A simple reversed-phase high-performance liquid chromatographic (HPLC) method has been developed for the simultaneous determination of the antiepileptic drugs (AEDs) zonisamide (ZNS), primidone (PRI), lamotrigine (LTG), phenobarbital (PB), phenytoin (PHT), oxcarbazepine (OXC), and carbamazepine (CBZ) and two of their active metabolites, monohydroxycarbamazepine (MHD) and carbamazepine 10,11-epoxide (CBZE) in human plasma. Plasma (100 μL) was pretreated by deproteinization with 300 μL methanol containing 20 μg mL−1 propranolol hydrochloride as internal standard. HPLC was performed on a C8 column (4.6 mm × 250 mm; particle size 5 μm) with methanol–acetonitrile–0.1% trifluoroacetic acid, 235:120:645 (v/v), as mobile phase at a flow rate of 1.5 mL min−1. ZNS, OXC, and CBZ were monitored by UV detection at 235 nm, and PRI, LTG, MHD, PB, PHT, and CBZE by UV detection at 215 nm. Relationships between response and concentration were linear over the concentration ranges 1–80 μg mL−1 for ZNS, 5–50 μg mL−1 for PRI, 1–25 μg mL−1 for LTG, 1–50 μg mL−1 for MHD, 5–100 μg mL−1 for PB, 1–10 μg mL−1 for CBZE, 0.5–25 μg mL−1 for OXC, 1–50 μg mL−1 for PHT, and 1–25 μg mL−1 for CBZ. Intra-day and inter-day reproducibility were adequate (coefficients of variation were ≤11.6%) and absolute recovery ranged from 95.2 ± 6.13 to 107.7 ± 7.76% for all the analytes; for the IS recovery was 98.69 ± 1.12%. The method was proved to be accurate, reproducible, convenient, and suitable for therapeutic monitoring of the nine analytes.  相似文献

Aidi injection is a clinical medicine used in China for the treatment of cancer. Calycosin-7-O-β-d-glucoside is the main effective components of the formulas. In this study, a high performance liquid chromatographic (LC) method was developed to quantify calycosin-7-O-β-d-glucoside in rat plasma using a liquid–liquid extraction and ultraviolet (UV) absorbance detection. LC analysis was performed on a Diamonsil C18 column (200 × 4.6 mm i.d., 5 μm particle size) with isocratic mobile phase consisting of acetonitrile–0.05% phosphoric acid (19.5:80.5, v/v) of a flow rate of 1.0 mL min−1. The linear range was 0.11–17.6 μg mL−1 and the low quantification limit was 0.11 μg mL−1 (S/N = 10). The intra- and inter-day relative standard deviations (RSD) in the measurement of quality control (QC) samples 0.11, 0.22, 1.32 and 8.80 μg mL−1 ranged from 4.1 to 6.3 and 4.3 to 6.2%, respectively. The accuracy was from −6.7 to 4.3% in terms of relative error (RE). Calycosin-7-O-β-d-glucoside was stable in storage at −20 °C for 2 weeks and stable after three freeze–thaw cycles in rat plasma. This method was validated for specificity, accuracy, precision and was successfully applied to pharmacokinetic study of calycosin-7-O-β-d-glucoside in rat plasma after intravenous administration of Aidi lyophilizer.  相似文献

A simple, rapid, and precise reversed-phase high-performance liquid chromatographic method has been developed for simultaneous determination of losartan potassium, ramipril, and hydrochlorothiazide. The three drugs were separated on a 150 mm × 4.6 mm i.d., 5 μm particle, Cosmosil C18 column. The mobile phase was 0.025 m sodium perchlorate–acetonitrile, 62:38 (v/v), containing 0.1% heptanesulphonic acid, pH adjusted to 2.85 with orthophosphoric acid, at a flow rate of 1.0 mL min−1. UV detection was performed at 215 nm. The method was validated for linearity, accuracy, precision, and limit of quantitation. Linearity, accuracy, and precision were acceptable in the ranges 35–65 μg mL−1 for losartan, 1.75–3.25 μg mL−1 for ramipril, and 8.75–16.25 μg mL−1 for hydrochlorothiazide.  相似文献

