Multimodal imaging is a highly desirable biomedical application since it can provide complementary information from each imaging modality. We propose a protein engineering-based strategy for the construction of a bimodal probe for fluorescence and magnetic resonance imaging. A recombinant protein was generated by the fusion of a supercharged green fluorescence protein (GFP36+) with a lanthanide-binding tag (dLBT) that can stably bind two Gd3+ ions. The GFP36+–dLBT fusion protein showed strong fluorescence and exhibited efficient contrast enhancement in magnetic resonance imaging. This protein probe improves the MR relaxation more efficiently than Gd-DTPA (gadopentetate dimeglumine). The superior cell-penetrating activity of GFP36+ allows the efficient cellular uptake of this fusion protein and it can thus be used as a cellular imaging probe. Dual imaging was conducted in vitro and in mice. This result indicates that the fusion of different functional domains is a feasible approach for making multi-modal imaging agents.  相似文献

A miniaturized multiplex biosensor exploiting a microfluidic oligonucleotide array and chemiluminescence (CL) lensless imaging detection has been developed for parvovirus B19 genotyping. The portable device consists of a reaction chip, comprising a glass slide arrayed with three B19 genotype-specific probes and coupled with a polydimethylsiloxane microfluidic layer, and a charge-coupled device camera modified for lensless CL imaging. Immobilized probes were used in DNA hybridization reactions with biotin-labeled targets, and then hybrids were measured by means of an avidin-horseradish peroxidase (HRP) conjugate and CL detection. All hybridization assay procedures have been optimized to be performed at room temperature through the microfluidic elements of the reaction chip, with sample and reagents delivery via capillary force exploiting adsorbent pads to drive fluids along the microchannels. The biosensor enabled multiplex detection of all B19 genotypes, with detectability down to 80 pmol?L?1 for all B19 genotype oligonucleotides and 650 pmol?L?1 for the amplified product of B19 genotype 1, which is comparable with that obtained in traditional PCR-ELISA formats and with notably shorter assay time (30 min vs. 2 h). The specificity of the assay has been evaluated by performing DNA–DNA hybridization reactions among sequences with different degrees of homology, and no cross hybridizations among B19 genotypes have been observed. The clinical applicability has been demonstrated by assaying amplified products obtained from B19 reference serum samples, with results completely consistent with the reference PCR-ELISA method. The next crucial step will be integration in the biosensor of a miniaturized PCR system for DNA amplification and for heat treatment of amplified products.

The translocator protein (TSPO) is an important target for imaging focal neuroinflammation in diseases such as brain cancer, stroke and neurodegeneration, but current tracers for non-invasive imaging of TSPO have important limitations. We present the synthesis and evaluation of a novel 3-fluoromethylquinoline-2-carboxamide, AB5186, which was prepared in eight steps using a one-pot two component indium(iii)-catalysed reaction for the rapid and efficient assembly of the 4-phenylquinoline core. Biological assessment and the implementation of a physicochemical study showed AB5186 to have low nanomolar affinity for TSPO, as well as optimal plasma protein binding and membrane permeability properties. Generation of [18F]-AB5186 through 18F incorporation was achieved in good radiochemical yield and subsequent in vitro and ex vivo autoradiography revealed the ability of this compound to bind with specificity to TSPO in mouse glioblastoma xenografts. Initial positron emission tomography imaging of a glioma bearing mouse and a healthy baboon support the potential for [18F]-AB5186 use as a radiotracer for non-invasive TSPO imaging in vivo.  相似文献

A target-driven DNA association was designed to initiate cyclic assembly of hairpins, which led to an enzyme-free amplification strategy for detection of a nucleic acid or aptamer substrate and flexible construction of logic gates. The cyclic system contained two ssDNA (S1 and S2) and two hairpins (H1 and H2). These ssDNA could co-recognize the target to produce an S1–target–S2 structure, which brought their toehold and branch-migration domains into close proximity to initiate the cyclic assembly of hairpins. The assembly product further induced the dissociation of a double-stranded probe DNA (Q:F) via toehold-mediated strand displacement to switch the fluorescence signal. This method could detect DNA and ATP as model analytes down to 21.6 pM and 38 nM, respectively. By designing different DNA input strands, the “AND”, “INHIBIT” and “NAND” logic gates could be activated to achieve the output signal. The proposed biosensing and logic gate operation platform showed potential applications in disease diagnosis.  相似文献

