The structural features of the 1H‐imidazo[4,5‐c]pyridine (ICPY) tautomers and homodimers of the most stable tautomers have been studied by quantum chemical methods. FTIR and Raman spectra of the ICPY were recorded in the range of 4000–60 cm?1 and 3500–5 cm?1. The predominant tautomer among four possible isomers of ICPY were determined. The optimized geometries and vibrational frequencies of possible ICPY tautomers and dimers were computed by B3LYP/DFT method with 6‐311++G(d,p) and 6‐31G(d) basis sets. All vibrational frequencies assigned in detail with the help of total energy distribution (TED) and isotopic shifts. ICPY dimeric forms were also characterized according to their hydrogen bonding interactions, and it has been found that the most stable ICPY homodimer establishes moderate strong N ? H …N type hydrogen bond. 1H NMR, 13C NMR, and 15N NMR properties have been calculated for all tautomeric forms using the gauge independent atomic orbital (GIAO) method. © 2010 Wiley Periodicals, Inc. One‐pot reaction of cyclic 1,3‐diketones, dimethylformamide dimethylacetal (DMFDMA) and 2‐(1H‐benzo[d ]imidaz‐2‐yl)acetonitrile was found to be a highly selective process leading to 4‐oxo‐1,2,3,4‐tetrahydrobenzo[4,5]imidazo[1,2‐a ]quinolin‐6‐yl cyanides. Optimized reaction conditions using water as solvent at room temperature or under microwave heating allowed high yields of the target products required no additional purification.  相似文献

A number of 4,5‐dihydrobenzo[e]imidazo[2,1‐c][1,4,2]diazaphosphinine derivatives were prepared by the direct phosphorylation of 1‐(4‐Chlorophenylcarboxamido)‐2‐(1H‐1‐imidazolyl)‐5‐trifluoromethylbenzene in basic medium with phosphorus(III) bromide and dibromophenylphosphine. The tricyclic compounds 6a, 6b , and 9 having a trivalent phosphorus atom undergo the diazaphosphinine ring opening upon treatment with secondary amines in the presence of sulfur. © 2002 John Wiley & Sons, Inc. Cyclization of N‐methyl‐ and N‐benzoylpyridylthioureas, prepared from the corresponding aminopy‐ridines, has been realized using various conditions. With bromine in acetic acid or potassium ferricyanide, the cyclization occurred on the nitrogen of the pyridine ring and pyridinium salts or 1,2,4‐fhiadiazolo[2,3‐a]pyridylidene systems were obtained. On the other hand, treatment of the thioureas with sodium methoxide in N‐methylpyrrolidinone (NMP) led to formation of fhiazolo[4,5‐b] and [5,4‐b]pyridines, which are interesting targets for biological evaluation.  相似文献

Thiazole 1 , when reacted with chloroacetyl chloride, afforded N‐(5‐acetyl‐4‐methylthiazol‐2‐yl) chloroacetamide 2 . It has been found that compound 2 reacted with α‐cyanocinnamonitrile derivatives 6a–c to afford reaction products 8a–c . Also, compound 2 coupled smoothly with benzenediazonium chloride afforded the phenylhydrazone 14 . Coupling of the sulfonium bromide 17 with diazotized aromatic amines or N‐nitrosoacetanilides afforded the arylhydrazones 20a,b . Synthesis of 2‐amino‐1‐methyl‐6‐phenylimidazo[4,5‐b]pyridine (PhIP), three structural isomers, and two desphenyl PhIP congeners has been carried out. Mutagenic potency was evaluated using S. typhimurium strain TA98 in the Ames test. Mutagenic potency increased in relation to structural features in these heterocyclic amines that allow extended resonance between the phenyl and imidazo[4,5‐b]pyridine N2‐amino substituents. By contrast, PhIP isomers, whose substitution disallows involvement of the phenyl group in their aminoimidazo resonance hybrids, and desphenyl congeners were from 86‐ to 234‐fold less mutagenic than PhIP.  相似文献

