3-Nitro-1,2,4-triazol-5-one and its monomethyl derivatives react with methyl vinyl ketone to give products of addition to the ring N1 and N4 atoms. The reaction with formaldehyde and N-methylolacetamide proceeds only at the N1 atom. The keto derivatives of 3-nitro-1,2,4-triazol-5-one undergo the Schmidt reaction to give the corresponding acetamides. A number of compounds that include functional groups in the N1-alkyl substituent of the 3-nitro ring were obtained by treatment of the bases of N1-substituted 3,5-dinitro-1,2,4-triazoles in aprotic media.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 552–558, April, 1981.  相似文献

The action of phenylmagnesium bromide on 6-p-chlorostyryl-3-thioxo-1,2,4-triazin-5-one ( 1 ) was studied whereby a mixture of seven products was obtained. These were separated and characterised, and a reasonable mechanism for their formation is presented and discussed.  相似文献

The reaction of amidoximes with cyanoguanidine in the presence of Lewis acids affords 3-substituted 5-guanidino-1,2,4-oxadiazoles. A study of the reaction of15N-labeled chloroacetamidoxime with cyanoguanidine showed that the formation of the oxadiazole ring occursvia the elimination of the amino group from the amidoxime fragment. 1,2,4-Oxadiazoles bearing the imidazole or pyrimidine moiety were synthesized.Translated fromIzvestiya Akademii Nauk. [structure: see text]. An efficient three-step synthesis of chiral 3H-quinazoline-4-one derivatives from commercial materials is disclosed. The Mumm reaction of imidoyl chloride with alpha-amino acids followed by reductive cyclization affords enantiomerically pure (ee >93%) quinazoline-4-ones in good overall yield. A comparison with existing approaches indicates that this method is superior for hindered substrates.  相似文献

Treatment of 5-nitro-2,4-dihydro-3H-1,2,4-triazol-3-one ( 1 ) with picryl fluoride (PkF) in 1-methyl-2-pyrrolidinone (NMP) gave a mixture of a monopicryl and a dipicryl derivative of 1 in a ratio of 2 :1 , respectively, regardless of the initial concentrations of 1 and PkF. The products were identified as 5-nitro-2-picryl-2,4-dihydro-3H-1,2,4-triazol-3-one ( 2 ) by X-ray crystallography and 5-nitro-2,4-dipicryl-2,4-dihydro-3H-1,2,4-triazol-3-one ( 4 ) by 15N labeling experiments.  相似文献

A novel procedure for preparing 1,2,4-triazol-3-ones is described. Various alkyl, aryl, and heterocyclic groups were introduced successfully at both the N2 and C5 positions. The triazolone ring was constructed through an intramolecular cyclization of a novel acyclic precursor, which in turn was synthesized by treating a mono protected hydrazine with an acyl isocyanate. Under conditions that remove the hydrazine protecting group, the intramolecular cyclization occurs rapidly, to deliver the 2,5-substituted 1,2,4-triazol-3-ones in excellent yields (79-99%) without column purification.  相似文献

A series of 1,5-dihydro-4H-pyrrolo[3,2-c]pyridin-4-one derivatives with 3-(4-hydroxy-2-oxo-2H-pyran-3-yl) groups were synthesized via a one-pot three-component reaction of 4-aminopyridin-2(1H)-ones, 2,2-dihydroxy-1-arylethan-1-ones, and 4-hydroxy-2H-pyran-2-ones in water. This strategy not only provides a valuable tool in the design and synthesis of new hydrogenated pyrrolo[3,2-c]pyridines but also has the advantages of inexpensive starting materials, atom and step economy, environmental friendliness, good to excellent yields, and operational simplicity. A total of 14 examples were examined to show a broad substrate scope and high yields.  相似文献

The Friedel-Crafts acetylation of 5-hydroxyindole derivativesla-c regioselectively produced 6-acetyl-5-hydroxy-indole derivatives2a-c which were condensed with cinnamic acid using POC13 and dry pyridine to afford 5-cinnamoyloxy-6-acetyl-indoles3a-c. These upon Baker-Venkataraman transformation furnished 6-cinnamoylacetyl-5-hydroxyindoles4a-c which were cyclised separately with AcOH/HCl and Ac2O/AcONa to 4H-pyrano-(2,3-f)indol-4-one derivatives5a-c and6a-c respectively.  相似文献
