The inhibitory effects of five hydroxyanthraquinones (HAQs) from root and rhizoma of Rheum officinale Baill, a traditional Chinese medicinal (TCM) herb, on Staphylococcus aureus growth were investigated by calorimetry. The power-time curves of S.aureus with and without HAQ were acquired and the extent and duration of inhibitory effects on the metabolism evaluated by growth rate constants (k1, k2), half inhibitory ratio (IC50), maximum heat output (Pmax) and peak time (tp). The value of k1 and k2 of S. aureus in the presence of the five HAQs decreased with the increasing concentrations of HAQs. Moreover, Pmax was reduced and the value of tp increased with increasing concentrations of the five drugs. The inhibitory activity varied for different drugs. IC50 of the five HAQs was 4 μg ml−1 for emodin, 3.5 μg ml−1 for rhein, 10 μg ml−1 for aloe-emodin, 1000 μg ml−1 for chrysophanol, 1600 μg ml−1 for physcion. The sequence of antimicrobial activity of the five HAQs: rhein > emodin > aloe-emodin > chrysophanol > physicion.  相似文献

A sensitive luminescent bioassay for the simultaneous detection of Salmonella Typhimurium and Staphylococcus aureus was developed using aptamer-conjugated magnetic nanoparticles (MNPs) for both recognition and concentration elements and using upconversion nanoparticles (UCNPs) as highly sensitive dual-color labels. The bioassay system was fabricated by immobilizing aptamer 1 and aptamer 2 onto the surface of MNPs, which were employed to capture and concentrate S. Typhimurium and S. aureus. NaY0.78F4:Yb0.2,Tm0.02 UCNPs modified aptamer 1 and NaY0.28F4:Yb0.70,Er0.02 UCNPs modified aptamer 2 further were bond onto the captured bacteria surface to form sandwich-type complexes. Under optimal conditions, the correlation between the concentration of S. Typhimurium and the luminescent signal was found to be linear within the range of 101–105 cfu mL−1 (R2 = 0.9964), and the signal was in the range of 101–105 cfu mL−1 (R2 = 0.9936) for S. aureus. The limits of detection of the developed method were found to be 5 and 8 cfu mL−1 for S. Typhimurium and S. aureus, respectively. The ability of the bioassay to detect S. Typhimurium and S. aureus in real water samples was also investigated, and the results were compared to the experimental results from the plate-counting methods. Improved by the magnetic separation and concentration effect of MNPs, the high sensitivity of UCNPs, and the different emission lines of Yb/Er- and Yb/Tm-doped NaYF4 UCNPs excited by a 980 nm laser, the present method performs with both high sensitivity and selectivity for the two different types of bacteria.  相似文献

The effect of a series of novel Schiff base compounds on Staphylococcus aureus was studied by microcalorimetric method at 37 °C The results showed that all of the organic compounds had the capacity to inhibit the growth of S. aureus in different extent. And the extent and duration of the inhibitory effect on the growth of S. aureus, judged from the rate constant (k), varied with the different structure of the Schiff base compounds. According to the power-time curves, the multiplication rate constant and inhibition ratio were calculated. The growth rate constant of S. aureus (in log phase) in the presence of Schiff base compounds decreased with the increasing of the concentrations of these compounds regularly. The experimental results revealed that the hydrophilicity of Schiff bases had a great influence on their antibacterial activity. Of these Schiff bases, the greater their hydrophilicity, the higher their antibacterial activity. The antibacterial structure-activity relationship (SAR) of Schiff base derivatives was also briefly discussed.  相似文献

The aim of this paper was to demonstrate a fluorescence measurement method for rapid detection of two bacterial count by using water-soluble quantum dots (QDs) as a fluorescence marker, and spectrofluorometer acted as detection apparatus, while Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) were as detection target bacteria. Highly luminescent water-soluble CdSe QDs were first prepared by using thioglycolic acid (TGA) as a ligand, and were then covalently coupled with target bacteria. The bacterial cell images were obtained using fluorescence microscopy. Our results showed that CdSe QDs prepared in water phase were highly luminescent, stable, and successfully conjugated with E. coli and S. aureus. The fluorescence method could detect 102-107 CFU/mL total count of E. coli and S. aureus in 1-2 h and the low detection limit is 102 CFU/mL. A linear relationship of the fluorescence peak intensity and log total count of E. coli and S. aureus have been established using the equation Y = 118.68X − 141.75 (r = 0.9907).  相似文献

