Human transferrin (Tf) is a model glycoprotein for congenital disorders of glycosylation (CDG) diagnosis. In the last few years, new CE-UV methods for intact Tf glycoforms analysis have been developed using nonvolatile BGEs and organic modifiers. However, the use of these BGEs does not allow the coupling of these procedures with electrospray MS (ESI-MS). In this study, a new CE-UV separation method of Tf glycoforms is developed, using a double-layer stable coating and a volatile BGE based on ammonium acetate. The separation method is optimized using standard Tf and their potential is demonstrated applying the method to the analysis of sera Tf from healthy individuals and CDG patients. The CE-UV separation method has been coupled to ESI-MS detection. Main parameters such as sheath liquid composition are optimized in order to obtain a good sensitivity. The CE-ESI-MS method has also been used in serum samples obtaining the separation of the different proteins present in serum and partial separation of Tf glycoforms. Different mass spectra and deconvoluted molecular masses were obtained for each sialoform, allowing unequivocal glycoform identification.  相似文献

The Dufour's gland content of workers of two ant species of the genus Messor has been analyzed by solid-phase microextraction and gas chromatography with mass spectrometry (GC/MS). The structures of the compounds in the pheromonal mixtures have been determined. In both cases only one intact gland, inserted in a properly dimensioned capillary vial, is sufficient to produce a clean and fully interpretable GC/MS profile. It is worth noting that, for the first time in Messor ants, farnesol has been detected as a minor component of glandular secretion in Messor capitatus.  相似文献

The analysis of complex matrices, such as perfumes, by means of gas chromatography-olfactometry (GC-O) can be rather imprecise due to the co-elutions, leading to a possible masking of odour-active trace-level compounds by major interferences or agglomeration of olfactive impressions resulting in unreliable olfactive characterization. To overcome these limits an innovative technique, comprehensive two-dimensional gas chromatography-olfactometry (GC x GC-O), was applied, revealing several relevant co-elutions, as in the linalool and linalyl acetate zones. A total of 177 compounds, out of these 135 odour-active, were detected by GC-O, while about 481 out of 818 compounds presented odour-activity through GC x GC-O analyses. In addition, GC/mass spectrometry (GC/MS) and GC x GC/MS analyses were also performed. Peak assignment was achieved by means of different information sources, such as GC/MS, GC x GC/MS, LRI, injection of standards and olfactive impressions.  相似文献

In this study, the investigation of the volatile compounds of dried rhizomes of Coptis chinensis Franch, C. deltoidea C. Y. Cheng et Hsiao, and C. teeta Wall was carried out to complete the chemical composition of these valuable natural products. Volatile profiles were established and compared after headspace solid-phase microextraction (HS-SPME) using a polydimethylsiloxane/divinylbenzene (PDMS/DVB, 65 μm) fibre coupled to comprehensive 2D gas chromatography time-of-flight mass spectrometry (GC×GC-TOFMS). Analyses were performed and compared on two column-phase combinations (non-polar/polar and polar/non-polar). The majority of the identified compounds eluted as well-separated (pure) components as a result of high-resolution capability of the GC×GC method, which significantly reduces co-elution. Therefore, this increases the likelihood that pure mass spectra can be obtained. More than 290 volatile and semi-volatile organic compounds were tentatively characterized by means of GC×GC in tandem with TOFMS detection. Improved result interpretations were obtained in terms of compound classification based on the organized structure of the peaks of structurally related compounds in the GC×GC contour plot. These compounds are distributed over the chemical groups of hydrocarbons, acids, alkenes, alkynes, aldehydes, ketones, alcohols, esters, furans, and terpenoids. Among all the chemical groups, terpenoids present the higher number of identified compounds (44), alkenes (41), and aldehydes and ketones (28). This study completed the volatile phytochemical analysis of the headspace composition of various Coptis species rhizomes, and should serve as a means to identify the difference between the rhizomes and may also be useful to confirm individual species based on their volatile chemical profile.  相似文献

