Abstract— –Photophysical properties of [26] porphyrin (26 P) were investigated in chloroform. The quantum yields of fluorescence, of S1→ T1 intersystem crossing and singlet oxygen formation were measured. The purity, stability, the strong absorption in the red (δmax= 783 nm; εmax= 28 000 M 1 cm-1) and the ability of singlet oxygen formation recommend 26 P as potential photosensitizer for tumor therapy.  相似文献

Abstract— The triplet state characteristics (spectrum, lifetime and quantum yield) for four dye sensi tisers [methylene blue (MB), erythrosin (ER), haematoporphyrin (HP) and riboflavin (RF)] were determined in methanol by laser flash photolysis and singlet oxygen yields (0.60 to 0.48) from time-resolved measurements of the 1270 nm near infrared emission. The reaction of singlet oxygen with four long chain unsaturated phenyl esters [oleate (18: 1), linoleate (18: 2), linolenate (18: 3) and arachidonate (20: 4)] was followed quantitatively using the singlet oxygen luminescence technique and also, after continuous420–700 nm irradiation, by HPLC and other analysis of the isomeric product monohydroperoxides. The overall quantum yield of photooxidation (∼10-2) was shown to be consistent with the observed singlet oxygen quenching constants(2–12 times 104 dm3 mol-1 s-1) for the four esters studied and the singlet oxygen lifetime in methanol (τ∼ 9 μs). The isomer product distribution was interpreted in terms of a dual singlet oxygen and radical mechanism, the radical contribution increasing with sensitiser in the order ER = MB < HP ≪ RF, but also showing some dependence on substrate unsaturation. Evidence is presented for singlet oxygen quenching by MB and RF ( kO = 1.6 and 6.0 times 107 dm3 mol-1 s-1) and for the accelerated photobleaching of the dye sensitisers in the presence of the unsaturated esters.  相似文献

The steady-state UVA (350 nm) photolysis of ( E )-β-ionone ( 1 ) in aerated toluene solutions was studied by 1H NMR spectroscopy. The formation of the 1,2,4-trioxane ( 2 ) and 5,8-endoperoxide ( 5 ) derivatives in the ratio of 4:1 was observed. Time-resolved laser induced experiments at 355 nm, such as laser-flash photolysis, photoacoustic and singlet oxygen 1O2 phosphorescence detection, confirmed the formation of the excited triplet state of 1 with a quantum yield Φ T = 0.50 as the precursor for the generation of singlet oxygen 1O2 ( Φ Δ = 0.16) and the isomeric α-pyran derivative ( 3 ), which was a reaction intermediate detected by NMR. In turn, the reaction of 1O2 with 1 and 3 occurred with rate constants of 1.0 × 106 and 2.5 × 108  m −1s−1 to yield the oxygenated products 5 and 2 , respectively, indicating the relevance of the fixed s-cis configuration in the α-pyran ring in the concerted [2+4] cycloaddition of 1O2.  相似文献

Abstract. Pulsed laser photolysis at 347nm has been used to study the transient spectroscopy of alloxazine, lumichrome, lumiflavin, and riboflavin in acidic (pH 2.2) aqueous solution and in ethanol. Intersystem crossing quantum yields (φISC) were determined by a modification of the comparative laser excitation method which utilizes the variation of the triplet yield with intensity in conjunction with a kinetic model for the various photophysical and photochemical processes occurring during the pulse. Fluorescence quantum yields and lifetimes are also reported. Correction for quenching of the excited singlet state by H+ ions shows that, in neutral aqueous solution, intersystem crossing for flavins is an efficient process (φISC˜ 0.7) which, in conjunction with fluorescence, accounts for the fate of all absorbed photons. For alloxazine (φISC˜ 0.45) and lumichrome (φISC˜ 0.7) the results are more difficult to interpret owing to interconversion between alloxazine and isoalloxazine structures in the singlet excited state. For all four compounds, the quantum yield of products derived from the singlet excited state is estimated as ˜0.04. There is evidence of biphotonic product formation at high laser energies. In ethanol, where φISC for lumichrome is about twice that of lumiflavin, internal conversion between the excited singlet and ground states appears to be a significant process. Complete triplet-triplet absorption spectra in the region 260–750nm are reported. For lumichrome at pH 2.2 there is spectral evidence for isomeric triplet states which appear to be in equilibrium.  相似文献