Serious bloodstream infections are a significant complication in critically ill patients. The treatment of these infections has become more difficult because of the increasing prevalence of multiresistant strains, especially methicillin-resistant Staphylococcus aureus (MRSA). Rapid differentiation of low number of MRSA from methicillin-susceptible S. aureus (MSSA) cells (101–102 cells mL−1) in blood is necessary for fast effective antibiotic therapy. Currently, three groups of techniques, phenotyping, genotyping, and mass spectrometry, are used for MRSA and MSSA strains differentiation. Most of these techniques are time-consuming. PCR and other molecular techniques allow the detection and differentiation between MSSA and MRSA directly from blood cultures. These methods alone are rapid and they have good reproducibility and repeatability. Potential disadvantages of the genotyping methods include their discrimination ability, technical complexity, financial costs, and difficult interpretation of the results.  相似文献

Inhibitors of biotin protein ligase (BPL) are novel antimicrobial compounds with the potential to treat infections caused by bacteria resistant to current antibiotics. A novel BPL inhibitor (12, Ki 1.4 μM) was synthesized from biotin acetylene and an azide-functionalized analogue of fluorescent nitrobenzofurazan by Cu(I) catalysed cycloaddition and also by template guided synthesis using wild-type BPL from Staphylococcus aureus. LC/HRMS-based detection provides improved sensitivity over previous reports using a mutant BPL, with demonstrated applicability to other BPLs. Super-imaging fluorescence microscopy demonstrated the accumulation of 12 in the cytoplasm of S. aureus, but not Escherichia coli. This novel fluorescent probe can be used to gain new insights into the mechanism of uptake, efflux and metabolism of BPL inhibitors in S. aureus.  相似文献

RNA is an important target for drug discovery efforts. Several clinically used aminoglycoside antibiotics bind to bacterial rRNA and inhibit protein synthesis. Aminoglycosides, however, are losing efficacy due to their inherent toxicity and the increase in antibiotic resistance. Targeting of other RNAs is also becoming more attractive thanks to the discovery of new potential RNA drug targets through genome sequencing and biochemical efforts. Identification of new compounds that target RNA is therefore urgent, and we report here on the development of rapid screening methods to probe binding of low molecular weight ligands to proteins and RNAs. A series of aminoglycosides has been immobilized onto glass microscope slides, and binding to proteins and RNAs has been detected by fluorescence. Construction and analysis of the arrays is completed by standard DNA genechip technology. Binding of immobilized aminoglycosides to proteins that are models for study of aminoglycoside toxicity (DNA polymerase and phospholipase C), small RNA oligonucleotide mimics of aminoglycoside binding sites in the ribosome (rRNA A-site mimics), and a large (approximately 400 nucleotide) group I ribozyme RNA is detected. The ability to screen large RNAs alleviates many complications associated with binding experiments that use isolated truncated regions from larger RNAs. These studies lay the foundation for rapid identification of small organic ligands from combinatorial libraries that exhibit strong and selective RNA binding while displaying decreased affinity to toxicity-causing proteins.  相似文献

The applicability of a capillary zone electrophoresis–electrospray ionisation tandem mass spectrometric (CZE–ESI-MS–MS) method for the separation of nine fluoroquinolones was investigated. Method optimisation involved systematic trouble-shooting starting with the type and duration of capillary pre-washing and conditioning, the choice of both the CE run buffer, MS sheath liquid, CE run potential, ESI spray voltage, sheath gas flow-rate, MS capillary voltage and CE capillary and MS capillary temperatures. Another extremely important factor was found to be the degree to which the CE capillary protrudes into the ESI chamber as well as whether or not sheath gas and spray voltage are employed during the CE injection or not. The importance of the latter has, to our knowledge, not been addressed elsewhere. Nine fluoroquinolones have been separated and detected in a single run by this technique.  相似文献