The chemical reaction of arthropods to their environment, i.e. their chemical ecology, can be studied particularly well with water beetles. Stenus comma, an aquatic beetle weighing only 2.5 mg, saves itself from drowning with the aid of an alkaloid, and the water beetle Ilybius fenestratus defends itself against small mammalian predators with a compound belonging to the same class. The water beetle Platambus maculatus employs a diterpene for precisely the same purpose and the whirligig beetle a norsesquiterpene, which also offers protection against troublesome microorganisms. As chemical artists, the ants can hardly be surpassed. In particular, the myrmecine ants guarantee their food supplies with plant growth substances. Since these compounds, depending upon concentration, can also act as inhibitors, we are confronted with an excellent example of an ecological equilibrium being established with the aid of organic chemicals. Even the little parasitic bombardier beetle Paussus favieri is tolerated, on account of its defensive chemistry, in the nest of the myrmecine ant Pheidole. In contrast, inorganic compounds are largely responsible for the stability of spiders' webs.  相似文献

Comprehensive two-dimensional gas chromatography coupled to time-of-flight mass spectrometry (GC×GC-TOFMS) was used for the characterization of aromatic compounds present in extra heavy gas oil (EHGO) from Brazil. Individual identification of EHGO compounds was successfully achieved in addition to group-type separation on the chromatographic plane. Many aromatic hydrocarbons, especially polycyclic aromatic hydrocarbons and sulfur compounds, were detected and identified, such as chrysenes, phenanthrenes, perylenes, benzonaphthothiophenes and alkylbenzonaphthothiophenes. In addition, triaromatic steroids, methyl-triaromatic steroids, tetrahydrochrysenes and tetraromatic pentacyclic compounds were present in the EHGO aromatic fractions. Considering the roof-tile effect observed for many of these compound classes and the high number of individual compounds identified, GC×GC-TOFMS is an excellent technique to characterize the molecular composition of the aromatic fraction from EHGO samples. Moreover, data processing allowed the quantification of aromatic compounds, in class and individually, using external standards. EHGO data were obtained in μgg(-1), e.g., benzo[a]pyrene were in the range 351 to 1164μgg(-1). Thus, GC×GC-TOFMS was successfully applied in EHGO quantitative analysis.  相似文献

Coal is a non renewable fossil fuel, used mainly as a source of electrical energy and in the production of coke. It is subjected to thermal treatment, pyrolysis, which produces coke as a main product, in addition to a condensed liquid by-product, called tar. Tar is a complex mixture of organic compounds which contains different chemical classes, presenting aromatic and sulphur heterocyclic compounds. In general, identification of these compounds requires steps of isolation and fractionation, mainly due to co-elution of these compounds with polyaromatic hydrocarbons (PAH). The objective of this work is to characterize the sulphur compounds present in the coal tar obtained via pyrolysis, using comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry detector (GC×GC/TOFMS). Coal samples from the State of Paraná, Brazil were subjected to laboratorial scale pyrolysis. Several experimental conditions were tested, such as sample weight (5, 10 and 15g), heating ramp (10, 25 and 100°C/min) and final temperature (500, 700 and 900°C). Samples were analyzed by one dimensional gas chromatography (1D-GC) coupled to a quadrupole mass spectrometry detector (GC/qMS) and two-dimensional gas chromatography with time-of-flight mass spectrometry detector (GC×GC/TOFMS). The higher amount of sulphur compounds was obtained at a final temperature of 700°C and a heating ramp of 100°C/min. The main classes observed in the color plot were thiophenes, benzothiophenes and alkylated dibenzothiophenes. GC×GC/TOFMS allowed the identification of the greater number of compounds and the separation of several sulphur compounds from one another. Moreover, separation of sulphur compounds from polyaromatic hydrocarbons and phenols was achieved, which was not possible by 1D-GC. Comparing GC×GC/TOFMS and 1D-GC (SIM mode) also showed that 1D-GC, one of the most employed quantification tools for sulphur compounds, can be misleading for detection, identification and quantification, as the number of isomers of sulphur compounds found was greater than theoretically possible.  相似文献

The development and use of a pre-concentrator-thermo-desorber/micro-gas chromatograph/mass spectrometer (μTD/μGC/MSD) coupling for the on-site analysis of VOCs in landfill gases are presented. The coupling has the advantage of analysing compounds with two detectors operated in series: the TCD (of the μGC) initially analyses the gas without destroying it, and then the MSD identifies the compounds. Due to the TCD response, the results were quantified with reference to toluene. The reliability of the analytical chain for quantitative analysis was validated by sampling two gaseous standards, including the EPA TO14 mixture, containing 39 compounds. With the OV1?μGC column, 24 compounds were identified and 16 correctly quantified. The repeatability of the measures estimated by their standard deviation was in the order of 1–2%. The detection limit was evaluated at 0.1?ppbv, for a 40?min pre-concentration on the Tenax of the μTD. The results of VOC analyses in the air of a landfill site obtained with the μTD/μGC/MSD coupling show its potential for on-site analyses: immediate results, high sensitivity, no storage for the samples, and measurements of pollution peaks.  相似文献

