Hydrazonoyl bromides 1a-c react with 5-amino-3-phenyl-1H-pyrazole, 5-amino-1H-1,2,4-triazole, 2-aminopyridine, and 2-aminobenzimidazole to afford the corresponding imidazol[1,2-b]pyrazoles 10, imidazo[1,2-b]-1,2,4-triazoles 11, imidazo[1,2-a]pyridines 16, imidazo[1,2-a]pyrimidines 17, and imidazo[1,2-a]benzimidazoles 20, respectively. 3-Substituted imidazo[1,2-a]pyridines, imidazo[1,2-a]pyrimidines, and imidazo[1,2-c]pyrimidine were obtained regiospecifically in yields of 35-92% in one pot by reaction of 2-aminopyridines or 2-(or 4-)aminopyrimidines, respectively, with 1,2-bis(benzotriazolyl)-1,2-(dialkylamino)ethanes.  相似文献

The reaction of phosphorus(III) halides with imidazo[1,2-a]pyridines in the presence of bases leads to the formation of 3-phosphorylated imidazo[1,2-a]pyridines. The reaction proceeds in high yield and requires no catalysts. In the compounds obtained, in contrast to phosphorylated indolizines, the phosphorus–heterocycle bond is stable and not cleaved by dry hydrogen chloride, alcohols, or water. Imidazo[1,2-a]pyridines with the phosphinic and phosphinous groups can be alkylated both at the phosphorus and at the nitrogen atom of the heterocycle, the alkylation direction being dependent on the strength of the alkylation reagent used. © 1995 John Wiley & Sons, Inc.  相似文献

Stable 3-acetyl derivatives of imidazo[1,2-a]benzimidazole were synthesized by the action of acetic anhydride on 2,9-disubstituted imidazo[1,2-a]benzimidazole. Imidazo[1,2-a]pyrazine-3,6-diones and imidazo[1,2-a]imidazo[1,2-d]pyrazine-3,8-diones can be produced by pyrolysis of simple amino acids. While such bicyclic and tricyclic amidines were detected and characterized by IR spectroscopy for some alpha-substituted amino acids, the parent systems composed of glycine fragments are unknown up to now. IR spectra for five amidines derived from glycine were calculated by using different semi-empirical (PM3, AM1, MNDO and MINDO/3), HF, and hybrid DFT (B3LYP, B3P86 and B3PW91) methods in conjunction with 6-31G(d) basis set (for HF and DFT). Vibration frequencies in the experimental IR spectra were predicted based upon the B3LYP data, by correcting the calculated wavenumbers by a scaling factor of 0.959. The behavior of most characteristic bands (nu(CX), nu(NH), etc.) and their shifts with respect to such bands in the spectra of alanine and alpha-aminoisobutyric acid derivatives studied before, are discussed. Performance of the semi-empirical methods was tested, bearing in mind possible future needs for IR spectra predictions for larger molecular systems of similar chemical nature; the use of MINDO/3 and MNDO is recommended. A basis set effect on the B3LYP fundamental vibration frequencies for hexahydroimidazo[1,2-a]pyrazine-3,6-dione was studied by varying Pople basis sets from minimal STO-3G to 6-311++G(d, p). No significant improvements were found beyond the 6-31G(d) basis set, which thus can be recommended to predict IR spectra for the amidines and similar molecules.  相似文献

The closeness of the electronic structures of the ions formed in the first act of disintegration of the ions is responsible for the monotypic character of the subsequent fragmentation of pyrrolo[1,2-a]benzimidazole and imidazo[1,2-a]benzimidazole derivatives. A new synthesis of imidazo[1,2-a]pyridine and imidazo[2,1-a]isoquinoline derivatives 4 and 8 , respectively by 1,5-dipolar cyclization reactions of stabilized pyridinium N-ylides 3a-e or isoquinolinium N-ylide 7 is described. The debenzylation of N-benzylimidazo[1,2-a]benzimidazoles with sodium in liquid ammonia was studied. The debenzylation of the 2-phenyl-substituted derivative is accompanied by hydrogenation of the outer imidazole ring to give 2-phenyl-2,3-dihydro-1H-imidazo[1,2-a]benzimidazole, the alkylation of which in alkaline media and neutral media, respectively, gives the 1-methyl derivative and the 9-methyl derivative. The 1-methyl derivative was subjected to quaternization and bromination; the latter proceeds in the condensed benzene ring. Esters of 1-alkyl- and 1-aralkyl-2-iminobenzimidazoline-3-acetic acids are converted to esters of 2,9-dimethylimidazo[1,2,-a]benzimidazole-3-carboxylic acid on heating with acetic anhydride. ,-Unsaturated ketones of the imidazo[1,2-a]benztmidazole series were synthesized from 3-formyl- and 3-acetyl-substituted imidazo[1,2-a]benzimidazoles by crotonic condensation in the presence of alkaline catalysts. The ,-unsaturated ketones can also be obtained by direct acylation of 3-unsubstituted imidazo[1,2-a]benzimidazoles with the chlorides of unsaturated acids. A series of unusual fused heterocyclic compound derivatives, consisting of a pyridine and a imidazole or pyrimidine core, with a bridgehead nitrogen, were successfully synthesized by a microwave-assisted, three-component domino reaction of aldehydes, enaminones, and malononnitrile. In this one-pot reaction, up to five new bonds were formed accompanied by generating the lactam group. This method has the advantages of short synthetic route, operational simplicity, increased safety for small-scale high-speed synthesis, and minimal environment impact.  相似文献
