High-performance liquid chromatography with tandem mass spectrometry has been used for rapid, specific, and sensitive analysis of busulfan in human plasma. Busulfan-d8 was used as internal standard. Analysis was performed on a C18 column (50 mm × 2.1 mm, 3.5-µm particles) with water–methanol 80:20 (v/v) as mobile phase at a flow-rate of 0.30 mL min−1. Detection was by tandem triple–quadrupole mass spectrometry with turbo ion-spray ionization. Linear calibration plots were obtained over the concentration range 1.096–1,096 ng mL−1. The assay is ideally suited to monitoring of busulfan and determination of its pharmacokinetic data.

A two-phase solvent system composed of a mixture of n-hexane–ethanol–water (6: 5: 1, v/v/v) was used and the separation conditions were optimized. In a typical run in less than 400 min, 100 mg of samples can be separated to yield 14 mg of ergosta-4,6,8(14),22-tetraen-3-one with 99.1% purity. The structure of this compound was elucidated by UV, EI-MS, 1H NMR and 13C NMR.

The method involves a two-step protein precipitation by liquid–liquid extraction. Phenytoin sodium was used as the internal standard. The separation was carried out on Acquity C18 column with acetonitrile:methanol:KH2PO4 buffer (adjusting pH to 4.6 with 85% o-phosphoric acid) (180/180/170, v/v/v) as the mobile phase at a flow rate of 0.4 mL min−1. Linear detection response was obtained for concentrations ranging from 50 to 5,000 ng mL−1. The limit of quantification (LOQ) was 50 ng mL−1. The method was validated successfully for the determination of carbamazepine and its active metabolite carbamazepine-10,11-epoxide, which can be applied through pharmacokinetics and bioequivalence studies.
相似文献A precise and sensitive LC method for determination of enantiomeric purity of trelagliptin has been developed and validated. Pre-column derivatization was performed before separation. Baseline separation with a resolution factor >2.5 was accomplished within 10 min by use of a Chiralpak AD column (250 × 4.6 mm; particle size 5 µm) and n-hexane–2-propanol (90:10 v/v) as mobile phase at a flow rate of 1 mL min−1. Eluted analytes were monitored by UV detection at 260 nm. The effects of mobile phase composition and temperature on enantiomeric selectivity and on resolution of enantiomers were thoroughly investigated. Calibration curves were plotted within the concentration range 0.005–2 mg mL−1 (n = 12), and recoveries between 98.23 and 101.34 % were obtained, with relative standard deviation (RSD) <1.39 %. LOD and LOQ for the trelagliptin derivative were 1.51 and 5.03 µg mL−1; those for its enantiomer were 1.49 and 4.94 µg mL−1, respectively. The method was evaluated and validated by analysis of bulk samples of trelagliptin of different enantiomeric purity. It was demonstrated that the method was accurate, robust, and sensitive, and enabled practical analysis of real samples.
相似文献A weak cation-exchange monolithic column has been prepared in stainless steel tubing and used as the solid-phase extraction material in quantitative analysis of caffeine and theophylline in urine. Column switching, with water as mobile phase, was used for on-line cleaning and screening of human urine samples. Reversed-phase high-performance liquid chromatography was then performed on a C18 column with methanol–water 30:70 (v/v) as mobile phase at a flow rate of 0.5 mL min−1. Ultraviolet detection was performed at 274 nm. Good linear relationships were obtained between response and concentrations of caffeine and theophylline in the range 0.1–50 μg mL−1. Absolute recovery ranged from 77.4 to 82.3% and inter-day and intra-day relative standard deviations were less than 5%. The method was suitable for analysis of caffeine and theophylline in human urine, because it eliminated tedious pretreatment and enabled rapid, economic, repeatable, and effective assay of traces of the drugs in biological samples.

Calibration curves ranged between 200–600 and 125–375 ng spot−1 for olmesartan and hydrochlorothiazide, respectively.

A C18 column was used and the mobile phase was 10 % (v/v) methanol and 90 % (v/v) water (0.1 % trifluoroacetic acid, v/v) at a flow rate of 0.2 mL min−1. The drift tube temperature was 40 °C. The pressure of nebulizing gas (N2) was 3.0 bar. The gain was 10. Good separation of K-222 from main related substances could be achieved. Excellent linearity (r 2 = 0.9995) was obtained over the range of 5–100 μg mL−1. The precision ranged from 0.68 to 5.16 % (RSD) and the accuracy ranged from −3.05 to 2.62 % (RE). The limit of detection was 2 μg mL−1. This method offers simple, rapid and quantitative detection of K-222, thus making it acceptable for routine determination.

Chromatography was performed on a 150 mm × 4.6 mm C18 column with acetonitrile–water 60:40 (v/v) as isocratic mobile phase at 1.0 mL min−1. Ultraviolet detection of dutasteride was at 210 nm. Its retention time was approximately 10 min and its peak was symmetrical. Response was a linear function of concentration over the range 0.2–1 μg mL−1 (R 2 = 0.997) and the limits of detection and quantitation were was 0.05 and 0.10 μg mL−1, respectively. The method was validated for linearity, precision, repeatability, sensitivity, and selectivity. Selectivity was validated by subjecting dutasteride stock solution to photolytic, acidic, basic, oxidative, and thermal degradation. The peaks from the degradation products did not interfere with that from dutasteride. The method was used to quantify dutasteride in pharmaceutical preparations.