This study evaluates comprehensive two-dimensional gas chromatography (GC×GC) coupled to time-of-flight mass spectrometry (GC×GC-ToF MS) for the simultaneous analysis of several classes of organobromines (OBs), including polybrominated diphenyl ethers (PBDEs), polybrominated biphenyls (PBBs), methoxylated PBDEs (MeO-PBDEs), several halogenated naturally produced compounds (HNPs) and eight novel brominated flame retardants (NBFRs), polybrominated hexahydroxanthene derivates (PBHDs), 2,4,6-tribromoanisole and a mixed halogenated compound (MHC-1), in bluefin tuna muscles. The proposed methodology maximised separation of both within and among OB families, and among these and other halogenated micropollutants detected in these samples and co-extracted matrix components. Special attention has been paid to solve co-elution problems observed during the analysis of OBs with one-dimensional GC-based techniques. Satisfactory separation among several relevant PBDEs and MeO-PBDEs has been obtained allowing their unambiguous determination in a single run. Additional studies were conducted to identify selected NBFRs and HNPs. 2,4-Dibromoanisole, a dibromophenol isomer and hexabromobenzene were identified in the investigated samples. Several new tri- and tetra-BHD derivates were also identified, indicating that these compounds could apparently exist as structured families in nature. In addition, a tetrabrominated diMeO-biphenyl and two tetrabrominated diMeO-BDEs were also tentatively identified.  相似文献

全二维气相色谱(GC×GC)是20世纪90年代发展起来的具有高分辨率、高灵敏度、高峰容量等优势的分离技术,在我国将其用于大气挥发性有机物(VOCs)研究方面才刚刚起步.本文将GC-GC与氢火焰离子化检测器(FID)联用,构建了用于测量大气有机物的热脱附-全二维气相色谱-氢火焰离子化分析系统(TD-GC×GC-FID).采用HP-5MS和HP-INNOWAX色谱柱,建立了C5-C15大气有机物分析方法,实现了一次分析过程同时分离非甲烷烃(NMHCs)、含氧挥发性有机物(OVOCs)和卤代烃等多种组分.利用标准物质和四级杆质谱(qMS)进行定性,外标法结合FID质量校正因子定量.目标物在GC-GC谱图中第一和第二维保留时间变化分别小于0.6s和0.02s,峰体积平均相对标准偏差为14.3%,其中烷烃和芳香烃为4.5%.标准曲线r2均值大于0.99,平均检出限为6.04ng,平均回收率为111%.利用该方法检测到2010年1月北京市区大气中400多种有机物(信噪比大于50),鉴定了其中的103种物质,包括烷烃、烯烃、芳香烃、卤代烃、醛、酮、酯、醇和醚等.所测定有机物平均总浓度为51.3×10-9V/V,其中OVOCs约占51%,芳香烃约占30%,烷烃约占15%,卤代烃和烯烃分别占3%和1%.平均浓度最高的前3个组分是乙醇(9.84×10-9V/V)、丙酮(6.72×10-9V/V)和甲苯(3.48×10-9V/V).  相似文献

Combining qualitative data from the chromatographic structure of 2-D gas chromatography with flame ionization detection (GC×GC-FID) and that from gas chromatography-mass spectrometry (GC/MS) should result in a more accurate assignment of the peak identities than the simple analysis by GC/MS, where coelution of analytes is unavoidable in highly complex samples (rendering spectra unsuitable for qualitative purposes) or for compounds in very low concentrations. Using data from GC×GC-FID combined with GC/MS can reveal coelutions that were not detected by mass spectra deconvolution software. In addition, some compounds can be identified according to the structure of the GC×GC-FID chromatogram. In this article, the volatile fractions of fresh and dehydrated pineapple pulp were evaluated. The extraction of the volatiles was performed by dynamic headspace extraction coupled to solid-phase microextraction (DHS-SPME), a technique appropriate for slurries or solid matrices. Extracted analytes were then analyzed by GC×GC-FID and GC/MS. The results obtained using both techniques were combined to improve compound identifications.  相似文献

