Bacitracin‐conjugated superparamagnetic iron oxide (Fe3O4) nanoparticles were prepared by click chemistry and their antibacterial activity was investigated. After functionalization with hydrophilic and biocompatible poly(acrylic acid), water‐soluble Fe3O4 nanoparticles were obtained. Propargylated Fe3O4 nanoparticles were then synthesized by carbodiimide reaction of propargylamine with the carboxyl groups on the surface of the iron oxide nanoparticles. By further reaction with N3‐bacitracin in a CuI‐catalyzed azide–alkyne cycloaddition, the magnetic Fe3O4 nanoparticles were modified with the peptide bacitracin. The functionalized magnetic nanoparticles were characterized by powder X‐ray diffraction, X‐ray photoelectron spectroscopy, TEM, zeta‐potential analysis, FTIR spectroscopy and vibrating‐sample magnetometry. Cell cytotoxicity tests indicate that bacitracin‐conjugated Fe3O4 nanoparticles show very low cytotoxicity to human fibroblast cells, even at relatively high concentrations. In view of the antibacterial activity of bacitracin, the biofunctionalized Fe3O4 nanoparticles exhibit an antibacterial effect against both Gram‐positive and Gram‐negative organisms, which is even higher than that of bacitracin itself. The enhanced antibacterial activity of the magnetic nanocomposites allows the dosage and the side effects of the antibiotic to be reduced. Due to the antibacterial effect and magnetism, the bacitracin‐functionalized magnetic nanoparticles have potential application in magnetic‐targeting biomedical applications.  相似文献

The development of magnetic nanoparticles (MNPs) with functional groups has been intensively pursued in recent years. Herein, a simple, versatile, and cost‐effective strategy to synthesize water‐soluble and amino‐functionalized MNPs, based on the thermal decomposition of phthalimide‐protected metal–organic precursors followed by deprotection, was developed. The resulting amino‐functionalized Fe3O4, MnFe2O4, and Mn3O4 MNPs with particle sizes of about 14.3, 7.5, and 6.6 nm, respectively, had narrow size distributions and good dispersibility in water. These MNPs also exhibited high magnetism and relaxivities of r2=107.25 mM?1 s?1 for Fe3O4, r2=245.75 mM?1 s?1 for MnFe2O4, and r1=2.74 mM?1 s?1 for Mn3O4. The amino‐functionalized MNPs were further conjugated with a fluorescent dye (rhodamine B) and a targeting ligand (folic acid: FA) and used as multifunctional probes. Magnetic resonance imaging and flow‐cytometric studies showed that these probes could specifically target cancer cells overexpressing FA receptors. This new protocol opens a new way for the synthesis and design of water‐soluble and amino‐functionalized MNPs by an easy and versatile route.  相似文献

A novel dispersive liquid–liquid microextraction method based on amine‐functionalized Fe3O4 magnetic nanoparticles was developed for the determination of six phenolic acids in vegetable oils by high‐performance liquid chromatography. Amine‐functionalized Fe3O4 was synthesized by a one‐pot solvothermal reaction between Fe3O4 and 1,6‐hexanediamine and characterized by transmission electron microscopy and Fourier transform infrared spectrophotometry. A trace amount of phosphate buffer solution (extractant) was adsorbed on bare Fe3O4‐NH2 nanoparticles by hydrophilic interaction to form the “magnetic extractant”. Rapid extraction could be achieved while the “magnetic extractant” on amine‐functionalized Fe3O4 nanoparticles was dispersed in the sample solution by vortexing. After extraction, the “magnetic extractant” was collected by application of an external magnet. Some important parameters, such as pH and volume of extraction and desorption solvents, the extraction and desorption time needed were carefully investigated and optimized to achieve the best extraction efficiency. Under the optimal conditions, satisfactory extraction recoveries were obtained for the six phenolic acids in the range of 84.2–106.3%. Relative standard deviations for intra‐ and inter‐day precisions were less than 6.3 and 10.0%, respectively. Finally, the established method was successfully applied for the determination of six phenolic acids in eight kinds of vegetable oils.  相似文献

