(3S,4R)-Bengamide E(2) was synthesized starting from D-glucono-δ-lactone(3) and the key deoxygenation step from 13 to 15 was achieved by the application of NaBH3CN and ZnI2.Compared with natural bengamide E(1),the synthetic compound(35,4R)-bengamide E(2) was inactive against the cell growth of HUVEC and cancer cells.These data represent the significance of the stereochemistry at C-3 and C-4 of bengamides for structural recognition and binding with the target(s).  相似文献

A concise total synthesis of (3R),(5R)-5-hydroxy-de-O-methyllasiodiplodin and its epimer have been synthesized from commercially available (R)-propylene oxide and Orsellinic acid as starting materials. 以(R)-( )-pu legone为起始原料,经1,4-加成,还原两步反应合成了手性辅助试剂( )-(1S,2S,5R)-8-联苯薄荷醇及其差向异构体(-)-(1R,2S,5R)-8-联苯薄荷醇,总产率95%。其结构经1H NMR,13C NMR,IR,MS和X-射线衍射仪表征。  相似文献

研究了一条新的路线用于他汀类药物的重要中间体(R)-4-氰基-3-羟基丁酸乙酯的合成. 以廉价、易得的L-(-)-苹果酸为起始原料, 经酯化、还原、溴代和氰化四步反应得到目标化合物(R)-4-氰基-3-羟基丁酸乙酯, 合成总收率为56.7%. 所有中间体和最终产物均由ESI-MS, 1H NMR和13C NMR光谱及比旋光度表征并与文献值比较. 该方法原料易得、操作简便、收率良好, 产物容易分离纯化, 是一条适合大规模制备(R)-4-氰基-3-羟基丁酸乙酯的新合成工艺路线.  相似文献

用KBH_4,CaCl_2为还原剂使D及L缬氨酸甲酯还原得到光学活性产物R-3-甲基-2-氨基丁醇(2a)及S-3-甲基-2-氨基丁醇(2b)。2a,2b同CS_2在KOH存在下反应得到(R)-4-异丙基四氢噻唑-2-硫酮(3a)及(S)-4-异丙基四氢噻唑-2-硫酮(3b)。肉桂酰氯分别同3a及3b在Et_3N存在下反应得到N-肉桂酰(R)-4-异丙基四氢噻唑-2-硫酮(4a)及N-肉桂酰(S)-4-异丙基四氢噻唑-2-硫酮(4b)。用半经验的量子化学PM3方法研究了反应物和产物的电子结构,得到了产物的最优构型和电荷键序分布以及反应焓变。  相似文献

价廉易得的L-亮氨酸先以苄基同时保护氨基及羧基得(S)-2-(二苄胺基)-4-甲基戊酸苄酯,进而在碱性条件下与乙腈发生亲核取代反应得(S)-4-(二苄胺基)-6-甲基-3-氧代庚腈,再经硼氢化钠选择性还原羰基得(3S,4S)-4-(二苄胺基)-3-羟基-6-甲基庚腈,用双氧水氧化得(3S,4S)-4-(二苄胺基)-3-羟基-6-甲基庚酸,最后在Pd(OH)2/C-H2作用下脱掉苄基得到(3S,4S)-4-氨基-3-羟基-6-甲基庚酸,即(3S,4S)-statine。整个合成路线总产率为33.6%。  相似文献

Synthesis of (2R,3S,4S)-4-aryl-3-hydroxyprolinols has been established starting from 2-benzyloxymethylpyrrolidin-2-one framework, which is derived from commercially available trans-(2S,4R)-4-hydroxyproline. The single diastereomer having a trans–cis relative configuration with C2 and C3 and C3 and C4 is constructed in two one-pot functional group transformations of Grignard addition/dehydration and epoxidation/isomerization as the key steps in moderate yield.  相似文献

离子液体中脂肪酶催化拆分外消旋烯丙酮醇反应   总被引:2,自引:0,他引:2  
 以离子液体为溶剂,考察了溶剂类型、水活度、温度、 pH值和共溶剂等因素对脂肪酶催化拆分外消旋烯丙酮醇(R,S)-4-羟基-3-甲基-2-(2-丙烯基)-2-环戊烯-1-酮反应的影响,并与常用于外消旋烯丙酮醇拆分的有机溶剂乙酸乙烯酯进行了比较. 结果表明,在离子液体[bmim]PF6中脂肪酶的催化性能较好,酶初始反应速率为18.48 μmol/(g·min), 半衰期为74.53 h, 高于在乙酸乙烯酯中的相应值(9.18 μmol/(g·min)和64.29 h). 但离子液体中拆分反应的转化率低于在乙酸乙烯酯中的转化率,可以通过向离子液体中补加酰基供体来提高外消旋烯丙酮醇的转化率. 两种反应介质中最佳酶反应条件均为水活度0.17, 温度40 ℃和pH=7, 但加入共溶剂后,离子液体中脂肪酶催化拆分外消旋烯丙酮醇的效率降低,而在乙酸乙烯酯中则有所提高.  相似文献

A new anti-inflammatory active phenylpropenoid, (R,E)-1-[4-(3-hydroxyprop-1-enyl) phenoxyl]-3-methylbutane-2,3-diol (1), isolated from the stem wood of Zanthoxylum integrifoliolum, has been synthesized for the first time using commercially available 4-hydroxy benzaldehyde (2). The key step involves the Sharpless asymmetric dihydroxylation of olefin (3).

An efficient and highly stereocontrolled convergent synthesis of fluvirucinine A1 is reported herein. In fluvirucinine A1 both C5-C13 and C1-C4 fragments were accessed from a common intermediate 6 derived from (S)-Roche ester in 15 and 7 steps, respectively. (3Z,9Z,6S,7R)-6,7-epoxy-3,9-octadecadiene (1) and (3Z,9Z,6R,7S)-6,7-epoxy-3,9-octadecadiene (2) have been stereoselectively synthesized in eight steps from 2-pentyn-1-ol with an overall yield of 8%. The key steps involved the Sharpless asymmetric dihydroxylation of (2E)-oct-2-en-5-yn-1-ol (6). The new synthetic method is suitable for multigram-scale preparation of 1 and 2 and might be used for producing sufficient quantities of the sex pheromone components for management of the pest of tea plantations.  相似文献
