Camera‐based fluorescence correlation spectroscopy (FCS) approaches allow the measurement of thousands of contiguous points yielding excellent statistics and details of sample structure. Imaging total internal reflection FCS (ITIR‐FCS) provides these measurements on lipid membranes. Herein, we determine the influence of the point spread function (PSF) of the optical system, the laser power used, and the time resolution of the camera on the accuracy of diffusion coefficient and concentration measurements. We demonstrate that the PSF can be accurately determined by ITIR‐FCS and that the laser power and time resolution can be varied over a wide range with limited influence on the measurement of the diffusion coefficient whereas the concentration measurements are sensitive to changes in the measurement parameters. One advantage of ITIR‐FCS is that the measurement of the PSF has to be performed only once for a given optical setup, in contrast to confocal FCS in which calibrations have to be performed at least once per measurement day. Using optimized experimental conditions we provide diffusion coefficients for over ten different lipid membranes consisting of one, two and three constituents, measured in over 200000 individual correlation functions. Using software binning and thus the inherent advantage of ITIR‐FCS of providing multiple observation areas in a single measurement we test the FCS diffusion law and show how they can be complemented by the local information provided by the difference in cross‐correlation functions (ΔCCF). With the determination of the PSF by ITIR‐FCS and the optimization of measurement conditions ITIR‐FCS becomes a calibration‐free method. This allows us to provide measurements of absolute diffusion coefficients for bilayers with different compositions, which were stable over many different bilayer preparations over a time of at least one year, using a single PSF calibration.  相似文献

Fluorescence correlation spectroscopy (FCS) measurements are widely used for determination of diffusion coefficients of lipids and proteins in biological membranes. In recent years, several variants of FCS have been introduced. However, a comprehensive comparison of these methods on identical systems has so far been lacking. In addition, there exist no consistent values of already determined diffusion coefficients for well-known or widely used membrane systems. This study aims to contribute to a better comparability of FCS experiments on membranes by determining the absolute diffusion coefficient of the fluorescent lipid analog 1,1'-dioctadecyl-3,3,3',3'-tetramethylindodicarbocyanine (DiD) in giant unilamellar vesicles (GUVs) made of dioleoylphosphatidylcholine (DOPC), which can in future studies be used as a reference value. For this purpose, five FCS variants, employing different calibration methods, were compared. Potential error sources for each particular FCS method and strategies to avoid them are discussed. The obtained absolute diffusion coefficients for DiD in DOPC were in good agreement for all investigated FCS variants. An average diffusion coefficient of D = 10.0 ± 0.4 μm(2) s(-1) at 23.5 ± 1.5 °C was obtained. The independent confirmation with different methods indicates that this value can be safely used for calibration purposes. Moreover, the comparability of the methods also in the case of slow diffusion was verified by measuring diffusion coefficients of DiD in GUVs consisting of DOPC and cholesterol.  相似文献

The fluorescence recovery after photobleaching (FRAP) method and the fluorescence correlation spectroscopy (FCS) have been applied on suspensions of highly charged colloidal spheres with a small content of rod-shaped tobacco mosaic virus (TMV) particles. Since these methods only determine the self-diffusion coefficient of the fluorescently labeled species, D(S) of the rods and the spheres could independently be measured. The ionic strength of the dispersion medium has been varied to measure self-diffusion of rods and spheres in dependence on the degree of order of the matrix spheres. In contrast to FRAP, which allows the determination of the long-time self-diffusion coefficient D(S) (L), FCS measures self-diffusion on a shorter time scale. Thus a comparison of the results that were obtained by FCS and FRAP, in combination with Brownian Dynamics simulations, gives insight into the time dependence of the self-diffusion coefficient of an interacting colloidal system. As the mean interparticle distance of the matrix is of the same order of magnitude as the length of a TMV rod, the rotational motion is influenced by the assembly of spheres around a TMV particle. Since FCS is sensitive both to translational and rotational motion, whereas FRAP, which probes the diffusion at much larger length scales, is only sensitive to the translational motion of TMV, the comparison of diffusion coefficients measured employing FRAP and FCS can give some insights in the rotational diffusion: the experimental data indicate a slowing down of the rotational motion of a TMV rod with increasing structural order of the matrix spheres.  相似文献