Abstract— Photophysical properties of two chlorin type molecules (CHLI) and (CHLII) were investigated in different solvents. Quantum yields of fluorescence φF of S, → T, intersystem crossing φT, and of singlet oxygen (1Δg) formation φΔ, as well as the Stern-Volmer constants for the quenching of the S, states by oxygen and the bimolecular rate constants of quenching of 1Δg by the chlorins were measured. The values of φT and φΛ can be given as 0.57 and 0.58 for CHLI and 0.69 and 0.58 for CHLII. The values of the fluorescence quantum yields, the strong absorption of the chlorins in the red (Λ > 630 nm) and the high values of the quantum yields for 1Δg formation recommend the chlorin derivatives as potential markers and photosensitizers for tumor therapy.  相似文献

Abstract— Fluorescence quantum yields and singlet lifetimes for a wide range of hydrophilic to hydrophobic porphyrins and metalloporphyrins have been determined in toluene, methanol or acetone. Photosensitized singlet oxygen yields have been determined in the same solvents. For some porphyrins, the same quantities were determined in an aqueous medium, through use of an amphiphilic polymer to solubilize the porphyrin sensitizer and target molecule, 1, 3-diphenylisobenzofuran. Because rate constants for the deactivation of singlet oxygen ( k d) and for its reaction with a target molecule (k a ) are unknown in such aqueous polymer systems, a new method was developed for evaluating yields of singlet oxygen formation that also provides a value for the ratio kd/ka. A variation observed in quantum yield of singlet oxygen production for the aqueous polymer system with variation in initial concentration of the target molecule is discussed.  相似文献

Abstract— The presence of different ground state species in ionic and/or keto s enol equilibrium has been demonstrated by studying NMR, IR and UV spectroscopic properties of kynurenic acid and related quinoline derivatives. The optical and emission properties of kynurenic acid and related compounds are thus very sensitive to pH conditions. Fluorescence and intersystem crossing (ISC) quantum yields have been measured in various solvents. The first excited singlet state is probably very short-lived in neutral aqueous solution. The triplet state is long-lived (10 μs) as shown by 353 nm laser flash spectroscopy. A high ISC quantum yield and a large molar extinction coefficient make it easy to detect the T1→ Tn transient (Δmax≃ 570 nm).  相似文献

The decay processes of the lowest excited singlet and triplet states of five heteropsoralens (HPS) were investigated by steady-state and shift-phase fluorometry and by laser-flash photolysis in different solvents. The emission spectra of HPS are detectable only in trifluoroethanol (TFE), where fluorescence lifetimes (τF) and quantum yields (φF) were measured. The triplet lifetimes (τT), triplet (φT) and singlet-oxygen production (φΔ) quantum yields were determined in benzene, ethanol and TFE by laser-flash photolysis. Semiempirical (INDO/1-CI) calculations allowed the nature of the lowest excited singlet and triplet states and transition probabilities to be obtained. Theoretical and experimental results indicate that the two lowest excited singlet states S1 and S2 of HPS are close-lying and different in nature (π,π* and n,π*). The "proximity effect" between these two states controls the photophysical properties of HPS as it does for the other furocoumarins. However, HPS have a peculiar behavior with respect to the related compounds because they are fluorescent and have, in three cases, detectable intersystem crossing only in TFE. This behavior can be tentatively explained by a different energy gap and/or order between the S1 and S2 states.  相似文献

