A series of chiral beta-aryl-gamma-amino acid ester derivatives were synthesized in high enantioselectivities (93-97% ee) via the Rh-catalyzed asymmetric hydrogenation of gamma-phthalimido-alpha,beta-unsaturated carboxylic acid esters using highly modular chiral BoPhoz-type phosphine-aminophosphine ligands. The method has been applied successfully to the synthesis of several chiral pharmaceuticals including (R)-baclofen and (R)-rolipram with high enantioselectivities.  相似文献

A new chiral ligand 6,6′‐dimethoxy‐2,2′‐diaminobiphenyl was successfully prepared from 3‐nitrophenol via iodination, Ullmann coupling, and reduction. The resolving reagent (2R, 3R)‐ or (2S,3S)‐2,3‐di (phenylaminocarbonyl)tartaric acid was prepared from commercially available tartaric acid in large scale and was used to resolve the racemic 6,6′‐dimethoxy‐2,2′‐diaminobiphenyl. The chiral 6,6′‐ dimethoxy‐2,2′‐diaminobiphenyl obtained was proved to be enantiomerically pure.  相似文献

New chiral, soluble binaphthyl derivatives that incorporate stilbenoid dendrons at the 6,6'-positions have been prepared. The synthesis of the new enantiopure dendrimers was performed in a convergent manner by Horner-Wadsworth-Emmons (HWE) reaction of the appropriately functionalized 1,1'-binaphthyl derivative (R)-1 and the appropriate dendrons (R)(2n)G(n)-CHO. Different electroactive units were incorporated in the peripheral positions of the dendrons in order to tune both the optical and electrochemical behavior of these systems. Fluorescence measurements on the chiral dendrimers reveal a strong emission with maxima between 409 and 508 nm depending upon the substitution pattern. Finally, the redox properties of the dendrimers were determined by cyclic voltammetry, showing the influence of the functional groups at the peripheral positions of the dendrimer on the redox behavior of these systems.  相似文献

Catalytic enantioselective Mannich-type reactions of silicon enolates with aldimines were investigated using chiral zirconium catalysts prepared from Zr(O(t)Bu)(4), N-methylimidazole, and newly designed multidentate BINOL derivatives. These new multidentate BINOL ligands were designed on the basis of an assumed transition state structure of a chiral zirconium catalyst derived from two molecules of (R)-6,6'-Br(2)-BINOL. Not only tetradentate BINOL 4 but also tridentate BINOL derivatives were found to be effective, and high enantioselectivities were attained. In a structural study of the most effective zirconium complex prepared from tridentate ligand 6e, several NMR experiments and DFT calculations were carried out. Consequently, the structure of an active catalyst and plausible mechanism of asymmetric induction were elucidated.  相似文献

A number of new chiral binaphthol (BN) derivatives with different substituents R,R' in the 6,6'-positions in open (BN-diethylethers) and bridged forms (BN-acetals) have been synthesized. The syntheses of the chiral 6,6'-disubstituted-2,2'-diethoxy-1,1'-binaphthyls (R,R' = -CH'CHCHO, -CH'CH-(p-BrPh), -CH'CH-(p-CHOPh)) and the chiral 9,14-disubstituted-dinaphtho [2,1-d:1',2'-f][1,3] dioxepins with R,R' = -CH'CH-(p-BrPh), -CH'CH-(p-CHOPh) are reported for the first time. The possible liquid crystalline properties and molar twisting powers (βM) in three different nematic liquid crystals (LCs) of the BN derivatives were investigated. Derivatives with spatially extended substituents in the 6,6'-positions (e.g. styryl or vinyl) show unusually high molar twisting power (up to 124.5μm-1). A direct correlation between the magnitude of βM and the length of the substituents was found. Bridged forms, in which the dihedral angle θ between the naphthyl moieties is ≊ 54°, show higher twisting power than the corresponding open forms, where θ is allowed to vary around 90° resulting in an equilibrium between transoid and cisoid forms. From the different temperature dependencies of βM of the open and bridged BNs, a molecular model was developed relating the molecular conformation and twisting power. Although no mesophase was found in any of the compounds synthesized, they can be considered as important precursors for the synthesis of potential chiral BN-containing LCs.  相似文献

