A sensitive and selective aptasensor using tri(2,2′-bipyridyl)ruthenium(II)-doped silica nanoparticles (Ru(bpy)32+-doped SNPs) as DNA tags for detection of thrombin is developed based on the target protein-induced strand displacement of the DNA probe. For the proposed aptasensor, the aptamer was assembled on the surface of the Au electrode through Au-S binding. The hybridization event between the DNA probe labeled by the Ru(bpy)32+-doped SNPs and the aptamer was evaluated by electrogenerated chemiluminescence (ECL) measurements. Then, the DNA probe was displaced by thrombin and the binding event between the thrombin and the aptamer was monitored by ECL measurements again. The difference of ECL intensity (ΔIECL) of the two events could be used to quantify the thrombin. Other proteins, such as bovine serum albumin and bovine hemoglobin, had almost negligible ΔIECL. Under the optimal conditions, the ΔIECL was linearly related to the concentration of the thrombin in the range of 10 fM to 10 pM and the detection limit was down to 1.0 fM since SNPs containing a large number of Ru(bpy)32+ molecules were labeled on the DNA probe.  相似文献

An ultrasensitive and signal‐on electrochemiluminescence (ECL) aptasensor to detect target protein (thrombin or lysozyme) was developed using the host‐guest recognition between a metallocyclodextrin complex and single‐stranded DNA (ss‐DNA). The aptasensor uses both the photoactive properties of the metallocyclodextrins named multi‐tris(bipyridine)ruthenium(II)‐β‐cyclodextrin complexes and their specific recognition with ss‐DNA, which amplified the ECL signal without luminophore labeling. After investigating the ECL performance of different multi‐tris(bipyridine)ruthenium(II)‐β‐cyclodextrin (multi‐Ru‐β‐CD) complexes, tris‐tris(bipyridine)‐ruthenium(II)‐β‐cyclodextrin (tris(bpyRu)‐β‐CD) was selected as a suitable host molecule to construct an atasensor. First, double‐stranded DNA (ds‐DNA) formed by hybridization of the aptamer and its target DNA was attached to a glassy carbon electrode via coupling interaction, which showed low ECL intensity with 2‐(dibutylamino) ethanol (DBAE) as coreactant, because of the weak recognition between ds‐DNA and tris(bpyRu)‐β‐CD. Upon addition of the corresponding protein, the ECL intensity increased when target ss‐DNA was released because of the higher stability of the aptamer‐protein complex than the aptamer‐DNA one. A linear relationship was observed in the range of 0.01 pmol/L to 100 pmol/L between ECL intensity and the logarithm of thrombin concentrations with a limited detection of 8.5 fmol/L (S/N=3). Meanwhile, the measured concentration of lysozyme was from 0.05 pmol/L to 500 pmol/L and the detection limit was 33 fmol/L (S/N=3). The investigations of proteins in human serum samples were also performed to demonstrate the validity of detection in real clinical samples. The simplicity, high sensitivity and specificity of this aptasensor show great promise for practical applications in protein monitoring and disease diagnosis.  相似文献

We developed an electrochemical thrombin aptasensor based on ZnO nanorods functionalized by electrostatically adsorption of 30‐mer DNA aptamers. The sensor surface was characterized by AFM and SEM. The surface layer assembling was optimized using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) with ferricyanide ions as redox markers. The peak current of the ferricyanide and the charge transfer resistance gradually decreased with increasing concentration of thrombin in the range from 3 pM to 100 nM due to formation of aptamer‐thrombin complexes and slower diffusion of the marker ions through the surface layer. At optimal conditions, a limit of detection (LOD) of 3 pM for EIS measurements and 10 pM for CV response was calculated from the S/N=3.  相似文献

A sensitive electrochemical aptasensor for detection of thrombin based on target protein‐induced strand displacement is presented. For this proposed aptasensor, dsDNA which was prepared by the hybridization reaction of the immobilized probe ssDNA (IP) containing thiol group and thrombin aptamer base sequence was initially immobilized on the Au electrode by self‐assembling via Au? S bind, and a single DNA labeled with CdS nanoparticles (DP‐CdS) was used as a detection probe. When the so prepared dsDNA modified Au electrode was immersed into a solution containing target protein and DP‐CdS, the aptamer in the dsDNA preferred to form G‐quarter structure with the present target protein resulting that the dsDNA sequence released one single strand and returned to IP strand which consequently hybridized with DP‐CdS. After dissolving the captured CdS particles from the electrode, a mercury‐film electrode was used for electrochemical detection of these Cd2+ ions which offered sensitive electrochemical signal transduction. The peak current of Cd2+ ions had a good linear relationship with the thrombin concentration in the range of 2.3×10?9–2.3×10?12 mol/L and the detection limit was 4.3×10?13 mol/L of thrombin. The detection was also specific for thrombin without being affected by the coexistence of other proteins, such as BSA and lysozyme.  相似文献

