual isomers of 1-vinyl-4,5,7-trimethyl-4,5,6,7-tetrahydropyrrolo[3,2-c]pyridines. Acetyl nitrate nitration of the cis isomer of this compound gave the 2- and 3-nitro derivatives. Similar nitration of 4,5,7-trimethyl-4,5,6,7-tetrahydropyrrolo[3,2-c]pyridine gave the 2-nitro-7-hydroxy derivative.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 10, pp. 1375–1380, October, 1991.  相似文献

1-[2-Arylthio(oxy)ethyl]-5-benzotriazolyl-2-pyrrolidinones 6a-e, 12 and 3-benzotriazolyl-2-[2-arylthio(oxy)ethyl]-1-isoindolinones 9a-f, 14 are readily available from reactions of benzotriazole (4), 2-(arylsulfanyl)ethylamines 3, or 2-phenoxyethylamine (11) with 2,5-dimethoxy-2,5-dihydrofuran (5) or 2-formylbenzoic acid (8). Lewis acid mediated cyclizations of 6 and 9 produced novel 1,4-benzothiazepines 7a-e and 10a-f, respectively. Cyclizations of 12 and 14 gave 1,4-benzoxazepines 13 and 15, respectively.  相似文献

A new synthesis of certain pyrimido[5,4-e]-as-triazine 4-oxides and their ring transformation to pyrrolo-[3,2-d]pyrimidines by the 1,3-dipolar cycloaddition reaction is described. Thus, reaction of 6-hydrazino-1,3-dimethyluracil ( 1 ) with triethyl orthoformate gave 6-ethoxymethylenehydrazino-1,3-dimethyluracil ( 2 ). Treatment of 2 with arylamines gave 6-arylaminomethylenehydrazino-1,3-dimethyluracils ( 3a-e ). Nitrosative cyclization of 3a-e with sodium nitrite afforded 3-arylaminofervenulin 4-oxides ( 6a-e ). Reaction of 6a-e with acetylenic esters yielded 7-alkoxycarbonyl-6-arylamino-1,3-dimethylpyrrolo[3,2-d]pyrimidine-2,4(1H,3H-diones ( 15a-e and 16 ).  相似文献

Photocyclization reactions were carried out on ethyl 2-(8-oxo-5,6,7,8-tetrahydro-1-naphthyloxy)acetates 1a-e and ethyl 2-(5-oxo-6,7,8,9-tetrahydro-5H-benzocyclohepten-4-yloxy)acetates 2a-e in acetonitrile. Irradiation of 1a-e gave naphtho[1,8-bc]furanols 3a-e and naphtho[1,8-bc]furans 4a-e in 33–83% yields and ethyl acrylates 5b-d were produced in 3–25% yields during irradiation of 1b-d . On the other hand, 2a-e afforded cyclohepta[ad|benzofuranols 6a-e and cyclohepta[ad]benzofurans 7a-e in 44–87% yields. Ethyl acrylates 8b-d were also produced in 7–43% yields from irradiation of 2b-d . Substituent effects on photocyclization and reaction pathways are discussed.  相似文献

New v-triazolo[1,5-α]- and v-triazolo[1,5-c]pyrimidinium salts 12a-e, 13a-c have been synthesized via oxidation (i.e. cyclodehydrogenation) of the appropriate pyrimidyl ketone arylhydrazones 3a-e, 6a-c using TBB (2,4,4,6-tetrabromocyclohexa-2,5-dien-1-one) as the reagent. The arylhydrazones were obtained by standard reactions; the Grignard reaction of 2-cyano- and 4-cyanopyrimidine 1a,b, 4a-c gave 2-pyrimidyl- and 4-pyrimidyl ketones 2a-e, 5a-c , which reacted with arylhydrazines to yield the desired ketone arylhydrazones 3a-e, 6a-c.  相似文献

Some novel Schiff bases have been prepared by reacting 6-azido-5-formyl-2-pyridone 1 with a series of aromatic amines 2a-f. 5-Arylaminomethylene-6-(E)-aryl-iminopyridones 3a-e were obtained by reaction of 1 with 2a-e at room temperature, whereas with 2f, the 6-azido-5-naphthalen-2-yl-iminomethylpyridone derivative 4 was formed. On the other hand, heating 1 with 2a-d at 140-150 degrees C yielded two sets of isomeric products, (E)-3a-d and (Z)-5a-d. Refluxing compounds (Z)-3a,c with hydroxyl-amine in methanol gave the corresponding hydroxyliminopyridones 8a,c. Heating of (E)-3a-d with excess POCl3 at reflux did not give the expected tricyclic compound 9, but rather the isomeric products (Z)-5a-d were obtained. The structures of all these products have been characterized using IR and 1H- and 13C-NMR spectroscopy.  相似文献