A rapid and sensitive liquid chromatography-tandem mass spectrometry assay was developed for the determination of a novel histone deacetylase inhibitor, cyclo{(2S)-2-amino-8-[(aminocarbonyl)hydrazono]decanoyl-1-l-tryptophyl-l-isoleucyl-(2R)-2-piperidinecarbonyl} (SD-2007), in rat serum. The mobile phase consisted of acetonitrile and ammonium formate (10 mM) (85:15 v/v), and the flow rate was 0.25 mL min−1. Chromatographic separations were achieved by isocratic elution on a C18 column. Multiple reaction monitoring was based on the transition of m/z = 681.8 → 83.6 for SD-2007 and 372.1 → 176.1 for trazodone (internal standard). A linearity was observed over a concentration range from 2 to 1,000 ng mL−1 (r 2 > 0.999), with the lower limit of quantification at 2 ng mL−1 with 100 μL of rat serum. The mean intra- and inter-day assay accuracy ranged from 98.5–109.7% to 95.2–102.7%, respectively, and the mean intra- and inter-day precision was between 4.3–11.3% and 2.9–13.3%. The developed assay was applied to a pharmacokinetic study of SD-2007 in rats after intravenous injection (dose 4 mg kg−1).  相似文献

An integrated solid-phase spectrophotometry–FIA method is proposed for simultaneous determination of the mixture of saccharin (1,2-benzisothiazol-3(2H)-one-1,1-dioxide; E-954) (SA) and aspartame (N-l-α-aspartyl-l-phenylalanine-1-methyl ester; E-951) (AS). The procedure is based on on-line preconcentration of AS on a C18 silica gel minicolumn and separation from SA, followed by measurement, at λ=210 nm, of the absorbance of SA which is transiently retained on the adsorbent Sephadex G-25 placed in the flow-through cell of a monochannel FIA setup using pH 3.0 orthophosphoric acid–dihydrogen phosphate buffer, 3.75×10–3 mol L−1, as carrier. Subsequent desorption of AS with methanol enables its determination at λ=205 nm. With a sampling frequency of 10 h−1, the applicable concentration range, the detection limit, and the relative standard deviation were from 1.0 to 200.0 μg mL−1, 0.30 μg mL−1, and 1.0% (80 μg mL−1, n=10), respectively, for SA and from 10.0 to 200.0 μg mL−1, 1.4 μg mL−1, and 1.6% (100 μg mL−1, n=10) for AS. The method was used to determine the amounts of aspartame and saccharin in sweets and drinks. Recovery was always between 99 and 101%. The method enabled satisfactory determination of blends of SA and AS in low-calorie and dietary products and the results were compared with those from an HPLC reference method.  相似文献

A new marine sediment certified reference material, NMIJ CRM 7306-a, for butyltin and phenyltin analysis has been prepared and certified by the National Metrological Institute of Japan at the National Institute of Advanced Industrial Science and Technology (NMIJ/AIST). Candidate sediment material was collected at a bay near industrial activity in Japan. After air-drying, sieving, and mixing the material was sterilized with γ-ray irradiation. The material was re-mixed and packaged into 250 glass bottles (15 g each) and these were stored in a freezer at −30 °C. Certification was performed by use of three different types of species-specific isotope-dilution mass spectrometry (SSID–MS)—SSID–GC–ICP–MS, SSID–GC–MS, and SSID–LC–ICP–MS, with 118Sn-enriched organotin compounds synthesized from 118Sn-enriched metal used as a spike. The 118Sn-enriched mono-butyltin (MBT), dibutyltin (DBT), and tributyltin (TBT) were synthesized as a mixture whereas the 118Sn-enriched di-phenyltin (DPhT) and triphenyltin (TPhT) were synthesized individually. Four different extraction methods, mechanical shaking, ultrasonic, microwave-assisted, and pressurized liquid extraction, were adopted to avoid possible analytical bias caused by non-quantitative extraction and degradation or inter-conversion of analytes in sample preparations. Tropolone was used as chelating agent in all the extraction methods. Certified values are given for TBT 44±3 μg kg−1 as Sn, DBT 51 ± 2 μg kg−1 as Sn, MBT 67 ± 3 μg kg−1 as Sn, TPhT 6.9 ± 1.2 μg kg−1 as Sn, and DPhT 3.4 ± 1.2 μg kg−1 as Sn. These levels are lower than in other sediment CRMs currently available for analysis of organotin compounds.  相似文献