The effect of detection chip geometry on chemiluminescence (CL) signal intensity of tris(1,10-phenanthroline)-ruthenium(II) peroxydisulphate system for analysis of chlorpheniramine maleate (CPM) in pharmaceutical formulations was investigated. It was observed that the design of the detection chip is very crucial and can play an important role in enhancing the CL signal intensity in this system. The CL signal intensity was enhanced 250% when a teardrop micromixer chip was used, compared to the commonly used serpentine chip geometry. The study was conducted using a multi-chip device. In this device, chip 1 was used to prepare and pump the reagent mixture, whereas chip 3 was used for pumping the sample. The two chips were connected to the teardrop chip (2) via silica capillary where detection took place. Non-linear regression curve fitting of the calibration data revealed that the calibration curves are best described by third order polynomial equation with excellent correlation coefficients (R2 = 0.9998) for the concentration range 7.69 × 10−8 to 5.12 × 10−5 mol L−1. A linear response is also observed over the range 7.69 × 10−8 to 1.28 × 10−5 mol L−1 (R2 = 0.9996) and the detection limit was found to be 5.49 × 10−8 mol L−1. The device was successfully used for the analysis of CPM in tablets and a multi-component cough syrup. Results were reproducible with relative standard deviation (RSD) of 0.6-1.1%.  相似文献

A novel enzyme-free amplification strategy was designed for sensitive electrochemical detection of deoxyribonucleic acid (DNA) based on Zn2+ assistant DNA recycling via target-triggered assembly of mutated DNAzyme. A gold electrode was used to immobilize molecular beacon (MB) as the recognition probe and perform the amplification procedure. In the presence of target DNA, the hairpin probe 1 was opened, and the DNAzyme was liberated from the caged structure. The activated DNAzyme first hybridized and then cleaved the MB in the presence of cofactor Zn2+. After cleavage, the MB was cleaved into two pieces and the ferrocene (Fc) labeled piece dissociated from the gold electrode, thus obviously decreasing the Fc signal and forming a free DNAzyme strand. Finally, each target-induced activated DNAzyme underwent many cycles to trigger the cleavage of many MB substrates. Therefore, the peak current of Fc dramatically decreased to approximately zero. The strategy showed a detection limit at 35 fM levels, which was about 2 orders of magnitude lower than that of the conventional hybridization without Zn2+-based amplification. The Zn2+ assistant DNA recycling offers a versatile platform for DNA detection in a cost-effective manner, and has a promising application in clinical diagnosis.  相似文献

Scanning electrochemical microscopy (SECM) and scanning chemiluminescence microscopy (SCLM) were used for imaging an enzyme chip with spatially-addressed spots for glucose oxidase (GOD) and uricase microspots. For the SECM imaging, hydrogen peroxide generated from the GOD and/or uricase spots was directly oxidized at the tip microelectrode in a solution containing glucose and/or uric acid (electrochemical (EC) detection). For the SCLM imaging, a tapered glass capillary (i.d. of 1∼2 μm) filled with luminol and horseradish peroxidase (HRP) was used as the scanning probe for generating the chemiluminescence (CL). The inner solution was injected from the capillary tip at 78 pl s−1 while scanning above the enzyme-immobilized chip. The CL generated when the capillary tip was scanned above the enzyme spots was detected using a photon-counter (CL detection). Two-dimensional mapping of the oxidation current and photon-counting intensity against the tip position affords images of which their contrast reflects the activity of the immobilized GOD and uricase. For both the EC and CL detections, the signal responses were plotted as a function of the glucose and uric acid concentrations in solution. The sensitivities for the EC and CL detection were found to be comparable.  相似文献

Sensitive, rapid and inexpensive chemiluminescence (CL) imaging has been developed based on molecular imprinted polymer (MIP) sensing elements. Imprinted uniform microspheres were synthesized by precipitation polymerization. Microtiter plates (96 wells) were coated with polymer microspheres imprinted with trans-resveratrol, which were fixed in place using poly(vinyl alcohol) (PVA) as glue. The amount of polymer-bound trans-resveratrol was quantified using imidazole (IMZ)-catalyzed peroxyoxalate chemiluminescence (PO-CL) reaction. The light produced was then measured with a high-resolution CCD camera. Calibration curve corresponding to analyte concentration ranging from 0.3 to 25 μg mL−1 was obtained with a limit of detection 0.1 μg mL−1. These results showed that the MIP-based CL imaging sensor can become a useful analytical tool for quick simultaneous detection of trans-resveratrol in a large number of real samples.  相似文献