The reaction of 1-methyl-4-piperidone ( 1 ) with amyl nitrate in the potassium tert-butoxidetetrahydrofuran system gave dipotassium 5-methyl-2-oxo-1,3-piperidinedinitronate ( 4 ) in 78% yield. Similar treatment of 1-methyl-2-pyrrolidinone ( 2 ) afforded potassium 3-methyl-2-oxopyrrolidinenitronate ( 7 ) in 85% yield. In contrast, the nitration of 1,3-dimethyl-2-pyrrolidinone ( 3 ) led to opening of the lactam ring with the formation of amyl 2-aza-2-methyl-5-nitrohexanoate ( 10 ) in 40% yield. Acidification of disalt 4 did not cause ring opening but gave the dipolar ion of 1-methyl-3-nitro 4-hydroxy-5-aci-nitro-Δ3-tetrahydropyridinium (6).  相似文献

Cyclization of acyclic C-glycoside derivatives 1a,b to 2a,b as the major isomers, and 4a,b as the minor isomers were carried out. The isopropylidene derivatives 3a,b were prepared, as well as the hydrazide derivative 6, which was condensed with a variety of aldehydes to give hydrazones 7a-e which were also prepared from the compounds 12a-e. Acetylation of 7a,d gave the corresponding acetyl derivatives 8a,d, respectively. In addition, the dicarbonyl compound 9 was prepared in the hydrate form, which reacted with a number of aroylhydrazines to give the corresponding bisaroylhydrazones 10a-d, which were cyclized into 1,3,4-oxadiazoles 11a-d. Furthermore, two of the prepared compounds were examined to show the ability to activate MAO-B. In addition a number of prepared compounds showed antibacterial and antiviral activities.  相似文献

2,4-Bis(4-methoxyphenyl)-1,3,2,4-dithiadiphosphetane-2,4-disulfide (Lawesson's Reagent, LR, 1 ) reacts with cycloalkylidenecyanothioacetamides ( 2 and 3 ) to give 5'-cyano-2'-(4-methoxyphenyl)spiro [cyclopentane(cyclohexane)-1,6'-perhydro-[1,3,2]thiazaphosphixane]-2',4'-disulfide ( 4 and 5 ). The reaction of compounds 4 and 5 with f -halo compounds led to the formation of the substituted thio-compounds 6a-e and 7a-e , respectively, these compounds, upon treatment with sodium ethoxide, produce the corresponding thienothiazaphosphixine derivatives 8a-e and 9a-e respectively. Compounds 8a-e and 9a-e react with LR under different reaction conditions to give polyfused heterocyclic compounds 10a-d and 11a-d respectively. Treatment of compounds 8b and 9b with CS 2 and (CH 3 ) 2 SO 4 gave the corresponding dithiocarbamate methyl ester derivatives 12 and 13 , respectively, which on treating with hydrazine hydrate yielded compounds 14 and 15 respectively. Compounds 14 and 15 reacted with LR to yield compounds 16a , b and 17a , b respectively.  相似文献

A novel general method for the synthesis of isomeric 3(4)-nitro-4(3)-R-furoxans is developed. 3-Nitro isomers were obtained by reaction of hydroximoyl chlorides with dinitromethane sodium salt followed by conversion of the resulting 1-substituted 1-hydroxyimino-2,2-dinitroethanes into dipotassium (or disodium salts) and their subsequent nitrosation with NaNO2 in AcOH or with N2O4. Thermal isomerization of 3-nitro isomers afforded 4-nitro isomers were prepared in high yields.  相似文献

[formula: see text] The bakers' yeast reduction of 3-(1,3-benzodioxol-5-yl)-6-propoxy-1H-inden-1-one 4 has been shown to give (S)-3-(1,3-benzodioxol-5-yl)-2,3-dihydro-6-propoxy-1H-indan-1-one 6 in 65% yield with high enantioselectivity (> 99.0% ee), a key intermediate for the synthesis of the endothelin receptor antagonist SB 217242. In addition, the substituted 3-arylinden-1-ones 10a-e gave equally high enantioselectivity for the 3-arylindan-1-one products 13a-e. Mechanistic studies of the reaction indicate the operative pathway to be an asymmetric conjugate reduction, wherein the hydride transfer from NAD(P)H occurs from the Re-face of the indenone substrate.  相似文献