The preparation of novel Staphylococcus aureus (S. aureus) amperometric immunosensing designs based on the covalent immobilization of RbIgG at gold electrodes using the heterobifunctional cross-linker 3,3-dithiodipropionic acid di(N-succinimidyl ester) (DTSP), are reported. Two different competitive immunosensing configurations have been tested and compared. In the first one, protein A-bearing S. aureus cells and HRP-labelled antiRbIgG compete for immobilized RbIgG binding sites, while in the second case HRP-labelled protein A was used. In both cases, the evaluation of the developed immunosensors performance was accomplished through the monitoring at 0.00 V (vs. Ag/AgCl) of the catalytic current originated after addition of hydrogen peroxide, using tetrathiafulvalene as redox mediator entrapped at the modified electrode surface by cross-linking with glutaraldehyde. Optimization of variables concerning the composition of the immunosensors as well as the detection conditions was carried out in 0.1 M NaAc/0.1 M NaCl buffer of pH 5.6. The configuration that employed antiRbIgG-HRP resulted in better analytical characteristics, with a detection limit of 1.4 × 104 cells mL−1 for S. aureus cells submitted to wall lyses by ultrasonic treatment. This immunosensor design was also evaluated using gold screen-printed electrodes in order to reduce the analysis time and cost. In this case, a limit of detection of 3.7 × 102 cells mL−1 and a dynamic range from 1.3 × 103 to 7.6 × 104 cells mL−1 was obtained. A RSD value of 10.5% was found for the responses to 9.6 × 103S. aureus cells mL−1 obtained with seven different Au/SPEs-immunosensors. These disposable immunosensors were applied to the quantification of S. aureus in milk spiked at two concentration levels, 1.2 × 103 and 4.8 × 103 cells mL−1, with good recoveries.  相似文献

This report describes a novel diagnostic assay for rapid detection of the Panton-Valentine Leukocidin (PVL) toxin of methicillin-resistant Staphylococcus aureus (MRSA) utilizing resistive pulse sensing (RPS), loop-mediated isothermal DNA amplification (LAMP) in combination with gold nanoparticles (AuNPs). The PVL DNA from MRSA was specifically amplified by LAMP using four primers at one temperature (65 °C). The DNA products with biotin were then conjugated to a first AuNP1 (55 ± 2 nm) through biotin–avidin binding. A second AuNP2 (30 ± 1.5 nm) coated with a specific DNA probe hybridized with the LAMP DNA products at the loop region to enhance assay sensitivity and specificity, to generate supra-AuNP1-DNA-AuNP2 assemblies. Scanning electron microscopy confirmed the presence of these supra-assemblies. Using RPS, detection and quantitation of the agglomerated AuNPs were performed by a tunable fluidic nanopore sensor. The results demonstrate that the LAMP-based RPS sensor is sensitive and rapid for detecting the PVL DNA. This technique could achieve a limit of detection (LOD) up to about 500 copies of genomic DNA from the bacteria MRSA MW2 and the detection can be completed within two hours with a straightforward signal-to-readout setup. It is anticipated that this LAMP-based AuNP RPS may become an effective tool for MRSA detection and a potential platform in clinical laboratory to report the presence or absence of other types of infectious agents.  相似文献

Surface-enhanced Raman scattering (SERS) spectroscopy was used to monitor DNA hybridization of a fragment of the BRCA1 breast cancer susceptibility gene on modified silver surfaces. Rhodamine B was covalently attached to a 5′-amino-labeled oligonucleotide sequence (23 mer) through a succinimidyl ester intermediate in methanol. The silver surfaces were prepared by depositing a discontinuous layer (9.0 nm) of silver onto glass slides, which had been etched with HF to form a microwell platform, and subsequently modified with a monolayer of mercaptoundecanoic acid. The complementary probe was covalently attached to the silver surfaces using a succinimidyl ester intermediate in acetonitrile. The silver island substrate allows a very large enhancement of the Raman signal of the DNA-Rhodamine B, and clear distinction between hybridized samples and controls on a microwell array sampling platform.  相似文献

Microarray technology has been proved to be greatly helpful for biomedical and biological diagnosis. And the evaluation of its biological applications lies in the detection sensitivity, which requires high intensity and stability of the signal. Recently, several nanomaterials, especially semiconductor nanomaterials, due to their excellent fluorescence properties, have been widely used to construct microarrays for biosensors. Here, we presented an approach for constructing CdSe/ZnS quantum dot (QD) microarray in microfluidic channels on a glass slide by photolithography. The conditions for immobilizing stable and uniform QD microarray on the glass slide were optimized. Several types of QD microarrays with different emission wavelengths and modified groups were constructed using silanization and lithography technology. Based on the fluorescence quenching effect of Cu2+ on QDs, the microfluidic chip with QD microarray was applied for the determination of Cu2+. 1 nmol/L Cu2+ could be detected by this method.  相似文献

