An Electron Spin Resonance (ESR) study of radiation induced free radicals in the commonly used detergent powder (Surf), was conducted to examine its potential application for retrospective/high level dosimetry, including possible dating of the accidents. The radical ions present in the irradiated detergent powder were identified as arising predominantly from sodium tri poly phosphate (Na3P5O10) and sodium sulphate (Na2SO4). The physical mixture of sodium tri poly phosphate and sodium sulphate in 4:1 ratio was found to give an ESR response, similar to that of the detergent powder. These results suggest that the detergent powder can be used as a dosimeter in the 20 Gy to 18 kGy dose range, spanning over 3 orders of magnitude. The second derivative ESR spectra of the detergent powder irradiated to different doses showed that the relative yield of the radicals is independent of irradiation dose. The decay pattern of radicals when followed as a function of post irradiation days, lead to the idea about the contrasting decay characteristics of the radicals. From the ESR signals of the irradiated detergent samples, it is shown that radiation dose can be evaluated and the radiation incident can be dated with an accuracy of ±10%.  相似文献

In view of an increasing demand for food irradiation technology, the development of a reliable means of detection for the control of irradiated foods has become necessary. Various vegetable food materials (dried cabbage, carrot, chunggyungchae, garlic, onion, and green onion), which can be legally irradiated in Korea, were subjected to a detection study using ESR spectroscopy. Correlation coefficients (R2) between absorbed doses (2.5–15 kGy) and their corresponding ESR signals were identified from ESR signals. Pre-established threshold values were successfully applied to the detection of 54 coded unknown samples of dried clean vegetables (chunggyungchae, Brassica camestris var. chinensis), both non-irradiated and irradiated. The ESR signals of irradiated chunggyungchae decreased over a longer storage time, however, even after 6 months of ambient storage, these signals were still distinguishable from those of non-irradiated samples. The most successful estimates of absorbed dose (5 and 8 kGy) were obtained immediately after irradiation using a quadratic fit with average values of 4.85 and 8.65 kGy being calculated.  相似文献

Four different techniques to detect the effect of irradiation in beans were investigated. Two types of Brazilian beans, Phaseolus vulgaris L., var. carioca and Vigna unguiculata (L.) Walp, var. macaçar, were irradiated using a 60Co source with doses ranging from 10.0 kGy. After 6 months storage at ambient temperature the detection tests were carried out. Firstly, germination tests showed markedly reduced root growth and almost totally retarded shoot elongation of irradiated beans as compared to non-irradiated beans. Secondly, DNA fragmentation was studied using a microgel electrophoresis. Irradiated cells produced typical comets with DNA fragments migrating towards the anode. DNA of non-irradiated cells exhibited a limited migration. Thirdly, electron spin resonance for detection of cellulose radicals was tested, since it was expected that these free radicals are quite stable in solid and dry foods. However, only in beans irradiated with 10 kGy a small signal could be detected. Fourtly, thermoluminescence, a method to analyze mineral debris adhering to food, turned out to be a good choice to detect irradiation effects in beans, even after 6 months of storage. The results indicate that three of these four techniques proposed, can be used to detect the effect of irradiation in these two varieties of Brazilian beans at a dose level useful for insect disinfestation (1 kGy).  相似文献

Three different techniques, photostimulated luminescence (PSL), electron spin resonance (ESR) and thermoluminescence (TL) were applied for the detection of dried anchovy and shrimp exposed to electron beam at 0–10 kGy. PSL values for irradiated samples were more than 5000 photon counts/60 s, upper threshold (T2), whereas those of non-irradiated samples were <700 counts (lower threshold, T1) in anchovy and intermediate values of T1T2 in shrimp. ESR measurements using both the whole samples did not show any signals specific to irradiation. However, in the case of anchovy it was possible to use bone for ESR detection, showing typical signals (g=2.002, 1.998). Minerals separated from both the samples for TL measurement showed that non-irradiated samples were characterised by glow curves situated at about 300°C with low intensity, while all irradiated samples showed glow peaked at about 200°C and its intensity was high enough to be discriminated from the non-irradiated ones. Furthermore, normalization by a re-irradiation enhanced the reliability of detection results of TL. In conclusion, a multi-step detection using different methods enhances confidence in the detection of irradiated food.  相似文献

Antioxidants quench oxidation by transferring hydrogen atoms to free radicals. In the present investigation, the effect of gamma irradiation on the natural antioxidants of irradiated cumin was studied. Cumin samples were purchased from retailers and then irradiated in a cobalt-60 irradiator to 0, 1, 3, 5 and 10 kGy at ambient temperature. The effect of irradiation on the antioxidant properties of the cumin seed were investigated by evaluating the radical-scavenging effect on the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, determination of ferric reducing antioxidant power (FRAP), total polyphenol content (TPC) and the antioxidant index by β-carotene/linoleic acid co-oxidation. Electron spin resonance (ESR) was performed to assess ionization of cumin seeds by gamma irradiation. Irradiation was found to nonsignificantly increase and/or maintain all antioxidant parameters, TPC and the ESR signal intensity was found to be increased in cumin seeds.  相似文献