Fe3O4 nanoparticles were indirectly implanted onto functionalized multi‐walled carbon nanotubes (MWCNTs) leading to a nanocomposite with stronger magnetic performance. Poly(acrylic acid) (PAA) oligomer was first reacted with hydroxyl‐functionalized MWCNTs (MWCNTs‐OH) forming PAA‐grafted MWCNTs (PAA‐g‐MWCNTs). Subsequently, Fe3O4 nanoparticles were attached onto the surface of PAA‐g‐MWCNTs through an amidation reaction between the amino groups on the surface of Fe3O4 nanoparticles and the carboxyl groups of PAA. Fourier transform infrared spectra confirmed that the Fe3O4 nanoparticles and PAA‐g‐MWCNTs were indeed chemically linked. The morphology of the nanocomposites was characterized using transmission electron microscope (TEM). The surface and bulk structure of the nanocomposites were examined using X‐ray diffraction, X‐ray photoelectron spectrometer (XPS), and thermogravimetric analysis (TGA). The magnetic performance was characterized by vibrating sample magnetometer (VSM) and the magnetic saturation value of the magnetic nanocomposites was 47 emu g?1. The resulting products could be separated from deionized water under an external magnetic field within about 15 s. Finally, the magnetorheological (MR) performances of the synthesized magnetic nanocomposites and pure Fe3O4 nanoparticles were examined using a rotational rheometer. © 2010 Wiley Periodicals, Inc. Au‐Fe3O4 nanoparticles were widely used as nanoplatforms for biologic applications through readily further functionalization. Dopamine (DA)‐coated superparamagnetic iron oxide (SPIO) nanoparticles (DA@Fe3O4) have been successfully synthesized using a one‐step process by modified coprecipitation method. Then 2–3 nm gold nanoparticles were easily conjugated to DA@Fe3O4 nanoparticles by the electrostatic force between gold nanoparticles and amino groups of dopamine to afford water‐soluble Au‐Fe3O4 hybrid nanoparticles. A detailed investigation by dynamic light scatting (DLS), transmission electron microscopy (TEM), fourier transform infrared (FT‐IR) and X‐ray diffraction (XRD) were performed in order to characterize the physicochemical properties of the hybrid nanoparticles. The hybrid nanoparticles were easily functionalized with a targeted small peptide A54 (AGKGTPSLETTP) and fluorescence probe fluorescein isothiocyanate (FITC) for liver cancer cell BEL‐7402 imaging. This simple approach to prepare hybrid nanoparticles provides a facile nanoplatform for muti‐functional derivations and may be extended to the immobilization of other metals or bimolecular on SPIO surface.  相似文献

Micron‐sized monodisperse superparamagnetic polyglycidyl methacrylate (PGMA) particles with functional amino groups were prepared by a process involving: (1) preparation of parent monodisperse PGMA particles by the dispersion polymerization method, (2) chemical modification of the PGMA particles with ethylenediamine (EDA) to yield amino groups, and (3) impregnation of iron ions (Fe2+ and Fe3+) inside the particles and subsequently precipitating them with ammonium hydroxide to form magnetite (Fe3O4) nanoparticles within the polymer particles. The resultant magnetic PGMA particles with amino groups were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), X‐ray diffractometry (XRD), and vibrating sample magnetometry (VSM). SEM showed that the magnetic particles had an average size of 2.6 μm and were highly monodisperse. TEM demonstrated that the magnetite nanoparticles distributed evenly within the polymer particles. The existence of amino groups in the magnetic polymer particles was confirmed by FTIR. XRD indicated that the magnetic nanoparticles within the polymer were pure Fe3O4 with a spinel structure. VSM results showed that the magnetic polymer particles were superparamagnetic, and saturation magnetization was found to be 16.3 emu/g. The Fe3O4 content of the magnetic particles was 24.3% based on total weight. © 2005 Wiley Periodicals, Inc. J Polym Sci Part A: Polym Chem 43: 3433–3439, 2005  相似文献