Abstract— –Pulse radiolysis has been used to excite the triplet states of β-carotene (τ# 9μ sec) and lycopene (τ= 8μsec) in hexane solution, both in the presence and absence of naphthalene as a triplet sensitiser. The absorption spectra of both triplets have been measured in the range 430–550 nm and have thus been extended into the region of the corresponding singlet absorptions. The overlap of the triplet and singlet spectra is discussed in relation to in vivo studies. Extinction coefficients of 1.3±0.1 × 105 l/mole cm for β-carotene triplet 515 nm and 3.9±0.2 × 105 l/mole cm for lycopene triplet at 525 nm were obtained. Isomerisation of the all- trans polyenes used was detected and preliminary measurements indicate that the yield of isomerisation was greater than the triplet yield. The rate of triplet energy transfer from naphthalene to β-carotene was estimated to be 1.5 × 1010 l/mole sec. The corresponding value for lycopene was 1.4× 1010 l/mole sec. The measured efficient quenching of triplet β-carotene by oxygen may occur by an energy transfer mechanism, leading to the formation of singlet oxygen (1Δg. This would suggest that the triplet energy level of β-carotene lies between 121 and 94 kJ mole-1.  相似文献

Abstract— The oxidation of purpurogalline (PPG) by alkaline solution of H2O2 pH 9–11 at 298°K is accompanied by chemiluminescence (CL) in the spectral range 400–600 nm with the maximum at 500 nm and quantum yield about 10-6. The optimal concentrations of reactants with respect to maximal intensity are: 2 × 10-4 M PPG, 10-2 M NaOH, 1 M H2O2. Activation energy calculated from the maximum intensity of CL is 8.1×0.4 kcal/mole. Light emission occurs only when OH-groups of the phenolic ring of PPG undergo oxidation and the blue anion of o -PPG-quinone is formed. The rate that determines step in the reaction associated with luminescence is the nucleophilic attack of OOH- ion on the blue anion of o -PPG-quinone. In this exergonic step (-ΔH = 63 to 230kcal/mole) the o - and/or p -quinone ring is opened and carbonyl derivatives of α-tropolone are produced. They display fluorescence in the region 400–600 nm. The fluorescence spectrum of the reaction mixture after oxidation of PPG is very close to that of CL. It is likely that carbonyl derivatives of α-tropolone are emitters of CL.  相似文献

The singlet oxygen quantum yield (φ1o2) of 11 purified fluorescein derivatives was determined by reaction with singlet oxygen acceptors in aqueous and ethanolic solutions; in both solvents φ1o2 was enhanced with increasing halogenation. Tryptophan and 2,2,6,6-tetramethylpiperidone were found to be unadapted for the determination of φ1o2, in our systems; however, the use of 9.10-dipropionic anthracene acid andp-nitrosodimethylaniline in conjunction with imidazole derivatives was suitable for 1O2 detection in water. Both methods lead to results in excellent agreement. As in ethanol. φ1o2, was equal to the triplet state quantum yield (φT), the comparison between the two solvents showed that φT in water was greater than in ethanol. The comparison between our values obtained with polychromatic light with published data obtained with monochromatic light suggests that the triplet quantum yield of fluorescein derivatives is wavelength independent.  相似文献

Abstract— The photophysical characteristics of two second-generation PDT photosensitizers, tin ethyl etiopurpurin I (SnET2) and tin octaethylbenzochlorin (SnOEBC), have been measured in homogeneous solution and when bound to bovine serum albumin (BSA). The ground state and triplet state absorption spectra have been characterized, as have triplet lifetimes and quantum yields for intersys-tem crossing, singlet oxygen formation and photobleaching. In total, these parameters provide a complete set of data that can be used to quantitatively compare the photosensitizing efficiencies of these molecules. The photo-bleaching quantum yield of SnET2 is increased dramatically when it is bound to BSA, thus limiting the production of singlet oxygen at incident fluences above 1 J/cm2. In contrast, the quantum yield of photobleaching of SnOEBC is at least an order of magnitude lower than that of SnET2 under these conditions and does not significantly limit the photosensitization process for typical in vivo or in vitro fluences. This difference is expected to play a significant role in determining the relative photosensitizing ability of these compounds in vivo.  相似文献