Cooked, frozen or chilled whole tails of Norway lobster were either not irradiated or given doses of 1, 2, 3, 4 or 5 kGy using a cobalt 60 source. The cuticle was removed, freeze-dried and ground before determination of the free radical concentration using electron spin resonance (ESR) spectroscopy. The ESR signal strength increased linearly with increasing irradiation dose. In comparison to the chilled samples, cooking before irradiation significantly increased (69%) signal intensity whereas cooking after irradiation decreased (27%) signal strength. Irradiating the samples in the frozen state did not significantly alter the free radical concentration in the cuticle.  相似文献

Fruit may be irradiated at rather low doses, below 1 kGy in combination treatments or for quarantine purposes. To improve the ESR detection sensitivity of irradiated fruit de Jesus et al. (Int. J. Food Sci. Technol. 34 (1999) 173.) proposed extracting the fruit pulp with 80% ethanol and measuring the residue with ESR using low power (0.25 mW) for detection of ‘cellulosic’ radicals. An improvement in ESR sensitivity using the extraction procedure could be confirmed in this paper for strawberries and papayas. In most cases, a radiation dose of 0.5 kGy could be detected in both fruits even after 2–3 weeks storage. In addition, some herbs and spices were also tested, but only for a few of them the ESR detection of the ‘cellulosic’ signal was improved by previous alcoholic extraction.

As an alternative to ESR measurements, other detection methods like DNA Comet Assay and thermoluminescence were also tested.  相似文献

The electron spin resonance (ESR) readout of radical concentration in irradiated L-alanine is well known as a transfer dosimetry system, however, the high cost of the ESR equipment is a serious handicap for large-scale routine application. In this study the reaction between irradiated L-alanine powder and aqueous solution of triphenyl tetrazolium chloride (TTC) was studied. The radical produced by irradiation of alanine reacts during the dissolution with TTC forming pink-colored formazans with absorption maxima around 401 and 487 nm. This readout method can be used in the 2–200 kGy dose range.  相似文献

Accidental exposure dose assessment by electron spin resonance (ESR) technique from the free radicals generated in a cotton handkerchief has been attempted in this investigation. The cotton handkerchief, a common material carried by individuals, was taken as the medium for free radical estimation. About 55 mg of the irradiated piece of cloth was loaded into a quartz tube and the dose dependence of the ESR signal at g=2.0026 was measured at room temperature, using a Bruker ESP-300 ESR spectrometer in X-band (9.74 GHz). The intensity of this signal was found to be proportional to the dose in the range of 1–1000 Gy. The stability of the free radicals with time of storage was followed. Dependence of dose rate as well as the presence of water on the yield of free radicals were also investigated.  相似文献

Formates and dithionates of 6Li, enriched and 7Li in natural composition of Li offer a possibility to measure the absorbed dose from photons and thermal neutrons in a mixed radiation field for instance at a boron neutron capture therapy (BNCT) facility. Tests with formates and dithionates of enriched 6Li and lithium compounds with natural composition have been performed at the BNCT facility at Studsvik, Sweden. Irradiations have been performed at 3 cm depth in a Perspex phantom in a fluence rate of thermal neutrons 1.8 x 10(9) n cm(-2) s(-1). The compounds were also irradiated in a pure X-ray field from a 4MV linear accelerator at 5 cm depth in a phantom with accurately determined absorbed doses. The signal intensity and shape was investigated within 3 h after the irradiation. A single line spectrum attributed to the CO2- radical was observed after irradiation of lithium formate. An increase in line width occurring after neutron irradiation in comparison with photon irradiation of the 6Li sample was attributed to dipolar broadening between CO2- radicals trapped in the tracks of the alpha particles. A spectrum due to the SO3- radical anion was observed after irradiation of lithium dithionate. The signal amplitude increased using the 6Li in place of the Li with natural composition of isotopes, in studies with low energy X-ray irradiation. Due to the decreased line width, caused by the difference in g(N) and I between the isotopes, the sensitivity with 6Li dithionate may be enhanced by an order of magnitude compared to alanine dosimetry. After comprehensive examination of the different combinations of compounds with different amounts of 6Li and 7Li regarding dosimetry, radiation chemistry and EPR properties these dosimeter material might be used for dose determinations at BNCT treatments and for biomedical experiments. Interesting properties of the radical formation might be visible due to the large difference in ionization density of neutrons compared to photons.  相似文献