Nitrodiene compound 1 was stirred with allylthiol for a long time and compound 3 was obtained. Compound 1 gave bis(thio)substituted 2-nitrodiene compound 4 and tris(thio)substituted 2-nitrodiene compound 5 with 2 moles allylthiol in the presence of NaOH in ethanol. Mono(allylthio)substituted diene compound 3 gave dibutadienyl substituted piperazine compounds with piperazine 6 in CH 2 Cl 2 (or diethylether). Compound 3 gave compound 9 with compound 8 . Compounds 11a-e have been obtained from the reaction of compound 3 with compounds 10a-e . Nach langem Rühren von der Verbindung 1 mit Allylthiol entsteht die Verbindung 3 . Die Verbindung 1 liefert mit zwei Mol von Allylthiol in ethanolischer Natronlunge die bis(thio)substituierte 2-Nitrodienverbindung 4 und tris(thio)substituierte Dienverbindung 5 . Mono(allylthio)substituierte Dienverbindung 3 ergibt mit Piperazine in CH 2 Cl 2 (oder in Ether) die dibutadienylsubstituierte Piperazinverbindung 7 . Die Verbindung 3 liefert mit der Verbindung 8 die Verbindung 9 . Durch die Reaktion von der Verbindungen 10a-c wurden die Verbindungen 11a-e erhalten.  相似文献

A new synthetic approach for the regiospecific alkylation of the 6-position of the tropane ring system has been developed. Alkylation, desulfonylation and deprotection of tropanes 5 and 12 furnished a series of 6-endo-alkyltropan-2-one derivatives 8a-e, 15a-e and 16a-e (R = Me, Et, n-Pr, n-Bu, Bn) stereoselectively in good yields. The 6-endo isomers 15a-e and 6-exo isomers 16a-e were easily obtained as pure diastereoisomers.  相似文献

Acid-catalyzed reaction of silylketene dithioacetal 1 with iminium salts or imines 3b,d-h gave the corresponding gamma,gamma-bis(ethylthio)allylamines 2a-h or 5b,d-h in generally high yields. A family of new coordination vanadium(IV) compounds supported by a terminal or bridged aryl imido ligand are reported. Reaction of V(NMe(2))(4) with anilines ArNH(2), where Ar = 2,6-i-Pr(2)-C(6)H(3), 2,6-Me(2)-C(6)H(3), Ph, 2,6-Cl(2)-C(6)H(3), and C(6)F(5), afforded the diamagnetic imido-bridged complexes [V(NAr)(NMe(2))(2)](2) (1a-e). Chlorination of 1a-e with trimethylchlorosilane afforded complexes 2a-e formulated as [V(=NAr)Cl(2)(NHMe(2))(x)()](n)(). One-pot reaction of V(NMe(2))(4) with ArNH(2) in the presence of an excess of trimethylchlorosilane gave the five-coordinate compound [V(=NAr)Cl(2)(NHMe(2))(2)] (3a-e). Reaction of 3a-e with pyridine, bipyridine (bipy), or N,N,N',N'-tetramethylethylenediamine (tmeda) gave respectively the six-coordinate tris- or bis(pyridine) adducts [V(=NAr)Cl(2)(Py)(3)] (4a-e) or [V(=NAr)Cl(2)(Py)(2)(NHMe(2))] (5a), bipyridine complexes [V(=NAr)Cl(2)(bipy)(NHMe(2))] (5a-e) and [V(=NAr)Cl(2)(bipy)(Py)] (9a), and tmeda adduct [V(=NAr)Cl(2)(tmeda)(NHMe(2))] (10a). Moreover, five-coordinate complexes free of NHMe(2) ligands, such as [V(=NAr)Cl(2)(Py)(2)] (5a), [V(=NAr)Cl(2)(bipy)] (8a), and [V(=NAr)Cl(2)(tmeda)] (11a), were directly prepared starting from precursors 2a-e. All compounds were totally characterized by spectroscopic methods (IR, (1)H NMR for diamagnetic complexes, and EPR for paramagnetic complexes), elemental analysis, magnetism, and single-crystal X-ray diffraction studies for 1b, 3a, 3d, 4b, 4d, 7c, 10a, and 11a.  相似文献

Photocyclization reactions were carried out on ethyl 2-formylphenoxyacetates 1a-e and ethyl 2-acetylphenoxyacetates 2a-e in acetonitrile. Irradiation of 1a-e gave dihydrobenzofuranols 3 in 20–46% yields. Similarly, irradiation of 2a-e afforded dihydrobenzofuranols 6 and their derivatives 7, 8 in 40–86% yields. Nitration of 2-benzylthiophene with nitric acid (molar ratio 1:1) in acetic anhydride gave only nitro-2-benzylthiophenes, indicating the benzene ring is unreactive to this electrophilic substitution. The main reaction product was 2-benzyl-5-nitrothiophene (about 87%) the amounts of the 3-nitro and 4-nitro isomers were about 10% and 3%, respectively. These percentages, determined by glc and nmr techniques, were found to be constant using various nitration conditions. The syntheses of the nitro-2-benzylthiophenes are described, together with some spectroscopic properties. The uv spectra confirm that the conjugation of the nitro group in the α position is greater than in the β position. The isomer structures have been assigned by nmr chemical shifts and coupling constant of the thiophene protons. Also infrared spectra are briefly considered.  相似文献