Human toxic responses are very often related to metabolism. Liver metabolism is traditionally studied, but other organs also convert chemicals and drugs to reactive metabolites leading to toxicity. When DNA damage is found, the effects are termed genotoxic. Here we describe a comprehensive new approach to evaluate chemical genotoxicity pathways from metabolites formed in situ by a broad spectrum of liver, lung, kidney and intestinal enzymes. DNA damage rates are measured with a microfluidic array featuring a 64-nanowell chip to facilitate fabrication of films of DNA, electrochemiluminescent (ECL) detection polymer [Ru(bpy)2(PVP)10]2+ {(PVP = poly(4-vinylpyridine))} and metabolic enzymes. First, multiple enzyme reactions are run on test compounds using the array, then ECL light related to the resulting DNA damage is measured. A companion method next facilitates reaction of target compounds with DNA/enzyme-coated magnetic beads in 96 well plates, after which DNA is hydrolyzed and nucleobase-metabolite adducts are detected by LC-MS/MS. The same organ enzymes are used as in the arrays. Outcomes revealed nucleobase adducts from DNA damage, enzymes responsible for reactive metabolites (e.g. cyt P450s), influence of bioconjugation, relative dynamics of enzymes suites from different organs, and pathways of possible genotoxic chemistry. Correlations between DNA damage rates from the cell-free array and organ-specific cell-based DNA damage were found. Results illustrate the power of the combined DNA/enzyme microarray/LC-MS/MS approach to efficiently explore a broad spectrum of organ-specific metabolic genotoxic pathways for drugs and environmental chemicals.  相似文献

Mass spectrometry (MS) has been widely used for enzyme activity assays. Herein, we propose a MALDI‐MS patterning strategy for the convenient visual presentation of multiple enzyme activities with an easy‐to‐prepare chip. The array‐based caspase‐activity patterned chip (Casp‐PC) is fabricated by hydrophobically assembling different phospholipid‐tagged peptide substrates on a modified ITO slide. The advantages of amphipathic phospholipids lead to high‐quality mass spectra for imaging analysis. Upon the respective cleavage of these substrates by different caspases, such as caspase‐1, ‐2, ‐3, and ‐8, to produce a mass shift, the enzyme activities can be directly evaluated by MALDI‐MS patterning by m/z‐dependent imaging of the cleavage products. The ability to identify drug‐sensitive/resistant cancer cells and assess the curative effects of anticancer drugs is demonstrated, indicating the applicability of the method and the designed chip.  相似文献

Simple but robust testing assays are essential for screening and diagnosis of individuals infected with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in COVID-19 pandemic. Here, we described a chemiluminescent imaging assay (CLIA) for sensitive and convenient detection of SARS-CoV-2 nucleocapsid protein (NP) by a target-induced enzyme activity regulation (T-EAR) strategy. The T-EAR used a pair of antibody-DNA probes to recognize SARS-CoV-2 NP and proximity-induce rolling circle amplification for mass-production of pyrophosphate to coordinate with Cu2+, which prevented the reduction of Cu2+ to Cu+ by sodium ascorbate as well as the Cu+-caused inactivation of horseradish peroxidase (HRP). The activity retention of HRP produced strong CL signal for the detection of SARS-CoV-2 NP by catalyzing the oxidation of luminol by H2O2. The T-EAR based CLIA showed a wide detection range from 1 pg/mL to 100 ng/mL (13 fM to 1.3 nM) with the requirement of only 0.75 μL of sample. This CLIA had advantages of good sensitivity, simple wash-free operation, acceptable accuracy, and high-throughput imaging detection, displaying potential applicability in screening assay of COVID-19 infection.  相似文献

Benzoyl-CoA epoxidase is a dinuclear iron enzyme that catalyzes the epoxidation reaction of the aromatic ring of benzoyl-CoA with chemo-, regio- and stereo-selectivity. It has been suggested that this enzyme may also catalyze the deoxygenation reaction of epoxide, suggesting a unique bifunctionality among the diiron enzymes. We report a density functional theory study of this enzyme aimed at elucidating its mechanism and the various selectivities. The epoxidation is suggested to start with the binding of the O2 molecule to the diferrous center to generate a diferric peroxide complex, followed by concerted O–O bond cleavage and epoxide formation. Two different pathways have been located, leading to (2S,3R)-epoxy and (2R,3S)-epoxy products, with barriers of 17.6 and 20.4 kcal mol–1, respectively. The barrier difference is 2.8 kcal mol–1, corresponding to a diastereomeric excess of about 99 : 1. Further isomerization from epoxide to phenol is found to have quite a high barrier, which cannot compete with the product release step. After product release into solution, fast epoxide–oxepin isomerization and racemization can take place easily, leading to a racemic mixture of (2S,3R) and (2R,3S) products. The deoxygenation of epoxide to regenerate benzoyl-CoA by a diferrous form of the enzyme proceeds via a stepwise mechanism. The C2–O bond cleavage happens first, coupled with one electron transfer from one iron center to the substrate, to form a radical intermediate, which is followed by the second C3–O bond cleavage. The first step is rate-limiting with a barrier of only 10.8 kcal mol–1. Further experimental studies are encouraged to verify our results.  相似文献