The method was accurate and precise with limits of detection and quantitation of 64 and 215 ng, respectively. Mean recovery was 100.71%. Sibutramine in the presence of its degradation products was analyzed using UV detection at 225 nm. Chromatography was performed on a reversed-phase C8 (150 × 4.0 mm I.D., 5 μm) analytical column under isocratic conditions. The mobile phase was composed of acetonitrile:water (aqueous phase containing 0.3% triethylamine and pH adjusted to 7.0) (75:25, v/v) at a flow-rate of 1.1 mL min−1. No chromatographic interference was found during the analysis. Light was the stress condition which most contributed to sibutramine degradation. The method showed a linear response (r > 0.999) from 30 to 90 μg mL−1. The mean recovery for capsules was 101.2%. Inter-day assays showed relative standard deviations of 0.42 and 1.62% for bulk and capsules, respectively. The developed method is able to separate sibutramine from its major degradation products and it may be used in the quality control of this active pharmaceutical ingredient in both bulk and capsules.

The mobile phase was 3% (v/v) acetic acid water solution–methanol (8:92, v/v) at a flow rate of 1.0 mL min−1. Detection was at 310 nm. Calibration curve was linear over the range of 0.25–50 μg mL−1 with intra- and inter-day precisions (RSD%) of less than 9.5%. The extraction recovery ranged from 87.0 to 90.2% (RSD 2.4–6.4%) for ginkgolic acids. The method was successfully applied to the pharmacokinetic study of ginkgolic acids in mice after oral dosing of 1.0 g kg−1.

The method was based on liquid–liquid extraction, followed by an LC procedure with a Gemini C-18, 50 mm × 2.0 mm (3 μm) column and using methanol:ammonium formate (97:3, v/v) as the mobile phase. Protonated ions formed by a turbo ionspray in positive mode were used to detect analytes and internal standard. MS–MS detection was by monitoring the fragmentation of 409.1 → 228.1 (m/z) for tamsulosin, 529.3 → 461.3 (m/z) for dutasteride and 373.2 → 305.3 (m/z) for finasteride (IS) on a triple quadrupole mass spectrometer. The lower limit of quantification for both tamsulosin and dutasteride was 1 ng mL−1. The proposed method enables the unambiguous identification and quantification of tamsulosin and dutasteride for clinical drug monitoring.

相似文献Auricularia polytricha is an edible mushroom, quite popular in China. Modern pharmacology research indicate that A. polytricha polysaccharides possess antitumor and immunomodulatory activities. In this paper, an antitumor Auricularia polytricha polysaccharide (APS)-2 was purified by radial flow chromatography (RFC) in a column of 500 mL bed-volume (40 cm i.d × 15 cm) packed with DEAE52-cellulose anion ion-exchange resin. The optimal separation conditions were: sample flow-velocity 15–20 mL min−1, sample volume 160–200 mL, and elution with distilled water and 0.2 M NaCl at a velocity of 35–45 mL min−1. The yield and content of APS-2 obtained under these conditions were 182 mg g−1 total polysaccharides and 98.35 %, respectively. The study provides guidelines for the separation and purification of polysaccharides using a radial flow chromatography column.
相似文献An LC-DAD method was developed for determination of lobeline from in vitro and in vivo cultures of Lobelia inflata. Samples were extracted with 0.1 N HCl–acetonitrile (1:1, v/v), and purified by solid-phase extraction. Optimized conditions resulted in high recovery. LC separations were performed on an Eurosphere C8 reversed-phase column using 30:70 (v/v) acetonitrile–0.1% trifluoroacetic acid as a mobile phase. Quantitative determination of lobeline was performed by external standard method at 250 nm, in the range of 2.4–80 μg mL−1. Validation studies proved that the repeatability of the method was good and the recovery was satisfactory. In vitro organized cultures contained considerable amount of lobeline (herb: 175 μg g−1, root: 100 μg g−1). When these cultures were transplanted into the open field, the lobeline content increased significantly (herb: 323 μg g−1, root: 833 μg g−1). Plants obtained from seed propagation contained 382 μg g−1 lobeline in the herb. For direct characterization of di-substituted piperidine alkaloids in extracts of L. inflata, tandem mass spectrometric method was developed using electrospray ionization. Analysis was performed in the positive ion mode on a triple quadropole LC–MS system. LC separations were achieved on Eurosphere C8 column with a modified mobile phase (acetonitrile–30 mM ammonium formate, pH 2.80) to ensure proper molecular ionization. The identification and structural elucidation of the alkaloids were performed by comparing their changes in molecular mass (ΔM), full-scan MS–MS spectra with those of lobeline, norlobelanine and lobelanidine. These alkaloids and ten other derivatives were identified in the plant extracts. Three piperidine alkaloids were reported in L. inflata for the first time.