An amino‐functionalized silica‐coated Fe3O4 nanocomposite (Fe3O4@SiO2/APTS) was synthesized. The Fe3O4@SiO2 microspheres possessed a well‐defined core–shell structure, uniform sizes and high magnetization. An immobilized ruthenium nanoparticle catalyst (Fe3O4@SiO2/APTS/Ru) was obtained after coordination and reduction of Ru3+ on the Fe3O4@SiO2/APTS nanocomposite. The Ru nanoparticles were not only ultra‐small with nearly monodisperse sizes but also had strong affinity with the surface of Fe3O4@SiO2/APTS. The obtained catalyst exhibited excellent catalytic performance for the hydrogenation of a variety of aromatic nitro compounds, even at room temperature. Moreover, Fe3O4@SiO2/APTS/Ru was easily recovered using a magnetic field and directly reused for at least five cycles without significant loss of its activity.  相似文献

A reversible addition‐fragmentation chain transfer (RAFT) agent was directly anchored onto Fe3O4 nanoparticles in a simple procedure using a ligand exchange reaction of S‐1‐dodecyl‐S′‐(α,α′‐dimethyl‐α″‐acetic acid)trithiocarbonate with oleic acid initially present on the surface of pristine Fe3O4 nanoparticles. The RAFT agent‐functionalized Fe3O4 nanoparticles were then used for the surface‐initiated RAFT copolymerization of N‐isopropylacrylamide and acrolein to fabricate structurally well‐defined hybrid nanoparticles with reactive and thermoresponsive poly(N‐isopropylacrylamide‐co‐acrolein) shell and magnetic Fe3O4 core. Evidence of a well‐controlled surface‐initiated RAFT copolymerization was gained from a linear increase of number‐average molecular weight with overall monomer conversions and relatively narrow molecular weight distributions of the copolymers grown from the nanoparticles. The resulting novel magnetic, reactive, and thermoresponsive core‐shell nanoparticles exhibited temperature‐trigged magnetic separation behavior and high ability to immobilize model protein BSA. © 2009 Wiley Periodicals, Inc. Thermal decomposition, as the main synthetic procedure for the synthesis of magnetic nanoparticles (NPs), is facing several problems, such as high reaction temperatures and time consumption. An improved a microwave‐assisted thermal decomposition procedure has been developed by which monodisperse Fe3O4 NPs could be rapidly produced at a low aging temperature with high yield (90.1 %). The as‐synthesized NPs show excellent inductive heating and MRI properties in vitro. In contrast, Fe3O4 NPs synthesized by classical thermal decomposition were obtained in very low yield (20.3 %) with an overall poor quality. It was found for the first time that, besides precursors and solvents, magnetic NPs themselves could be heated by microwave irradiation during the synthetic process. These findings were demonstrated by a series of microwave‐heating experiments, Raman spectroscopy and vector‐network analysis, indicating that the initially formed magnetic Fe3O4 particles were able to transform microwave energy into heat directly and, thus, contribute to the nanoparticle growth.  相似文献

In this study, Fe3O4@TiO2 nanoparticles were synthesized as a new Positron Emission Tomography/Magnetic Resonance Imaging (PET/MRI) hybrid imaging agent and radiolabeled with 89Zr. In addition, Fe3O4 nanoparticles were synthesized and radiolabeled with 89Zr. Df-Bz-NCS was used as bifunctional ligand. The nanoconjugates were characterized with transmission electron microscopy, scanning electron microscopy, and dynamic light scattering. Radiolabeling yields were 100%. Breast and prostate cancer cell affinities and cytotoxicity were determined using in vitro cell culture assays. The results demonstrated that Fe3O4@TiO2 nanoparticles are promising for PET/MR imaging. Finally, unlike Fe3O4 nanoparticles, Fe3O4@TiO2 nanoparticles showed a fluorescence spectrum at an excitation wavelength of 250 nm and an emission wavelength of 314 nm. Therefore, in addition to bearing the magnetic properties of Fe3O4 nanoparticles, Fe3O4@TiO2 nanoparticles display fluorescence emission. This provides them with photodynamic therapy potential. Therefore multimodal treatment was performed with the combination of PDT and RT by using human prostate cancer cell line (PC3). The development of 89Zr-Df-Bz-NCS-Fe3O4@TiO2 nanoparticles as a new multifunctional PET/MRI agent with photodynamic therapy and hyperthermia therapeutic ability would be very useful.  相似文献