Chemiluminescence (CL) emission from luminol–tetrachloroaurate ([AuCl4]?) system studied in presence of monosaccharide sugars such as glucose and fructose was investigated on a microfluidic chip fabricated by the soft lithography technique. CL emission from the luminol–[AuCl4]? system at 430?nm was intensified remarkably by the catalytic activity of glucose and fructose at room temperature. Under optimized conditions, the CL emission intensity of the system was found to be linearly related to the concentration of the sugars. Based on this observation, nonenzymatic determination of total sugar (glucose, fructose, or hydrolyzable sucrose) was performed in a rapid and sensitive analytical method. The results revealed that the linearity ranged from 9 to 1,750?μM for glucose and 80 to 1,750?μM for fructose, with a limit of detection of 0.65 and 0.69?μM, respectively. The relative standard deviations determined at 250?μM based on six repetitive injections were 1.13 and 1.15?% for glucose and fructose, respectively. The developed method was successfully applied for determination of the total sugar concentration in food and beverages.

We presented a new strategy for ultrasensitive detection of DNA sequences based on the novel detection probe which was labeled with Ag+ using metallothionein (MT) as a bridge. The assay relied on a sandwich-type DNA hybridization in which the DNA targets were first hybridized to the captured oligonucleotide probes immobilized on Fe3O4@Au composite magnetic nanoparticles (MNPs), and then the Ag+-modified detection probes were used to monitor the presence of the specific DNA targets. After being anchored on the hybrids, Ag+ was released down through acidic treatment and sensitively determined by a coupling flow injection–chemiluminescent reaction system (Ag+–Mn2+–K2S2O8–H3PO4–luminol) (FI–CL). The experiment results showed that the CL intensities increased linearly with the concentrations of DNA targets in the range from 10 to 500 pmol L−1 with a detection limit of 3.3 pmol L−1. The high sensitivity in this work may be ascribed to the high molar ratio of Ag+–MT, the sensitive determination of Ag+ by the coupling FI–CL reaction system and the perfect magnetic separation based on Fe3O4@Au composite MNPs. Moreover, the proposed strategy exhibited excellent selectivity against the mismatched DNA sequences and could be applied to real samples analysis.  相似文献

Various strategies have been proposed for the detection of disease protein biomarkers; however, most methods are too expensive, cumbersome or limited in sensitivity for clinical use. Here, we report that a fabricated complex can be used as a powerful tool to detect trace proteins in complex samples. In this strategy, a DNA–protein complex that comprises of one target molecule and two or more deoxyribozyme-containing probes can exhibit autonomous cleavage behavior on the surface of the substrate DNA modified electrode. In the meantime, the complex can remove the cleaved DNA fragment from the electrode surface by taking advantage of the proximity effect. The proposed approach allows one-step and highly sensitive detection of a variety of targets based on the changes of the direct electrochemical readout. Moreover, this method may also have considerable advantages over the commonly reported DNA amplification-assisted immunoassays, particularly in terms of assay simplicity and cost, which may hold great potential for application in resource-constrained regions.  相似文献

The effective removal and selective detection of explosive and toxic pollutant trinitrophenol (TNP) is an attractive but challenging field. Herein, a double-cavity nor-seco-cucurbit[10]uril (ns-Q[10])-based supramolecular assembly 8-HQ@ns-Q[10] was fabricated and its structure was characterized by X-ray single crystal diffraction. In this assembly, the stoichiometric ratio of ns-Q[10] and 8-hydroxyquinoline (8-HQ) is 1:2, which is also attributed to the special double-cavity structure of ns-Q[10]. The luminescence sensing experiments showed that 8-HQ@ns-Q[10] can be used as a good fluorescence-enhanced sensing material (enhanced 27-fold) for the rapid detection of explosives and the aqueous contaminant TNP, with a limit of detection (LOD) of 2.07 × 10−5 mol/L, without interference from other phenolic compounds. Furthermore, TNP can be efficiently removed in the presence of assembly 8-HQ@ns-Q[10], and the removal efficiency is more than 89%. Therefore, the supramolecular assembly 8-HQ@ns-Q[10], as a fluorescence-enhanced luminescence sensor and adsorption material, has rich research value and potential application prospect when applied to the detection and removal of TNP in aqueous environment.  相似文献