A number of thiosemicarbazones of 2‐acetyl‐imidazo[4,5‐b]pyridine were prepared in order to investigate their in vitro antineoplastic activities. Three compounds: (i) 2‐acetylimidazo[4,5‐b]pyridin‐4‐ sec ‐butyl‐3‐thiosemicarbazone [(A7), NSC674098], (ii) 2‐acetylimidazo[4,5‐b]pyridin‐4‐tert‐butyl‐3‐thiosemi‐carbazone [(A9), NSC674099], (iii) 2‐acetylimidazo[4,5‐b]pyridin‐4‐cyclohexyl‐3‐thiosemicarbozone [(A11), NSC674101] showed remarkable activity against some of the cell lines tested. The Biological Evaluation Committee of N.C.I. determined that further secondary testing should be carried out (these compounds were tested against prostate cancer).  相似文献

A quantitative structure–activity relationship (QSAR) of 3‐(9‐acridinylamino)‐5‐hydroxymethylaniline (AHMA) derivatives and their alkylcarbamates as potent anticancer agents has been studied using density functional theory (DFT), molecular mechanics (MM+), and statistical methods. In the best established QSAR equation, the energy (ENL) of the next lowest unoccupied molecular orbital (NLUMO) and the net charges (QFR) of the first atom of the substituent R, as well as the steric parameter (MR2) of subsituent R2 are the main independent factors contributing to the anticancer activity of the compounds. A new scheme determining outliers by “leave‐one‐out” (LOO) cross‐validation coefficient (q) was suggested and successfully used. The fitting correlation coefficient (R2) and the “LOO” cross‐validation coefficient (q2) values for the training set of 25 compounds are 0.881 and 0.829, respectively. The predicted activities of 5 compounds in the test set using this QSAR model are in good agreement with their experimental values, indicating that this model has excellent predictive ability. Based on the established QSAR equation, 10 new compounds with rather high anticancer activity much greater than that of 34 compounds have been designed and await experimental verification. © 2006 Wiley Periodicals, Inc. Benzo[4,5]imidazo[1,2‐a]pyrimidine derivatives were synthesized via the three‐component reaction of aldehyde, β‐dicarbonyl compound and 2‐aminobenzimidazole in water under microwave irradiation and without catalyst conditions. A series of 2,5,6‐trisubstituted imidazo[2,1‐b][1,3,4]thiadiazoles via Mannich reaction of imidazo[2,1‐b][1,3,4]thiadiazoles with morpholine and formaldehyde were synthesized. Structures of all the newly synthesized compounds are well supported by spectral data such as IR, NMR, Mass, and Elemental analysis. Compound 3a has been confirmed by X‐ray diffraction studies. J. Reaction of 6‐amino‐2‐methylthiouracil and 6‐amino‐1,3‐dimethyluracil with equimoler amounts of cyclic ketones or cyclic 1,3‐diketones and the appropriate aromatic aldehydes yielded regioselectivity a series of polycyclic pyrimido[4,5‐b]quinoline and pyrido[2,3‐d]pyrimidine derivatives in good yields.  相似文献

A series of novel imidazo[4,5‐b]pyridine derivatives were designed and synthesized. The structures of all the newly synthesized compounds were identified by spectroscopic data NMR, MS, and elemental analysis. Bioassay showed that the compounds exhibited potent fungicidal activities against Erysiphe graminis, Puccinia polysora, and so forth. Particularly, 2‐chloro‐5‐((5‐methoxy‐2‐(2‐(trifluoromethyl)phenyl)‐3H‐imidazo[4,5‐b]pyridin‐3‐yl)methyl)thiazole ( 9b ) displayed fungicidal potency against Ppolysora. Its EC50 value: 4.00 mg/L is comparable with that of tebuconazole. The structure–activity relationship for the target compounds is discussed.  相似文献