We describe the three‐step synthesis of a new polymeric fluoride ion conductor based on the fully aromatic polymer polysulfone (PSU). In the first step, PSU is chloromethylated (CM‐PSU) using reagents (i.e., stannic chloride, paraformaldehyde, and trimethylchlorosilane) that are less toxic than those used in the standard procedure. In the second step, CM‐PSU reacts with a tertiary amine (trimethylamine or 1,4‐diazabicyclo[2.2.2]octane) to form quaternary ammonium groups fixed on the PSU backbone and mobile chloride counter‐anions. The chloride ions can, in a third step, be exchanged with fluoride ions by immersion of the ionomer in NaF solution. The fluoride ion conductivity reaches 3–5 mS cm?1 at 25 °C and 5–10 mS cm?1 at 40 °C. We determined the F? diffusion coefficient in these ionomers by pulsed gradient spin‐echo (PGSE) high‐resolution magic angle spinning (HRMAS) nuclear magnetic resonance (NMR) spectroscopy and by impedance spectroscopy using the Nernst–Einstein relation. The diffusion coefficients determined by the two methods are in good agreement, ranging from 2 to 4×10?10 m2 s?1. The porosity and tortuosity of the ionomer membranes can be estimated.  相似文献

In this work cellular bovine prion protein (PrPc) was incorporated in supported lipid membranes and its lateral diffusion was studied by single-dye tracking (SDT) and a complementary ensemble method, fluorescence recovery after photobleaching (FRAP). PrPc was purified from calf brain with its native glycosylphosphatidylinositol (GPI) anchor and reconstituted into DMPC lipid vesicles. Homogeneous spreading on solid supports over macroscopic areas was confirmed with fluorescence microscopy. FRAP results demonstrated very high mobile fractions of up to 94%, confirming that most of the GPI-anchored PrPc are freely diffusive in the fluid supported membrane matrix. Moreover, the lateral diffusivity of PrPc significantly depends on the pH of the buffer, suggesting that the conformation of PrPc and thus the frictional drag exerted to the protein molecule (and thus the effective hydrodynamic radius) is influenced by the effective net charge. To complement the ensemble results obtained by FRAP, the statistical variation of lateral diffusion coefficients of individual PrPc molecules in the supported membranes were measured with SDT. Simulation-based statistical analysis indicated that in addition to the expected statistical scatter there is a significant spread of diffusion coefficients, while the average of the diffusion coefficients of individual proteins obtained by SDT is in excellent agreement with those measured by ensemble FRAP. In further experiments, PrPc was laterally concentrated in the membrane by the application of tangential electric fields (membrane electrophoresis). However, the equilibrium concentration profile reached after 20 min was different from an exponential gradient. This finding suggests that PrPc purified from bovine brain possesses non-uniform net charges. As the lateral diffusion coefficient of proteins in two-dimensional lipid membranes sensitively depends upon the frictional drag, the combination of SDT, ensemble FRAP, and membrane electrophoresis can be used as a powerful tool to gain insights into protein–protein binding and oligomer formation that would play a crucial role in infectious protein transmitted diseases such as BSE.  相似文献

Fluorescence correlation spectroscopy (FCS) is a powerful technique for measuring diffusion coefficients of small fluorescent molecules at pico- to nanomolar concentrations. Recently, a modified version of FCS, dual-focus FCS (2fFCS), was introduced that significantly improves the reliability and accuracy of FCS measurements and allows for obtaining absolute values of diffusion coefficients without the need of referencing again a known standard. It was shown that 2fFCS gives excellent results for measuring the diffusion of small molecules. However, when measuring colloids or macromolecules, the size of these objects can no longer be neglected with respect to the excitation laser focus. Here, we analyze how 2fFCS data evaluation has to be modified for correctly taking into a count these finite size effects. We exemplify the new method of measuring the absolute size of polymeric particles with simple and complex fluorophore distributions.  相似文献