The quick separation and simultaneous determination of d-amphetamine and diphenhydramine in the quick-acting anti-motion capsules was investigated by capillary zone electrophoresis. The influence of different parameters (internal standard, injection modes, pH, concentration of the running buffer and applied voltage) was systematically studied. The two compounds could be well separated within 2.0 min in a 40.2 cm fused-silica capillary at a separation voltage of 20 kV in a 50 mM phosphate–12.5 mM borate buffer adjusted to pH 5.5. Correlation coefficients for calibration curves in the range 0.50–1.50 μg mL−1 for d-amphetamine and 2.75–8.25 μg mL−1 for diphenhydramine were higher than 0.999. The limits of detection of d-amphetamine and diphenhydramine were 10.0 and 5.5 ng mL−1 and the recoveries of the compounds in the QAAMC were 99.80 and 99.85%, respectively. The authors L. Zhang and Y. Chen equally contributed to this work.  相似文献

Liquid chromatography–fluorescence (LC–FLS), liquid chromatography–tandem mass spectrometry (LC–MS/MS) and inductively coupled plasma–mass spectrometry (ICP-MS) methods were developed and validated for the evaluation of motexafin gadolinium (MGd, Xcytrin) pharmacokinetics and biodistribution in plasma and tissues. The LC–FLS method exhibited the greatest sensitivity (0.0057 μg mL−1), and was used for pharmacokinetic, biodistribution, and protein binding studies with small sample sizes or low MGd concentrations. The LC–MS/MS method, which exhibited a short run time and excellent selectivity, was used for routine clinical plasma sample analysis. The ICP–MS method, which measured total Gd, was used in conjunction with LC methods to assess MGd stability in plasma. All three methods were validated using human plasma. The LC–FLS method was also validated using plasma, liver and kidneys from mice and rats. All three methods were shown to be accurate, precise and robust for each matrix validated. For three mice, the mean (standard deviation) concentration of MGd in plasma/tissues taken 5 hr after dosing with 23 mg kg−1 MGd was determined by LC–FLS as follows: plasma (0.025±0.002 μg mL−1), liver (2.89±0.45 μg g−1), and kidney (6.09±1.05 μg g−1). Plasma samples from a subset of patients with brain metastases from extracranial tumors were analyzed using both LC–MS/MS and ICP–MS methods. For a representative patient, ≥90% of the total Gd in plasma was accounted for as MGd over the first hour post dosing. By 24 hr post dosing, 63% of total Gd was accounted for as MGd, indicating some metabolism of MGd.  相似文献

Summary A simple and rapid liquid chromatographic method has been developed for the determination of therapeutic levels of piperacillin (I) and ceftazidime (II) in human plasma. Plasma and p-propionamidophenol (internal standard) were precipitated with methanol (I) or 20% trichloroacetic acid (II). The supernatant was analysed on a 5 μm Spherisorb ODS C18 column with acetonitrile-0.05 M phosphate buffer pH 3.8 as mobile phase and ultraviolet detection at 254 nm. The calibration graph was linear from 10 to 250 μg mL−1, for (I), and from 5 to 200 μg mL−1 for (II). Intra and inter-day CV did no exceed 2.29% for (I), and were 10.76–11.13%–2.00–5.62 for (II) at concentrations of 10 μg mL−1 and 250 μg mL−1.  相似文献

Summary A high-performance liquid chromatographic method, with 9-anthryldiazomethane as derivatizing agent, has been developed for the simultaneous determination ofN-carbamoyl aspartate andl-dihydroorotate in serum. Sample preparation for 1 mL serum was by simple liquid-liquid extraction and then derivatization. The compounds were separated on a Luna C18(2) column by use of a gradient prepared from acetonitrile and 10 mM sodium acetate buffer, pH 6.0, and fluorimetric detection was performed at excitation and emission wavelengths of 365 nm and 412 nm, respectively. The response was found to be linearly dependent on concentration between 0.8 and 60 μg mL−1 forl-dihydrooratate and between 0.9 and 90 μg mL−1 forN-carbamoyl aspartate; the mean recovery rates were 50 and 51%, respectively. The limits of detection and quantification were 0.33 μg mL−1 and 0.6 μg mL−1, respectively, forl-dihydroorotate and 0.4 μg mL−1 and 0.7 μg mL−1 forN-carbamoyl aspartate. This method can be used to assess accumulation ofN-carbamoyl aspartate andl-dihydroorotate in body fluids in situations where cellular pyrimidine de novo synthesis is impaired.  相似文献