This work reports a newly designed pH-activatable and aniline-substituted aza-boron-dipyrromethene as a trifunctional photosensitizer to achieve highly selective tumor imaging, efficient photodynamic therapy (PDT) and therapeutic self-monitoring through encapsulation in a cRGD-functionalized nanomicelle. The diethylaminophenyl is introduced in to the structure for pH-activatable near-infrared fluorescence and singlet oxygen (1O2) generation, and bromophenyl is imported to increase the 1O2 generation efficiency upon pH activation by virtue of its heavy atom effect. After encapsulation, the nanoprobe can target αvβ3 integrin-rich tumor cells via cRGD and is activated by physiologically acidic pH for cancer discrimination and PDT. The fascinating advantage of the nanoprobe is near-infrared implementation beyond 800 nm, which significantly improves the imaging sensitivity and increases the penetration depth of the PDT. By monitoring the fluorescence decrease in the tumor region after PDT, the therapeutic efficacy is demonstrated in situ and in real time, which provides a valuable and convenient self-feedback function for PDT efficacy tracking. Therefore, this rationally designed and carefully engineered nanoprobe offers a new paradigm for precise tumor theranostics and may provide novel opportunities for future clinical cancer treatment.  相似文献

An improved microfluidic chip-based sequential-injection trapped-droplet array liquid-liquid extraction system with chemiluminescence (CL) detection was developed in this work. Two recess arrays were fabricated on both sides of the extraction channel to produce droplet arrays of organic extractant. A chip integrated monolithic probe was fabricated at the inlet of the extraction channel on the glass chip instead of the capillary probe connected to the microchannel, in order to improve the system stability and reliability. A slotted-vial array system coupled with the monolithic probe was used to sequentially introduce sample and different solvents and reagents into the extraction channel for extraction and CL detection. The performance of the system was demonstrated in the determination of Al3+ using Al3+-dihydroxyazobenzene (DHAB) and tributyl phosphate (TBP) extraction system. The operation conditions, including extraction time, concentration and flow rate of the CL reagents, were optimized. Within one analysis cycle of 12 min, an enrichment factor of 85 was obtained in the extraction stage with a sample consumption of 1.8 μL. The consumption of CL reagent, bis(2-carbopentyloxy-3,5,6-trichlorophenyl)oxalate (CPPO), was 120 nL/cycle. The detection limit of the system for Al3+ was 1.6 × 10−6 mol/L with a precision of 4.5% (R.S.D., n = 6).  相似文献

A mimic-hybridization chain reaction (mimic-HCR) amplified strategy was proposed for sensitive electrochemically detection of DNA methylation and methyltransferase (MTase) activity In the presence of methylated DNA, DNA-gold nanoparticles (DNA-AuNPs) were captured on the electrode by sandwich-type assembly. It then triggered mimic-HCR of two hairpin probes to produce many long double-helix chains for numerous hexaammineruthenium (III) chloride ([Ru(NH3)6]3+, RuHex) inserting. As a result, the signal for electrochemically detection of DNA MTase activity could be amplified. If DNA was non-methylated, however, the sandwich-type assembly would not form because the short double-stranded DNAs (dsDNA) on the Au electrode could be cleaved and digested by restriction endonuclease HpaII (HapII) and exonuclease III (Exo III), resulting in the signal decrement. Based on this, an electrochemical approach for detection of M.SssI MTase activity with high sensitivity was developed. The linear range for M.SssI MTase activity was from 0.05 U mL−1 to 10 U mL−1, with a detection limit down to 0.03 U mL−1. Moreover, this detecting strategy held great promise as an easy-to-use and highly sensitive method for other MTase activity and inhibition detection by exchanging the corresponding DNA sequence.  相似文献

Sandwich ELISA methods have been widely used for biomarker and pathogen detection because of their high specificity and sensitivity. However, the main drawbacks of this assay are the cost, the time-consuming procedure for the isolation of antibodies and their poor stability. To overcome these restrictions, we herein fabricated artificial antibodies based on imprinting technology and developed a sandwich ELISA for pathogen detection. Both the capture and detection antibodies were obtained via an in situ method, with simplicity, rapidity and low cost. The peroxidase mimics, the CeO2 nanoparticles, as signal generators were integrated with the detection antibody. The fabricated artificial antibodies exhibited not only natural antibody-like binding affinities and selectivities, but also superior stability and reusability. The detection limit was about 500 CFU mL–1, which is much lower than that of traditional ELISA methods (104 to 105 CFU mL–1). Furthermore, the capture antibody can disinfect pathogens in situ.  相似文献
