An effective method for the synthesis of dialkyl [2-(polyfluoroalkoxy)ethyl]phosphonates by free radical hydrophosphorylation of fluoroalkyl vinyl ethers with dialkyl (H)-phosphonates was developed. The reaction proceeds in the presence of catalytic amounts of azabisisobutyric acid dinitrile (AIBN) (150°C, 2 h, portionwise addition of AIBN) to afford the target fluoroalkyl phosphonates in up to 85% isolated yield.  相似文献

A simple and efficient stereoselective synthesis of fluoroalkyl substituted aziridine-2-phosphine oxides and -phosphonates by diastereoselective addition of methoxide, imidazole, benzenethiol, and Grignard reagents to functionalized ketoxime-phosphine oxides and -phosphonates is described. Aziridines are used as intermediates for the regioselective synthesis of fluorine containing beta-amino phosphine oxides and beta-amino phosphonates. Amino phosphorus derivatives can also be obtained from ketoximes derived from phosphine oxides and phosphonates with sodium borohydride.  相似文献

1,2-Diaza-1,3-butadienes derived from phosphine oxides and phosphonates and with optically active substituents on N-1 and C-3 are obtained by 1,4-elimination from chlorohydrazonoalkyl-phosphine oxides and -phosphonates in the presence of bases. The Ritter reaction of enantiopure 2-(1-aminoalkyl)aziridines 1 with different nitriles afford enantiopure tetrasubstituted imidazolines 2. 1-Ehtoxycarbonyl-1, 2-dihydroquinoline-2-phosphonates (3) were treated with n-butyllithium followed by alkyl halides to afford the corresponding 4-alkylated phosphonates (7) with complete regioselectivity in 67–97% yields. The phosphonates (7) were converted to 4-alkylquinolines (8) in ca. 50% yields by treatment with sodium iodide in HMPA or by alkaline hydrolysis in aqueous ethanol.  相似文献

Various disubstituted phosphonates including alkylnucleoside 3′-phosphonates were converted rapidly into the corresponding phosphorochloridites by use of tris(2, 4, 6-tribromophenoxy) dichlorophosphorane (BDCP) as a chlorinating reagent. Addition of diethyl lithiodifluoromethylphosphonate to enantiopure sulfinimines afforded the corresponding N-sulfinyl α,α-difluoro-β-amino phosphonates with good diastereoselectivity. Aziridines are important synthetic intermediates which readily undergo ring-opening reactions. It is demonstrated that electron-rich phosphines are efficient catalysts for the regioselective rearrangement of N-acylaziridines to oxazolines. The reactions occur in excellent yield under neutral conditions. Evidence is provided for an addition/elimination mechanism by generation of a phosphonium intermediate. Similar intermediates may be useful for the development of alternate aziridine ring-opening processes and stereoselective synthesis with enantiopure phosphines.  相似文献

The data are presented on the synthesis of podands with terminal quinoxaline fragments of rings and their influence on both the micellization properties of cetyltrimethylammonium bromide in a water—DMF solution and kinetics of basic hydrolysis of O-p-nitrophenyl O-ethyl chloromethylphosphonate and O-p-nitrophenyl O-hexyl chloromethylphosphonate in the absence and presence of surfactants. The mechanism of the podand effect on the reaction rate depends on the structures of phosphonate and podand. 1,8-Bis(3-ethyl-1,2-dihydro-2-oxoquinoxalin-1-yl)-3,6-dioxaoctane inhibits the basic hydrolysis of the substrates to 3—4 times. In a micellar solution of the surfactant, an approximately 20-fold acceleration of the reaction rate constant is observed. The observed rate constant decreases when podand is added to a micellar solution. The catalytic effect of the polycomponent system is due to concentrating of the reactants. The micellar microenvironment can exert both positive and negative effects on the reactivity of phosphonates.  相似文献

Preparation of enantiomerically pure, highly oxygenated decalins via tandem Wittig-type Diels–Alder reactions from the corresponding sugar-derived dieno-phosphoranes and/or -phosphonates and sugar aldehydes is described. Application of the phosphonates is more convenient than phosphoranes since the former can be prepared in much higher yields and react with aldehydes under milder conditions. The intermediate trienes resulting from the Wittig-type process undergo spontaneous and highly stereoselective cyclization to the cis-decalins under the reaction conditions.  相似文献

Aziridines are highly useful compounds as building blocks for the synthesis of important organic compounds. Amino acid synthesis by aziridine ring opening reaction is a good example to the use of aziridines. Although this reaction is studied by many groups, the synthesis of amino phosphonic acids is less explored. In this study, we have carried out the ring opening reaction of aziridinyl phosphonates with a variety of alcohols including the more functional propargylic and allylic alcohols. These reactions provided functionalized α‐amino‐β‐alkoxyphosphonates in 40–91 % yield.  相似文献

[Reaction: see text]. The reaction of chiral 2-(1-aminoalkyl)aziridines 1 with different thiols, in the presence of BF3*Et2O, is reported. The obtained products were dependent on the structure of the starting amino aziridines 1. Thus, enantiopure (2S,3S)-2-(alkylthio)alkane-1,3-diamines 2 were obtained from aziridines with C-2 substituents with lower steric congestion and partially racemized (2S,3S)-2,3-bis(alkylthio)alkan-1-amines 3 (ee = 56-66%) from aziridines with larger C-2 subtituents. In both cases, the opening of the nonactivated aziridine ring at C-2 took place with retention of configuration and proceeded with regio- and stereoselectivity at C-2. In the synthesis of 3, 2 equiv of thiol reacts with 1 and the opening of aziridine ring at C-2 was followed by an unusual displacement of the dibenzylamino group by a second equivalent of thiol. The regiochemistry and relative configuration of compounds 3 was established by single-crystal X-ray analysis. A mechanism is proposed to explain the results obtained.  相似文献

Abstract

A series of unactivated racemic and chiral aziridine-2-phosphonates were synthesized by modified Gabriel-Cromwell reaction. Ring opening reaction of the synthesized phosphonates by gaseous HCl provided access to a wide range of biologically interesting novel β-chloro-α-aminophosphonates. All reactions toward each of the above-mentioned products can be conducted regioselectively in high yields.  相似文献

An efficient and general synthesis of aziridines by the reaction of imines derived from p-toluenesulfonamides with in situ generated iodomethyllithium, with a simple and rapid experimental protocol, is reported for the first time. The reaction with the chiral aldimine derived from phenylalaninal allowed access to (2R,1'S)-2-(1'-aminoalkyl)aziridine with very high diastereoselectivity, in enantiopure form.
