Incorporations of [1-13C]?, [2-13C]?, [1,2-13C2]-acetates and [13C]-methionine into anditomin, a metabolite of Aspergillus variecolor, indicate its formation by a mixed polyketide-terpenoid biosynthetic pathway similar to that elucidated for andibenin; observations are made on the possible biosynthetic relationship of the A. variecolor metabolites with austin and terretonin, mycotoxins recently isolated from A. ustus and A. terreus respectively.  相似文献

diendo- and diexo-2-Methylamino and 2-benzylamino-3-hydroxymethylbicyclo [2.2.1]heptanes and the corresponding bicycio [2.2.1] heptenes (5a -d, 7a -d) were synthesized from β-amino acid esters containing the norbornane or norbornene skeleton (3a-d). The aminoalcohols were converted to 5,8- methano -3,1-benzoxazines by reaction with formaldehyde. As established by 1H and 13C NMR spectroscopy, the predominant conformation is endo - boat (B) for the diexo, and exo - boat (E) for the diendo derivatives.  相似文献

By the self-immolative asymmetric synthesis, optically pure key intermediate β-piperonyl-γ-lactone (R)-(+)-6 was found to be prepared in reasonable yield from the easily available chiral γ-lactone synthon (1) which had been also reported to give an antipode (S)-(-)-6. The optically pure (R)-(+)-6 was shown to be converted successfully into several optically pure natural lignan lactones.  相似文献

X-Ray and 1H N.M.R. studies on pyranoid rings 1,2-cis-fused to dioxolane rings in acetylated D-gluco- and D--galactopyranose derivatives demonstrate that the configuration of the dioxolane ring influences the conformation of the pyranoid ring in the D-gluco but not in the D-galactopyranose series. The crystal structure of 3,4,6-tri-O-acetyl-1,2-O-(R)--(l-cyano-ethylidene)-α-D-glucopyranose (1) and 3,4,6-tri-O-acetyl-1,2-O-(R)-(1-cyano-ethylidene)-α-D-galactopyranose (2)have been determined by X-ray analysis. Lattice parameters for 1 are a=20.6021 (11), b=8.0438 (2), c=5.5541 (1) Å and β= 95.588 (3)° for a cell with P21 symmetry. These parameters for 2 are a=20.3361 (7), b=10.0907 (2), c=18.9115 (5) Å, β =112.399 (2)°, C2, with two crystallographycally independent molecules. The conformation of the pyranoid ring in both compounds can be described as flattened 4C1 and that of the dioxolane ring as distorted E1. The importance of the torsion angles for describing problems of configuration is remarked and the use of relative configurational angles is stressed. The 1H N.M.R. spectra of 1 and 2 and 3,4,6-tri-O-acetyl-1,2-O-(S)- and (R)-ethylidene-α-D-glucopyranose (5 and 7), 3,4,6-tri-O-acetyl--1,2-O-(S)- and (R)-ethylidene-α-D-galactopyranose (6 and 8), and 3,4,6-tri-O-acetyl-1,2-O-(S)-and (R)-benzylidene-α-D-glucopyranose (9 and 10) have been analyzed by using iterative computer methods and N.O.E. measurements. The results indicate that the major solution conformation of the pyranoid ring of the derivatives in the D-gluco series 1, 5 and 9 may be described as flattened 4C1 and that of 7 and 10 as 2S5. The major solution conformation of the pyranoid ring in all compounds in the D-galacto series (2,4,6,8) may be described as flattened 4C1.  相似文献

Evidence is presented that glycosylation of a ribose-OH group in nucleosides results in a significant downfield shift for the appended 13C nucleus and smaller upfield displacements for the adjacent carbons, providing an efficient tool for differentiating between 2′-o-, 3′-o- and 5′-o-glycosyl-ribonucleosides. Therewith, the products formed on enzymatic galactosylation of uridine, inosine, and adenosine are unequivocally assigned β(1→3)-glycosidic linkages (3a3c).  相似文献

Pentafluoroformanilide was prepared by a new synthetical route. dl-[2-13C]Leucine was prepared by condensing the sodium salt of ethyl acetamido-[2-13C]cyanoacetate with isobutylbromide in hexamethylphosphoroustriamide followed by acid hydrolysis. N-Boc-dl-[2-13C]Leucine was prepared and incorporated into [8-dl-[2-13C]leucine]oxytocin by total synthesis. The 13C-labeled hormone derivative [8-[2-13C]leucine]oxytocin was separated from its 8-position diastereoisomer by partition chromatography. The specifically 13C-labeled peptide hormone diastereoisomeric analog [3-dl-[2-13C]leucine]oxytocin also was prepared by solid phase peptide synthesis. No suitable solvent system for partition chromatography separation of the latter diastereoisomeric peptide mixture could be found. However an excellent preparative separation of the diastereoisomers could be obtained by reverse phase high pressure liquid chromatography on a partisil 10 M9 ODS column using the solvent system 0.05 M ammonium acetate (pH 4.0), acetonitrile (81:19, vv) to give pure [3-(2-13C]leucine]oxytocin and [3-D-(2-13C]leucine]oxytocin. An excellent separation of [8[2-13C]leucine]oxytocin and the corresponding 8-D-leucine diastereoisomer derivative could also be accomplished by high pressure liquid chromatography.  相似文献

Reaction of cyclo-(Gly-L-Cys-Gly)3 with 1,3,5-tris-bromomethylbenzene yields a tris thioether of the cyclic nonapeptide in 28% yield. Both 1H and 13C NMR spectra are consistent with a molecule of 3-fold symmetry; the temperature dependences of chemical shifts of the amide hydrogens are consistent with a structure composed of three β-turns.  相似文献

Abstract  

The synthesis of [1-15N,2-13C]-difloxacin, an arylfluoroquinolone antibacterial agent, is reported. As a crucial initial step, the starting materials ethyl 2,4,5-trifluorobenzoylacetate, [formyl-13C]-triethyl orthoformate, and [15N]-4-fluoroaniline were reacted to ethyl [15N,3-13C]-3-(4-fluoroanilino)-2-(2,4,5-trifluorobenzoyl)acrylate. After cyclization and ester cleavage, the resulting intermediate was reacted with 1-methylpiperazine to [1-15N,2-13C]-1-(4-fluorophenyl)-6-fluoro-7-(4-methyl-1-piperazinyl)-1,4-dihydro-4-oxoquinoline-3-carboxylate, i.e., [1-15N,2-13C]-difloxacin. The overall yield was 62% based on the non-labeled and 43% based on the labeled starting materials (both used in 1.4 molar excess). The product was identified by 1H-, 13C-, and 15N-NMR spectroscopy and by cochromatography (TLC, HPLC) with an authentic reference; its purity (HPLC) was above 98%. Prior to synthesis of [1-15N,2-13C]-difloxacin, non-labeled difloxacin was synthesized in order to optimize procedures and to identify and characterize all intermediates.  相似文献

The facile reactions of CF3NCF2O with a variety of non-fluorinated nucleophiles are discussed. CF3NCF2O + Nu- → CF3N(Nu)C(O)F + F- Reactions with alcohols, carboxylic acids, mercaptans, amines, KSCN, KCN, NaN3 and NaOCN were carried out. All except NaN3 and NaOCN were reactive and the expected compounds were isolated in most cases. Subsequent reaction of CF3N(Nu)C(O)F with additional nucleophile was observed in many cases forming CF3N(Nu)C(O)Nu. With CH3CO2H, the latter reaction forms CH3C(O)F and the novel amine CF3N(H)OC(O)CH63. Ten new compounds are reported with characterization by IR, NMR and physical properties.  相似文献