The photoinduced reaction of a mixture of (Z)‐α‐cyano‐β‐bromomethylcinnamide (1) and (E)‐α‐cyano‐β‐bromomethylcinnamide (2) with 1‐benzyl‐1, 4‐dihydronicotinamide produces a mixture of the (E)‐ and (Z)‐ isomers of α‐cyano‐β‐methylcinnamide (3 and 4). Using spin‐trapping technique for monitoring reactive intermediate, it is shown that the reaction proceeds via electron transfer‐debromination‐H abstraction mechanism. The thermal reaction of the same substrate with BNAH at 60°C in the dark gives three products: the (E)‐ and (Z)‐isomers of α‐cyano‐β‐methylcinnamide and a dehydrodimeric product; 2, 7‐dicyano‐3, 6‐diphenylocta‐2, 4, 6‐trien‐1, 8‐dioic amide (7). Based on product analysis, scavenger experiment and cyclic voltammetry, an electron transfer‐debromination‐disproportionation mechanism is proposed.  相似文献

A novel G‐quadruplex‐based DNAzymes aptasensor for the amplified electrochemical detection of thrombin has been described. The aptasensor utilized a combination of hemin and guanine‐rich thrombin‐binding aptamer (TBA) to form horseradish peroxidase (HRP)‐mimicking DNAzymes with peroxidase catalytic activity. In the presence of thrombin, the enzyme activity could be extensively promoted, thereby providing the amplified electrochemical readout signals for detecting thrombin. This aptasensor exhibited high sensitivity and selectivity for thrombin determination, which enabled the analysis of thrombin with a detection limit of 6×10–11 M. On the basis of results, this method could have broad applications in the detection of proteins and other biomolecules.  相似文献

The biomimic reactions of N‐phosphoryl amino acids, which involved intramolecular penta‐coordinate phosphoric‐carboxylic mixed anhydrides, are very important in the study of many biochemical processes. The reactivity difference between the α‐COOH group and β‐COOH in phosphoryl amino acids was studied by experiments and theoretical calculations. It was found that the α‐COOH group, and not β‐COOH, was involved in the ester exchange on phosphorus in experiment. From MNDO calculations, the energy of the penta‐coordinate phosphoric intermediate containing five‐member ring from α‐COOH was 35 kJ/mol lower than that of the six‐member one from β‐COOH. This result was in agreement with that predicted by HF/6‐31G** and B3LYP/6‐31G** calculations. Theoretical three‐dimensional potential energy surface for the intermediates predicted that the transition states 4 and 5 involving α‐COOH or β‐COOH group had energy barriers of ΔE=175.8 kJ?mol?1 and 210.4 kJ?mol?1, respectively. The effect of aptamer structure and immobilization platform on the efficiency of thrombin binding and its detection using electrochemical impedance spectroscopy (EIS) characteristics was investigated with aptasensors based on glassy carbon electrodes covered with multiwalled carbon nanotubes (MWNTs). Aptamers with one or two binding sequences GGTTGGTGTGGTTGG specific for thrombin and poly(dA) and poly(dT) tags able to form dimeric products (aptabodies) were used to establish significance of steric and electrostatic factors in aptasensor performance. We have shown that electropolymerization of methylene blue onto MWNTs significantly improved electrochemical characteristics and sensitivity of thrombin detection against bare MWNTs. Charge transfer resistance and capacitance of the surface layer were measured in the presence of redox probe [Fe(CN)6]3?/4?. Aptasensors make it possible to detect thrombin in the concentration range 1 nM–1 µM with the limit of detection of 0.7 nM (monitoring resistance changes) and 0.5 nM (capacitance changes), respectively.  相似文献