Abstract— Weak luminescence was detected using photon counting equipment, from oxygenated, liquid cultures of Escherichia coli during two stages of its growth cycle. The first period of emission occurred during the exponential phase of growth and comprised a UV(210–330 nm) band and a visible region(450–620 nm) band, the total intensity being (1.65 ± 0.12) x 103 counts s-1. The second period of emission occurred during the stationary phase of growth and comprised only a visible region(450–620 nm) band of intensity (8.72 ± 0.15) x 103 counts s-1. When the growth temperature was raised from 306.15 to 310.15 K, the above emission intensities were approximately halved, but the spectra were not changed significantly. No luminescence was observed at either temperature when the E. coli was grown anaerobically. The visible region luminescence was attributed to excited carbonyl groups and excited singlet O2 dimers formed during the decomposition of lipid peroxides. The UV component was tentatively assigned to oxidative side reactions accompanying the synthesis of proteins.  相似文献

Abstract— The 1-anilinonaphthalene-8-sulfonic acid solubilized in dodecylammonium propionate reversed micellar cyclohexanic solutions, emitted a strong fluorescence, and was photooxidized under aerobic conditions. Carbon tetrachloride (CCl4) highly quenched the fluorescence and remarkably enhanced the oxidation reaction. The fluorescence quenching obeyed the Stern-Volmer relation, and the photooxidation was caused by the singlet oxygen generated by the photosensitization of the dye. From the kinetic analysis, it was known that the intersystem crossing rate from the dye excited singlet to triplet was enhanced by CCl4. Carbon tetrachloride did not quench the triplet state. The ratio of quantum yields for the oxidation in the presence and absence of CCl4 was independent of the oxygen concentration in the reaction mixture. The fluorescence quenching constant and the intersystem crossing rate were obtained at various solubilized water contents.  相似文献

Abstract— The relative reactivity of singlet molecular oxygen, 02(1Δg), α-,β-,Γ-and δ with -tocopherol (vitamin E) was investigated using microwave discharge generation as a uniquely clean source of singlet oxygen and using a hydrocarbon solvent to approximate the membrane environment. The relative efficiencies of the tocopherols for O2(1Δg) were found to decrease in the order: D-α-tocopherol > D-β-tocopheroI > D–Γ-tocopherol > D-δ-tocopherol. The reaction products in all cases were found to be mixtures of quinone and quinone epoxides apparently resulting from decomposition of the primary product, the hydroperoxydienone.  相似文献

The highly reversible thermally stable photochromic system consisting of heterocoerdianthrone-endoperoxide (HCDPO) and its parent compounds HCD and 02 is proposed as a new reusable liquid chemical actinometer in the UV region 248 ≤λ≤ 334 nm. The foremost advantageous features of this new system are: (1) high reproducibility and accuracy, (2) high sensitivity, (3) almost wavelength independent quantum yields, (4) no loss in accuracy even after 100 repeated actinometric cycles and (5) very easy handling and straightforward calculability of the radiation quantum flux.  相似文献