A novel and practical asymmetric synthesis of chiral glycidic acid derivatives involving methyl (2R,3S)-3-(4-methoxyphenyl)glycidate ((2R,3S)-2a), a key intermediate for diltiazem hydrochloride (1), was developed. Treatment of methyl (E)-4-methoxycinnamate ((E)-3a) with chiral dioxirane, generated in situ from a catalytic amount (5 mol %) of an 11-membered C(2)-symmetric binaphthyl ketone (R)-7a, provided (2R,3S)-2a in 92% yield and 80% ee. Other cinnamic acid esters and amides were epoxidized by the use of the same procedure to give the corresponding chiral glycidic acid derivatives with up to 95% yield and 92% ee. Higher enantioselectivities in the asymmetric epoxidation of (E)-cinnamates than that of (E)-stilbene derivatives were observed and were proposed to be attributed to a dipole-dipole repulsion between oxygen atoms of an ester group in the cinnamates and those of the lactone moieties in the binaphthyl dioxirane.  相似文献

The axially chiral dopants (R)-5,5'-, 5,6'-, and 6,6'-diheptyloxy-2,2'-spirobiindan-1,1'-dione ((R)-2, -3, and -4) were synthesized in optically pure form, and their absolute configurations were assigned by the exciton chirality method using circular dichroism spectroscopy. These new compounds were doped in four achiral liquid crystal hosts to give chiral smectic C* (SmC*) phases with spontaneous polarizations (Ps) that vary with the core structure of the host. The spontaneous polarization induced by the 5,5'-dialkoxy derivative (R)-2 is uniformly positive, whereas that induced by the 6,6'-dialkoxy derivative (R)-4 is uniformly negative and shows a different trend in host dependence. Polarization power (delta(p)) values range from +21 nC/cm2 for (R)-2 in 2',3'-difluoro-4-heptyl-4' '-nonyl-p-terphenyl (DFT) to -1037 nC/cm2 for (R)-4 in 4-(4'-heptyl[1,1'-biphen]-4-yl)-1-hexylcyclohexanecarbonitrile (NCB76). The unsymmetrical dopant (R)-3 behaves like a hybrid of the two symmetrical isomers, with lower absolute values of delta(p), on average, and varying signs of Ps. 2H NMR spectra of the doped mixtures using racemic mixtures of 2-4 with -OCD2C6H13 side-chains, in combination with phase diagrams, show that relatively minor changes in the dopant structure, that is, moving the alkoxy side-chains from the 5,5' to the 6,6' positions of the spirobiindandione core, have profound effects on dopant-host compatibility, and on the propensity of the dopant to exert chiral perturbations in the host environment. The variations in sign and magnitude of delta(p) as a function of alkoxy group positions are rationalized based on an analysis of zigzag conformations that conform to the binding site of the SmC host according to the Boulder model.  相似文献

Catalytic asymmetric aldol reactions of silyl enol ethers with aldehydes (Mukaiyama aldol reactions) have been performed using novel chiral zirconium catalysts. The reactions proceeded in high yields under mild conditions, and anti-adducts were obtained in high diastereo- and enantioselectivities. The catalysts were first prepared from zirconium(IV) tert-butoxide (Zr(O(t)Bu)(4)), (R)-3,3'-diiodo-1,1'-binaphthalene-2,2'-diol ((R)-3,3'-I(2)BINOL), a primary alcohol, and a small amount of water. It was revealed that the primary alcohol played an important role in completing the catalytic cycle and that a small amount of water was essential for obtaining high selectivities. Moreover, activities of the chiral zirconium catalysts were enhanced by using new ligands, (R)-3,3'-I(2)-6,6'-X(2)BINOL (X = Br, I, C(2)F(5)), and it has been shown that even aldol reactions of less reactive substrates proceeded smoothly using the novel zirconium catalysts. Finally, NMR studies of these catalysts were performed, which suggested that the catalyst would form a dimeric structure and that the water affected the catalyst formation.  相似文献