Dried herbal samples consisting of root, rhizome, cortex, fruit, peel, flower, spike, ramulus, folium, and whole plant of 20 different medicinal herbs were investigated using pulsed photostimulated luminescence (PPSL), thermoluminescence (TL), and electron spin resonance spectroscopy (ESR) to identify γ-ray irradiation treatment. Samples were irradiated at 0–50 kGy using a 60Co irradiator. PPSL measurement was applied as a rapid screening method. Control samples of 19 different herbs had photon counts less than the lower threshold value (700 counts 60 s−1). The photon counts of non-irradiated clematidis radix and irradiated evodia and gardenia fruits were between the lower and upper threshold values (700–5,000 counts 60 s−1). TL ratios, i.e., integrated areas of the first glow (TL1)/the second glow (TL2), were found to be less than 0.1 in all non-irradiated samples and higher than 0.1 in irradiated ones providing definite proof of radiation treatment. ESR spectroscopy was applied as an alternative rapid method. In most of the irradiated samples, mainly radiation-induced cellulosic, sugar, and relatively complicated carbohydrate radical ESR signals were detected. No radiation-specific ESR signal, except one intense singlet, was observed for irradiated scrophularia and scutellaria root and artemisiae argyi folium. Figure PPSL can be used as a rapid simple preliminary screening method and a combination of ESR and TL tests for a definite proof of gamma irradiation treatment of medicinal herbs.  相似文献

The representative ESR spectrum of the pepper composed of a sextet centered at g=2.0, a singlet at the same g-value and a singlet at g=4.0. This reflects the evidence of three independent radicals in the pepper before irradiation. Upon gamma ray irradiation, a new pair of signals appeared. The progressive saturation behavior (PSB) at various microwave power levels indicates quite different relaxation behaviors of those signals. For the evaluation of radiation-induced radicals and irradiation effects we propose a new protocol using the PSB method. We found various free radicals in a commercially available pepper in Japan before and after irradiation using electron spin resonance (ESR) spectroscopy. The typical ESR spectrum of the pepper consists of a sextet centered at g = 2.0, a singlet at the same g-value and a singlet at g = 4.0. Upon gamma ray irradiation, a new pair of signals appeared in the pepper. The progressive saturation behavior (PSB) at various microwave power levels indicated quite different relaxation behaviors of those radicals. Namely, the peak intensity of the organic free radical component decreases in a monotonic fashion, whereas the Mn2+ and Fe3+ ESR signals substantially keep constant. This reflects the evidence of three independent radicals in the pepper before irradiation. The PSB of the pair peaks as induced by irradiation possessed quite different PSB from that of the free radical located at g = 2.0. We proposed a new protocol for the ESR detection of irradiated foods by the PSB method at different microwave power levels. This would call for a major modification of the CEN protocol in European Union.  相似文献

Suspensions of Propionibacterium acnes were UV irradiated and the induced radicals were measured at 77 K by electron spin resonance (ESR) spectrometry. Two types of radical were formed during irradiation and stabilized in the frozen suspensions. The relative yield of each radical was studied as a function of irradiation wavelength. The first radical, which was a singlet with a peak-to-peak width of 20 G, was insensitive to the deoxygenation of the samples and to the exchange of solvent water by heavy water. The action spectrum was similar to the absorption spectrum of NADPH. The second type of radical was not formed in deoxygenated samples and the shape of the ESR spectrum was characteristic of the superoxide radical. This radical was only formed at wavelengths below 340 nm.  相似文献

Using electron spin resonance (ESR) spectroscopy, we revealed the presence of four radical species in gamma-ray irradiated ginseng (Agaliaceae). Before irradiation, the representative ESR spectrum of ginseng is composed of a sextet centered at g = 2.0, a sharp singlet at the same g-value, and a singlet at about g = 4.0. The first one is attributable to a hyperfine (hf) signal of Mn2+ ion (hf constant: 7.4 mT). The second one is due to an organic free radical. The third one is originated from Fe3+. Upon gamma-ray irradiation, a new ESR (the fourth) signal was detectable in the vicinity of g = 2.0 region. The progressive saturation behaviors of the ESR signals at various microwave power levels were indicative of different relaxation time for those radicals. The anisotropic ESR spectra were detected by the angular rotation of the sample tube. This is due to the existence of anisotropic microcrystalline in the ginseng powder sample.  相似文献