Summary A reliable and sensitive high-performance liquid chromatographic method for the determination of the recent antidepressant citalopram and two metabolites in human plasma has been developed. Fluorescence detection at 300 nm was used, exciting at 238 nm. Separation was obtained using a reversed-phase column (C18, 250 × 3.0 mm i.d., 5 μm) and a mobile phase. 40% acetonitrile: 60% aqueous tetramethylammonium perchlorate (pH 1.9). Calibration curves were linear over a working range: 5–300 ng mL−1 for citalopram, 2.5–150.0 ng mL−1 for desmethylcitalopram and 2.5–50.0 ng mL−1 for didesmethylcitalopram. The limits of quantitation (LOQ) were 1.5 ng mL−1 for citalopram and desmethylcitalopram and 2.0 ng mL−1 for didesmethylcitalopram. Precision data, as well as accuracy, were satisfactory and no interference from different psychotropic drugs was found. The method was therefore suitable for therapeutic drug monitoring of citalopram and its active metabolites in plasma of depressed patients.  相似文献

A stability-indicating reversed-phase liquid chromatographic (RPLC) method has been established for analysis of ramipril (RAM) and moexipril hydrochloride (MOEX.HCl) in the presence of the degradation products generated in studies of forced decomposition. The drug substances were subjected to stress by hydrolysis (0.1 m NaOH and 0.1 m HCl), oxidation (30% H2O2), photolysis (254 nm), and thermal treatment (80 °C). The drugs were degraded under basic and acidic conditions and by thermal treatment but were stable under other stress conditions investigated. Successful separation of the drugs from the degradation products was achieved on a cyanopropyl column with 40:60 (v/v) aqueous 0.01 m ammonium acetate buffer (pH 6)–methanol as mobile phase at a flow rate of 1 mL min−1. Detection was by UV absorption at 210 nm. Response was a linear function of concentration over the range 5–50 μg mL−1 (r > 0.9995), with limits of detection and quantitation (LOD and LOQ) of 0.04 and 0.09 μg mL−1, respectively, for RAM and 0.014 and 0.32 μg mL−1, respectively, for moexipril. The method was validated for specificity, selectivity, solution stability, accuracy, and precision. Statistical analysis proved the method enabled reproducible and selective quantification of RAM and MOEX as the bulk drug and in pharmaceutical preparations. Because the method effectively separates the drugs from their degradation products, it can be used as stability-indicating.  相似文献