The diffusion coefficients of nine fluorescently labeled antibodies, antibody fragments, and antibody complexes have been measured in solution very close to supported planar membranes by using total internal reflection with fluorescence correlation spectroscopy (TIR-FCS). The hydrodynamic radii (3-24 nm) of the nine antibody types were determined by comparing literature values with bulk diffusion coefficients measured by spot FCS. The diffusion coefficients very near membranes decreased significantly with molecular size, and the size dependence was greater than that predicted to occur in bulk solution. The observation that membrane surfaces slow the local diffusion coefficient of proteins in a size-dependent manner suggests that the primary effect is hydrodynamic as predicted for simple spheres diffusing close to planar walls. The TIR-FCS data are consistent with predictions derived from hydrodynamic theory. This work illustrates one factor that could contribute to previously observed nonideal ligand-receptor kinetics at model and natural cell membranes.  相似文献

The self‐diffusion (Dc) coefficients of various lanthanum(III) diamagnetic analogues of open‐chain and macrocyclic complexes of gadolinium used as MRI contrast agents were determined in dilute aqueous solutions (3–31 mM ) by pulsed‐field‐gradient (PFG) high‐resolution 1H‐NMR spectroscopy. The self‐diffusion coefficient of H2O (Dw) was obtained for the same samples to derive the relative diffusion constant, a parameter involved in the outersphere paramagnetic‐relaxation mechanism. The results agree with an averaged relative diffusion constant of 2.5 (±0.1)×10?9 and of 3.3 (±0.1)×10?9 m2 s?1 at 25 and 37°, respectively, for 'small' contrast agents (Mr 500–750 g/mol), and with the value of bulk H2O (2.2×10?9 and 2.9×10?9 m2 s?1 at 25° and at 37°, respectively) for larger complexes. The use of the measured values of Dc for the theoretical fitting of proton NMRD curves of gadolinium complexes shows that the rotational correlation times (τR) are very close to those already reported. However, differences in the electronic relaxation time (τSO) at very low field and in the correlation time (τV) related to electronic relaxation were found.  相似文献

The complex shape and plasticity of cells is an intricate issue for the measurement of molecular diffusion in plasma membranes by fluorescence correlation spectroscopy (FCS). An important precondition for accurate diffusion measurements is a sufficient flatness of the membrane over the considered region and the absence of non‐membrane‐bound fluorescence diffusion. A method is presented to identify axial motion components caused by a non‐ideal geometry of the membrane based on simultaneous measurement of the fluorescence emitted above and below the critical angle of the specimen/glass interface. Thereby, two detection volumes are generated that are laterally coincident, but differ in their axial penetration of the specimen. The similarity between the intensity tracks of the supercritical angle fluorescence (SAF) and the undercritical angle fluorescence (UAF) strongly depends on the membrane flatness and intracellular fluorescence, and can help to avoid sample‐related artifacts in the diffusion measurement.  相似文献

New graphene oxide (GO)‐tethered–CoII phthalocyanine complex [CoPc–GO] was synthesized by a stepwise procedure and demonstrated to be an efficient, cost‐effective and recyclable photocatalyst for the reduction of carbon dioxide to produce methanol as the main product. The developed GO‐immobilized CoPc was characterized by X‐ray diffraction (XRD), FTIR, XPS, Raman, diffusion reflection UV/Vis spectroscopy, inductively coupled plasma atomic emission spectroscopy (ICP‐AES), thermogravimetric analysis (TGA), Brunauer–Emmett–Teller (BET), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). FTIR, XPS, Raman, UV/Vis and ICP‐AES along with elemental analysis data showed that CoII–Pc complex was successfully grafted on GO. The prepared catalyst was used for the photocatalytic reduction of carbon dioxide by using water as a solvent and triethylamine as the sacrificial donor. Methanol was obtained as the major reaction product along with the formation of minor amount of CO (0.82 %). It was found that GO‐grafted CoPc exhibited higher photocatalytic activity than homogeneous CoPc, as well as GO, and showed good recoverability without significant leaching during the reaction. Quantitative determination of methanol was done by GC flame‐ionization detector (FID), and verification of product was done by NMR spectroscopy. The yield of methanol after 48 h of reaction by using GO–CoPc catalyst in the presence of sacrificial donor triethylamine was found to be 3781.8881 μmol g?1 cat., and the conversion rate was found to be 78.7893 μmol g?1cat. h?1. After the photoreduction experiment, the catalyst was easily recovered by filtration and reused for the subsequent recycling experiment without significant change in the catalytic efficiency.  相似文献