We investigated sucrose radicals produced by heavy-ion irradiation with various linear energy transfer (LETs) and the possibility for a sucrose electron spin resonance (ESR) dosimeter. The impact of heavy ions on sucrose produced sucrose radicals, which were measured by ESR. The obtained spectral pattern was the same as that for helium (He) ions, carbon (C) ions, neon (Ne) ions, iron (Fe) ions, and gamma-ray irradiation. Identical spectra were measured after 1 year, but the initial intensities decreased by a few percent when the samples were kept in ESR tubes with the caps at ambient temperature. The total spin concentration obtained by heavy-ion irradiation had a linear relation with the absorbed dose, and correlated logarithmically with the LET. Qualitative ESR analyses showed that the production of sucrose radicals depended on both the particle identity and the LET at the same dose. The production of spin concentration by He ions was the most sensitive to LET. Empirical relations between the LET and the spin yield for various particles imply that the LET at a certain dose can be estimated by the spin concentration. Therefore, the present ESR results imply that sucrose can be used to monitor the absorbed dose and the LET of particle irradiation.  相似文献

Two ethylene–octene copolymers (POE) were irradiated with 60Co gamma radiation and influence of irradiation atmosphere, absorbed dose and heat treatment of samples on the crosslinking were studied. Thermal properties and crystalline morphology of non-irradiated and irradiated POE were determined by using differential scanning calorimetry (DSC) and wide-angle X-ray diffraction (WAXS), respectively. The Charlesby–Pinner equation was used to describe the relationship between absorbed dose and sol fraction. The gel fraction of irradiated POE was lower and decreased with the increase of octene content when irradiated in oxygen, compared to irradiation in nitrogen atmosphere. The gel fraction increased significantly with the increasing of absorbed dose for the two copolymers. Heat treatment of samples prohibited the crosslinking of irradiated POE. The DSC results indicated that a subtle change of thermal properties of POE was observed before and after gamma irradiation at low dose. No change was found from the WAXS spectra of non-irradiated and irradiated POE. For heat-treating samples, the Charlesby–Pinner equation can not fit perfectly with the relationship between the sol fraction and absorbed dose, but it fits well with the crosslinking reaction of POE pellets.  相似文献

The nature and behavior of free radicals induced in acetylated cotton celluloses irradiated with γ-rays have been studied by electron spin resonance (ESR) spectroscopy. Dehydrogenation and deacetylation appear to be responsible for the free radicals observed from samples irradiated at 77°K. The degree of substitution enhanced the yield of acetyl radicals when the samples were irradiated at 77°K and adversely affected the overall radical concentration when irradiation was done at 300°K. In addition, the ESR spectra of samples irradiated under vacuum at 300°K were more intense than those obtained from samples irradiated in air. The nature, yield, and post-irradiation behavior of the primary radicals are discussed in the light of the ultimate chemical effects observed.  相似文献

In some countries, clearance has been given for treating certain types of shellfish by ionizing radiation in order to increase the shelf-life and to reduce health hazards which might be caused by contaminating microorganisms. In the present study, thermoluminescence (TL) analysis was used to examine the irradiation status of shellfish products purchased from local suppliers. For analysis minerals were isolated from the guts of the animals. Although on none of the examined products an irradiation treatment prior to analysis could be shown, the results obtained on non-irradiated and irradiated products have revealed that irradiation within the commercially used dose range can clearly be detected. Already first glow TL intensities of minerald indicated irradiation treatments. Normalized TL signals of non-irradiated and irradiated samples were clearly separated. By calculation of differences of TL intensities and TL signals between non-irradiated and irradiated samples in dependency of integration temperature an optimized integration area for glow curves was determined. The result of this study agree well with results obtained by two large-scale intercomparisons between food control laboratories to detect irradiation treatment of spices and herbal products as well as of fruit and vegetables by TL analysis of contaminating minerals.  相似文献

The effects of high dose γ-irradiation on six herbal medicines were investigated using gas chromatography–mass spectrometry (GC/MS) and high-performance liquid chromatography (HPLC). Herbal medicines were irradiated at 0–50 kGy with 60Co irradiator. HPLC was used to quantify changes of major components including glycyrrhizin, cinnamic acid, poncirin, hesperidin, berberine, and amygdalin in licorice, cinnamon bark, poncirin immature fruit, citrus unshiu peel, coptis rhizome, and apricot kernel. No significant differences were found between gamma-irradiated and non-irradiated samples with regard to the amounts of glycyrrhizin, berberine, and amygdalin. However, the contents of cinnamic acid, poncirin, and hesperidin were increased after irradiation. Volatile compounds were analyzed by GC/MS. The relative proportion of ketone in licorice was diminished after irradiation. The relative amount of hydrocarbons in irradiated cinnamon bark and apricot kernel was higher than that in non-irradiated samples. Therefore, ketone in licorice and hydrocarbons in cinnamon bark and apricot kernel can be considered radiolytic markers. Three unsaturated hydrocarbons, i.e., 1,7,10-hexadecatriene, 6,9-heptadecadiene, and 8-heptadecene, were detected only in apricot kernels irradiated at 25 and 50 kGy. These three hydrocarbons could be used as radiolytic markers to distinguish between irradiated (>25 kGy) and non-irradiated apricot kernels.  相似文献