This paper describes a new and rapid method for accurate quantification of the six ergot alkaloids, ergometrine, ergotamine, ergosine, ergocristine, ergocryptine, and ergocornine, by liquid chromatography–tandem mass spectrometry (LC–MS–MS). The six ergot alkaloids studied have been defined by the European Food Safety Authority (EFSA) as among the most common and physiologically active ones. In addition, the method enables the quantification of the corresponding six epimers (ergo-inines) of these ergot alkaloids. This is of considerable importance in terms of the differences in toxicity of the isomeric forms. The method involves extraction under alkaline conditions using a mixture of acetonitrile and ammonium carbonate buffer followed by a rapid clean-up using dispersive solid-phase extraction with PSA (primary secondary amine) and a short chromatographic LC-run (21 min) with subsequent MS–MS detection. The method was developed and validated using ten different cereal and food samples. The major strength of the new method compared with previously published techniques is the simplicity of the clean-up procedure and the short analysis time. The limits of quantification were 0.17 to 2.78 μg kg−1 depending on the analyte and matrix. Recovery values for the 12 ergot alkaloids spiked into ten different matrices at levels of 5, 50, and 100 μg kg−1 were between 69 and 105% for 85 of 90 recovery measurements made over six days. Measurement uncertainty values were highly satisfactory. At a concentration level of 5 μg kg−1 the expanded measurement uncertainty ranged from ±0.56 to ±1.49 μg kg−1, at a concentration level of 100 μg kg−1 the expanded measurement uncertainty ranged from ±8.9 to ±20 μg kg−1. Both LOQs and measurement uncertainties were dependent on the analyte but almost independent of the matrix. The method performance was satisfactory when tested in a mini-intercomparison study between three laboratories from three different countries. A high-performance liquid chromatography–UV method for determining DCJW concentration in rat plasma was developed. The method described was applied to a pharmacokinetics study of intramuscular injection in rats. The plasma samples were deproteinized with acetonitrile in a one-step extraction. The HPLC assay was carried out using a VP-ODS column and the mobile phase consisting of acetonitrile–water (80:20, v/v) was used at a flow rate of 1.0 mL min−1 for the effective eluting DCJW. The detection of the analyte peak area was achieved by setting a UV detector at 314 nm with no interfering plasma peak. The method was fully validated with the following validation parameters: linearity range 0.06–10 μg mL−1 (r > 0.999); absolute recoveries of DCJW were 97.44–103.46% from rat plasma; limit of quantification, 0.06 μg mL−1 and limit of detection, 0.02 μg mL−1. The method was further used to determine the concentration–time profiles of DCJW in the rat plasma following intramuscular injection of DCJW solution at a dose of 1.2 mg kg−1. Maximum plasma concentration (C max) and area under the plasma concentration–time curve (AUC) for DCJW were 140.20 ng mL−1 and 2405.28 ng h mL−1.  相似文献

A simple, rapid and reproducible HPLC method was developed and validated for the simultaneous determination of olmesartan (OLM) medoxomil and hydrochlorothiazide (HCT) in combined tablets. Chromatography was carried out on a 4.6 mm I.D × 200 mm, 5 μm cyano column with methanol–10 mM phosphoric acid containing 0.1% triethylamine (pH 2.5, 50:50 v/v) at a flow rate of 1.0 mL min−1 and UV detector was set at 260 nm. Valsartan (VAL) was used as internal standard (IS). A linear response was observed in the range of 0.2–6 μg mL−1 (r 2 = 0.9998) for OLM and 0.1–4 μg mL−1 (r 2 = 0.9999) for HCT, respectively. The method showed good recoveries (99.56% for OLM and 99.48% for HCT) and the relative standard deviation (RSD) values for intra- and inter-day precision were 0.70–1.59 and 0.80–2.00% for OLM and 1.20–1.37 and 1.63–1.93% for HCT, respectively. The developed method was applied successfully for quality control assay of OLM and HCT in combined tablets and in vitro dissolution studies.  相似文献

Summary A simple and rapid capillary zone electrophoretic method with UV detection has been developed for determination of tosufloxacin and trovafloxacin. The separation was performed in fused-silica capillaries (57 cm length × 75μm i.d.); the running buffer was 35mm borate + 35mm phosphate buffer solution, pH 8.6, containing 6% (v/v) acetonitrile. The applied potential was 15 kV, the temperature 30°C, and detection was at 262 nm. Piromidic acid was used as the internal standard. Response was linearly dependent on concentration in the range 1.0–120.0 μg mL−1 and the detection limit was 0.2 μg mL−1 for both compounds. The analysis was highly reproducible (RSD between 3.41 and 1.25%). The method was applied to the determination of tosufloxacin and trovafloxacin in human and rat urine. The method was validated by using HPLC as a reference method. Recovery was between 96.8 and 102%.  相似文献