A fast gas chromatography (GC)-MS method has been developed and validated for the simultaneous screening of different classes of drugs of abuse in urine. Tetrahydrocannabinol metabolite, cocaine, opiates such as morphine, O-6-monoacetylmorphine (O-6-MAM), codeine, opioids such as buprenorphine, methadone, pentazocine, fentanyl and analogues and their main metabolites can be detected and quantified after a simple liquid-liquid extraction in alkaline conditions and derivatisation to obtain the corresponding trimethylsilyl derivatives. The chromatographic separation is performed in a total time of 6 min, using a short GC column (5% phenyl methyl silicone, 10-m length × 0.18-mm internal diameter). The Limits of Detection are satisfactory for forensic purposes for all the substances; the repeatability of concentrations (percent coefficients of variation) are always lower than 15% at high and low concentration levels, and accuracy, intended as % error on the true value, is always lower than 15% for all the analytes. The method can successfully be applied for screening analyses in many fields of forensic toxicology.  相似文献

2,6-dialkylpiperideines found in the venom of Solenopsis (Hymenoptera, Formicidae) fire ants are a range of compounds possessing various biological activities. A series of racemic 2-methyl-6-alkyl-Δ1,6-piperideines were synthesized for chemical confirmation of the natural products found in fire ant venom, and the evaluation of their biological activity. Synthetic Δ1,6-piperideines and the natural compounds in the cis-alkaloid fraction of Solenopsis invicta had identical mass spectra and retention times. Their insecticidal activities against the third-instar larvae of cotton bollworm (Helicoverpa armigera) were evaluated by using injection and topical application methods. All three compounds exhibited no lethal effect at concentrations of 0.05–0.4 mol/L by topical treatment, but moderate lethal effect at 0.4 mol/L through injection treatment. Compound 6a showed significantly higher activity than the natural insecticide nicotine. The differences in activity among compounds 6b, 6c and nicotine were not significant. The elongation of the carbon chain at the 6-position of the piperideine ring appears to decrease insecticidal activity.  相似文献

Two ionic liquids (ILs), namely (S,S)-1-butyl-3-(2'-hydroxy-cyclohexyl)-3H-imidazol-1-ium tetrafluoroborate and (S,S)-1-butyl-3-(2'-acetyl-cyclohexyl)-3H-imidazol-1-ium tetrafluoroborate have been employed as stationary phases in capillary gas chromatography. These new phases exhibit a column efficiency of 1,600 and 2,100 plates m(-1) for IL 1 and IL 2, respectively, a wide operating temperature range and good thermal stability (bleeding temperature of 250 °C for IL 1 and 160 °C for IL 2). Inverse gas chromatography (GC) analyses were used to study the solvation properties of these ILs through a linear solvation energy model. The application of these ILs as new GC stationary phases was studied. These stationary phases exhibited unique selectivity for many organic substances, such as alkanes, ketones, esters, and aromatic compounds. The efficient separation of several mixtures containing compounds of different polarities and the good separation of fatty acid methyl esters (FAMEs) and cis/trans isomers indicate that these ILs may be applicable as a new type of GC stationary phases.  相似文献

Gas chromatographic (GC) profiles of cuticular hydrocarbon extracts obtained from individual and pooled ant samples were analyzed using pattern recognition techniques. Clustering according to the biological variables of social caste and colony were observed. Pooling individual extracts enhanced the recognition of patterns in the GC profile data characteristic of colony. Evidently, the contribution of the ant's individual pattern to the overall hydrocarbon profile pattern can obscure information about colony in the GC traces of cuticular hydrocarbon extracts obtained from red fire ants. Re-analysis of temporal caste and time period data on the cuticular hydrocarbon patterns demonstrates that sampling time and social caste must be taken into account to avoid unnecessary variability and possible confounding. This and the fact that foragers could not be separated from reserves and brood-tenders in all five laboratory colonies studied suggests that cuticular hydrocarbons as a class of sociochemicals cannot model every facet of nestmate recognition in Solenopsis invicta which in turn suggests a potential role for other compounds in the discrimination of alien conspecifics from nestmates.  相似文献