The chiral monosubstituted derivatives of spiropentane, spiropentylcarboxylic acid methyl ester, 1, and spiropentyl acetate, 2, have been synthesized in optically active form. Configurational and conformational analysis of 1 and 2 has been carried out using infrared (IR) and vibrational circular dichroism (VCD) spectroscopies. Analysis of the experimental IR and VCD spectra has been carried out using ab initio density functional theory (DFT). For both 1 and 2, DFT predicts two populated conformations. Comparison to experiment of the conformationally averaged IR and VCD spectra of 1 and 2, predicted using DFT, provides unequivocal evidence of the predicted conformations and yields the absolute configurations R(-)/S(+) for 1 and R(+)/S(-) for 2. These absolute configurations are consistent with the R(-)/S(+) absolute configuration of spiropentylcarboxylic acid, assigned previously via X-ray crystallography of its alpha-phenylethylammonium salt.  相似文献

The key steps include lithium ion-activated SN2-type alkynylation of the tricyclic N,O-acetal leading to exclusive formation of the (6S)-ethynylpiperidine and ring-closing olefin metathesis of the (2R,6S)-cis-2,6-dialkenylpiperidine for constructing the bridged azabicyclononane.  相似文献

A series of chiral beta-aryl-substituted gamma-amino butyric acid derivatives were synthesized in good enantioselectivities via the Cu-catalyzed asymmetric conjugate reduction of gamma-phthalimido-alpha,beta-unsaturated carboxylic acid esters using Cu(OAc)2 x H2O as a catalyst precursor, (S)-BINAP as a ligand, PMHS as a hydride source, and t-BuOH as an additive. Treatment of V(NMe(2))(4) or M(NMe(2))(5) (M = Nb, Ta) with 1 equiv of C(2)-symmetric amidate ligands 1H(2), 2H(2), 3H(2), 4H(2), and 5H(2), or Schiff base ligands 6H(2), 7H(2) and 8H(2) at room temperature gives, after recrystallization from a benzene, toluene or n-hexane solution, the vanadium amides (1)V(NMe(2))(2) (11), (2)V(NMe(2))(2) (14), (3)V(NMe(2))(2) (17), (5)V(NMe(2))(2) (22), (6)V(NMe(2))(2) (23) and (7)V(NMe(2))(2) (24), and niobium amides (1)Nb(NMe(2))(3) (12), (2)Nb(NMe(2))(3) (15), (3)Nb(NMe(2))(3) (18), (4)Nb(NMe(2))(3) (20) and [2-(3-Me(3)C-2-O-C(6)H(3)CHN)-2'-(N)-C(20)H(12)][2-(Me(2)N)(2)CH-6-CMe(3)-C(6)H(3)O]NbNMe(2)·C(7)H(8) (25·C(7)H(8)), and tantalum amides (1)Ta(NMe(2))(3) (13), (2)Ta(NMe(2))(3) (16), (3)Ta(NMe(2))(3) (19) and (4)Ta(NMe(2))(3) (21) respectively, in good yields. Reaction of V(NMe(2))(4) or M(NMe(2))(5) (M = Nb, Ta) with 2 equiv of C(1)-symmetric amidate ligands 9H or 10H at room temperature gives, after recrystallization from a toluene or n-hexane solution, the chiral bis-ligated vanadium amides (9)(2)V(NMe(2))(2)·3C(7)H(8) (27·3C(7)H(8)) and (10)V(NMe(2))(2) (28), and chiral bis-ligated metallaaziridine complexes (10)(2)M(NMe(2))(η(2)-CH(2)NMe) (M = Nb (29), Ta (30)) respectively, in good yields. The niobium and tantalum amidate complexes are stable in a toluene solution at or below 160 °C, while the vanadium amidate complexes degrade via diemthylamino group elimination at this temperature. For example, heating the complex (2)V(NMe(2))(2) (14) in toluene at 160 °C for four days leads to the isolation of the complex [(2)V](2)(μ-NMe(2))(2) (26) in 58% yield. These new complexes have been characterized by various spectroscopic techniques, and elemental analyses. The solid-state structures of complexes 12, 13, and 15-30 have further been confirmed by X-ray diffraction analyses. The vanadium amides are active chiral catalysts for the asymmetric hydroamination/cyclization of aminoalkenes, affording cyclic amines in moderate to good yields with good ee values (up to 80%), and the tantalum amides are outstanding chiral catalysts for the hydroaminoalkylation, giving chiral secondary amines in good yields with excellent ee values (up to 93%).  相似文献