Metabolism of four tobacco-specific N-nitrosamines (TSNAs), N′-nitrosonornicotine (NNN), N′-nitrosoanatabine (NAT), N′-nitrosoanabasine (NAB), and 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) has been studied by solid-phase extraction (SPE) and liquid chromatography–tandem mass spectrometry (LC–MS–MS). 4-(Methylnitrosamino)-4-(3-pyridyl)-1-butanol (iso-NNAL) was used as internal standard. SPE and LC–MS–MS was found to be a rapid, simple, sensitive, and selective method for analysis of TSNAs in rabbit serum. The relative standard deviation (R.S.D., n = 6) for analysis of 5 ng mL−1 and 0.5 ng mL−1 standards and of serum sample spiked with 5 ng mL−1 standards of five TSNAs was 2.1–11% and recovery of 5 ng mL−1 standards from serum was 100.2–112.9%. A good linear relationship was obtained between peak area ratio and concentration in the range of 0.2–100 ng mL−1 for NNAL and 0.5–100 ng mL−1 for other four TSNAs, with correlation coefficients (R 2) >0.99 (both linear and log–log regression). Detection limits for standards in solvent were between 0.04 and 0.10 ng mL−1. Doses of TSNAs administered to rabbits via the auricular vein were 4.67 μg kg−1 and 11.67 μg kg−1, in accordance with the different levels in cigarettes. Metabolic curves were obtained for the four TSNAs and for 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL), a metabolite of NNK; on the basis of these curves we modeled metabolic kinetic equations for these TSNAs by nonlinear curve fitting.  相似文献

Summary A method was developed for the separation and quantification of the warfare nerve agent sarin (O-isopropylmethylphosphonoflouridate), its metabolite methylphosphonic acid, the anti nerve agent drug pyridostigmine bromide (PB;3-dimethylaminocarbonyloxy-N-methyl pyridinium bromide) and its metaboliteN-methyl-3-hydroxypyridinium bromide in rat plasma and urine. The method involved using solid phase extraction and high performance liquid chromatography (HPLC) with reversed phase C18 column, and UV detection at 280 nm. The compounds were separated using gradient of 1% to 55% acetonitrile in 0.1% triflouroacetic acid water solution (pH 3.20) at flow rate of 0.9 ml/min in a period of 15 min. The retention times ranged from 4.4–12.1 min. The limits of detection were 50 ng mL−1 for PB andN-methyl-3-hydroxypyridinium bromide, and 10 μg mL−1 for sarin and methylphosphonic acid, while limits of quantitation were between 100 ng mL−1–12 μg mL−1. Average percentage recovery of five spiked samples from plasma were 84.6±8.4, 86.5±9.0, 76.4±8.5, 81.3±8.2, and from urine 78.5±7.9, 76.4±7.8, 74.4±8.4, 80.6±6.8 for sarin, methylphosphonic acid, pyridostigmine bromide andN-methyl-3-hydroxypyridinium bromide, respectively. This method was applied to analyze the above chemicals and metabolites following combined administration in rats.  相似文献

Simple, rapid, sensitive and selective methods for the determination of Cr(III) and W(VI) with flavonol derivatives in the presence of surface-active agents are proposed. In the pH ranges 3.4–4.2 and 1.9–2.5, the molar absorptivities of Cr(III)-morin-emulsifier S (EFA) and W(VI)-morin-polyvinylpyrrolidone (PVP) systems are 1.13×105 and 2.13×104 L mol−1 cm−1 at 435 and 415 nm, respectively. The Cr(III)-quercetin-PVP and W(VI)-quercetin-cetylpyridinium bromide (CPB) systems are formed in the pH ranges 4–4.6 and 2.2–2.8 with molar absorptivities 1.02×105 and 9.02×104 L. mol−1 cm−1 at 441 and 419 nm, respectively. The linear dynamic ranges for the determination of Cr(III) and W(VI) with morin in the presence of EFA and PVP are 0.03–0.46 and 0.71–8.1 μg mL−1, respectively. The corresponding ranges with quercetin are 0.04–0.54 and 0.14–2.1 μg mL−1 of Cr(III) and W(VI), respectively. The r.s.d (n = 10) for the determination of 0.25 and 3.7 μg mL−1 of Cr(III) and W(VI) with morin and their detection limits are 0.88 and 0.99% and 0.016 and 0.63 μg mL−1, respectively. Using quercetin, the r.s.d (n = 10) for 0.22 and 1.2 μg mL−1 of Cr(III) and W(VI) and their detection limits are 0.92 and 0.91% and 0.015 and 0.08 μg mL−1, respectively. The critical evaluation of the proposed methods is performed by statistical analysis of the experimental data. The proposed methods are applied to determine Cr in steel, non-ferrous alloys, wastewater and mud filtrate and to the determination of W in steel. Received March 8, 1999.