Fluorescence correlation spectroscopy (FCS) is a powerful tool to measure useful physical quantities such as concentrations, diffusion coefficients, diffusion modes or binding parameters, both in model and cell membranes. However, it can suffer from severe artifacts, especially in non-ideal systems. Here we assess the potential and limitations of standard confocal FCS on lipid membranes and present recent developments which facilitate accurate and quantitative measurements on such systems. In particular, we discuss calibration-free diffusion and concentration measurements using z-scan FCS and two focus FCS and present several approaches using scanning FCS to accurately measure slow dynamics. We also show how surface confined FCS enables the study of membrane dynamics even in presence of a strong cytosolic background and how FCS with a variable detection area can reveal submicroscopic heterogeneities in cell membranes.  相似文献

Peroxynitrite, ONOO? also so‐called PON, is a very powerful oxidant and cytotoxic agent produced in biological systems by the recombination of nitric oxide and superoxide anion radical. Most of the techniques for assaying PON (chemiluminescence, fluorescence, UV‐visible spectroscopy, immunochemistry and EPR ) use indirect methods relying on measurements of secondary species. We report in this study the calibration of a chemically modified Pt ultramicroelectrode by electropolymerized manganese tetraaminophthalocyanine film (MnTAPc) for the determination of PON in aqueous solution. The obtained result allows showing for the first time a real‐time calibration curve of the amperometric determination of stable PON in aqueous solution. The sensitivity of the sensor is 14.6 nA mM?1 and its detection limit is 5 μM.  相似文献

An LC–MS/MS method was developed and validated for the simultaneous quantification of edaravone and taurine in beagle plasma. The plasma sample was deproteinized using acetonitrile containing formic acid. Chromatographic separations were achieved on an Agilent Zorbax SB‐Aq (100 × 2.1 mm, 3.5 μm) column, with a gradient of water (containing 0.03% formic acid) and methanol as the mobile phase at a flow rate of 0.3 mL/min. The analyte detection was carried out in multiple reaction monitoring mode and the optimized precursor‐to‐product transitions of m/z [M+H]+ 175.1 → 133.0 (edaravone), m/z [M+H]+ 189.1 → 147.0 (3‐methyl‐1‐p‐tolyl‐5‐pyrazolone, internal standard, IS), m/z [M–H]? 124.1→80.0 (taurine), and m/z [M–H]? 172.0 → 80.0 (sulfanilic acid, IS) were employed to quantify edaravone, taurine, and their corresponding ISs, respectively. The LOD and the lower LOQ were 0.01 and 0.05 μg/mL for edaravone and 0.66 and 2 μg/mL for taurine, respectively. The calibration curves of these two analytes demonstrated good linearity (r > 0.99). All the validation data including the specificity, precision, recovery, and stability conformed to the acceptable requirements. This validated method has successfully been applied in the pharmacokinetic study of edaravone and taurine mixture in beagle dogs.  相似文献