A novel fluorescence aptasensor based on DNA charge transport for sensitive protein detection has been developed. A 15nt DNA aptamer against thrombin was used as a model system. The aptamer was integrated into a double strand DNA (dsDNA) that was labeled with a hole injector, naphthalimide (NI), and a fluorophore, Alexa532, at its two ends. After irradiation by UV light, the fluorescence of Alexa532 was bleached due to the oxidization of Alexa532 by the positive charge transported from naphthalimide through the dsDNA. In the presence of thrombin, the binding of thrombin to the aptamer resulted in the unwinding of the dsDNA into ssDNA, which led to the blocking of charge transfer and the strong fluorescence emission of Alexa532. By monitoring the fluorescence signal change, we were able to detect thrombin in homogeneous solutions with high selectivity and high sensitivity down to 1.2 pM. Moreover, as DNA charge transfer is resistant to interferences from biological contexts, the aptasensor can be used directly in undiluted serum with similar sensitivity as that in buffer. This new sensing strategy is expected to promote the exploitation of aptamer-based biosensors for protein assays in complex biological matrixes.  相似文献

In this work, a dual-signaling electrochemical aptasensor based on exonuclease-catalyzed target recycling was developed for thrombin detection. The proposed aptasensor coupled “signal-on” and “signal-off” strategies. As to the construction of the aptasensor, ferrocene (Fc) labeled thrombin binding aptamer (TBA) could perfectly hybridize with the methylene blue (MB) modified thiolated capture DNA to form double-stranded structure, hence emerged two different electrochemical signals. In the presence of thrombin, TBA could form a G-quadruplex structure with thrombin, leading to the dissociation of TBA from the duplex DNA and capture DNA formed hairpin structure. Exonuclease could selectively digest single-stranded TBA in G-quadruplex structure and released thrombin to realize target recycling. As a consequence, the electrochemical signal of MB enhanced significantly, which realized “signal on” strategy, meanwhile, the deoxidization peak current of Fc decreased distinctly, which realized “signal off” strategy. The employment of exonuclease and superposition of two signals significantly improved the sensitivity of the aptasensor. In this way, an aptasensor with high sensitivity, good stability and selectivity for quantitative detection of thrombin was constructed, which exhibited a good linear range from 5 pM to 50 nM with a detection limit of 0.9 pM (defined as S/N = 3). In addition, this design strategy could be applied to the detection of other proteins and small molecules.  相似文献

Human serum is one of the effective samples for point-of-care testing (POCT). Sensitive and quick determination of thrombin content in human serum samples is important. An electrochemical aptasensor based on Prussian blue and Au nanoparticles loaded MoS2 nanoflowers (PB−Au@MoS2) hybrid was constructed. By using PB−Au@MoS2 as both a substrate and a signal reporter, this aptasensor could demonstrate excellent performance for thrombin detection with a detection linear range from 0.01 pM to 30 nM and detection limit down to 1 fM. This work may provide a strategy to establish effective and sensitive sensing devices for thrombin in clinical diagnosis.  相似文献

It has been demonstrated that potentiometric and polarographic data coming from either acid? base or ligand titrations can be evaluated by the same set of equations and mathematical procedures involving mass‐balance equations written for any metal? ligand model, including polynuclear species. It is shown that the concept of the complex formation curves, used previously in modeling and refinement of stability constants in acid‐base titration, is of general nature and can be successfully used in polarographic and potentiometric experiments conducted as a function of pH or an excess of a ligand. It appears that the linear relationship ΔE vs. log [M], typical for potentiometric studies with ISE, holds also in the case of the study of kinetically mixed metal‐ligand system studied by polarography when the corrected shift in the polarographic signal is used. The relationship ΔE vs. log [M] applies equally to the acid‐base and ligand titration for both experimental techniques employed (potentiometry and polarography). The significance of the corrected shift is discussed and its meaning in the study of kinetically fast or slow metal? ligand systems is elaborated. Advantages of the acid‐base titration over the ligand titration are discussed. The generalized mathematical data treatment was successfully employed in the study of the CdII? (N‐(2‐hydroxyethyl)iminodiacetic acid) system. Results obtained from both analytical techniques (potentiometry with the use of an ion selective electrode, and two polarographic techniques) and analytical procedures (acid? base and ligand titrations) compare well with each other and with the literature data (the formation and stability constants of ML and ML2). In addition, a new complex M(HL) was identified and its stability constant is reported.  相似文献