Abstract— Triplet-and singlet-related photoprocesses of pyrene-1-aldehyde (PA) in various solvents have been investigated in detail using 337.1 and 355 nm laser flash photolysis in conjunction with time-correlated determination of fluorescence lifetimes (τF) and steady-state photochemical and absorption-emission spectral measurements. In benzene, the lowest triplet of PA (43 < ET < 46 kcal/mol) has a lifetime of about 50 µs (τT) and displays the absorption maximum at 443 nm with a maximum extinction coefficient (εmax) of 21000 M -1cm-1; the corresponding ketyl radical has a sharp absorption maximum at 428 nm (εmax≥ 25000 M -1cm-1). The quantum yields (φT) of lowest triplet occupation are high in nonprotic solvents (0.6–0.8), decrease in protic solvents (alcohols) as the polarity of the latter is increased, and maintain a complementary relationship with the quantum yields (φF) of fluorescence. Quantum yields (φPC) of loss of PA due to photoreactions in some solvents have also been determined under conditions of steady irradiation at 366 nm; φPC is in the range 0.1–0.2 in electron-rich olefinic solvents such as cyclohexene and tetramethylethylene. These results concerning τF, τT, φF. φT and φPC as well as the effects of 1,2,4-trimethoxybenzene and 2,5-dimethyl-2,4-hexadiene as quenchers for fluorescence, triplet yield, and photochemistry are discussed in the light of possible state orders for PA in polar and nonpolar environments.  相似文献

Abstract— Several ozone-biomolecule reactions have previously been shown to generate singlet oxygen in high yields. For some of these orone-biomolecule reactions, we now show that the apparent singlet-oxygen yields determined from measurements of 1270 nm chemiluminescence were artifactually elevated by production of gas-phase singlet oxygen. The gas-phase singlet oxygen results from the reaction of gas-phase ozone with biomolecules near the surface of the solution. Through the use of a flow system that excludes air from the reaction chamber, accurate singlet-oxygen yields can be obtained. The revised singlet-oxygen yields (mol 1O2 per mol O3) for the reactions of ozone with cysteine, reduced glutathione, NADH, NADPH, human albumin, methionine, uric acid and oxidized glutathione are 0.23 ± 0.02, 0.26 ± 0.02, 0.48 ± 0.04, 0.41 ± 0.01, 0.53 ± 0.06, 1.11 ± 0.04, 0.73 ± 0.05 and 0.75 ± 0.01, respectively. These revised singlet-oxygen yields are still substantial.  相似文献

Abstract— The quantum yield of the photodynamic inactivation of lysozyme increases in the sequence acridine orange, methylene blue, proflavine and acriflavine (1:5:6:12). At least up to protein concentrations of 0.1 m M , singlet oxygen is exclusively responsible for the inactivation of the enzyme. For methylene blue, acriflavine and proflavine the quantum yields decrease considerably with increasing dye concentrations. From measurements in H2O and D2O buffer solutions it was concluded that in the case of methylene blue the effect is mainly caused by the quenching of singlet oxygen [rate constant (3–4) × 108 M −1 s−1]. For the acridine sensitizers both singlet oxygen and dye triplet quenching processes have to be taken into consideration. It has been found that all sensitizers act as competitive inhibitors of the enzymatic reaction of lysozyme. However, the dye-protein interaction near the active center cannot be responsible for the observed dye self-quenching effect.  相似文献

Abstract The results of a nanosecond laser flash photolysis investigation of the UVA sunscreen Mexoryl* SX in various solvent environments and within a commercial sunscreen formulation are reported. To the best of our knowledge this is the first laser flash photolysis study of a commercial suncare formulation. In each of these environments kinetic UV-visible absorption measurements following nanosecond 355 nm laser excitation reveals a short-lived species with a solvent-dependent absorption maximum around 470–500 nm and a solvent-dependent lifetime of 50–120 ns. This transient absorption is attributed to the triplet state of Mexoryl* SX on the basis that it is quenched by molecular oxygen leading to the formation of singlet oxygen in acetonitrile. The singlet oxygen quantum yield (φΔ), determined by comparative time-resolved near-infrared luminescence measurements and extrapolated to the limit of complete triplet state quenching, is estimated as 0.09 ± 0.03 in acetonitrile. In aqueous solution the shorter triplet state lifetime combined with lower ambient oxygen concentrations precludes significant triplet state quenching. For the commercial sunscreen formulation there was no observable difference in the measured triplet lifetime between samples exposed to oxygen or argon, suggesting that the singlet oxygen quantum yield in such environments is likely to be orders of magnitude lower than that measured in acetonitrile.  相似文献