Novel ionic‐liquid‐functionalized Fe3O4 magnetic nanoparticles were synthesized by the thiol‐ene click reaction. The prepared functionalized Fe3O4 nanoparticles possessed multiple interactions, such as electrostatic, hydrophobic, and π–π interactions. The functionalized Fe3O4 nanoparticles were characterized by using Fourier transform infrared spectroscopy, X‐ray diffraction, vibrating sample magnetometry, and transmission electron microscopy. Four kinds of linear alkylbenzene sulfonates, namely, sodium decylbenzenesulfonate, sodium undecylbenzene sulfonate, sodium dodecylbenzenesulfonate, and sodium tridecylbenzenesulfonate, were selected as model compounds to evaluate the applicability of adsorbents for extraction and subjected to high‐performance liquid chromatography analysis. In addition, the effects of various parameters, such as sorbent amount, pH value, ionic strength, sample volume, extraction time, and elution conditions on extraction efficiency were studied in detail. Under the optimum conditions, good linearities were attained, with correlation coefficients between 0.9912 and 0.9968. The proposed method exhibited limits of detection ranging from 0.061 to 0.099 μg/L for all the target analytes. The spiked recoveries of the target analytes in real water samples ranged from 86.3 to 107.5%, with relative standard deviations lower than 7.96%. The enrichment factors of the analytes ranged from 364 to 391, indicating that the obtained functionalized Fe3O4 nanoparticles can effectively extract trace target analytes from environmental water samples.  相似文献

For the first time, a magnetic solid‐phase extraction with high‐performance liquid chromatography detection method using Zr functionalized Fe3O4 magnetic material to enrich ribavirin was successfully established. Zr components that modified in Fe3O4 nanoparticles via a simple one‐step hydrothermal method was selected in this work to specifically capture ribavirin by the strong chemical bonding between Zr components of Zr functionalized Fe3O4 magnetic material and cis‐hydroxyl of ribavirin, which was confirmed by pseudo‐second‐order kinetic model. And Fe3O4 components were selected in this work to achieve simple operation. Under the optimal experimental conditions, proposed magnetic solid‐phase extraction with high‐performance liquid chromatography detection method along with Zr functionalized Fe3O4 magnetic material offered a wide range linearity at 10–200 µg/L with correlation coefficient of 0.9978 with low detection limit of 2.68 µg/L for ribavirin. The relative standard deviations obtained from nine parallel extractions of 100 µg/L ribavirin were 4.41% and revealed good repeatability. This established method was successfully applied to detect real samples including chicken liver, egg, and shrimp with satisfactory recoveries of 74.13–92.9%.  相似文献

We report a facile fabrication of a host–metal–guest coordination‐bonding system in a mesostructured Fe3O4/chitosan nanoparticle that can act as a pH‐responsive drug‐delivery system. The mesostructured Fe3O4/chitosan was synthesized by a solvothermal approach with iron(III) chloride hexahydrate as a precursor, ethylene glycol as a reducing agent, ammonium acetate as a porogen, and chitosan as a surface‐modification agent. Subsequently, doxorubicin (DOX), acting as a model drug (guest), was loaded onto the mesostructured Fe3O4/chitosan nanoparticles, with chitosan acting as a host molecule to form the NH2? ZnII? DOX coordination architecture. The release of DOX can be achieved through the cleavage of coordination bonds that are sensitive to variations in external pH under weakly acidic conditions. The pH‐responsive nature of the nanoparticles was confirmed by in vitro releases and cell assay tests. Furthermore, the relaxation efficiency of the nanoparticles as high‐performance magnetic resonance imaging contrast agents was also investigated. Experimental results confirm that the synthesized mesostructured Fe3O4/chitosan is a smart nanovehicle for drug delivery owing to both its pH‐responsive nature and relaxation efficiency.  相似文献