An easy to use and low time consuming capillary electrophoresis (CE) method was developed and applied to the simultaneous determination of six antibiotics (ampicillin, amoxicillin, cloxacillin, penicillin, tetracycline and chloramphenicol) in spiked milk samples. Samples of milk were cleaned up by solid-phase extraction (with a C18 cartridge) after protein precipitation. Analysis was performed by CE and results compared with the obtained via HPLC, both coupled to a UV-vis detector (210 nm). CE employed a 58.5 cm long fused-silica capillary (50 cm to detector), 75 μm i.d., a 2.7 × 10−2 M KH2PO4, 4.3 × 10−2 M Na2B4O7 separation buffer, pH 8; an applied voltage of 18 kV; a hydrostatic injection of 0.5 psi during 3 s; and a run temperature of 25 °C. Under the described conditions, amoxicillin was not separated by HPLC, while CE was able to separate, and, therefore, allow detection. Regardless of amoxicillin, comparable results were obtained by HPLC and CE. The average recoveries of antibiotics, from milk fortified at 2.5 and 5 μg/mL, was over 72% with R.S.D.s within 5%. Recovery levels were essentially dictated by the used SPE cartridge.  相似文献

Two enantiomerically pure B-ring modified analogues of (−)-epicatechin gallate were synthesised and their modulation of β-lactam resistance using three strains of methicillin resistant Staphylococcus aureus (BB 568, EMRSA-15 and EMRSA-16) evaluated. Sub-inhibitory concentrations (12.5 and 25 mg/L) of the two analogues fully sensitised each of the three MRSA strains to oxacillin, reducing the MIC to less than 0.5 mg/L, identical to levels achieved with ECg. Lower concentrations demonstrated that the position and degree of hydroxylation of the B-ring is important for activity.  相似文献

In this study we propose a chromogenic platform for rapid analysis of organophosphate (OP) and carbamate (CM) insecticide residues, based on recombinant Drosophila melanogaster acetylcholinesterase (R-DmAChE) as enzyme and indoxyl acetate as substrate. The visible chromogenic strip had the advantages identical to those of commonly used lateral flow assays (LFAs) with utmost simplicity in sample loading and result observation. After optimization, depending on the color intensity (CI) values, the well-established assay has the capabilities of both qualitative measurement via naked eyes and quantitative analysis by colorimetric reader with the desirable IC50 values against the tested six insecticides (0.06 μg mL−1 of carbofuran, 0.28 μg mL−1 of methomyl, 0.03 μg mL−1 of dichlorvos, 31.6 μg mL−1 of methamidophos, 2.0 μg mL−1 of monocrotophos, 6.3 μg mL−1 of omethoate). Acceptable matrix effects and satisfactory detection performance were confirmed by in-parallel LC–MS/MS analysis in different vegetable varieties at various spiked levels of 10−3 to 101 μg g−1. Overall, the testified suitability and applicability of this novel platform meet the requirements for practical use in food safety management and environmental monitoring, especially in the developing world.  相似文献

Incorporation of labeled acetate and l-serine into PF1140 in Eupenicillium sp. indicated that the skeleton of PF1140 is derived from five acetates and a l-serine. Upon administration of [1,3-13C2]glycerol, a precursor of biotransformation into l-[1,3-13C2]serine, the isotopic labels became contiguous in the resultant 2-pyridone of PF1140. Based on the feeding experiments, a novel and potentially general biosynthetic pathway for a 2-pyridone framework has been proposed, in which an acyl tetramic acid precursor could be converted via a ring expansion with loss of the hydroxyl group.  相似文献

Herein a quantitative method for the determination of seven penicillins in bovine plasma and veterinary drugs has been developed. Amoxicillin (AMO), ampicillin (AMP), penicillin G (PENG), penicillin V (PENV), oxacillin (OXA), cloxacillin (CLO) and dicloxacillin (DICLO) were separated on a Perfectsil ODS‐2 (250×4 mm, 5 μm) column, using gradient elution, with a mobile phase of 0.1% v/v TFA and ACN–methanol (90:10 v/v). PDA detection was used at 240 nm. Penicillins were isolated from bovine plasma by SPE on Lichrolut RP‐18 cartridges with mean recoveries from 85.7 to 113.5%. Colchicine (3 ng/μL) was used as an internal standard. The developed method was validated in terms of selectivity, linearity, accuracy, precision, stability and sensitivity. Repeatability (n = 5) and between‐day precision (n = 5) revealed RSD < 12%. The detection limits in the bovine plasma were estimated as 18 ng for AMO and AMP, 25 for PENG, PENV and OXA, 3 ng for CLO and 12 ng for DICLO. Spiked plasma samples were stable for 1 wk, except for AMP and CLO, which were stable for 3 wk and OXA for 4 wk. AMO, PENG and PENV were stable for two freeze–thaw cycles, OXA, CLO and DICLO for four, while AMP only for one.  相似文献