Fully synthetic jet fuel (FSJF) produced via Fischer-Tropsch (FT) technology was recently approved by the international aviation fuel authorities. To receive approval, comparison of FSJF and crude-derived fuel and blends on their qualitative and quantitative hydrocarbon composition was of utmost importance. This was performed by comprehensive two-dimensional gas chromatography (GC×GC) in the reversed phase mode. The hydrocarbon composition of synthetic and crude-derived jet fuels is very similar and all compounds detected in the synthetic product are also present in crude-derived fuels. Quantitatively, the synthetic fuel consists of a higher degree of aliphatic branching with less than half the aromatic content of the crude-derived fuel. GC×GC analyses also indicated the presence of trace levels of hetero-atomic impurities in the crude-derived product that were absent in the synthetic product. While clay-treatment removed some of the impurities and improved the fuel stability, the crude-derived product still contained traces of cyclic and aromatic S-containing compounds afterwards. Lower level of aromatics and the absence of sulphur are some of the factors that contribute to the better fuel stability and environmental properties of the synthetic fuel. GC×GC was further applied for the analysis of products during Jet Fuel Thermal Oxidation Testing (JFTOT), which measures deposit formation of a fuel under simulated engine conditions. JFTOT showed the synthetic fuel to be much more stable than the crude-derived fuel.  相似文献

Several novel synthetic organic compounds were successfully analyzed with a unique type of GC-MS titled Supersonic GC-MS following a failure in their analysis with standard GC-MS. Supersonic GC-MS is based on interfacing GC and MS with a supersonic molecular beam (SMB) and on electron ionization of sample compounds as vibrationally cold molecules while in the SMB, or by cluster chemical ionization. The analyses of novel synthetic organic compounds significantly benefited from the extended range of compounds amenable to analyses with the Supersonic GC-MS. The Supersonic GC-MS enabled the analysis of thermally labile compounds that usually degrade in the GC injector, column and/or ion source. Due to the high carrier gas flow rate at the injector liner and column these compounds eluted without degradation at significantly lower elution temperatures and the use of fly-through EI ion source eliminated any sample degradation at the ion source. The cold EI feature of providing trustworthy enhanced molecular ion (M+), complemented by its optional further confirmation with cluster CI was highly valued by the synthetic organic chemists that were served by the Supersonic GC-MS. Furthermore, the provision of extended mass spectral structural, isomer and isotope information combined with short (a few minutes) GC-MS analysis times also proved beneficial for the analysis of unknown synthetic organic compounds. As a result, the synthetic organic chemists were provided with both qualitative and quantitative data on the composition of their synthetic mixture, and could better follow the path of their synthetic chemistry. Ten cases of such analyses are demonstrated in figures and discussed.  相似文献

In this work capillary electrophoresis (CE) with UV detection has been applied to the analysis of different natural toxins produced in the aquatic environment. This technique is presented as an alternative to other chemical techniques such as HPLC, and the optimisation of analytical methodologies was carried out for diverse marine toxins including Paralytic and Amnesic and some polyether toxins, such as Yessotoxins, as well as for certain microcystin toxins produced by cyanobacteria present in freshwaters. Sample preparation steps were optimised and adequate electrophoretic conditions developed for achieving a complete separation of compounds with similar structures involved in such contamination. The influence of the biological matrices where they are involved has also been studied and the potential use of CE-UV as a tool for monitoring these aquatic toxins is also discussed.  相似文献

Thermochemical conversion processes play a crucial role in all routes from fossil and renewable resources to base chemicals, fuels and energy. Hence, a fundamental understanding of these chemical processes can help to resolve the upcoming challenges of our society. A bench scale pyrolysis set-up has been used to study the thermochemical conversion of rapeseed oil methyl ester (RME), i.e. a mixture of fatty acid methyl esters. A GC×GC, equipped with both a flame ionization detector (FID) and a time-of-flight mass spectrometer (TOF-MS), allows quantitative and qualitative characterization of the reactor feed and product. Analysis of the latter is accomplished using a dedicated high temperature on-line sampling system. Temperature programmed analysis, starting at -40°C, permits effluent characterization from methane up to lignoceric acid methyl ester (C(25)H(50)O(2)), in a single run of the GC×GC. The latter combines a 100% dimethylpolysiloxane primary column with a 50% phenyl polysilphenylene-siloxane secondary column. Modulation is started when the oven temperature reaches 40°C, thus dividing the chromatogram in a conventional 1D and a comprehensive 2D part. The proposed quantification approach allows to combine the quantitative GC×GC analysis with 2 other on-line 1D GC analyses, resulting in a complete and detailed product composition including the measurement of CO, CO(2), formaldehyde and water. The GC×GC reveals that the product stream contains a huge variety of valuable products, such as linear alpha olefins, unsaturated esters and aromatics, that could not have been identified and quantified accurately with conventional 1D GC because of peak overlap.  相似文献
