Reaction of diazonium salts with solutions of 1,2‐diaminocyclohexane mixed with formaldehyde affords the 1‐[2‐aryl‐1‐diazenyl]‐3‐({3‐[2‐aryl‐1‐diazenyl]perhydrobenzo[d]imidazol‐1‐yl}methyl)perhydrobenzo‐[d]imidazoles ( 6 ), a new series of bis‐triazenes with different connectivity than any previous type of bis‐triazene reported. The products have been characterized principally by NMR and IR spectroscopy, elemental analysis and unequivocally by X‐ray crystallography. The methylene protons of the perhydroimidazole rings are diastereotopic giving rise to a doublet of doublets pattern in the 1H NMR spectra. However, detailed analysis of the NMR spectra shows that there is more than one set of doublet‐of‐doublet signals, suggesting the presence of different rotameric forms of the products. The 13C NMR spectral assignments were assisted by COSY and DEPT experiments with selected compounds.  相似文献

1,2,3‐Triazole has become one of the most important heterocycles in contemporary medicinal chemistry. The development of the copper‐catalyzed Huisgen cycloaddition has allowed the efficient synthesis of 1‐substituted 1,2,3‐triazoles. However, only a few methods are available for the selective preparation of 2‐substituted 1,2,3‐triazole isomers. In this context, we decided to develop an efficient flow synthesis for the preparation of various 2‐aryl‐1,2,3‐triazoles. Our strategy involves a three‐step synthesis under continuous‐flow conditions that starts from the diazotization of anilines and subsequent reaction with malononitrile, followed by nucleophilic addition of amines, and finally employs a catalytic copper(II) cyclization. Potential safety hazards associated with the formation of reactive diazonium species have been addressed by inline quenching. The use of flow equipment allows reliable scale up processes with precise control of the reaction conditions. Synthesis of 2‐substituted 1,2,3‐triazoles has been achieved in good yields with excellent selectivities, thus providing a wide range of 1,2,3‐triazoles.  相似文献

Starting from pyrimido[4,5‐e][1,3,4]oxadiazines ( 3a , 3b , 3c ) , a synthetic pathway to [1,2,4]triazolo[4′,3′:1,2]pyrimido[4,5‐e][1,3,4]oxadiazines ( 5a , 5b , 5c , 5d , 5e , 5f , 5g , 5h , 5i ) is described. The reaction of pyrimido[4,5‐e][1,3,4]oxadiazines ( 3a , 3b , 3c ) with hydrazine hydrate afforded the corresponding hydrazino derivatives ( 4a , 4b , 4c ) . Further treatment of these compounds with different orthoesters in acetic acid gave the corresponding [1,2,4]triazolo[4′,3′:1,2]pyrimido[4,5‐e][1,3,4]oxadiazines ( 5a , 5b , 5c , 5d , 5e , 5f , 5g , 5h , 5i ) . Compound ( 3a ) and ( 5b ) , as examples, were tested on different cancer cell lines including HeLa, MCF‐7, and HepG2. Malignant cells were cultured in DMEM medium and incubated with different concentrations of the titled compounds. Cell viability was quantitated by MTT assay. J. Heterocyclic Chem., (2010).  相似文献