In soft matter, thermal energy causes molecules to continuously translate and rotate, even in crowded environments, thereby impacting the spatial organization and function of most molecular assemblies, such as lipid membranes. Directly measuring the orientation and spatial organization of large collections (>3000 molecules μm?2) of single molecules with nanoscale resolution remains elusive. In this paper, we utilize SMOLM, single‐molecule orientation localization microscopy, to directly measure the orientation spectra (3D orientation plus “wobble”) of lipophilic probes transiently bound to lipid membranes, revealing that Nile red's (NR) orientation spectra are extremely sensitive to membrane chemical composition. SMOLM images resolve nanodomains and enzyme‐induced compositional heterogeneity within membranes, where NR within liquid‐ordered vs. liquid‐disordered domains shows a ≈4° difference in polar angle and a ≈0.3π sr difference in wobble angle. As a new type of imaging spectroscopy, SMOLM exposes the organizational and functional dynamics of lipid‐lipid, lipid‐protein, and lipid‐dye interactions with single‐molecule, nanoscale resolution.  相似文献

NMR spectroscopy is a very important and useful method for the structural analysis of oligosaccharides, despite its low sensitivity. We first applied conventional measuring methods, 2D DQF COSY, 1H–13C HSQC, and 1H–13C HMBC, and also the Double Pulsed Field Gradient Spin Echo (DPFGSE)‐TOCSY and DPFGSE‐NOESY/ROESY techniques to analyze a branched mannose pentasaccharide as a model of high mannose type N‐glycans in natural abundance. The NMR spectra of the model compound are very complex and difficult to analyze owing to overlapping signals. The superior selective irradiation capability of the DPFGSE technique is useful for fine structural and conformational analyses of such complex oligosaccharides. We here introduce a novel technique called DPFGSE‐Double‐Selective Population Transfer (SPT)‐Difference and DPFGSE‐NOE/ROE‐SPT‐Difference spectroscopy. The DPFGSE‐Double‐SPT‐Difference method involves irradiation of two peaks from one proton and the subtraction of higher and lower peaks from each spectrum. The DPFGSE‐NOE/ROE‐SPT‐Difference method involves the transfer of the magnetization polarized by NOE/ROE from the nuclei to the spin‐coupled nuclei through scalar spin–spin interaction using the SPT method. Even if the signals in the NMR spectra overlap, each signal can be accurately assigned. In particular, DPFGSE‐NOE/ROE‐SPT‐Difference is very useful for identifying sugar connectivity. An electrochemical study of the anthelmintic drug bithionol using edge plane pyrolytic graphite electrode (EPPGE) is presented for the first time by applying different electrochemical techniques, such as cyclic voltammetry (CV), square‐wave voltammetry (SWV), square‐wave adsorptive stripping voltammetry (SWAdSV), and alternating current (AC) impedance spectroscopy. Mechanistic aspects of the electrode reaction were studied implying a quasireversible electrode reaction from an adsorbed state of the reactant, coupled with a follow‐up chemical reaction to a final electroinactive product. The overall mechanism appears totally irreversible under conditions of CV at moderate scan rate, while being quasireversible under conditions of the fast SWV. Furthermore, an optimisation of the analytical procedure for quantitative determination of bithionol was conducted by applying SWV in an adsorptive stripping mode. The calibration curve was constructed in the concentration range of 0.1–1.0 μmol L?1 (R2=0.9984) with a sensitivity of 3.6 μA L μmol?1 and LOD of 26.7 nmol L?1. The simple and sensitive SWAdSV procedure was proved to be suitable for the analysis of spiked urine samples.  相似文献

