[Hmim]TFA catalyzed multicomponent reaction: direct,mild, and efficient procedure for the synthesis of 1,2-dihydroquinazoline derivatives

4-Substituted-spiro-1,2-dihydroquinazolines and related compounds were synthesized by direct reaction of 2-aminobenzophenones, isatin, or 1,2-diketone derivatives and ammonium acetate in the presence of dual role catalyst-solvent ionic liquid, [Hmim]TFA. Excellent conversion of starting materials was achieved to the desired 1,2-dihydroquinazoline products.     

Conformational Analysis of (S)-4,5,6,7-Tetrahydro-5-Methylimidazo [4,5,1-jk][1,4]-Benzodiazepin-2(1H)-one (R78362)

The solution and solid state conformation of (S)-4,5,6,7-tetrahydro-5-methylimidazo [4,5,1-jk][1,4]-benzodiazepin-2(1H)-one (R78362) have been investigated by low temperature NMR and x-ray diffraction studies. The ¹H NMR spectrum of R78362 shows no evidence of the presence of multiple conformers in the temperature range 340K - 177K. Molecular mechanisms and semiempirical molecular orbital calculations suggest that the nitrogen and ring inversion barriers of R78362 are small and thus a time-averaged ¹H NMR spectrum is probably occurring at 298K. The x-ray diffraction data indicated that there were two independent molecules in the asymmetric unit of the crystal. The two molecules had similar conformations with the benzoimidazole ring being planar and the diazepine ring in a “half-chair” conformation.

New methods for the synthesis of spiro compounds containing dihydrofuran and coumarin moieties from 3-bromo-4-hydroxycoumarins as cyclic α-halo compound based on a pyridinium-ylide-assisted three-component tandem reaction is investigated. To the best of our knowledge, employing cyclic α-halo compounds for the synthesis of dihydrofuran-fused coumarin via a pyridinium ylide has not been reported yet.

A regioselective multicomponent protocol for the synthesis of novel bioactive 4-hydroxyquinolin-2(1H)-one grafted monospiropyrrolidine and thiapyrrolizidine hybrids

An expedient route toward the synthesis of 4-hydroxyquinolone grafted spiropyrrolidines or pyrrolizidines has been accomplished through 1,3-dipolar cycloaddition reaction of various azomethine ylides derived from isatin or acenaphthalene and sarcosine with 4-hydroxyquinolone derivatives as dipolarophile. The regio and stereo chemical outcome of the cycloaddition reaction is ascertained by X-ray crystallographic studies and spectroscopic techniques of the cycloadducts. Furthermore, cytotoxicity evaluation of selected compounds showed significant inhibition of cell proliferation against cervical as well as colon cancer cell lines.

Summary Protonation of the reactive intermediates produced in the reaction between alkyl(aryl) isocyanides and dialkyl acetylenedicarboxylates by indan-1,3-dione leads to vinylnitrilium cations, which undergo carbon centered Michael type addition with the conjugate base of the CH-acid to produce functionalized 5-oxo-4,5-dihydroindeno[1,2-b]pyrans.

An expeditious, straightforward and efficient synthesis of diversely naphtho[1,2-e][1,3]oxazines via one-pot condensation reaction of ??- naphthol, 3-amino-5-methylisoxazole and arylaldehydes catalyzed by bismuth(III) trifluoromethanesulfonate is described. The reaction preferentially afforded 1,3-trans oxazines.