Interaction between alpha‐eleostearic acid (α‐ESA) and calf thymus DNA in Tris‐HCl buffer (pH = 7.4) using neutral red (NR) dye as a spectral probe was investigated using UV–Vis absorption and fluorescence spectroscopy. Spectral data matrix of the complexed reaction between α‐ESA and NR with DNA was processed with an alternative least‐squares (ALS) algorithm, the obtained concentration profiles and the corresponding pure spectra for species (NR, DNA–NR, and DNA–NR–ESA) demonstrated three kinds of reactions might occur in the system. The major groove binding between α‐ESA and DNA was further validated using circular dichroism, viscosity, DNA melting, and ionic strength effect measurements. Moreover, the calculated values of thermodynamic parameters, such as enthalpy (ΔHθ, ?22.04 kJ/mol) and entropy change (ΔSθ, 91.52 J K?1 mol?1), suggested binding between α‐ESA and DNA was mainly driven by hydrophobic interactions and hydrogen bonds without electrostatic force.  相似文献

3‐Methyl‐3‐(3‐pentyl)‐1,2‐dioxetane 1 and 3‐methyl‐3‐(2,2‐dimethyl‐1‐propyl)‐1,2‐dioxetane 2 were synthesized in low yield by the α‐bromohydroperoxide method. The activation parameters were determined by the chemiluminescence method (for 1 ΔH‡ = 25.0 ± 0.3 kcal/mol, ΔS‡ = −1.0 entropy unit (e.u.), ΔG‡ = 25.3 kcal/mol, k1 (60°C) = 4.6 × 10−4s−1; for 2 ΔH‡ = 24.2 ± 0.2 kcal/mol, ΔS‡ = −2.0 e.u., ΔG‡ = 24.9 kcal/mol, k1 (60°C) = 9.2 × 10−4s−1. Thermolysis of 1–2 produced excited carbonyl fragments (direct production of high yields of triplets relative to excited singlets) (chemiexcitation yields for 1: ϕT = 0.02, ϕS ≤ 0.0005; for 2: ϕT = 0.02, ϕS ≤ 0.0004). A new technique is proposed for detecting interactions between DNA and DNA autoimmune antibodies using a potentiometric sensor based on a glassy-carbon electrode modified with polyaniline and native DNA from chicken erythrocytes. It is shown that the DNA-antibody interaction changes the rate of polyaniline doping in transferring the DNA sensor from an alkaline (pH 7.5) solution, which is optimum for the immunochemical reaction, to an acidic (pH 3.0) solution. The dynamics of the variation of the DNA sensor potential depends on the titer of antibodies and their origin. The dependence of the DNA sensor signal on the dilution of the blood sera from systemic lupus erythematosus and autoimmune thyroiditis patients shows that DNA antibodies can be diagnosed by the characteristic maximum in the dilution curve found in the range of serum dilutions from 1: 20 to 1: 50.  相似文献

Synthesis of α‐C‐ethylene phosphate and phosphonate as well as α‐C‐methylene phosphate analogues of N‐acetyl‐α‐d‐glucosamine 1‐phosphate is reported starting from the common perbenzylated 2‐acetamido‐2‐deoxy‐α‐C‐allyl glucoside. Anomerisation of the corresponding amino α‐C‐glucosyl aldehyde to the β‐aldehyde was observed. Thus, both amino α‐ and β‐C‐glucosyl methanol were obtained after reduction.  相似文献

A reaction mechanism that describes the substitution of two imino protons in a thymine:thymine (T:T) mismatched DNA base pair with a HgII ion, which results in the formation of a (T)N3‐HgII‐N3(T) metal‐mediated base pair was proposed and calculated. The mechanism assumes two key steps: The formation of the first HgII? N3(T) bond is triggered by deprotonation of the imino N3 atom in thymine with a hydroxo ligand on the HgII ion. The formation of the second HgII? N3(T) bond proceeds through water‐assisted tautomerization of the remaining, metal‐nonbonded thymine base or through thymine deprotonation with a hydroxo ligand of the HgII ion already coordinated to the thymine base. The thermodynamic parameters ΔGR=?9.5 kcal mol?1, ΔHR=?4.7 kcal mol?1, and ΔSR=16.0 cal mol?1 K?1 calculated with the ONIOM (B3LYP:BP86) method for the reaction agreed well with the isothermal titration calorimetric (ITC) measurements by Torigoe et al. [H. Torigoe, A. Ono, T. Kozasa, Chem. Eur. J. 2010 , 16, 13218–13225]. The peculiar positive reaction entropy measured previously was due to both dehydration of the metal and the change in chemical bonding. The mercury reactant in the theoretical model contained one hydroxo ligand in accord with the experimental pKa value of 3.6 known for an aqua ligand of a HgII center. The chemical modification of T:T mismatched to the T‐HgII‐T metal‐mediated base pair was modeled for the middle base pair within a trinucleotide B‐DNA duplex, which ensured complete dehydration of the HgII ion during the reaction.  相似文献