A novel electrochemical immunosensor based on a magnetic glassy carbon electrode (MGCE) was developed for the quantitative determination of human immunoglobulin G (IgG). The immunosensing interface was fabricated by initially depositing silver nanoparticles on the MGCE surface and then immobilizing anti‐human IgG antibodies via the magnetic force between MGCE and Fe3O4 nanoparticles. The antibodies were covalently bonded to the amine‐functionalized Fe3O4 nanoparticles. Under optimal conditions, the magnetism‐assisted immunosensor exhibited a wide linear range from 0.1 pg/mL to 1.0 µg/mL with the detection limit of 0.05 pg/mL. Furthermore, the immunosensor displayed the advantages of good reproducibility and satisfactory stability.  相似文献

A renewable potentiometric immunosensor for detection of immunoglobulin G (IgG) has been developed by magnetic force attraction of Fe3O4 nanoparticles immobilized goat‐anti‐human IgG antibody. For preparing sensitive film of the sensor, cysteine was bonded on the nano‐Fe3O4 particles surface. The cysteine functionalized magnetic nanoparticles was attracted on a solid paraffin carbon paste electrode surface to covalently immobilize of anti‐immunoglobulin G (anti‐IgG) by employing a conventional glutaraldehyde‐crosslinking method. The immunosensor showed a specific response to human immunoglobulin G in the range of 0.1–1.2 ng/mL with a detection limit of 0.023 ng/mL. The immunosensor based on the magnetic nanoparticles was made easily by this method. It can be used expediently, renewed easily and low‐cost relatively. The renewable potentiometric immunosensor with better stability and higher sensitivity can be employed extensively in clinical diagnosis, monitoring of disease and environmental studies and etc.  相似文献

Surface functionalization of magnetic nanoparticles is an elegant way to bridge the gap between heterogeneous and homogeneous catalysis. We have conveniently loaded sulfonic acid groups on amino‐functionalized Fe3O4 nanoparticles affording sulfamic acid‐functionalized magnetic Fe3O4 nanoparticles (MNPs/DAG‐SO3H) as an active and stable magnetically separable acidic nanocatalyst, which was characterized using X‐ray diffraction, Fourier transform infrared and energy‐dispersive X‐ray spectroscopies, scanning and transmission electron microscopies, vibrating sample magnetometry and elemental analysis. The catalytic activity of MNPs/DAG‐SO3H was probed through one‐pot synthesis of N‐substituted pyrroles from γ‐diketones and primary amines in aqueous phase at room temperature. The heterogeneous catalyst could be recovered easily by applying an external magnet device and reused many times without significant loss of its catalytic activity. This study describes the synthesis and characterization of ethylenediaminetetraacetic acid (EDTA) functionalized magnetic nanoparticles of 20 nm in size – Fe3O4@SiO2‐EDTA – which were used as a novel magnetic adsorbent for Cd(II) and Pb(II) binding in aqueous medium. These nanoparticles were obtained in two‐stage synthesis: covering by tetraethyl orthosilicate and functionalization with EDTA derivatives. Nanoparticles were characterized using TEM, FT‐IR, and XPS methods. Metal ions were detected under optimized experimental conditions using Differential Pulse Anodic Stripping Voltammetry (DPASV) and Hanging Mercury Drop Electrode (HDME) techniques. We compared the ability of Fe3O4@SiO2‐EDTA to bind cadmium and lead in concentration of 553.9 μg L?1 and 647.5 μg L?1, respectively. Obtained results show that the adsorption rate of cadmium binding was very high. The equilibrium for Fe3O4@SiO2‐EDTA‐Cd(II) was reached within 19 min while for the Fe3O4@SiO2‐EDTA‐Pb(II) was reached within 25 minutes. About 2 mg of nanoparticles was enough to bind 87.5 % Cd(II) and 54.1 % Pb(II) content. In the next step the binding capacity of Fe3O4@SiO2‐EDTA nanoparticles was determined. Only 1.265 mg of Fe3O4@SiO2‐EDTA was enough to bind 96.14 % cadmium ions while 5.080 mg of nanoparticles bound 40.83 % lead ions. This phenomenon proves that the studied nanoparticles bind Cd(II) much better than Pb(II). The cadmium ions binding capacity of Fe3O4@SiO2‐EDTA nanoparticles decreased during storage in 0.5 M KCl solution. Two days of Fe3O4@SiO2‐EDTA storage in KCl solution caused the 32 % increase in the amount of nanoparticles required to bind 60 % of cadmium while eight‐days storage caused further increase to 328 %. The performed experiment confirmed that the storage of nanoparticles in solution without any surfactants reduced their binding capacity. The best binding capacity was observed for the nanoparticles prepared directly before the electrochemical measurements.  相似文献

