Don't get trapped : The effect of conjugating electron‐withdrawing groups and α‐anion‐stabilizing heteroatom substituents on configurational stability of chiral carbanions through a double bond was examined on the basis of extent of chirality transfer in intramolecular trapping in [2,3]‐Wittig rearrangement of chiral 3‐substituted 1‐propenyloxy‐1‐phenyl‐2‐propen‐1‐yl carbanions (see scheme).

Small libraries of 3,5‐unsubstituted 4‐substituted‐6‐aryl‐3,4‐dihydropyridin‐2(1H)‐ones derivatives were synthesized from the condensation‐products of aldehydes with Meldrum's acid, aromatic ketones and ammonium acetate using acetic acid as energy transferring‐agent under microwave irradiation without catalyst. This method has the advantages of excellent yields (65‐90%), short reaction time (5‐10 min) and being environmentally friendly. It aimed to provide new series of potential biologically active compounds for biomedical screening.  相似文献

New synthetic approaches to 3‐substituted‐5′‐(N‐pyridiniummethyl)‐4′,5′‐dihydropsoralens are described. The novel pathways presented utilize appropriately substituted coumarins and 4′,5′‐dihydropsoralens. The synthesis and characterisation of a series of novel 4‐acylamino and 4‐alkylamino‐N‐1,8‐naphthalimides is described. The UV‐visible absorption and emission properties of the compounds are reported. Significant solvent effects are noted for 4‐n‐butyl‐9‐n‐butyl‐1,8‐naphthylimide. The incorporation of acetyl and chloroacetyl groups into the 4‐substituent markedly increases the fluorescence quantum yield compared with 4‐alkylamino substituemnts.  相似文献

Addition reactions of thioamide dianions that were derived from N‐arylmethyl thioamides to imines and aziridines were carried out. The reactions of imines gave the addition products of N‐thioacyl‐1,2‐diamines in a highly diastereoselective manner in good‐to‐excellent yields. The diastereomeric purity of these N‐thioacyl‐1,2‐diamines could be enriched by simple recrystallization. The reduction of N‐thioacyl‐1,2‐diamines with LiAlH4 gave their corresponding 1,2‐diamines in moderate‐to‐good yields with retention of their stereochemistry. The oxidative‐desulfurization/cyclization of an N‐thioacyl‐1,2‐diamine in CuCl2/O2 and I2/pyridine systems gave the cyclized product in moderate yield and the trans isomer was obtained as the sole product. On the other hand, a similar cyclization reaction with antiformin (aq. NaClO) as an oxidant gave the cis isomer as the major product. The reactions of N‐tosylaziridines gave the addition products of N‐thioacyl‐1,3‐diamines with low diastereoselectivity but high regioselectivity and in good‐to‐excellent yields. The use of AlMe3 as an additive improved the efficiency and regioselectivity of the reaction. The stereochemistry of the obtained products was determined by X‐ray diffraction.  相似文献

Unambiguous characterization of 5‐substituted‐4‐thiopyrimidine nucleosides (ribonucleosides and 2'‐deoxynucleosides) was performed using NMR spectroscopy. Assignments of all proton and carbon signals of 5‐bromo‐4‐thiouridine and related nucleosides were systematically carried out and firmly established by COSY and HMQC techniques. The NMR data of various 4‐thiopyrimidine nucleosides are compared, and the key contributing factors discussed. The approach presented here is applicable to other modified nucleosides and nucleotides, as well as nucleobases. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

A fast, efficient and convenient reaction of 6,7,8‐trimethoxy‐4‐chloroquinazoline and aryl (or benzyl) amines was achieved under microwave irradiation in isopropyl alcohol, providing a simple method for synthesis of novel 6,7,8‐trimethoxy N‐substituted‐4‐aminoquinazoline compounds in good yield in short time. The title compounds were evaluated for their in vitro anti‐proliferative activities against PC3 cell by MTT method.  相似文献

The asymmetric conjugate addition of malonate esters to α,β‐unsaturated N‐sulfonyl imines is catalyzed by PyBOX/La(OTf)3 complexes in the presence of 4 Å MS. The reaction gives the corresponding E enamines bearing a stereogenic center at the allylic position with good yields and enantiomeric ratios up to 97:3. This reaction provides a synthetic entry to chiral δ‐aminoesters and piperidones.  相似文献

1‐Substituted‐3‐dimethylaminopropenones 1a‐d reacted with acetylacetone and with ethyl acetoacetate to yield regioselectively 2,3,6‐trisubstituted pyridines. Refluxing 1a‐d in acetic acid/ammonium acetate resulted in the formation of 6‐substituted‐3‐aroylpyridines, whereas refluxing in acetic acid alone afforded 1,3,5‐triaroylbenzene.  相似文献

We report herein the synthesis of new synthons obtained by reductive opening of 3‐glycosyl‐5‐substituted‐2‐isoxazolines. Different cleavage products have been obtained depending on the substituents (aliphatic or aromatic) in position five of the heterocycle ring. In this paper the regiospecific transformations of methyl 2‐(benzyloxycarbonyl)amino‐3‐dimethylaminopropenoate ( 1 ) with hydrazine, alkyl‐, aryl‐ and heteroaryl‐substituted hydrazines via the corresponding hydrazones 12‐16 into pyrazoles 17‐25 are described. Heteroaryl‐substitued hydrazones 13‐16 afforded by oxidation with bromine or lead tetraacetate the corresponding substituted (1,2,4‐triazolo[4,3‐b]pyridazin‐3‐yl)glycinates 27‐30 . Alkyl 2‐(2,2‐disubstituted‐1‐ethenyl)amino‐3‐dimethylaminopropenoates 31‐33 gave with hydrazines alkyl 2‐[2,2‐(disubstituted)ethenyl]amino‐3‐heteroarylhydrazonopropanoates 40‐48 and 2‐alkyl 2,3‐bis((hetero)arylhydrazono)propanoates 51‐55 .  相似文献

Enantiomerically enriched α,α‐disubstituted phenylacetonitriles have been readily prepared by stereoselective quaternization of 2‐alkyl‐2‐[2‐(p‐tolylsulfinyl)phenyl]acetonitriles with different alkylating electrophiles in the presence of bases. The use of potassium hexamethyldisilazane (KHMDS)/[18]crown‐6 ether and NHMDS with alkyl halides afforded S,SS and R,SS diastereoisomers, respectively, in high enantiomeric purities, thus providing stereodivergent processes for synthesizing both isomers. The dependence of the stereochemical course of the reactions on the experimental conditions (mainly on the counterion) has been rationalized by assuming a planar or pyramidal structure for the benzylic carbanions. This hypothesis has been supported by NMR spectroscopic studies, which permit one to assign a chelated pyramidal structure to the sodium benzylic carbanions and an almost planar naked carbanionic structure to the potassium benzylic carbanions generated in the presence of [18]crown‐6 ether.  相似文献