The volatile composition of different apple varieties of Malus domestica Borkh. species from different geographic regions at Madeira Islands, namely Ponta do Pargo (PP), Porto Santo (PS), and Santo da Serra (SS) was established by headspace solid‐phase microextraction (HS‐SPME) procedure followed by GC‐MS (GC‐qMS) analysis. Significant parameters affecting sorption process such as fiber coating, extraction temperature, extraction time, sample amount, dilution factor, ionic strength, and desorption time, were optimized and discussed. The SPME fiber coated with 50/30 μm divinylbenzene/carboxen/PDMS (DVB/CAR/PDMS) afforded highest extraction efficiency of volatile compounds, providing the best sensitivity for the target volatiles, particularly when the samples were extracted at 50°C for 30 min with constant magnetic stirring. A qualitative and semi‐quantitative analysis between the investigated apple species has been established. It was possible to identify about 100 of volatile compounds among pulp (46, 45, and 39), peel (64, 60, and 64), and entire fruit (65, 43, and 50) in PP, PS, and SS apples, respectively. Ethyl esters, terpenes, and higher alcohols were found to be the most representative volatiles. α‐Farnesene, hexan‐1‐ol and hexyl 2‐methylbutyrate were the compounds found in the volatile profile of studied apples with the largest GC area, representing, on average, 24.71, 14.06, and 10.80% of the total volatile fraction from PP, PS, and SS apples. In PP entire apple, the most abundant compounds identified were α‐farnesene (30.49%), the unknown compound m/z (69, 101, 157) (21.82%) and hexyl acetate (6.57%). Regarding PS entire apple the major compounds were α‐farnesene (16.87%), estragole (15.43%), hexan‐1‐ol (10.94), and E‐2‐hexenal (10.67). α‐Farnesene (30.3%), hexan‐1‐ol (18.90%), 2‐methylbutanoic acid (4.7%), and pentan‐1‐ol (4.6%) were also found as SS entire apple volatiles present in a higher relative content. Principal component analysis (PCA) of the results clustered the apples into three groups according to geographic origin. Linear discriminant analysis (LDA) was performed in order to detect the volatile compounds able to differentiate the three kinds of apples investigated. The most important contributions to the differentiation of the PP, PS, and SS apples were ethyl hexanoate, hexyl 2‐methylbutyrate, E,E‐2,4‐heptadienal, p‐ethyl styrene, and E‐2‐hexenal.  相似文献

The elastic moduli El of the crystalline regions of α‐chitin and chitosan in the direction parallel to the chain axis were measured by X‐ray diffraction. The El values were 41 GPa for α‐chitin, and 65 GPa for chitosan, respectively, at 20°C. The contracted skeletons of α‐chitin and chitosan are the key factor for the low El values compared with that (138 GPa) of cellulose I. The El value of α‐chitin was constant at 41 GPa both at −190°C and 150°C, which indicates that the molecular chain of α‐chitin is stable against heat within the temperature and stress range studied. © 1999 John Wiley & Sons, Inc. Densities have been measured for the electrolyte (NaCl, NaBr and NaI)‐monosaccharide (D ‐mannose and D‐ribose)‐water solutions at 298.15 K. These data have been used to calculate the apparent molar volumes of the saccharides (VΦ,S) and electrolytes (VΦ,E) in the studied solutions. Infinite dilution apparent molar volumes, VΦ,S0 and VΦ,E0, have been evaluated, together with the standard transfer volumes of the saccharides (ΔtVS0) from water to aqueous electrolyte solutions and those of the electrolytes (ΔtVE0) from water to aqueous saccharide solutions. It was shown that both the ΔtVS0 and ΔtVE0 values are positive and increase with increasing molalities of sodium halides and saccharides, respectively. Overall, the ΔtVS0 and ΔtVE0 values have the order of NaCl > NaBr > NaI except for NaI‐ribose and NaI‐ribose. Volumetric interaction parameters for the electrolyte‐monosaccharide pairs in water were obtained and interpreted by the stereochemistry of the monosaccharide molecules and the structural interaction model.  相似文献