In this study, a novel multifunctional poly(arylene ether nitriles)(PEN)/carbon nanotubes/Fe3O4 nanocomposite with high tensile strength, magnetic, and electrical properties was investigated. First, we synthesized the monodisperse Fe3O4 nanoparticles on the surface of the multiwalled carbon nanotubes and then the hybrid material was compounded with PEN through the solution‐casting method. The SEM and TEM images indicated that the monodisperse Fe3O4 nanoparticles, with the diameters of 70∼80 nm, were self‐assembled along CNTs via the covalent bond method, which was confirmed by FTIR and XRD. The results of tensile properties showed that the tensile strength and modulus reached their highest values at the CNTs/Fe3O4 loading content of 1 wt % and both were greatly enhanced after heat treatment. Electrical conductivity of the polymer was dramatically enhanced at the low loading level of CNTs/Fe3O4; the electrical percolation of was in the range of 5∼8 wt % of CNTs/Fe3O4. The magnetic study showed that the saturation magnetization (Ms) of PEN/CNTs/Fe3O4 nanocomposites increased with the increase of CNTs/Fe3O4 loading content, and the coercive force (Hc) of the nanocomposite was independent of the CNTs/Fe3O4 content. © 2011 Wiley Periodicals, Inc. The extraction adsorbent was fabricated by immobilizing the highly specific recognition and binding of aptamer onto the surface of Fe3O4 magnetic nanoparticles, which not only acted as recognition elements to recognize and capture the target molecule berberine from the extract of Cortex phellodendri , but also could favor the rapid separation and purification of the bound berberine by using an external magnet. The developed solid‐phase extraction method in this work was useful for the selective extraction and determination of berberine in Cortex phellodendri extracts. Various conditions such as the amount of aptamer‐functionalized Fe3O4 magnetic nanoparticles, extraction time, temperature, pH value, Mg2+ concentration, elution time and solvent were optimized for the solid‐phase extraction of berberine. Under optimal conditions, the purity of berberine extracted from Cortex phellodendri was as high as 98.7% compared with that of 4.85% in the extract, indicating that aptamer‐functionalized Fe3O4 magnetic nanoparticles‐based solid‐phase extraction method was very effective for berberine enrichment and separation from a complex herb extract. The applicability and reliability of the developed solid‐phase extraction method were demonstrated by separating berberine from nine different concentrations of one Cortex phellodendri extract. The relative recoveries of the spiked solutions of all the samples were between 95.4 and 111.3%, with relative standard deviations ranging between 0.57 and 1.85%.  相似文献