Branched sulfonated poly(ether ketone sulfone)s (Br‐SPEKS) were prepared with bisphenol A, bis(4‐fluorophenyl)sulfone, 3,3′‐disodiumsulfonyl‐4,4′‐difluorobenzophenone, and THPE (1,1,1‐tris‐p‐hydroxyphenylethane), respectively, at 180 °C using potassium carbonate in NMP (N‐methylpyrrolidinone). THPE, as a branching agent, was used with 0.4 mol % of bisphenol A to synthesize branched copolymers. Copolymers containing 10–50 mol % disulfonated units were cast from dimethylsulfoxide solutions to form films. Linear sulfonated poly(ether ketone sulfone)s (SPEKS) were also synthesized without THPE. The films were converted from the salt to acid forms with dilute hydrochloric acid. A series of copolymers were studied by Fourier transform infrared, 1H‐NMR spectroscopy, differential scanning calorimetry (DSC), and thermogravimetric analysis (TGA). Sorption experiments were conducted to observe the interaction of sulfonated polymers with water and methanol. The ion‐exchange capacity (IEC), a measure of proton conductivity, was evaluated. The synthesized Br‐SPEKS and SPEKS membranes exhibit conductivities (25 °C) from 1.04 × 10?3 to 4.32 × 10?3 S/cm, water swell from 20.18 to 62.35%, IEC from 0.24 to 0.83 mequiv/g, and methanol diffusion coefficients from 3.2 × 10?7 to 4.7 × 10?7 cm2/S at 25 °C. © 2008 Wiley Periodicals, Inc. We report the fabrication of a Ni nanoparticle modified BDD electrode and its application in the electrocatalysis of primary alcohol electrooxidation. Modification was achieved via electrodeposition from Ni(NO3)2 dissolved in sodium acetate solution (pH 5). Characterization of the Ni‐modified BDD (Ni‐BDD) was performed using ex situ atomic force microscopy (AFM) and high resolution scanning electron microscopy (SEM) coupled with energy dispersive X‐ray spectroscopy (EDX). Large nanoparticles of nickel were observed on the BDD surface ranging 5 to 690 nm in height and 0.18 μm?3 in volume, and an average number density of ca. 13×106 nanoparticles cm?2 was determined. The large range of sizes suggests progressive rather than instantaneous nucleation and growth. Electrocatalysis of ethanol and glycerol, was conducted in an alkaline medium using an unmodified BDD, Ni‐BDD and a bulk Ni macro electrode. The Ni‐BDD electrode gave the better electrocatalytic performance, with glycerol showing the greatest sensitivity. Linear calibration plots were obtained for the ethanol and glycerol additions over concentration ranges of 2.8–28.0 mM and 23–230 μM respectively. This gave an ethanol limit of detection of 1.7 mM and sensitivity of 0.31 mA/M, and the glycerol a limit of detection of 10.3 μM with a sensitivity of 35 mA/M.  相似文献

In this article a sensitive differential pulse stripping voltammetry technique on Nafion‐coated bismuth‐film electrode (NCBFE) was studied for the simultaneous determination of zinc, cadmium, and lead ions in blood samples at ultra trace levels. The measurement results were in excellent agreement with those obtained from atomic absorption spectroscopy. Various operational parameters were investigated and discussed in terms of their effect on the measurement signals. Under optimal conditions, calibration curves for the simultaneous determination of zinc, cadmium, and lead ions were achieved, based on three times the standard deviation of the baseline, the limits of detection were 0.09 μg L?1 for Cd(II), 0.13 μg L?1 for Pb(II), and 0.97 μg L?1 for Zn(II) respectively.  相似文献

A manual method for the determination of hydrogen chloride in air, based on diffusion/denuder tube separation from particulate chloride aerosol is described. When air is drawn through a tube coated with a selective absorbent (sodium fluoride), separation is achieved because gaseous hydrogen chloride diffuses much more rapidly to the tube walls than particulate chloride aerosol, which passes through virtually unabsorbed. After the sampling period (the length of which depends on the concentration of gaseous hydrogen chloride expected), the sorbed hydrogen chloride is washed from the tube and measured with a highly sensitive chloride ion-selective electrode with a mercury (I) chloride membrane. The method is examined theoretically and experimentally. The experimentally derived absorption efficiencies of the diffusion/denuder tubes were > 90% and the standard deviation of the method was 0.023 μg m?3 for hydrogen chloride concentrations of 0.16–0.55 μg m?3. Interference from particulate chloride salts was negligible; this was confirmed by tests with artificially generated aerosol particles from an aerosol generator. The diffusion/denuder tubes have high capacity; level as high as 330 μg m?3 hydrogen chloride can be sampled for 60 min without affecting performance. A detection limit of (50/t) μg m?3 can be achieved, where t is the sampling rime (min); e.g., 1μg m?3 hydrogen chloride can be detected with a sampling period of 50 min.  相似文献