A new type of efficient chiral catalyst has been elaborated for asymmetric C-alkylation of CH acids under PTC conditions. Sodium alkoxides formed from chiral derivatives of tartaric acid and aminophenols (TADDOL's 2a-e and NOBIN's 3a-h) can be used as chiral catalysts in the enantioselective alkylation, as exemplified by the reaction of Schiff's bases 1a-e derived from alanine esters and benzaldehydes with active alkyl halides. Acid-catalyzed hydrolysis of the products formed in the reaction afforded (R)-alpha-methylphenylalanine, (R)-alpha-naphthylmethylalanine, and (R)-alpha-allylalanine in 61-93% yields and with ee 69-93%. The procedure could be successfully scaled up to 6 g of substrate 1b. When (S,S)-TADDOL or (R)-NOBIN are used, the (S)-amino acids are formed. A mechanism rationalizing the observed features of the reaction has been suggested.  相似文献

Novel optically active 2-(pyrazol-1-yl)pyridines have been prepared using resolved the O-methyl ether of atrolactic acid as a source of the adjacent quaternary carbon stereocenter. Different regioisomers were formed selectively in the reaction of 2-hydrazinopyridines with the chiral 1,3-diketone and in the nucleophilic substitution of 2-chloropyridines with the potassium salt of the chiral pyrazole. The second route gave 2-(pyrazol-1-yl)pyridines with the stereogenic center neighboring the coordinating nitrogen in the pyrazole ring. Also, new C2-symmetric chiral ligands based on 2,6-bis(pyrazolyl)pyridine and 6,6′-bis(pirazolyl)-2,2′-bipyridine structures were obtained.  相似文献

A series of mono- and bidentate phosphites was prepared with (S)-5,5',6,6'-tetramethyl-3,3'-di-tert-butyl-1,1'-biphenyl-2,2'-dioxy [(S)-BIPHEN] as a chiral auxiliary and screened in the asymmetric hydroformylation of allyl cyanide. These hydroformylation results were compared with those of two existing chiral ligands, Chiraphite and BINAPHOS, whose utility in asymmetric hydroformylation has been previously demonstrated. Bisphosphite 11 with a 2,2'-biphenol bridge was found to be the best overall ligand for asymmetric hydroformylation of allyl cyanide with up to 80% ee and regioselectivities (branch-to-linear ratio, b/l) of 20 with turnover frequency of 625 [h(-)(1)] at 35 degrees C. BINAPHOS gave enantioselectivities up to 77% ee when the reaction was conducted in either acetone or neat but with poor regioselectivity (b/l 2.8) and activities 7 times lower than that of 11. The product of allyl cyanide hydroformylation using (R,R)-11 was subsequently transformed into (R)-2-methyl-4-aminobutanol, a useful chiral building block. Single-crystal X-ray structures of (S,S)-11 and its rhodium complex 19 were determined.  相似文献

The key construction in the preparation of as-indacene-bridged bis(alpha-amino acid) derivatives was effected by a Ru(II)-catalyzed RCM cascade reaction of appropriately substituted triynes. The latter were available after stepwise and stereocontrolled alkynylations of (2R)-2,5-dihydro-2-isopropyl-3,6-dimethoxypyrazine as chiral auxiliary. The regio- and stereochemical transformations have been verified by a single-crystal X-ray analysis.  相似文献