An efficient one‐pot synthesis of 3‐[(4,5‐dihydro‐1H‐pyrrol‐3‐yl)carbonyl]‐2H‐chromen‐2‐one (=3‐[(4,5‐dihydro‐1H‐pyrrol‐3yl)carbonyl]‐2H‐1‐benzopyran‐2‐one) derivatives 4 by a four‐component reaction of a salicylaldehyde 1 , 4‐hydroxy‐6‐methyl‐2H‐pyran‐2‐one, a benzylamine 2 , and a diaroylacetylene (=1,4‐diarylbut‐2‐yne‐1,4‐dione) 3 in EtOH is reported. This new protocol has the advantages of high yields (Table), and convenient operation. The structures of these coumarin (=2H‐1‐benzopyran‐2‐one) derivatives, which are important compounds in organic chemistry, were confirmed spectroscopically (IR, 1H‐ and 13C‐NMR, and EI‐MS) and by elemental analyses. In this study phenylselenocyanate and some of its derivatives (o‐Cl, p‐Cl, p‐Br, o‐NO2, p‐NO2, o‐CH3, p‐CH3, o‐COOH, p‐COOH, p‐OCH3 substituted) were synthesized ( 3a–3j ). The synthesized compounds were converted to 5‐aryl‐1H‐tetrazole ( 4a–4j ), by Et3N ċ HCl‐NaN3 in toluene, which are a new series of phenylselanyl‐1H‐tetrazoles. A series of substituted N‐(4‐substituted‐benzoyl)‐N‐[3‐(1‐methyl‐1H‐imidazol‐2‐yl)propyl]amines ( 13 ) and N‐arylsulfonyl‐N‐[3‐(1‐methyl‐1H‐imidazol‐2‐yl)propyl]amines ( 14 ) were prepared from the reaction of 3‐(1‐methyl‐1H‐imidazol‐2‐yl)propan‐1‐amine ( 7 ) with substituted benzoyl chloride or substituted‐benzene sulfonyl chloride respectively. By means of regioselective S‐alkylation of 1H‐1,2,4‐triazole‐3‐thiol ( 1 ), a series of S‐substituted derivatives 2a‐j were synthesized. In certain conditions, the reaction of 2 with arylsulfochlorides, arylisocyanates, and quaternary ammonium salts of azines corresponding compounds were obtained 1‐arylsulfonyl‐ (3a‐d) , 1‐arylcarbonamido‐ ( 4a,b ), and 1‐azinyl‐1,2,4‐ ( 6a‐p ) triazoles. Starting from 1H‐pyrazol, a wide number of 1‐alkyl‐1H‐pyrazol‐4‐yl and 1‐alkyl‐1H‐pyrazol‐5‐ylboronic acids and their pinacol esters were synthesized and characterized. The key step in the described methodology is the regioselective lithiation of the pyrazole ring. The synthesized pinacolates are stable under prolonged storage and can be used as convenient reagents in organic synthesis.  相似文献

The title compound, C19H14N5+·ClO4?·H2O, contains planar C19H14N5+ cations, perchlorate anions and water mol­ecules. The two closest parallel cations (plane‐to‐plane distance of 3.41 Å), together with two neighbouring perchlorate anions and two water mol­ecules, form an electrically neutral quasi‐dimeric unit. Two acidic H atoms of the cation, both H atoms of the water mol­ecule, the N atoms of the imidazole rings and three of the four O atoms of the perchlorate anion are involved in the hydrogen‐bonding network within the dimeric unit. The remaining third acidic H atom of the imidazole rings and the water mol­ecules complete a two‐dimensional network of hydrogen bonds, thus forming puckered layers of dimers. The angle between the planes of two neighbouring dimeric units in the same layer is 33.25 (3)°.  相似文献

A simple and versatile methodology to synthesise 4‐hydroxy‐1H‐[1,2,4]triazino[4,5‐a]quinoline‐1,6(2H)‐dione from methyl 6‐fluoro‐4‐oxo‐1,4‐dihydro‐2‐quinolinecarboxylate has been developed. It involves car‐bohydrazide formation followed by a condensation with triphosgene to form the fused [1,2,4]triazino ring. A novel facile photoconversion of 4‐hydroxy‐1,2‐bezothiazine 1,1‐dioxides ( 3a‐e ) into 4‐oxo‐1,3‐2H‐benzothiazine 1,1‐dioxides ( 4a‐e ) and 4‐hydroxy‐2‐methyl‐N‐(pyridin‐2‐yl)‐2H‐1,2‐benzothiazine‐3‐carboxamide 1,1‐dioxide (PRX) into N‐methyl saccharin ( 2 ) upon 254 nm irradiation in methanol or acetonitrile is reported. A series of 50 novel 7‐[2‐hydroxy‐3‐(1,2,3‐triazol‐1‐yl)propyloxy]‐3‐alkyl‐4‐methylcoumarins had been designed and synthesized in good to excellent yields via Cu(I)‐catalyzed 1,3‐dipolar cycloaddition reaction “click chemistry” of 7‐(3‐azido‐2‐hydroxypropyloxy)‐3‐alkyl‐4‐methylcoumarins with variety of acetylene derivatives. In turn, the precursor compound, that is, 7‐(3‐azido‐2‐hydroxypropyloxy)‐3‐alkyl‐4‐methylcoumarin, was synthesized by condensation of epichlorohydrin with 7‐hydroxy‐3‐alkyl‐4‐methylcoumarins followed by opening of the epoxide ring in the resulted 7‐epoxymethoxy‐3‐alkyl‐4‐methylcoumarins with sodium azide. All the synthesized compounds were unambiguously identified on the basis of their spectral data analyses (IR, 1H‐NMR, 13C‐NMR spectra, and HRMS).  相似文献
